    Influence of sulfide on the evaluation of methane production through the degradation of sugarcane vinasse

    Anaerobic digestion is a widely used effluent and organic waste treatment practice, in which it is possible to minimize and control environmental problems, associating the reduction of environmental impacts with energy recovery. Low methane production and process instability are often found in anaerobic digestion reactors, preventing this technique from being widely applied. Inhibitory substances, such as sulfides resulting from sulfate conversion by the sulfur reducing bacteria, are one of the causes of inhibition or malfunctioning of anaerobic digesters if they are present in the effluent to be treated. The objective of this work is to evaluate the effect of sulfide at two different values of pH (7.0 and 7.5) using sulfide concentrations of 0 to 1000 mg S2-L-1. All the tests were performed in batches and performed at mesophilic conditions. For the concentrations of 50 mg S2-L-1 and 1000 mg S2-L-1, the inhibitions of the methanogenic activity at pH 7.0 were in the order of 38.5% to 59.8% and at pH 7.5 in the order of 67% to 94%, respectively. Concerning the test at pH 7.0, the removal of COD in the experiment without addition of any concentration of S2- was 93.3%, and it reached a 49.14% COD removal at concentrations of 1000 mg S2-L-1. At pH 7.5 under the same conditions, the COD removals were respectively 80.7% and 9.6%. The concentrations of 50, 75 and 100 mg S2-L-1 of S2- initially tested at the two aforementioned pH values promoted the greatest increase in the reduction of SMA. When the experiments were carried out at pH 7.0 the reductions were 37.96%, 41.70%, and 46.06% respectively for the same concentrations.     Phylogenetic and functional diversity of metagenomic libraries of phenol degrading sludge from petroleum refinery wastewater treatment system

    In petrochemical refinery wastewater treatment plants (WWTP), different concentrations of pollutant compounds are received daily in the influent stream, including significant amounts of phenolic compounds, creating propitious conditions for the development of particular microorganisms that can rapidly adapt to such environment. In the present work, the microbial sludge from a refinery WWTP was enriched for phenol, cloned into fosmid vectors and pyrosequenced. The fosmid libraries yielded 13,200 clones and a comprehensive bioinformatic analysis of the sequence data set revealed a complex and diverse bacterial community in the phenol degrading sludge. The phylogenetic analyses using MEGAN in combination with RDP classifier showed a massive predominance of Proteobacteria, represented mostly by the genera Diaphorobacter, Pseudomonas, Thauera and Comamonas. The functional classification of phenol degrading sludge sequence data set generated by MG-RAST showed the wide metabolic diversity of the microbial sludge, with a high percentage of genes involved in the aerobic and anaerobic degradation of phenol and derivatives. In addition, genes related to the metabolism of many other organic and xenobiotic compounds, such as toluene, biphenyl, naphthalene and benzoate, were found. Results gathered herein demonstrated that the phenol degrading sludge has complex phylogenetic and functional diversities, showing the potential of such community to degrade several pollutant compounds. This microbiota is likely to represent a rich resource of versatile and unknown enzymes which may be exploited for biotechnological processes such as bioremediation

    Leishmania donovani is a protozoan parasite that causes visceral leishmaniasis, provoking liver and spleen tissue destruction that is lethal unless treated. The parasite replicates in macrophages and modulates host microbicidal responses. We have previously reported that neutrophil elastase (NE) is required to sustain L. donovani intracellular growth in macrophages through the induction of interferon beta (IFN-ÎČ). Here, we show that the gene expression of IFN-ÎČ by infected macrophages was reduced by half when TLR4 was blocked by pre-treatment with neutralizing antibodies or in macrophages from tlr2 (-/-) mice, while the levels in macrophages from myd88(-/-) mice were comparable to those from wild-type C57BL/6 mice. The neutralization of TLR4 in tlr2 (-/-) macrophages completely abolished induction of IFN-ÎČ gene expression upon parasite infection, indicating an additive role for both TLRs. Induction of type I interferon (IFN-I), OASL2, SOD1, and IL10 gene expression by L. donovani was completely abolished in macrophages from NE knock-out mice (ela2 (-/-)) or from protein kinase R (PKR) knock-out mice (pkr (-/-)), and in C57BL/6 macrophages infected with transgenic L. donovani expressing the inhibitor of serine peptidase 2 (ISP2). Parasite intracellular growth was impaired in pkr (-/-) macrophages but was fully restored by the addition of exogenous IFN-ÎČ, and parasite burdens were reduced in the spleen of pkr (-/-) mice at 7 days, as compared to the 129Sv/Ev background mice. Furthermore, parasites were unable to grow in macrophages lacking TLR3, which correlated with lack of IFN-I gene expression. Thus, L. donovani engages innate responses in infected macrophages via TLR2, TLR4, and TLR3, via downstream PKR, to induce the expression of pro-survival genes in the host cell, and guarantee parasite intracellular development

    Cosmological constraints from lensing statistics and supernovae on the cosmic equation of state

    We investigate observational constraints from lensing statistics and high-z type Ia supernovae on flat cosmological models with nonrelativistic matter and an exotic fluid with equation of state, px=(m/3−1)ρxp_x=(m/3 -1)\rho_x. We show that agreement with both tests at the 68% confidence level is possible if the parameter mm is low (mâ‰Č0.85m \lesssim 0.85) and 0.24â‰ČΩm0â‰Č0.380.24 \lesssim \Omega_{m0} \lesssim 0.38 with lower values of Ωm0\Omega_{m0} corresponding to higher mm. We find that a conventional cosmological constant model with Ωm0≃0.33\Omega_{m0}\simeq 0.33 is the best fit model of the combined likelihood.Comment: 7 pages, 4 postscript figures, revtex, submitted to Phys.     Accuracy comparison among 3D-printing technologies to produce dental models

    Background: Little is known on accuracy comparisons among 3D-printing trueness and precision to produce dental models. The purpose of this study was to investigate the accuracy of different 3D-printing technologies (SLA; DLP; LCD poly and monochromatic; and Polyjet). Methods: Fifteen models were manufactured by the five different 3D printers used in this study. The manufactured models were physically measured in the corresponding lengths for trueness and reproducibility. Means and standard deviations were obtained for the five computer-aided manufacturing (CAM) methods and compared. Results: No significant difference was found in the comparison between observers in all measurements performed with the different model algorithms. PairwiseWilcoxon non-parametric test comparisons of trueness between 3D-printers revealed statistically significant differences between Stratasys versus Anycubic (p = 0.025) and Anycubic versus Form 2 (p = 0.048). Conclusion: The present findings suggest that the 3D-printing methods may have small significant discrepancies when compared to the original digital files, which may not be clinically relevant. In addition, there were no significant discrepancies among median measurements of each printing method (within 3D-printer analysis), which suggests that, for dental casts, all of the studied devices performed similarly.peer-reviewe

    Taxonomic and Functional Microbial Signatures of the Endemic Marine Sponge Arenosclera brasiliensis

    The endemic marine sponge Arenosclera brasiliensis (Porifera, Demospongiae, Haplosclerida) is a known source of secondary metabolites such as arenosclerins A-C. In the present study, we established the composition of the A. brasiliensis microbiome and the metabolic pathways associated with this community. We used 454 shotgun pyrosequencing to generate approximately 640,000 high-quality sponge-derived sequences (∌150 Mb). Clustering analysis including sponge, seawater and twenty-three other metagenomes derived from marine animal microbiomes shows that A. brasiliensis contains a specific microbiome. Fourteen bacterial phyla (including Proteobacteria, Cyanobacteria, Actinobacteria, Bacteroidetes, Firmicutes and Cloroflexi) were consistently found in the A. brasiliensis metagenomes. The A. brasiliensis microbiome is enriched for Betaproteobacteria (e.g., Burkholderia) and Gammaproteobacteria (e.g., Pseudomonas and Alteromonas) compared with the surrounding planktonic microbial communities. Functional analysis based on Rapid Annotation using Subsystem Technology (RAST) indicated that the A. brasiliensis microbiome is enriched for sequences associated with membrane transport and one-carbon metabolism. In addition, there was an overrepresentation of sequences associated with aerobic and anaerobic metabolism as well as the synthesis and degradation of secondary metabolites. This study represents the first analysis of sponge-associated microbial communities via shotgun pyrosequencing, a strategy commonly applied in similar analyses in other marine invertebrate hosts, such as corals and algae. We demonstrate that A. brasiliensis has a unique microbiome that is distinct from that of the surrounding planktonic microbes and from other marine organisms, indicating a species-specific microbiome

    The contribution of 7q33 copy number variations for intellectual disability

    Copy number variations (CNVs) at the 7q33 cytoband are very rarely described in the literature, and almost all of the cases comprise large deletions affecting more than just the q33 segment. We report seven patients (two families with two siblings and their affected mother and one unrelated patient) with neurodevelopmental delay associated with CNVs in 7q33 alone. All the patients presented mild to moderate intellectual disability (ID), dysmorphic features, and a behavioral phenotype characterized by aggressiveness and disinhibition. One family presents a small duplication in cis affecting CALD1 and AGBL3 genes, while the other four patients carry two larger deletions encompassing EXOC4, CALD1, AGBL3, and CNOT4. This work helps to refine the phenotype and narrow the minimal critical region involved in 7q33 CNVs.
