    Non-gaussianity in the strong regime of warm inflation

    The bispectrum of scalar mode density perturbations is analysed for the strong regime of warm inflationary models. This analysis generalises previous results by allowing damping terms in the inflaton equation of motion that are dependent on temperature. A significant amount of non-gaussianity emerges with constant (or local) non-linearity parameter fNL∌20f_{NL}\sim 20, in addition to the terms with non-constant fNLf_{NL} which are characteristic of warm inflation.Comment: 15 pages, 3 figures.     Evaluating Native American Bird Use and Bird Assemblage Variability along the Oregon Coast

    Native American use of birds on the Oregon coast is not well known and has never been synthesized to present a regional understanding. We rectify this by analyzing data from 26 zooarchaeological assemblages, including three previously unpublished bird assemblages: Umpqua/Eden (35DO83), Whale Cove (35LNC60), and the Dunes Site (35CLT27). We employ a series of non-parametric randomization tests to directly evaluate patterns of taxonomic diversity, correlations with nearby breeding colonies, and broader procurement strategies discussed in ethnohistorical accounts. We compare the assemblages to contemporary surveys of naturally beached birds as observed by COASST (Coastal Observation Seabird Survey Team) and evaluate whether archaeological specimens were scavenged from the beach. While 71% of the identified bird remains belong to just three families (Anatidae, Alcidae, and Procellariidae), closer analysis reveals the incredible diversity of birds used by Oregon coast Native Americans. The assemblages vary considerably in terms of taxonomic diversity and composition, leading us to conclude that people used birds opportunistically, likely incorporating multiple strategies, including hunting, collecting beached carcasses and targeting cormorant colonies. We hope that the methods and approaches employed here will inspire other archaeologists to devote more attention to bird assemblages, and how their study can inform conservation efforts

    Effectiveness and limitations of ÎČ-blocker therapy in congenital long-QT syndrome

    Background—ÎČ-blockers are routinely prescribed in congenital long-QT syndrome (LQTS), but the effectiveness and limitations of ÎČ-blockers in this disorder have not been evaluated. Methods and Results—The study population comprised 869 LQTS patients treated with ÎČ-blockers. Effectiveness of ÎČ-blockers was analyzed during matched periods before and after starting ÎČ-blocker therapy, and by survivorship methods to determine factors associated with cardiac events while on prescribed ÎČ-blockers. After initiation of ÎČ-blockers, there was a significant (P<0.001) reduction in the rate of cardiac events in probands (0.97±1.42 to 0.31±0.86 events per year) and in affected family members (0.26±0.84 to 0.15±0.69 events per year) during 5-year matched periods. On-therapy survivorship analyses revealed that patients with cardiac symptoms before ÎČ-blockers (n=598) had a hazard ratio of 5.8 (95% CI, 3.7 to 9.1) for recurrent cardiac events (syncope, aborted cardiac arrest, or death) during ÎČ-blocker therapy compared with ..

    COMPLEMENT FACTOR B IS A DETERMINANT OF BOTH METABOLIC AND CARDIOVASCULAR FEATURES OF METABOLIC SYNDROME

    CFB (complement factor B) is elevated in adipose tissue and serum from patients with type 2 diabetes mellitus and cardiovascular disease, but the causal relationship to disease pathogenesis is unclear. Cfb is also elevated in adipose tissue and serum of the spontaneously hypertensive rat, a well-characterized model of metabolic syndrome. To establish the role of CFB in metabolic syndrome, we knocked out the Cfb gene in the spontaneously hypertensive rat. Cfb−/− rats showed improved glucose tolerance and insulin sensitivity, redistribution of visceral to subcutaneous fat, increased adipocyte mitochondrial respiration, and marked changes in gene expression. Cfb−/− rats also had lower blood pressure, increased ejection fraction and fractional shortening, and reduced left ventricular mass. These changes in metabolism and gene expression, in adipose tissue and left ventricle, suggest new adipose tissue-intrinsic and blood pressure-independent mechanisms for insulin resistance and cardiac hypertrophy in the spontaneously hypertensive rat. In silico analysis of the human CFB locus revealed 2 cis-regulated expression quantitative trait loci for CFB expression significantly associated with visceral fat, circulating triglycerides and hypertension in genome-wide association studies. Together, these data demonstrate a key role for CFB in the development of spontaneously hypertensive rat metabolic syndrome phenotypes and of related traits in humans and indicate the potential for CFB as a novel target for treatment of cardiometabolic disease

Sandia continues to pursue innovative CSP concepts with the goal of reducing the cost of CSP while improving efficiency and performance. In this pursuit, Sandia has developed many tools for the analysis of CSP performance. The following capabilities document highlights Sandia's extensive experience in the design, construction, and utilization of large-scale testing facilities for CSP and the tools that Sandia has created for the full characterization of heliostats.     A prevalence of dynamo-generated magnetic fields in the cores of intermediate-mass stars

    Magnetic fields play a part in almost all stages of stellar evolution. Most low-mass stars, including the Sun, show surface fields that are generated by dynamo processes in their convective envelopes. Intermediate-mass stars do not have deep convective envelopes, although 10 per cent exhibit strong surface fields that are presumed to be residuals from the star formation process. These stars do have convective cores that might produce internal magnetic fields, and these fields might survive into later stages of stellar evolution, but information has been limited by our inability to measure the fields below the stellar surface. Here we report the strength of dipolar oscillation modes for a sample of 3,600 red giant stars. About 20 per cent of our sample show mode suppression, by strong magnetic fields in the cores, but this fraction is a strong function of mass. Strong core fields occur only in red giants heavier than 1.1 solar masses, and the occurrence rate is at least 50 per cent for intermediate-mass stars (1.6–2.0 solar masses), indicating that powerful dynamos were very common in the previously convective cores of these stars

    Untersuchungen zum Einfluss von thrombozytÀren Wachstumsfaktoren auf den zellvermittelten Abbau eines nanopartikulÀren Knochenersatzstoffes auf Hydroxylapatitbasis : eine experimentelle Studie am Miniaturschwein

    Ziel der vorliegenden tierexperimentellen Studie am Miniaturschwein war es, den Einfluss von plĂ€ttchenreichem Plasma (PRP) auf den zellvermittelten Abbau eines nanopartikulĂ€ren Hydroxylapatits (HA) in der FrĂŒhphase der Knochendefektheilung zu untersuchen. Hierzu wurden 26 mĂ€nnliche Miniaturschweine der Rasse Mini-Lewe in drei Versuchsgruppen eingeteilt und jeweils ein standardisierter Knochendefekt in der Intercondylarregion des rechten Femurs angelegt. Die Defekte wurden entweder mit dem Knochenersatzstoff (Gruppe I/PRP-,n = 11) oder dem Knochenersatzstoff kombiniert mit PRP (Gruppe II/PRP+, n = 11) befĂŒllt. In einer Kontrollgruppe (n = 4) blieben die Defekte unbefĂŒllt. WĂ€hrend der Implantationsoperation wurden bei sechs Tieren jeweils 250 ml Vollblut entnommen, aus dem anschließend durch fraktionierte Zentrifugation plĂ€ttchenreiches Plasma gewonnen wurde. Die enthaltenen Thrombozyten wurden durch den Zusatz von Thrombin und Kalziumglukonat zur Degranulation angeregt, wodurch die enthaltenen Wachstumsfaktoren aus den alpha-Granula freigesetzt wurden. Zu diesen Wachstumsfaktoren gehören Platelet Derived Growth Factor AB und BB (PDGF AB, BB), Transforming Growth Factor ß 1 (TGF beta1), Vascular Endothelial Growth Factor (VEGF) und basic Fibroblast Growth Factor (bFGF). Die Konzentration der genannten Wachstumsfaktoren wurde mit Hilfe der ELISA-Technik bestimmt. Sie lagen zwischen Faktor 1,6 fĂŒr TGF-beta1 und Faktor 24,4 fĂŒr bFGF. 20 Tage post operationem fand die Explantation der operierten distalen Femura statt. Zur lichtmikroskopischen Untersuchung fanden die Knochen-Implantat-Proben Eingang in unterschiedliche Techniken der Einbettung (Paraffin-, Kunststoffeinbettungen), PrĂ€paration (Paraffinschnitte, Kunststoffschnitte und SchliffprĂ€parationen), FĂ€rbung (Toluidinblau, Haematoxylin-Eosin, Safranin) und Histochemie (Enzym-, Immunhistochemie). DarĂŒber hinaus wurden transmissionselektronenmikroskopische und computergestĂŒtzte histomorphometrische Untersuchungen durchgefĂŒhrt. Wie die Ergebnisse der Licht- und Transmissionselektronenmikroskopie aufgezeigt haben, erfolgt in den mit Knochenersatzmaterial behandelten Versuchsgruppen, unabhĂ€ngig von der PRP-Applikation, die HA-Degradation hydrolytisch und Makrophagen-vermittelt. Die Makrophagen-Population wird durch Riesenzellen vom Langhans-Typ reprĂ€sentiert. Diese polarisierten Polykaryen adhĂ€rieren ĂŒber ihre apikale MembrandomĂ€ne an den ImplantatoberflĂ€chen. Das subplasmalemmale Zytoplasma ist immunhistochemisch durch Vimentin-Kondensationen gekennzeichnet. Nicht-adhĂ€rente, frei im Granulationsgewebe lokalisierte Polykaryen zeigen dagegen ein homogenes Vimentin-Verteilungsmuster im Zytoplasma. Der zellulĂ€re Abbau des HA erfolgt mittels Phagozytose, indem die Polykaryen den "Fremdkörper" mit pseudopodienartigen ZytoplasmaauslĂ€ufern umschließen und in ihr Zytoplasma inkorporieren. Diese Art der Degradation wird durch den post implantationem stattfindenden Zerfall des Knochenersatzmaterials in zahlreiche kleine Partikel unterstĂŒtzt. Die hieraus resultierende VergrĂ¶ĂŸerung der ImplantatoberflĂ€che bietet einer Vielzahl von Zellen die Möglichkeit zur Haftung. Die festgestellten Expressionsmuster des CD44- Membranglykoproteins verweisen auf dessen funktionelle Rolle im Rahmen der Fusion mononukleĂ€rer Makrophagen zu multinukleĂ€ren Riesenzellen. Die darĂŒber hinaus beobachtete Umverteilung von CD44 von der apikalen zur basalen MembrandomĂ€ne bei Implantatassoziierten Polykaryen ist als transientes Geschehen im Zuge der AdhĂ€sion zu interpretieren. Der hohe AktivitĂ€tsstatus der adhĂ€renten Polykaryen ist immunhistochemisch durch eine intensive Kathepsin K-Expression gekennzeichnet. Die vergleichende histomorphometrische Auswertung der mit HA aufgefĂŒllten Defekte dokumentiert eine Verdopplung der Anzahl von Polykaryen in der Gruppe "Knochenersatzstoff mit PRP". Ein auf Basis der Messergebnisse durchgefĂŒhrter Wilcoxon-Rangsummentest verweist auf den hochsignifikanten Einfluss (p < 0,01) des Faktors PRP auf die Ausdehnung Tartrat-resistenter saurer Phosphatase-positiver Areale in den PrĂ€paraten. Diese Effekte können sowohl auf den im PRP angereicherten Wachstumsfaktoren als auch auf dem homologen Charakter der PRP-Zubereitung beruhen. Die beobachteten Polykaryen – sogenannte "Fremdkörperriesenzellen" – sind auch immer Indikatoren einer stattfindenden EntzĂŒndungsreaktion. Die histomorphometrisch dargestellte, deutlich verstĂ€rkte Fremdkörperreaktion in Gruppe II/PRP+ kann auf die PRP-Applikation zurĂŒckgefĂŒhrt werden. Im weiteren Heilungsverlauf kann dies zu einer Verzögerung der knöchernen Konsolidierung der Defekte fĂŒhren.Aim of the current experimental study in Minipigs was to examine the effects of homologous platelet-rich plasma (PRP) on the cell-mediated degradation of a nanoparticulate hydroxyapatite (HA) during the early phase of bone defect healing. Twenty-six male "Lewe" minipigs were divided into three groups. Standardized bone defects were created in the intercondylar region of the right femur of each pig and were filled with HA (Group I/PRP-, n = 11) or HA + PRP (Group II/PRP+, n = 11). The defects of the control group (n = 4) were left empty. During the implantation procedure blood was drawn from six minipigs (250 ml each). PRP was isolated from these blood samples after several centrifugation steps. After the addition of thrombin and calcium gluconate growth factors were released from the alpha-granules of the thrombocytes which were enriched within the PRP. Some of these growth factors are Platelet Derived Growth Factor AB and BB (PDGF AB, BB), Transforming Growth Factor ß 1 (TGF beta1), Vascular Endothelial Growth Factor (VEGF) and basic Fibroblast Growth Factor (bFGF). The level of enrichment of these growth factors was controlled by the ELISA technique. Growth factor enrichment within the PRP ranged from 1.6 fold (TGF-beta1) to 24.4 fold (bFGF). After 20 days the treated distal femura were explanted. For light microscopical examination different tissue embedding methods (paraffine, plastic, resin), sectioning techniques (paraffine sections, plastic and resin sections, sawing and grinding sections), staining procedures (toluidine blue, hematoxylin eosin, safranin) and histochemical methods (enzyme- and immunohistochemistry) were performed. Additionally transmission electron microscopy and computer-assisted histomorphometry were used. The results of light microscopy and transmission electron microscopy showed that regardless of the addition of PRP, the HA is degraded by hydrolysis and macrophages. The population of macrophages consists of Langhans-type giant cells. The adhesion of the polarized polykaryons at the surfaces of the implant is mediated by the apical domain of the plasmamembranes. Vimentin condensations of the cytoplasm are attached to the apical plasmalemma. In contrast, non-adherent polykaryons of the granulation tissue reveal a homogeneous Vimentin distribution pattern within their cytoplasma. As shown ultrastructurally, the implant is degraded by means of phagocytosis. The implant particles are encircled by pseudopodia of the polykaryons and become incorporated into the cytoplasma. The degradation process is supported by disintegration of the bone substitute into numerous small particles after implantation. This disintegration causes enlargement of the implant surface and increases the probability of phagocyte adhesion. The pattern of CD44 expression points towards a functional role of the molecule during fusion of mononucleated macrophages into multinucleated giant cells. Implant-associated polykaryons show CD44 immunoreactivity only along the basal domains of the cytomembrane. This pattern can be interpreted as a temporal event during adhesion. Adherent polykaryons are further characterized by strong cathepsin K expression. The histomorphometric examination demonstrates twice as much foreign body giant cells in "Group II/PRP+" as in "Group I/PRP-". Based on these results, a Wilcoxon-signed-rank test was performed and a highly significant effect (p < 0.01) of PRP on the expansion of tartrate resistent acid phophatase (TRAP)-positive areas within bone defects could be demonstrated. This effect could be a result of the substution of PRP or of its homologous character. The polykaryons descibed in this work - so-called Foreign Body Giant Cells - are also indicators of inflammation. The enhanced cellular reaction observed in Group II/PRP+ must be interpreted as a strong foreign body reaction, triggered by the addition of PRP. It cannot be excluded that the strong inflammation reaction will lead to delayed bone formation in the course of healing

    Reduced Risk of Malaria Parasitemia Following Household Screening and Treatment: A Cross-Sectional and Longitudinal Cohort Study

    BACKGROUND: In regions of declining malaria transmission, new strategies for control are needed to reduce transmission and achieve elimination. Artemisinin-combination therapy (ACT) is active against immature gametocytes and can reduce the risk of transmission. We sought to determine whether household screening and treatment of infected individuals provides protection against infection for household members. METHODOLOGY/PRINCIPAL FINDINGS: The study was conducted in two areas in Southern Province, Zambia in 2007 and 2008/2009. To determine the impact of proactive case detection, households were randomly selected either to join a longitudinal cohort, in which participants were repeatedly screened throughout the year and those infected treated with artemether-lumefantrine, or a cross-sectional survey, in which participants were visited only once. Cross-sectional surveys were conducted throughout the year. The prevalence of RDT positivity was compared between the longitudinal and cross-sectional households at baseline and during follow-up using multilevel logistic regression. In the 2007 study area, 174 and 156 participants enrolled in the cross-sectional and longitudinal groups, respectively. In the 2008/2009 study area, 917 and 234 participants enrolled in the cross-sectional and longitudinal groups, respectively. In both study areas, participants and households in the longitudinal and cross-sectional groups were similar on demographic characteristics and prevalence of RDT positivity at baseline (2007: OR = 0.97; 95% CI:0.46, 2.03 | 2008/2009: OR = 1.28; 95% CI:0.44, 3.79). After baseline, the prevalence of RDT positivity was significantly lower in longitudinal compared to cross-sectional households in both study areas (2007: OR = 0.44; 95% CI:0.20, 0.96 | 2008/2009: OR = 0.16; 95% CI:0.05, 0.55). CONCLUSIONS/SIGNIFICANCE: Proactive case detection, consisting of screening household members with an RDT and treating those positive with ACT, can reduce transmission and provide indirect protection to household members. A targeted test and treat strategy could contribute to the elimination of malaria in regions of low transmission

    Genotype-Phenotype Correlation in the Long-QT Syndrome

    Background —The congenital long-QT syndrome (LQTS) is caused by mutations on several genes, all of which encode cardiac ion channels. The progressive understanding of the electrophysiological consequences of these mutations opens unforeseen possibilities for genotype-phenotype correlation studies. Preliminary observations suggested that the conditions ("triggers") associated with cardiac events may in large part be gene specific. Methods and Results —We identified 670 LQTS patients of known genotype (LQT1, n=371; LQT2, n=234; LQT3, n=65) who had symptoms (syncope, cardiac arrest, sudden death) and examined whether 3 specific triggers (exercise, emotion, and sleep/rest without arousal) differed according to genotype. LQT1 patients experienced the majority of their events (62%) during exercise, and only 3% occurred during rest/sleep. These percentages were almost reversed among LQT2 and LQT3 patients, who were less likely to have events during exercise (13%) and more likely to have events during rest/sleep (29% and 39%). Lethal and nonlethal events followed the same pattern. Corrected QT interval did not differ among LQT1, LQT2, and LQT3 patients (498, 497, and 506 ms, respectively). The percent of patients who were free of recurrence with ÎČ-blocker therapy was higher and the death rate was lower among LQT1 patients (81% and 4%, respectively) than among LQT2 (59% and 4%, respectively) and LQT3 (50% and 17%, respectively) patients. Conclusions —Life-threatening arrhythmias in LQTS patients tend to occur under specific circumstances in a gene-specific manner. These data allow new insights into the mechanisms that relate the electrophysiological consequences of mutations on specific genes to clinical manifestations and offer the possibility of complementing traditional therapy with gene-specific approaches

    Vision-related quality of life and Appearance concerns are associated with anxiety and depression after eye enucleation: A cross-sectional study

Aims: To investigate the association of demographic, clinical and psychosocial variables with levels of anxiety and depression in participants wearing an ocular prosthesis after eye enucleation. Methods: This cross-sectional study included 195 participants with an enucleated eye who were attending an ophthalmic clinic for prosthetic rehabilitation between July and November 2014. Demographic and clinical data, and self-reported feelings of shame, sadness and anger were collected. Participants also completed the National Eye Institute Visual Function Questionnaire, the Facial Appearance subscale of the Negative Physical Self Scale, and the Hospital Anxiety and Depression Scale. Regression models were used to identify the factors associated with anxiety and depression. Results: The proportion of participants with clinical anxiety was 11.8% and clinical depression 13.8%. More anxiety and depression were associated with poorer vision-related quality of life and greater levels of appearance concerns. Younger age was related to greater levels of anxiety. Less educated participants and those feeling more angry about losing an eye are more prone to experience depression. Clinical variables were unrelated to anxiety or depression. Conclusions: Anxiety and depression are more prevalent in eye-enucleated patients than the general population, which brings up the issues of psychiatric support in these patients. Psychosocialrather than clinical characteristics were associated with anxiety and depression. Longitudinal studies need to be conducted to further elucidate the direction of causality before interventions to improve mood states are developed. Copyright
