    Dynamic topic modeling using social network analytics

    First Online: 03 September 2021Topic modeling or inference has been one of the well-known problems in the area of text mining. It deals with the automatic categorisation of words or documents into similarity groups also known as topics. In most of the social media platforms such as Twitter, Instagram, and Facebook, hashtags are used to define the content of posts. Therefore, modelling of hashtags helps in categorising posts as well as analysing user preferences. In this work, we tried to address this problem involving hashtags that stream in real-time. Our approach encompasses graph of hashtags, dynamic sampling and modularity based community detection over the data from a popular social media engagement application. Further, we analysed the topic clusters’ structure and quality using empirical experiments. The results unveil latent semantic relations between hashtags and also show frequent hashtags in a cluster. Moreover, in this approach, the words in different languages are treated synonymously. Besides, we also observed top trending topics and correlated clusters

    Impact of amoeba and scuticociliatidia infections on the aquaculture European sea bass (Dicentrarchus labrax L.) in Portugal

    In this work, a survey of sea bass, Dicentrarchus labrax, for amoebae and scuticociliatidia infections was carried out to evaluate their effects on the aquaculture of this fish species. The study was conducted in two different fish farms, one using seawater and the other brackish water. Infection with parasitic amoebae was found to be fairly high (prevalence: 43-73%), being more frequent in sea bass from the brackish water system. Although it was never found to cause outbreaks of disease or mortality in the surveyed fish, amoebic gill disease (AGD) histopathological signs, i.e., hyperplasia, secondary lamellae fusion and cavity formation (interlamellar vesicles), were observed in fish manifesting no macroscopic lesions. Furthermore, some evidence was found that amoebae affects the fish's general state of health and growth rate. These results indicate that cautious and detailed surveys to detect this sort of infection, and thus carefully plan its control, are fully justified. Compared with amoebic infection, the prevalence of scuticociliatosis was found to be low (7-13%). No outbreaks of disease or mortality were ever recorded, even when scuticociliatidia was present in turbot raised in the same water system, leading to serious outbreaks of disease and mortalities in that species. This suggests that sea bass is far more resistant than turbot to such infections, and if this is the case, the former fish may be a good farming alternative when scuticociliatidia is present

    Distrofia Muscular de Duchenne (DMD) é uma doença neuromuscular que acomete 1:3.000 nascidos vivos do sexo masculino cuja herança é recessiva ligada ao cromossomo X e causada por mutação no gene que codifica a proteína distrofina. Essas alterações genéticas causam manifestações clínicas que comprometem o desenvolvimento neurológico do paciente. Os sintomas iniciam-se na primeira infância e o principal é a fraqueza muscular progressiva, cujas complicações são capazes de provocar morte prematura.Desta forma, torna-se relevante informar a situação epidemiológica da população que sofre de DMD, podendo ser base para realização de políticas públicas para essa população.Na metodologia foram coletados os dados referentes ao diagnóstico de DMD nos prontuários de um hospital de referência e de pacientes vinculados à Associação Cearense de Distrofia Muscular (ACDM). Este estudo foi devidamente autorizado e aprovado pelo comitê de ética de Pesquisa do Hospital que atende os pacientes.Nos resultados, para o município de Fortaleza, foi encontrada uma prevalência de 0,9/100.000 (IC95%: 0,53 –1,31), e o estado do Ceará apresentou prevalência de 0,44/100.000 (IC95%: 0,31 – 0,60). Apesar da DMD ser considerada uma doença genética rara, seu tratamento envolve a expertise de uma equipe multidisciplinar e medicações de alto custo para alguns casos, fazendo com que essa terapia não seja acessível à população mais carente do país, usuária do Sistema Único de Saúde. Recentemente o governo brasileiro tem se preocupado com a acessibilidade desses pacientes a um tratamento de qualidade. Esse fato impulsionou nosso grupo a determinar a epidemiologia da doença no estado do Ceará, onde a prevalência calculada da DMD foi abaixo da mundial (1–9/100.000). Calculamos a prevalência mínima da doença, porém é importante determinar se há outros casos a serem incluídos no estudo, que venham de outros grupos que fazem a terapia. Supomos que essa doença seja sub-diagnosticada, principalmente no interior. Para corroborar com os dados encontrados nesse estudo, sugerimos a elaboração de pesquisas que utilizem a dosagem de CPK neonatal como indicador para avaliar a presença de DMD precoce. Apoiamos a inclusão desta análise no protocolo de triagem neonatal como lei municipal. Tal fato levaria a uma maior celeridade nos diagnósticos dessa enfermidade e proporcionaria uma série de benefícios para a qualidade de vida dessas crianças. A partir desse estudo pretendemos iniciar uma campanha conjunta com o Hospital Infantil AIbert Sabin (HIAS) e a ACDM no estado do Ceará para identificarmos o maior número de casos de DMD e poder realizar o diagnóstico precoce, a fim de evitar o tratamento tardio e dar a essa população os recursos terapêuticos adequados

    School environment associates with lung function and autonomic nervous system activity in children : a cross-sectional study

    Children are in contact with local environments, which may affect respiratory symptoms and allergic sensitization. We aimed to assess the effect of the environment and the walkability surrounding schools on lung function, airway inflammation and autonomic nervous system activity. Data on 701 children from 20 primary schools were analysed. Lung function, airway inflammation and pH from exhaled breath condensate were measured. Pupillometry was performed to evaluate autonomic activity. Land use composition and walkability index were quantified within a 500 m buffer zone around schools. The proportion of effects explained by the school environment was measured by mixed-effect models. We found that green school areas tended to be associated with higher lung volumes (FVC, FEV1 and FEF25-75%) compared with built areas. FVC was significantly lower in-built than in green areas. After adjustment, the school environment explained 23%, 34% and 99.9% of the school effect on FVC, FEV1, and FEF25-75%, respectively. The walkability of school neighbourhoods was negatively associated with both pupil constriction amplitude and redilatation time, explaining -16% to 18% of parasympathetic and 8% to 29% of sympathetic activity. Our findings suggest that the environment surrounding schools has an effect on the lung function of its students. This effect may be partially mediated by the autonomic nervous system.Peer reviewe

    Phylogenetic and syntenic data support a single horizontal transference to a Trypanosoma ancestor of a prokaryotic proline racemase implicated in parasite evasion from host defences

    Abstract\ud \ud Background\ud Proline racemase (PRAC) enzymes of Trypanosoma cruzi (TcPRAC), the agent of Chagas disease, and Trypanosoma vivax (TvPRAC), the agent of livestock trypanosomosis, have been implicated in the B-cells polyclonal activation contributing to immunosuppression and the evasion of host defences. The similarity to prokaryotic PRAC and the absence in Trypanosoma brucei and Trypanosoma congolense have raised many questions about the origin, evolution, and functions of trypanosome PRAC (TryPRAC) enzymes.\ud \ud \ud Findings\ud We identified TryPRAC homologs as single copy genes per haploid genome in 12 of 15 Trypanosoma species, including T. cruzi and T. cruzi marinkellei, T. dionisii, T. erneyi, T. rangeli, T. conorhini and T. lewisi, all parasites of mammals. Polymorphisms in TcPRAC genes matched T. cruzi genotypes: TcI-TcIV and Tcbat have unique genes, while the hybrids TcV and TcVI contain TcPRACA and TcPRACB from parental TcII and TcIII, respectively. PRAC homologs were identified in trypanosomes from anurans, snakes, crocodiles, lizards, and birds. Most trypanosomes have intact PRAC genes. T. rangeli possesses only pseudogenes, maybe in the process of being lost. T. brucei, T. congolense and their allied species, except the more distantly related T. vivax, have completely lost PRAC genes.\ud \ud \ud \ud Conclusions\ud The genealogy of TryPRAC homologs supports an evolutionary history congruent with the Trypanosoma phylogeny. This finding, together with the synteny of PRAC loci, the relationships with prokaryotic PRAC inferred by taxon-rich phylogenetic analysis, and the absence in trypanosomatids of any other genera or in bodonids or euglenids suggest that a common ancestor of Trypanosoma gained PRAC gene by a single and ancient horizontal gene transfer (HGT) from a Firmicutes bacterium more closely related to Gemella and other species of Bacilli than to Clostridium as previously suggested. Our broad phylogenetic study allowed investigation of TryPRAC evolution over long and short timescales. TryPRAC genes diverged to become species-specific and genotype-specific for T. cruzi and T. rangeli, with resulting genealogies congruent with those obtained using vertically inherited genes.     Prevalência de anticorpos séricos para hantavírus em população rural do extremo sul do Estado de Santa Catarina, Brasil

    Introduction: Rodent-borne hantaviruses cause severe human diseases. We completed a serological survey of hantavirus infection in rural inhabitants of Turvo County, in the southern state of Santa Catarina, Brazil, in which seropositivity for hantavirus was correlated to previous disease in the participants. Methods: The levels of IgG antibodies to hantavirus Araraquara in the sera of 257 individuals were determined using an immunoenzymatic assay. Results: IgG antibodies to hantavirus were found in 2.3% of the participants. All seropositive participants reported previous disease with symptoms suggestive of hantavirus cardiopulmonary syndrome. Conclusions: Human infections causing unreported cardiopulmonary syndrome probably occur in the southern state of Santa Catarina.INTRODUÇÃO: Hantaviroses são zoonoses de roedores silvestres que causam doenças graves em seres humanos. Este trabalho mostra inquérito sorológico para hantavírus em população rural e periurbana do município de Turvo, Estado de Santa Catarina e a correlação da presença de anticorpos anti-hantavírus com a história mórbida pregressa dos indivíduos. \ud MÉTODOS: Investigaram-se os 257 participantes do estudo quanto à presença de anticorpos IgG séricos anti-hantavírus Araraquara, por método imunoenzimático. \ud RESULTADOS: A prevalência de anticorpos anti-hantavírus na população foi de 2,3%.     Genomic comparison of Trypanosoma conorhini and Trypanosoma rangeli to Trypanosoma cruzi strains of high and low virulence

    Abstract Background Trypanosoma conorhini and Trypanosoma rangeli, like Trypanosoma cruzi, are kinetoplastid protist parasites of mammals displaying divergent hosts, geographic ranges and lifestyles. Largely nonpathogenic T. rangeli and T. conorhini represent clades that are phylogenetically closely related to the T. cruzi and T. cruzi-like taxa and provide insights into the evolution of pathogenicity in those parasites. T. rangeli, like T. cruzi is endemic in many Latin American countries, whereas T. conorhini is tropicopolitan. T. rangeli and T. conorhini are exclusively extracellular, while T. cruzi has an intracellular stage in the mammalian host. Results Here we provide the first comprehensive sequence analysis of T. rangeli AM80 and T. conorhini 025E, and provide a comparison of their genomes to those of T. cruzi G and T. cruzi CL, respectively members of T. cruzi lineages TcI and TcVI. We report de novo assembled genome sequences of the low-virulent T. cruzi G, T. rangeli AM80, and T. conorhini 025E ranging from ~ 21–25 Mbp, with ~ 10,000 to 13,000 genes, and for the highly virulent and hybrid T. cruzi CL we present a ~ 65 Mbp in-house assembled haplotyped genome with ~ 12,500 genes per haplotype. Single copy orthologs of the two T. cruzi strains exhibited ~ 97% amino acid identity, and ~ 78% identity to proteins of T. rangeli or T. conorhini. Proteins of the latter two organisms exhibited ~ 84% identity. T. cruzi CL exhibited the highest heterozygosity. T. rangeli and T. conorhini displayed greater metabolic capabilities for utilization of complex carbohydrates, and contained fewer retrotransposons and multigene family copies, i.e. trans-sialidases, mucins, DGF-1, and MASP, compared to T. cruzi. Conclusions Our analyses of the T. rangeli and T. conorhini genomes closely reflected their phylogenetic proximity to the T. cruzi clade, and were largely consistent with their divergent life cycles. Our results provide a greater context for understanding the life cycles, host range expansion, immunity evasion, and pathogenesis of these trypanosomatids

    Impact of safety-related dose reductions or discontinuations on sustained virologic response in HCV-infected patients: Results from the GUARD-C Cohort

    BACKGROUND: Despite the introduction of direct-acting antiviral agents for chronic hepatitis C virus (HCV) infection, peginterferon alfa/ribavirin remains relevant in many resource-constrained settings. The non-randomized GUARD-C cohort investigated baseline predictors of safety-related dose reductions or discontinuations (sr-RD) and their impact on sustained virologic response (SVR) in patients receiving peginterferon alfa/ribavirin in routine practice. METHODS: A total of 3181 HCV-mono-infected treatment-naive patients were assigned to 24 or 48 weeks of peginterferon alfa/ribavirin by their physician. Patients were categorized by time-to-first sr-RD (Week 4/12). Detailed analyses of the impact of sr-RD on SVR24 (HCV RNA <50 IU/mL) were conducted in 951 Caucasian, noncirrhotic genotype (G)1 patients assigned to peginterferon alfa-2a/ribavirin for 48 weeks. The probability of SVR24 was identified by a baseline scoring system (range: 0-9 points) on which scores of 5 to 9 and <5 represent high and low probability of SVR24, respectively. RESULTS: SVR24 rates were 46.1% (754/1634), 77.1% (279/362), 68.0% (514/756), and 51.3% (203/396), respectively, in G1, 2, 3, and 4 patients. Overall, 16.9% and 21.8% patients experienced 651 sr-RD for peginterferon alfa and ribavirin, respectively. Among Caucasian noncirrhotic G1 patients: female sex, lower body mass index, pre-existing cardiovascular/pulmonary disease, and low hematological indices were prognostic factors of sr-RD; SVR24 was lower in patients with 651 vs. no sr-RD by Week 4 (37.9% vs. 54.4%; P = 0.0046) and Week 12 (41.7% vs. 55.3%; P = 0.0016); sr-RD by Week 4/12 significantly reduced SVR24 in patients with scores <5 but not 655. CONCLUSIONS: In conclusion, sr-RD to peginterferon alfa-2a/ribavirin significantly impacts on SVR24 rates in treatment-naive G1 noncirrhotic Caucasian patients. Baseline characteristics can help select patients with a high probability of SVR24 and a low probability of sr-RD with peginterferon alfa-2a/ribavirin

    Risk portfolio on modern finance has become increasingly technical, requiring the use of sophisticated mathematical tools in both research and practice. Since companies cannot insure themselves completely against risk, as human incompetence in predicting the future precisely that written in Al-Quran surah Luqman verse 34, they have to manage it to yield an optimal portfolio. The objective here is to minimize the variance among all portfolios, or alternatively, to maximize expected return among all portfolios that has at least a certain expected return. Furthermore, this study focuses on optimizing risk portfolio so called Markowitz MVO (Mean-Variance Optimization). Some theoretical frameworks for analysis are arithmetic mean, geometric mean, variance, covariance, linear programming, and quadratic programming. Moreover, finding a minimum variance portfolio produces a convex quadratic programming, that is minimizing the objective function ðð¥with constraintsð ð 𥠥 ðandð´ð¥ = ð. The outcome of this research is the solution of optimal risk portofolio in some investments that could be finished smoothly using MATLAB R2007b software together with its graphic analysis
