    Quadricipite architecture evaluation with ultrasound, in an elderly population

    Aging is associated with a progressive loss of neuromuscular function that often leads to a progressive disability and loss of independence. Degenerative processes begin to limit autonomy and functionality. Those processes is sarcopenia, defined as a gradual decline in muscle mass The aim of this study was to evaluate the muscle architecture, in an elderly population that practice exercise and a sedentary group, with the purpose of investigate the process of sarcopenia. Methods: Elderly people were evaluated, this evaluation was performed with an ultrasound, with 12 Mhz transducer. We evaluate both quadricipite muscles in contraction and in rest. A questionnaire was also per- formed for each individual in both samples. Informed consent was done. Main Results: Our results showed some differences in the muscle architecture in both groups. The main alteration was in the pennation angle that was higher in the exercised group. These differences altered the muscle performance. Conclusions: Muscular architecture is affected with disuse and sarcopenia. It is well known a relationship between performance, contractions and muscle architecture. Our results suggested that the atrophy directly affect muscles by reducing the volume and pennation angle. Thus, the effectiveness of strength training improves the structural and functional parameters of the skeletal muscle of elderly.info:eu-repo/semantics/publishedVersio

    Synthesis, Characterization and use of Nb2 O5 based Catalysts in Producing Biofuels by Transesterification, Esterification and Pyrolysis

    Nb2O5/HX (X = HSO4-, H2PO4-, NO3-) compounds were obtained from the treatment of niobium acid (Nb2O5·xH2O) with sulfuric, phosphoric, and nitric acids as well as Nb2O5 and Nb2O5·xH2O have been investigated as catalysts for the transesterification, esterification and pyrolysis of vegetable oils. The compounds were characterized by thermal analysis (DTA-TGA), spectroscopy (DRX, FT-IR and FT-Raman), surface area (BET) and the acidity (Ho) determined by n-butylamine titration using the Hammet´s indicator method. It was observed that after the acid treatment both the surface area and the acidity decreased as compared to the starting Nb2O5·xH2O. The only exception was a higher acidity verified when nitric acid was used. Among the catalyst investigated, the Nb2O5/H3PO4 presented the highest activity in the alcoholysis of soybean oil with different mono-alcohols (methanol, ethanol, 2-propanol, n-butanol). All tested solids seemed to stabilize the carboxylic acids formed during the pyrolysis, yielding higher acid numbers for the obtained products. Finally, the use of Nb2O5/H3PO4 and Nb2O5/H2SO4 as catalysts for the esterification showed better activity than Nb2O5·xH2O and Nb2O5

    An Henbest reduction, followed by the preparation of a silyl enol ether and oxidation in situ with m-CPBA has led to the neurosteroids 3[alpha]-hydroxy- and 3[alpha],21-dihydroxy-5[alpha]-pregnanolones. Using testosterone as starting material, a new short synthesis of an aromatase inhibitor, 4-OHA, has been achieved through hydroboration/oxidation followed by a Swern type oxidation and epimerization.     Antimicrobial, Antiproliferative and Proapoptotic Activities of Extract, Fractions and Isolated Compounds from the Stem of Erythroxylum caatingae Plowman

    In the study, we have examined the antitumor and antimicrobial activities of the methanol extract, the fractions, a fraction of total alkaloids and two alkaloids isolated from the stem of Erythroxylum caatingae Plowman. All test fractions, except the hexane fractions, showed antimicrobial activity on gram-positive bacteria and fungi. The acetate: methanol (95:5), acetate, chloroform and hexane fractions show the highest cytotoxicity activity against the NCI-H292, HEp-2 and K562 cell lines using MTT. The absence of hemolysis in the erythrocytes of mice was observed in these fractions and 6β-Benzoyloxy-3α-(3,4,5- trimethoxybenzoyloxy) tropane (catuabine B). Staining with Annexin V-FITC and JC-1 was used to verify the mechanism of action of the compounds of E. caatingae that showed cytotoxicity less than 30 μg/mL in leukemic cells. After 48 h of incubation, we observed that the acetate: methanol (95:5), acetate, and chloroform fractions, as well as the catuabine B, increased in the number of cells in early apoptosis, from 53.0 to 74.8%. An analysis of the potential of the mitochondrial membrane by incorporation of JC-1 showed that most cells during incubation of the acetate: methanol (95:5) and acetate fractions (63.85 and 59.2%) were stained, suggesting the involvement of an intrinsic pathway of apoptosis

    Body composition, strength static and isokinetic, and bone health: comparative study between active adults and amateur soccer players

AOW is supported by São Paulo Research Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) process: 2017/27234-2.Objective: To compare tissue composition, total and regional bone mineral content and bone mineral density, static hand grip and knee joint isokinetic strength between amateur soccer players and Control Group. Methods: Cross-sectional study. Air displacement plethysmography was used to estimate body volume and, in turn, density. Body composition, bone mineral content and bone mineral density were assessed for the whole body and at standardized regions using dual energy X-ray absorptiometry. Static grip strength was assessed with an adjustable dynamometer, and peak torque derived from isokinetic strength dynamometer (concentric muscular knee actions at 60°/s). Magnitude of the differences between groups was examined using d-Cohen. Results: Compared to healthy active adults, soccer players showed larger values of whole body bone mineral content (+651g; d=1.60; p<0.01). In addition, differences between groups were large for whole body bone mineral density (d=1.20 to 1.90; p<0.01): lumbar spine, i.e. L1-L4 (+19.4%), upper limbs (+8.6%) and lower limbs (+16.8%). Soccer players attained larger mean values in strength test given by static hand grip protocol (+5.6kg, d=0.99; p<0.01). Conclusion: Soccer adequately regulates body composition and is associated better bone health parameters (bone mineral content and density at whole-body and at particular sites exposed to mechanical loadings).Objetivo: Comparar a composição de tecidos, o conteúdo mineral ósseo e a densidade mineral óssea totais e por regiões do corpo, a força de preensão manual estática, e força isocinética da articulação do joelho, entre um grupo de jogadores de futebol amadores e um Grupo Controle. Métodos: Estudo transversal utilizando pletismografia de ar deslocado para estimar o volume corporal, para subsequente cálculo da densidade corporal. A composição de tecidos, o conteúdo mineral ósseo e a densidade mineral óssea foram avaliados para o corpo todo e regiões padronizadas através da absorciometria de raios-X de dupla energia. A força de preensão manual estática foi avaliada por um dinamômetro ajustável. Os momentos máximos de força das ações musculares concêntricas para os extensores e flexores do joelho foram avaliados pela dinamometria isocinética (60°/s). Foi calculado o valor d-Cohen para apreciar a magnitude do efeito das diferenças entre grupos. Resultados: Os futebolistas apresentaram níveis superiores de conteúdo mineral ósseo em comparação com os adultos ativos do Grupo Controle (+651g; d=1,60; p<0,01) e obtiveram valores superiores de densidade mineral óssea (d: 1,20 a 1,90; p<0,01) para a coluna lombar, L1-L4 (+19,4%), membros superiores (+8,6%) e membros inferiores (+16,8%). Para a força de preensão (estática) a diferença foi moderada (d=0,99; p<0,01) com valores mais elevados apresentados pelo futebolistas (+5,6kg; d=0,99; p<0,01). Conclusão: A prática de futebol promove uma regulação adequada da composição corporal (tecidos magro e gordo) e ganhos na densidade mineral óssea, mais acentuada em partes do corpo com maior exposição aos impactos mecânicos da atividade motora.info:eu-repo/semantics/publishedVersio

    New heterogeneous metal-oxides based catalyst for vegetable oil trans-esterification

    Neste trabalho são apresentados a preparação de novos materiais obtidos da co-precipitação de alumina e óxidos de estanho e zinco e seu uso como sistemas catalíticos ativos para alcoólise de óleos vegetais. Foi observado que estes óxidos metálicos do tipo (Al2O3)X(SnO)Y(ZnO) Z são ativos para a alcoólise do óleo de soja usando diversos álcoois, incluindo ramificados. Os melhores resultados foram observados utilizando metanol, sendo atingidas conversões acima de 80% em 4 horas. Foi também possível reciclar estes materiais sem perda aparente de sua atividade catalítica.The preparation of new materials obtained from the co-precipitation of aluminum, tin and zinc oxides and their use as catalytic system activities for vegetable oils alcoholysis are reported herein. It was observed that these metal-oxides of the type (Al2O3)X(SnO)Y(ZnO) Z are active for soybean oil alcoholysis, uzing several alcohols, including branched ones. Best result was achieved using methanol, with conversion yields up to 80% in 4 h. It was also possible to recycle the catalysts without apparent loss of activity

Although ADCs are in the spotlight, they still present significant engineering challenges. Therefore, there is an urgent need to develop more stable and effective ADCs. Most rabbit light chains have an extra disulfide bridge, that links the variable and constant domains, between Cys80 and Cys171, which is not found in the human or mouse. Thus, to develop a new generation of ADCs, we explored the potential of rabbit-derived VL-single-domain antibody scaffolds (sdAbs) to selectively conjugate a payload to Cys80. Hence, a rabbit sdAb library directed towards canine non-Hodgkin lymphoma (cNHL) was subjected to in vitro and in vivo phage display. This allowed the identification of several highly specific VL-sdAbs, including C5, which specifically target cNHL cells in vitro and present promising in vivo tumor uptake. C5 was selected for SN-38 site-selective payload conjugation through its exposed free Cys80 to generate a stable and homogenous C5-DAB-SN-38. C5-DAB-SN-38 exhibited potent cytotoxicity activity against cNHL cells while inhibiting DNA-TopoI activity.
