Angiotensin II and tumour necrosis factor α as mediators of ATP-dependent potassium channel remodelling in post-infarction heart failure

Aims Angiotensin II (Ang II) and tumour necrosis factor α (TNFα) are involved in the progression from compensated hypertrophy to heart failure. Here, we test their role in the remodelling of ATP-dependent potassium channel (KATP) in heart failure, conferring increased metabolic and diazoxide sensitivity. Methods and results We observed increased expression of both angiotensinogen and TNFα in the failing rat myocardium, with a regional gradient matching that of the KATP subunit Kir6.1 expression. Both angiotensinogen and TNFα expression correlated positively with Kir6.1 and negatively with Kir6.2 expression across the post-infarction myocardium. To further identify a causal relationship, cardiomyocytes isolated from normal rat hearts were exposed in vitro to Ang II or TNFα. We observed increased Kir6.1 and SUR subunit and reduced Kir6.2 subunit mRNA expression in cardiomyocytes cultured with Ang II or TNFα, similar to what was observed in failing hearts. In patch-clamp experiments, cardiomyocytes cultured with Ang II or TNFα exhibited responsiveness to diazoxide, in terms of both KATP current and action potential shortening. This was not observed in untreated cardiomyocytes and resembles the diazoxide sensitivity of failing cardiomyocytes that also overexpress Kir6.1. Ang II exerted its effect through induction of TNFα expression, because TNFα-neutralizing antibody abolished the effect of Ang II, and in failing hearts, regional expression of angiotensinogen matched TNFα expression. Finally, Ang II and TNFα regulated KATP subunit expression, possibly through differential expression of Forkhead box transcription factors. Conclusion This study identifies Ang II and TNFα as mediators of the remodelling of KATP channels in heart failur

Photo-initiated chemical vapor deposition as a scalable particle functionalization technology (a practical review)

ABSTRACT: Chemical vapor deposition (CVD), and its variants, is a more viable technology than the addition of surface active agents to modify nanoparticle surfaces. While thermally-activated CVD simply works by initiating the monomers using heat, some other techniques are more powerful and versatile. Indeed, higher energy CVD methods open up possibilities to a wider range of monomers. Unfortunately, different terminologies and classifications due to parallel work have led to confusion. This paper presents and explains the different techniques as well as their equivalent terminologies to clarify the big picture. While the demand for functionalized nanoparticles grows rapidly, current functionalization methods are still too expensive for most applications. This paper is intended to be a practical review of the gas phase methods available in order to identify a potential candidate for large scale functionalization of nanoparticles. This study identifies photo-initiated CVD (PICVD) as an ideal solution for scalable particle functionalization technology

Método para reconhecimento de íris baseado na sua região interna

    Get PDF

PESQUISA CIENTÍFICA EM CONTABILIDADE GERENCIAL NOS ENANPADs DE 2003 A 2008

The objective of this work is to know some bibliometrics aspects of Management Accounting (MA) in the EnANPADs from 2003 to 2008. In this empirical research 251 articles were analyzed based on the model of Shields (1997). To do so, descriptive statistical methods were used, besides the application of the law of Lotka. It was verified that the Brazilian authors follow the trend of replying the international studies in MA, in which the subject Management Control System (MCS) was predominant. These authors have explored more the new areas of the MA, corroborating Faro and Silva (2008) and Shields (1997). Women have extended their interest for the research in MA. It was perceived that an abandonment of the individual research has occurred, perhaps due to the pressure of CAPES for publications. The own articles of the authors were mentioned few times to base their later research, something distinct of what the international researchers do. Finally, with the estimated coefficient above Lotka's standard, it can be said that MA Brazilian authors do not publish with effectiveness in the EnANPAD, either due to the severity in the evaluation or because some of them only publishes works deriving of dissertations and thesis, not becoming acting researchers. Then, it was concluded that apparently there is a lack of scientific research in MA in the EnANPADs.O estudo objetiva conhecer alguns aspectos bibliométricos da Contabilidade Gerencial (CG) nos EnANPADs entre os anos de 2003-2008.

BackgroundThe incidence of chronic diseases, including diabetes mellitus (DM), heart failure (HF) and chronic obstructive pulmonary disease (COPD) is on the rise. The existing health care system must evolve to meet the growing needs of patients with these chronic diseases and reduce the strain on both acute care and hospital-based health care resources. Paramedics are an allied health care resource consisting of highly-trained practitioners who are comfortable working independently and in collaboration with other resources in the out-of-hospital setting. Expanding the paramedic's scope of practice to include community-based care may decrease the utilization of acute care and hospital-based health care resources by patients with chronic disease.Methods/designThis will be a pragmatic, randomized controlled trial comparing a community paramedic intervention to standard of care for patients with one of three chronic diseases. The objective of the trial is to determine whether community paramedics conducting regular home visits, including health assessments and evidence-based treatments, in partnership with primary care physicians and other community based resources, will decrease the rate of hospitalization and emergency department use for patients with DM, HF and COPD. The primary outcome measure will be the rate of hospitalization at one year. Secondary outcomes will include measures of health system utilization, overall health status, and cost-effectiveness of the intervention over the same time period. Outcome measures will be assessed using both Poisson regression and negative binomial regression analyses to assess the primary outcome.DiscussionThe results of this study will be used to inform decisions around the implementation of community paramedic programs. If successful in preventing hospitalizations, it has the ability to be scaled up to other regions, both nationally and internationally. The methods described in this paper will serve as a basis for future work related to this study.Trial registrationClinicalTrials.gov: NCT02034045. Date: 9 January 2014
