Humaria setimarginata (Pyronemataceae, Ascomycota), una nueva especie de México
Background and Aims: Humaria (Pyronemataceae, Ascomycota) is an ectomycorrhizal genus of fungi, mainly distributed in temperate forest. It is characterized by cupuliform to discoid apothecia that are covered with abundant hairs throughout the exterior of the brown ascomata, and by ellipsoid, hyaline, warty ascospores. There are 66 accepted species of which only one has been recorded in Mexico. The present study aims to describe a new species of Humaria with morphological, ecological and molecular data, found in Quercus forests from Tamaulipas, Mexico.
Methods: The specimens were collected in 2019 in the Victoria municipality, Tamaulipas, Mexico. Description and morphological studies were performed according to traditional mycological techniques. Studied material was deposited in the José Castillo Tovar Mycological Herbarium of the Instituto Tecnológico de Ciudad Victoria (ITCV) and the Escuela Nacional de Ciencias Biológicas Herbarium (ENCB) of the Instituto Politécnico Nacional. Phylogenetic analyses were performed using ITS and LSU sequences of the nuclear rDNA.
Key results: Humaria setimarginata sp. nov. is described and illustrated. This species is well differentiated by its morphological, molecular and ecological characteristics. It forms typically cupuliform apothecia with straight margin, abundant dark brown colored short hairs at the margin, and a greyish-white hymenium. It presents a dextrinoid reaction in the ectal excipulum in contact with Melzer´s reagent. It grows around Quercus rysophylla and Q. polymorpha in oak forests.
Conclusions: Humaria setimarginata is the second species of this genus reported in Mexico, after Humaria hemisphaerica. There are several collections under the name Humaria sp. that need a revision, to expand the diversity knowledge of this genus in this country. It is possible that several species exist, considering that this genus is ectomycorrhizal and the diversity of its potential hosts is high.
Sequencing of Non-model Plants for Understanding the Physiological Responses in Plants
From a genomic point of view, plants are complex organisms. Plants adapt to the environment, by developing different physiological and genetic properties, changing their genomic and expression profiles of adaptive factors, as exemplified by polyploidy studies. These characteristics along with the presence of duplicated genes/genomes make sequencing with early low-throughput DNA sequencing technologies in plants a challenging task. With the development of new technologies for molecular analysis, including transcriptome, proteome or microarray profiling, a new perspective in the genomic analysis was open, making possible to programs in species without genomic maps. The opportunity to extend molecular studies from laboratory model scale toward naturally occurring plant populations made it possible to precisely answer the longstanding important ecological and evolutionary questions. Some plant species have unique properties that could help to understand their adaptability to environment, crop production, pest protection or other biological processes. Molecular studies on non-model plants, including algae, mosses, ferns and plants with very specific characteristics are ongoing
Pediatric Chagas disease in the non-endemic area of Madrid: A fifteen-year review (2004-2018)
Background: Chagas disease (CD) has become an emerging global health problem in association with the immigration of individuals from endemic areas (in LatinAmerica) to other countries.Spain is the country in Europe with the highest number of CD cases. Concerning pediatric CD, treatment is not only better tolerated by younger children but also has greater cure possibilities. The aim of this study was to describe clinical and epidemiological aspects of CD in a pediatric population diagnosed of 10 hospitals in the Community of Madrid during the 2004-2018 period, as well as the safety and efficacy of CD treatment on this population. Methodology/principal findings: A multicenter, retrospective, descriptive study was conducted. The studied population included all identified children under the age of 18 with a diagnosis of CD. Diagnosis was performed with a positive parasitological test (with subsequent confirmation) or confirmed persistence of positive serology beyond 9 months, for children younger than one year-old, and with two different positive serological tests, for children older than one. Fifty-one children were included (59% male; 50.9% born in Spain). All mothers were from Latin America. The median age at diagnosis was 0.7 months for those under one year of age, and 11.08 years for those older than one year-old. Only one case presented a symptomatic course (hydrops faetalis, haemodynamic instability at birth, ascites, anaemia). For 94% treatment was completed. Considering patients who received benznidazole (47), AE were recorded in 48,9%. Among the 32 patients older than one year-old treated with benznidazole, 18 (56.25%) had adverse events whereas in the 15 under one year, 5(33,3%) did. Eigtheen (78.2%) of the patients with benznidazole AE were older than one year-old(median age 11.4 years). Of the patients treated with nifurtimox (9), AE were reported in 3 cases (33,3%). Cure was confirmed in 80% of the children under one year-old vs 4.3% in those older (p<0.001). Loss to follow- up occurred in 35.3% of patients. Conclusions/significances: Screening programs of CD since birth allow early diagnosis and treatment, with a significantly higher cure rate in children treated before one year of age, with lower incidence of adverse events. The high proportion of patients lost to follow-up in this vulnerable population is of concern.S

Efficacy of nimotuzumab according to inflammatory indices in patients with advanced non-small cell lung cancer
Introduction: The response to therapies in advanced lung cancer could be related to certain prognostic factors such as inflammatory indices.
Objective: To evaluate the efficacy of the humanized monoclonal antibody nimotuzumab in patients with advanced non-small cell lung cancer according to inflammatory indices.
Method: A retrospective longitudinal evaluation study was carried out in a universe of 498 patients older than 18 years, with a cytohistological diagnosis of non-small cell lung cancer, in advanced stages, after the first line of oncological therapy, including in multicenter clinical trials promoted by the Center for Molecular Immunology from 2002 to 2018. Descriptive statistics were applied, the x-tile 3.6.1 software was used for the Kaplan Meier test, significant differences were considered when p< 0,05.
Results: In the patients analyzed, nimotuzumab showed therapeutic benefit in the group of patients who did not progress to the first line of treatment with chemotherapy or chemoradiotherapy, when they had a lower neutrophil-lymphocyte index (p= 0,017 and p= 0,027) and a lower platelet-lymphocyte index (p= 0,030 and p= 0,009).
Conclusion: Selecting a patient with a lower inflammatory index benefits the efficacy of treatment with the humanized mAb nimotuzumab in advanced non-small cell lung cancer, which becomes a predictive tool for response to treatment
Natural History of MYH7-Related Dilated Cardiomyopathy
BACKGROUND Variants in myosin heavy chain 7 (MYH7) are responsible for disease in 1% to 5% of patients with dilated cardiomyopathy (DCM); however, the clinical characteristics and natural history of MYH7-related DCM are poorly described. OBJECTIVES We sought to determine the phenotype and prognosis of MYH7-related DCM. We also evaluated the influence of variant location on phenotypic expression. METHODS We studied clinical data from 147 individuals with DCM-causing MYH7 variants (47.6% female; 35.6 +/- 19.2 years) recruited from 29 international centers. RESULTS At initial evaluation, 106 (72.1%) patients had DCM (left ventricular ejection fraction: 34.5% +/- 11.7%). Median follow-up was 4.5 years (IQR: 1.7-8.0 years), and 23.7% of carriers who were initially phenotype-negative developed DCM. Phenotypic expression by 40 and 60 years was 46% and 88%, respectively, with 18 patients (16%) first diagnosed at <18 years of age. Thirty-six percent of patients with DCM met imaging criteria for LV noncompaction. During follow-up, 28% showed left ventricular reverse remodeling. Incidence of adverse cardiac events among patients with DCM at 5 years was 11.6%, with 5 (4.6%) deaths caused by end-stage heart failure (ESHF) and 5 patients (4.6%) requiring heart transplantation. The major ventricular arrhythmia rate was low (1.0% and 2.1% at 5 years in patients with DCM and in those with LVEF of <= 35%, respectively). ESHF and major ventricular arrhythmia were significantly lower compared with LMNA-related DCM and similar to DCM caused by TTN truncating variants. CONCLUSIONS MYH7-related DCM is characterized by early age of onset, high phenotypic expression, low left ventricular reverse remodeling, and frequent progression to ESHF. Heart failure complications predominate over ventricular arrhythmias, which are rare. (C) 2022 The Authors. Published by Elsevier on behalf of the American College of Cardiology Foundation
