    Gaia Eclipsing Binary and Multiple Systems. A study of detectability and classification of eclipsing binaries with Gaia

    In the new era of large-scale astronomical surveys, automated methods of analysis and classification of bulk data are a fundamental tool for fast and efficient production of deliverables. This becomes ever more imminent as we enter the Gaia era. We investigate the potential detectability of eclipsing binaries with Gaia using a data set of all Kepler eclipsing binaries sampled with Gaia cadence and folded with the Kepler period. The performance of fitting methods is evaluated with comparison to real Kepler data parameters and a classification scheme is proposed for the potentially detectable sources based on the geometry of the light curve fits. The polynomial chain (polyfit) and two-Gaussian models are used for light curve fitting of the data set. Classification is performed with a combination of the t-SNE (t-distrubuted Stochastic Neighbor Embedding) and DBSCAN (Density-Based Spatial Clustering of Applications with Noise) algorithms. We find that approximately 68% of Kepler Eclipsing Binary sources are potentially detectable by Gaia when folded with the Kepler period and propose a classification scheme of the detectable sources based on the morphological type indicative of the light curve, with subclasses that reflect the properties of the fitted model (presence and visibility of eclipses, their width, depth, etc.).Comment: 9 pages, 18 figures, accepted for publication in Astronomy & Astrophysic

    Collective cell migration in a fibrous environment:a hybrid multi-scale modelling approach

    International audienceThe specific structure of the extracellular matrix (ECM), and in particular the density and orientation of collagen fibres, plays an important role in the evolution of solid cancers. While many experimental studies discussed the role of ECM in individual and collective cell migration, there are still unanswered questions about the impact of nonlocal cell sensing of other cells on the overall shape of tumour aggregation and its migration type. There are also unanswered questions about the migration and spread of tumour that arises at the boundary between different tissues with different collagen fibre orientations. To address these questions, in this study we develop a hybrid multi-scale model that considers the cells as individual entities and ECM as a continuous field. The numerical simulations obtained through this model match experimental observations, confirming that tumour aggregations are not moving if the ECM fibres are distributed randomly, and they only move when the ECM fibres are highly aligned. Moreover, the stationary tumour aggregations can have circular shapes or irregular shapes (with finger-like protrusions), while the moving tumour aggregations have elongate shapes (resembling to clusters, strands or files). We also show that the cell sensing radius impacts tumour shape only when there is a low ratio of fibre to non-fibre ECM components. Finally, we investigate the impact of different ECM fibre orientations corresponding to different tissues, on the overall tumour invasion of these neighbouring tissues

    Simulating Photodynamic Therapy for the Treatment of Glioblastoma using Monte Carlo Radiative Transport

    Significance: Glioblastoma (GBM) is a rare but deadly form of brain tumor with a low median survival rate of 14.6 months, due to its resistance to treatment. An independent simulation of the INtraoperative photoDYnamic therapy for GliOblastoma (INDYGO) trial, a clinical trial aiming to treat the GBM resection cavity with photodynamic therapy (PDT) via a laser coupled balloon device, is demonstrated. Aim: To develop a framework providing increased understanding for the PDT treatment, its parameters, and their impact on the clinical outcome. Approach: We use Monte Carlo radiative transport techniques within a computational brain model containing a GBM to simulate light path and PDT effects. Treatment parameters (laser power, photosensitizer concentration, and irradiation time) are considered, as well as PDT’s impact on brain tissue temperature. Results: The simulation suggests that 39% of post-resection GBM cells are killed at the end of treatment when using the standard INDYGO trial protocol (light fluence = 200 J∕cm 2 at balloon wall) and assuming an initial photosensitizer concentration of 5 μM. Increases in treatment time and light power (light fluence = 400 J∕cm 2 at balloon wall) result in further cell kill but increase brain cell temperature, which potentially affects treatment safety. Increasing the p hotosensitizer concentration produces the most significant increase in cell kill, with 61% of GBM cells killed when doubling concentration to 10 μM and keeping the treatment time and power the same. According to these simulations, the standard trial protocol is reasonably well optimized with improvements in cell kill difficult to achieve without potentially dangerous increases in temperature. To improve treatment outcome, focus should be placed on improving the photosensitizer. Conclusions: With further development and optimization, the simulation could have potential clinical benefit and be used to help plan and optimize intraoperative PDT treatment for GBM.</p

    Impaired value-based decision-making in Parkinson’s Disease Apathy

    Apathy is a common and disabling complication of Parkinson’s disease characterized by reduced goal-directed behaviour. Several studies have reported dysfunction within prefrontal cortical regions and projections from brainstem nuclei whose neuromodulators include dopamine, serotonin and noradrenaline. Work in animal and human neuroscience have confirmed contributions of these neuromodulators on aspects of motivated decision-making. Specifically, these neuromodulators have overlapping contributions to encoding the value of decisions, and influence whether to explore alternative courses of action or persist in an existing strategy to achieve a rewarding goal. Building upon this work, we hypothesized that apathy in Parkinson’s disease should be associated with an impairment in value-based learning. Using a four-armed restless bandit reinforcement learning task, we studied decision-making in 75 volunteers; 53 patients with Parkinson’s disease, with and without clinical apathy, and 22 age-matched healthy control subjects. Patients with apathy exhibited impaired ability to choose the highest value bandit. Task performance predicted an individual patient’s apathy severity measured using the Lille Apathy Rating Scale (R = −0.46, P &lt; 0.001). Computational modelling of the patient’s choices confirmed the apathy group made decisions that were indifferent to the learnt value of the options, consistent with previous reports of reward insensitivity. Further analysis demonstrated a shift away from exploiting the highest value option and a reduction in perseveration, which also correlated with apathy scores (R = −0.5, P &lt; 0.001). We went on to acquire functional MRI in 59 volunteers; a group of 19 patients with and 20 without apathy and 20 age-matched controls performing the Restless Bandit Task. Analysis of the functional MRI signal at the point of reward feedback confirmed diminished signal within ventromedial prefrontal cortex in Parkinson’s disease, which was more marked in apathy, but not predictive of their individual apathy severity. Using a model-based categorization of choice type, decisions to explore lower value bandits in the apathy group activated prefrontal cortex to a similar degree to the age-matched controls. In contrast, Parkinson’s patients without apathy demonstrated significantly increased activation across a distributed thalamo-cortical network. Enhanced activity in the thalamus predicted individual apathy severity across both patient groups and exhibited functional connectivity with dorsal anterior cingulate cortex and anterior insula. Given that task performance in patients without apathy was no different to the age-matched control subjects, we interpret the recruitment of this network as a possible compensatory mechanism, which compensates against symptomatic manifestation of apathy in Parkinson’s disease.</p

    Participation in medical decision-making across Europe: an international longitudinal multicenter study

    Background: The purpose of this paper was to examine national differences in the desire to participate in decision-making of people with severe mental illness in six European countries. Methods: The data was taken from a European longitudinal observational study (CEDAR; ISRCTN75841675). A sample of 514 patients with severe mental illness from the study centers in Ulm, Germany, London, England, Naples, Italy, Debrecen, Hungary, Aalborg, Denmark and Zurich, Switzerland were assessed as to desire to participate in medical decision-making. Associations between desire for participation in decision-making and center location were analyzed with generalized estimating equations. Results: We found large cross-national differences in patients’ desire to participate in decision-making, with the center explaining 40% of total variance in the desire for participation (p<0.001). Averaged over time and independent of patient characteristics, London (mean=2.27), Ulm (mean=2.13) and Zurich (mean=2.14) showed significantly higher scores in desire for participation, followed by Aalborg (mean=1.97), where scores were in turn significantly higher than in Debrecen (mean=1.56). The lowest scores were reported in Naples (mean=1.14). Over time, desire for participation in decision-making increased significantly in Zurich (b=0.23) and decreased in Naples (b=-0.14). In all other centers, values remained stable. Conclusions: This study demonstrates that patients’ desire for participation in decisionmaking varies by location. We suggest that more research attention be focused on identifying specific cultural and social factors in each country to further explain observed differences across Europe

    Public drinking water supply and egg laying by Aedes aegypti

    OBJETIVO: Analisar o efeito da qualidade da água de reservatórios domésticos de abastecimento público na resposta oviposicional de fêmeas de Aedes aegypti. MÉTODOS: Estudo conduzido em laboratório a partir de imaturos de Ae. aegypti, coletados em caixas d'água do município de Potim, SP, 2009. Foram disponibilizados simultaneamente três tipos de água por gaiola para a deposição dos ovos: ovipositor (A) com água coletada em caixa d'água de Taubaté, SP; ovipositor (B) com água destilada (controle); e ovipositor (C) com água coletada em caixa d'água de Potim. Foram analisados parâmetros fisicoquímicos. O teste de Kruskall-Wallis foi utilizado para analisar a média de ovos nas diferentes amostras de água e nas comparações posteriores, o teste Dwass-Steel-Chritchlow-Flingner. O índice de atividade de oviposição foi adotado para avaliar a resposta oviposicional. RESULTADOS: Foi observada diferença significativa no número de ovos entre as soluções líquidas testadas (H = 45; p < 0,0001). O número de ovos na água de caixas d'água de abastecimento público de captação em poços profundos (C) foi estatisticamente superior a amostras de água de caixas d'água de abastecimento público de superfície (A) (p < 0,0001) e do Controle (B) (p < 0,0001). Não houve diferença significante entre o número de ovos do Controle (B) e água de superfície (A). A primeira postura foi a mais produtiva em todas as soluções testadas nas três gaiolas. A amostra de água (C) produziu índice positivo (0,54), i.e., atrativo para oviposição. CONCLUSÕES: A qualidade da água influiu na oviposição de Ae. aegypti. As elevadas concentrações de nitrogênio amoniacal na água de abastecimento público sugerem que esse componente químico foi o responsável pela atratividade de fêmeas grávidas de Ae. aegypti para a oviposição nesses criadouros.OBJECTIVE: To evaluate the effect of the quality of publicly supplied water in domestic water tanks on egg laying by female Aedes aegypti. METHODS: Laboratory study on immature Ae. aegypti, collected from water-tanks in the municipality of Potim, SP, Southeastern Brazil. Each cage contained three types of water in which eggs could be laid: Three choice per test were simultaneously used to deposit the eggs, ovipositor (A) with water collected from a water tank in Taubaté, ovipositor (B) with distilled water (control) and ovipositor (C) water collected from a water tank in the municipality of Potim. Physiochemical parameters were analyzed. The Kruskall-Wallis test was used to analyze the mean number of eggs in each water sample and the Dwass-Steel-Critchlow-Fligner test was used in making comparisons. To evaluate egg laying, an ovipositional activity index was adopted. RESULTS: A significant difference in the number of eggs was found between the liquid solutions tested (H = 45; p < 0.0001). The number of eggs found in water tank samples originating from deep wells (C), was statistically superior to water samples from water tanks originating from superficial wells (A) (p < 0.0001) and from the Control (C) p < 0.0001. There was no significant difference between the number of eggs in Control (B) and the surface water (A). In all three tests, the first position was the most productive in all tested solutions. Only water sample (C) produced a positive index (0.54), i.e., attractive to egg laying. CONCLUSIONS: Water quality influences egg laying by Aedes aegypti. The high concentrations of ammonium nitrate in public water supplies suggest that this chemical component was responsible for attracting pregnant female Aedes aegypti to lay eggs in these environments.OBJETIVO: Analizar el efecto de la calidad de agua de reservatorios domésticos de abastecimiento público en la respuesta de oviposición por hembras de Aedes aegypti. El número de huevos en el agua de los tanques para abastecimiento público, de captación en pozos profundos (C) fue estadísticamente superior a las muestras de tanques para abastecimiento público de superficie (A) (p˂0,0001) y del Control (B) (p˂0,0001). No hubo diferencia significativa entre el número de huevos del Control (B) y el agua de superficie (A). La primera postura fue la más productiva en todas las soluciones evaluadas en las tres jaulas. La muestra de agua (C) produjo índice positivo (0,54), es decir, atractivo para oviposición. CONCLUSIONES: La calidad de agua influyó en la oviposición de Ae. aegypti. Las elevadas concentraciones de nitrógeno amoniacal en el agua de abastecimiento público sugieren que dicho componente químico fue el responsable por la atracción de hembras preñadas de Ae. aegypti para la oviposición en tales criaderos

    Whole genome sequencing and imputation in isolated populations identify genetic associations with medically-relevant complex traits

    Next-generation association studies can be empowered by sequence-based imputation and by studying founder populations. Here we report ∼9.5 million variants from whole-genome sequencing (WGS) of a Cretan-isolated population, and show enrichment of rare and low-frequency variants with predicted functional consequences. We use a WGS-based imputation approach utilizing 10,422 reference haplotypes to perform genome-wide association analyses and observe 17 genome-wide significant, independent signals, including replicating evidence for association at eight novel low-frequency variant signals. Two novel cardiometabolic associations are at lead variants unique to the founder population sequences: chr16:70790626 (high-density lipoprotein levels beta −1.71 (SE 0.25), P=1.57 × 10−11, effect allele frequency (EAF) 0.006); and rs145556679 (triglycerides levels beta −1.13 (SE 0.17), P=2.53 × 10−11, EAF 0.013). Our findings add empirical support to the contribution of low-frequency variants in complex traits, demonstrate the advantage of including population-specific sequences in imputation panels and exemplify the power gains afforded by population isolates
