Specification of spatial relationships in directed graphs of cell signaling networks
Graph theory provides a useful and powerful tool for the analysis of cellular signaling networks. Intracellular components such as cytoplasmic signaling proteins, transcription factors and genes are connected by links, representing various types of chemical interactions that result in functional consequences. However, these graphs lack important information regarding the spatial distribution of cellular components. The ability of two cellular components to interact depends not only on their mutual chemical affinity but also on co-localization to the same subcellular region. Localization of components is often used as a regulatory mechanism to achieve specific effects in response to different receptor signals. Here we describe an approach for incorporating spatial distribution into graphs, and for the development of mixed graphs where links are specified by mutual chemical affinity as well as colocalization. We suggest that such mixed graphs will provide more accurate descriptions of functional cellular networks and their regulatory capabilities and aid in the development of large-scale predictive models of cellular behavior

Contributo da análise ergonómica do trabalho para o estudo de acidentes de trabalho em enfermeiros no serviço de urgência
Mestrado em Segurança e Higiene no Trabalho.O presente estudo pretende identificar os contributos da Análise Ergonómica Trabalho
(AET) para a compreensão dos Acidentes de Trabalho (AT) com implicações a nível da coluna lombar em enfermeiros do serviço de urgência (SU). Na maioria dos casos, os resultados da análise dos AT (AAT) revelam-se insuficientes, quer na gestão do
risco, quer na identificação dos motivos que impedem a aprendizagem com os AT. Metodologia: O estudo é de carácter exploratório/descritivo e transversal e integra todos os enfermeiros do SU de uma UH. A recolha de dados assenta na identificação dos últimos 10 AT no SU, recorrendo aos resultados da AAT e comparando-os com a aplicação da metodologia da AET (questionários, entrevistas, observação directa do local do AT, presencial, com registos em fotografia e/ou filme e confrontação dos acidentados).
Resultados esperados: a AAT relaciona-se essencialmente com causas activas, o que
é visível, atribuindo “culpas ao trabalhador” pelo acidente ocorrido. Em oposição, a AET identifica condições latentes da actividade de LMTD, designadamente a nível da concepção e organização do espaço de trabalho, da (in)adequação dos equipamentos, dos rácios doente/enfermeiro, da presença de stress, etc., que estão na origem da ocorrência de AT, frequentemente “ocultas ou diluídas” no contexto e, por isso, invisíveis, e associa-as às causas activas (ex. posições corporais, decisões e acções). Só através desta associação se consegue efectuar um diagnóstico das situações reais de trabalho. Espera-se um contributo para uma abordagem de gestão de risco mais assertiva e, sobretudo, com maior probabilidade de prevenção desses AT. ABSTRACT - This study aims to identify the contributions of Ergonomic Work Analysis (EWA) for the
understanding of Occupational Accidents (OA) with implications for the lumbar spine in
nurses at the emergency room department (ER). In most cases, the results of the
Occupational Accidents Analysis (OAA) are insufficient, either in risk management, or
in identifying the reasons which prevent the learning with the OA. Methodology: The study is an exploratory/descriptive and transversal that integrates all the nurses of the ER from one Hospital. The data collection is based on the identification from the last 10 OA occurred in the ER, using the results of the OAA comparing them with the EWA methodology (questionnaires, interviews, direct observation from the workplace where OA occurred, photography/movie records, and workers confrontation). Expected Results: The OAA is primarily related with active causes, what is visible, assigning "blame to worker" for the accident occurred. In opposition EWA identifies latent conditions of the manual patients handling (MPH) activity particularly, the design and layout from the workplace, the (in)adequacy of equipment, the ratios of patient/nurses, the presence of stress, etc., which are the source of OA, often "hidden or diluted" in the context and, therefore invisible, so those are associated with the active causes (eg, body positions, decisions and actions). Only through this association is possible to make a diagnosis of real work situations. It is expected to contribute to a more assertive risk management approach and, especially, to prevent OA related with manual patient handling
Fractional Brownian Motion in Finance
Mestrado em Matemática FinanceiraAlgumas das propriedades estatísticas dos dados financeiros são comuns a uma ampla variedade de mercados: a propriedade de memória longa, as caudas pesadas, assimetria (ganho / perda de assimetria), saltos, agrupamento de volatilidade, etc. A necessidade de procurar novos modelos de produtos financeiros tem aumentado nas últimas décadas devido à incapacidade dos actuais modelos explicarem algumas dessas propriedades estatísticas. Este trabalho tem como objetivo dar uma visão geral de alguns estudos que foram feitos relativamente à aplicação às finanças do movimento Browniano fracionário, em particular o trabalho de Paolo Guasoni e Cheridito Patrick, que mostram que, se assumirmos certas restrições, podemos eliminar oportunidades de arbitragem. Além disso, também são apresentados estudos empíricos com dados de mercado, com o objectivo de mostrar como se pode obter um estimador para o índice Hurst (o parâmetro do movimento Browniano fracionário). Para este fim, foram utilizados dois métodos, o método Rescaled Range e o método modificado do Rescaled Range. Este estudo permite-nos discutir o efeito de memória nas séries temporais de alguns índices de mercado.Some of the statistical properties of the financial data are common to a wide variety of markets: long-range dependence properties, heavy tails, skewness (gain/loss asymmetry), jumps, volatility clustering, etc. The need to seek new models for financial products has increased in recent decades due to the inability of current models to explain some of these facts. One of these models is fractional Brownian motion. This work aims to give an overview of some studies that were done on the financial applications of fractional Brownian motion, in particular the work of Paolo Guasoni and Patrick Cheridito which shows that if we assume certain restrictions, we can eliminate arbitrage opportunities. Moreover, we also present empirical studies with market data, in order to show how to obtain an estimator for the Hurst index (the fractional Brownian motion parameter). Exploring the impacts of distance higher education on adult learners' lives and reclaiming lifelong learning as a human development process
This article intends to launch discussion and reflection on two main themes: lifelong learning and digital
literacy in nowadays societies. In looking for the intersections between these concepts to connote them with
a more humanistic and holistic perspective, we explore the potentials of distance learning in the lives of adult
learners. The empirical basis for this exploration is a survey applied to 706 (143 respondents) graduates in
the education of the Portuguese Open University to know their future projects and the impact of this degree
in the various dimensions of their personal, social and professional lives. The conclusions of this analysis
point to recognition, by the respondents, of the positive impacts that distance higher education has in their
personal and social lives. However, these impacts are not as visible in professional terms since the answers
show little significant professional progressions. In this sense, the article concludes that it is important to
rethink the founding ideas of the concept of lifelong learning from a humanistic perspective and to approach
it with a holistic and transversal conception of what is now defined as digital literacy. Distance education, for
adult learners, is a scenario that not only strengthens the personal, social and professional development of
individuals, but also the development of competencies applied not only to the digital world but also to each

O transporte vertical de momento linear num caso de convecção profunda
Mestrado em Meteorologia e Oceanografia FísicaO objectivo deste trabalho científico é o estudo do transporte vertical de
momento linear horizontal (CMT) realizado pelas nuvens cumulus (Cu) em
sistemas de nuvens convectivos sobre a região tropical dos oceanos. Para
realizar este estudo, foram utilizadas as simulações tridimensionais do modelo
explícito de nuvens (CRM) para os quatro meses de duração da campanha
observacional TOGA COARE nas águas quentes do Pacífico ocidental.
A aplicação de uma análise diária a vários campos simulados de modo a
comparar, para um período de 10 dias, duas simulações em dois domínios
espaciais diferentes: 64x64 e 256x256 nós, estabeleceu que o CMT não
depende do domínio espacial. O teste realizado à parameterização do
percurso de mistura colocou em evidência os fracos resultados desta
parameterização na troposfera média, indicando que algumas
parameterizações do CRM, nomeadamente do fluxo de massa da corrente
ascendente da nuvem no CRM devem ser melhoradas.
ABSTRACT: The aim of the proposed research is the investigation of vertical cumulus
momentum transport (CMT) in tropical oceanic convective cloud systems. Our
approach is to use a unique four month three-dimensional cloud resolving
model (CRM) simulations of TOGA COARE - a major field experiment over the
warm waters of the west Pacific.
For the purpose of testing the CRM sensitivity to the spatial domain resolution,
a daily mean analysis centered in a strong convection episode with substantial
mean vertical shear was performed over several simulated fields, establishing
that the CMT is insensitive to the domain size. The test applied to the skill of a
downgradient mixing-length parameterization highlighted very poor results in
the mid-troposphere, suggesting that some CRM parameterizations might be
responsible for these results, namely in updraft cloud mass flux field, and
should be refined

Fast-to-fed shift in glucose homeostasis: clues to an earlier detection of human prediabetic states
FCM: UC Bioquímica I - PhD ThesisA acção da insulina está associada à libertação da substância hepática sensibilizadora da insulina
(HISS), que aumenta o aporte de glucose periférico. No estado pós-prandial, a libertação da HISS é
máxima, diminuindo com o período de jejum. O controlo prandial da acção da HISS é mediado pelo
sistema parassimpático hepático/óxido nítrico (NO) e pelo glutationo (GSH) hepático. Os actuais
métodos utilizados para avaliar a sensibilidade à insulina são realizados no estado de jejum. A presente dissertação destaca a hipótese de que o mecanismo dependente da HISS existe em humanos, e pode ser manipulado. Em humanos, uma robusta ferramenta para caracterizar a acção da insulina dependente da HISS,
não só no estado de jejum, mas também após uma refeição, o teste rápido de sensibilidade à insulina (RIST), foi desenvolvido. O RIST pode ser realizado com reproductibilidade, e sem intra e intervariabilidade. A diminuição da sensibilidade à insulina observada no jejum é potenciada após uma refeição, e a
administração de atropina, suprime este efeito. A inibição parcial da sensibilidade à insulina induzida
pela refeição, é consistente com a hipótese de que um “sinal prandial” dependente do sistema
parassimpático hepático é necessário para a libertação hepática da HISS.
Quando voluntários magros e com excesso de peso foram submetidos a um período de 24h de
jejum, a acção da insulina per se, foi similar em ambos os grupos estudados. Contudo, quando
avaliados no estado pós-prandial, os resultados apresentados nesta dissertação mostraram que, a
potenciação induzida pela refeição era inferior nos voluntários com excesso de peso, estando esta
associada a uma alteração da componente da acção da insulina dependente da HISS. Estes resultados
indicam a importância da avaliação da sensibilidade à insulina no estado pós-prandial, e sugerem também que este estado de pré-diabetes apenas pode ser detectado após a ingestão de uma refeição. Níveis elevados de glucagina estão associados à diabetes tipo 2. Sabendo que a glucagina leva a
uma diminuição da síntese de GSH, e que o GSH é fundamental para a libertação da HISS, foi avaliado
o efeito da glucagina na via da HISS. Em animais, tanto a administração de um análogo do cAMP,
como de glucagina, produziram um decréscimo da sensibilidade à insulina, sendo este efeito
dependente da dose. A resistência à insulina dependente da HISS observada aquando da
administração de um inibidor do sintetase do NO, não se agravou com a posterior administração de
glucagina. Estes resultados sugerem então que a glucagina induz um decréscimo da sensibilidade à insulina, sendo este dependente da via da HISS, e não por acção de outra via. Estas duas observações indicam que a glucagina, através da via de finalização do cAMP, leva à diminuição dos níveis hepáticos de GSH e, consequentemente, a uma alteração da via da HISS. Esta alteração poderá ser a responsável por um estado precoce de resistência à insulina
