    Occupational Injuries on Thoroughbred Horse Farms: A Description of Latino and Non-Latino Workers\u27 Experiences

    Animal production is a dangerous industry and increasingly reliant on a Latino workforce. Within animal production, little is known about the risks or the occupational hazards of working on farms involved in various aspects of thoroughbred horse breeding. Extant research suggests that horse workers are at risk of musculoskeletal and respiratory symptoms, kicks, and other injuries. However, limited known research has examined the experiences of the industry\u27s workers, including immigrant workers, despite their prominence and increased vulnerability. Using data collected from thoroughbred farm representatives via a phone-administered survey, a 2-hour face-to-face semi-structured interview, and farm injury logs, this article identifies and describes types of injuries experienced by workers (N = 284) and their surrounding circumstances. Results indicate that general injuries and musculoskeletal strains, sprains, and tears account for a majority of injuries among workers on thoroughbred farms. Upper limbs and extremities are most frequently injured, while direct contact with the horse accounted for over half of all injuries. No differences in the diagnoses or distribution of injury were found by ethnicity; however, Latinos were more often struck by or trampled by a horse while non-Latinos were more often injured by an insect or plant. Implications and opportunities for future research are discussed

    Reciprocal co-regulation of EGR2 and MECP2 is disrupted in Rett syndrome and autism

    Mutations in MECP2, encoding methyl-CpG-binding protein 2 (MeCP2), cause the neurodevelopmental disorder Rett syndrome (RTT). Although MECP2 mutations are rare in idiopathic autism, reduced MeCP2 levels are common in autism cortex. MeCP2 is critical for postnatal neuronal maturation and a modulator of activity-dependent genes such as Bdnf (brain-derived neurotropic factor) and JUNB. The activity-dependent early growth response gene 2 (EGR2), required for both early hindbrain development and mature neuronal function, has predicted binding sites in the promoters of several neurologically relevant genes including MECP2. Conversely, MeCP2 family members MBD1, MBD2 and MBD4 bind a methylated CpG island in an enhancer region located in EGR2 intron 1. This study was designed to test the hypothesis that MECP2 and EGR2 regulate each other’s expression during neuronal maturation in postnatal brain development. Chromatin immunoprecipitation analysis showed EGR2 binding to the MECP2 promoter and MeCP2 binding to the enhancer region in EGR2 intron 1. Reduction in EGR2 and MeCP2 levels in cultured human neuroblastoma cells by RNA interference reciprocally reduced expression of both EGR2 and MECP2 and their protein products. Consistent with a role of MeCP2 in enhancing EGR2, Mecp2-deficient mouse cortex samples showed significantly reduced EGR2 by quantitative immunofluorescence. Furthermore, MeCP2 and EGR2 show coordinately increased levels during postnatal development of both mouse and human cortex. In contrast to age-matched Controls, RTT and autism postmortem cortex samples showed significant reduction in EGR2. Together, these data support a role of dysregulation of an activity-dependent EGR2/MeCP2 pathway in RTT and autism

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    ‘The Way of the Rain’: Towards a conceptual framework for the retrospective examination of historical American and Australian ‘rain follows the plow/plough’ messages

    History establishes that even before the advent of modern media, erroneous climate messages were disseminated to the public. A folk belief captured by the phrase ‘rain follows the plow’ (RFTP) is a prime example of such misinformation. This belief, popular in the late nineteenth century, maintained that cultivation of arid lands in the United States beyond the 100th meridian west would boost precipitation, creating a climate more favourable for farming. Encouraged by this narrative, homesteaders cultivated arid lands west of the meridian. Rain did not follow the plough and many farms in the Great Plains failed. RFTP was also invoked in South Australia in support of agricultural settlement north of Goyder’s Line, a geographical boundary delineating the limits of reliable rainfall in the colony. This article revisits the origins of the doctrine and places RFTP messaging in its historical context by examining articles and poetry published in American and Australian settlement-era newspapers. The results of two newspaper database surveys reveal that a number of historical RFTP stories and an environmental poem with religious overtones appeared first in US newspapers and were later republished in newspapers throughout Australia. One of the surveys also reveals that, from 1876 to 1898, reports of parliamentary discussions or debates referencing the slogan were published in South Australia. The dissemination of inaccurate climate information in settlement-era America and Australia is discussed in relation to a proposed conceptual framework based upon contemporary theories of science communication that might provide a basis for the analysis of historical science messaging

    Understanding work organisation factors on thoroughbred farms in southeastern United States

    There is little reported on the work environment of thoroughbred breeding operations. As a first step toward minimizing risk in this hazardous industry, this study documents farm and workforce characteristics, employment conditions, and organisational and job factors on thoroughbred farms in one southeastern state in the U.S. Data were collected via a phone-administered survey with a convenience sample of management representatives (owner, manager, or human resource personnel) from 32 thoroughbred breeding farms. Farms chiefly employed a full-time, non-native, low-wage labour force that worked long hours year-round, but that was offered numerous benefits. Seasonal workers, also commonly employed, received low wages, few benefits, and experienced low retention. Future research is necessary to determine how the interplay between work organisation factors influences farmworkers' risk of injury and illness as well as their subsequent health outcomes
