    "Jnking” atherosclerosis

Abstract.: Numerous studies in animal models established a key role of the C-jun N-terminal kinase (JNK) family (JNK1, JNK2 and JNK3) in numerous pathological conditions, including cancer, cardiac hypertrophy and failure, neurodegenerative disorders, diabetes, arthritis and asthma. A possible function of JNK in atherosclerosis remained uncertain since conclusions have mainly been based on in vitro studies investigating endothelial cell activation, T-effector cell differentiation and proliferation of vascular smooth muscle cells, all of which represent crucial cellular processes involved in atherosclerosis. However, recent experiments demonstrated that macrophage-restricted deletion of JNK2 was sufficient to efficiently reduce atherosclerosis in mice. Furthermore, it has been shown that JNK2 specifically promotes scavenger receptor A-mediated foam cell formation, an essential step during early atherogenesis, which occurs when vascular macrophages internalize modified lipoproteins. Thus, specific inhibition of JNK2 activity may emerge as a novel and promising therapeutic approach to attenuate atheroma formation in the future. In this review, we discuss JNK-dependent cellular and molecular mechanisms underlying atherosclerosi

Narratives from the Online Frontier: A K-12 Student’s Experience in an Online Learning Environment

    Get PDF
Despite a large increase in the number of students enrolled in online courses, published research on student experiences in these environments is minimal. This article reports the narrative analysis of a series of interviews conducted with a female student at a brick-and-mortar school enrolled in a single virtual school course. Her narratives describe a student who often struggled with the content in her online course and was reluctant to interact with her online teacher. When she interacted with people online, it was using text, because she was shy and the hardware often did not work. Darlene’s experiences, likely typical of many K-12 online students, highlight a system in need of better strategies for the design and delivery of its educational opportunities

Effect of higher salinities on growth and survival of pacific white shrimp, Litopenaeus vannamei (Boone, 1931)

The growth and survival of Litopenaeus vannamei post larvae was measured in controlled different salinities condition 35ppt (T1), 40ppt (T2), 45ppt (T3) and 50ppt (T4) were maintained. Group of Shrimp post larvae (weight 0.032 g ± 0.002) were stocked at a density of 35 nos. /aquarium in above salinity ranges. Animals were fed with commercial feed (35% Crude Protein) @ 5% of body weight four times a day. The results indicate that higher SGR was observed in T2 (1.99±0.08) followed by T1 (1.75±0.07), T3 (1.54±0.06) and T4 (1.49±0.17). Highest survival (100 %) was recorded in T1 followed by T2 (96.42%), T3 (94.99%) and T4 (74.21%). From the results of the present study it could be seen that higher salinity significantly reduced the growth and survival of L. vannamei but will also open study area of physiological adaption of animal at higher saline water in performance of organisms

Mechanistic insights into the transcriptional arrest in the presence of Double Strand Breaks

Double-strand breaks (DSBs) occur frequently in the genome during genome replication or by DNA damaging agents. DNA lesions affect fundamental DNA-dependent nuclear processes, such as replication and transcription. We have developed an experimental system where DSBs are induced at coding regions of RNA polymerase II transcribing genes. We have started to study the kinetics of RNA polymerase II transcription inhibition in the presence of DNA breaks. We observed that induction of the break led to transcription inhibition and the restoration of transcription closely followed the dynamics of the repair of breaks. We confirmed by chromatinimmunoprecipitation that the break induction led to displacement of RNA polymerase II affecting both the elongation and the initiation of transcription. Our results show that this is dependent on one of the major kinase in DNA damage repair called DNAPKcs. We also investigated the downstream steps of RNA polymerase II removal and we claimed that it was a multistep process involving additional kinases and ubiquitin ligases NEDD4 and CUL3. At the last step of break dependent transcriptional silencing the RNA polymerase II is targeted for proteasome dependent degradation. These data demonstrate that the DNA damage repair complexes and proteasomal system have a synergistic and active role in transcriptional silencing during the DSB repair by removing the RNA pol II from the transcribing region. We show here that DNA lesions occurring at transcribed regions cause a transient repression until the lesion is repaired. This is probably a cell defense mechanism to avoid production of truncated or mutated transcripts in essential genes whose alterations in their gene expression would endanger cell viability. Understudying the role of DNAPKcs, in preventing RNA pol II bypassing a DSB might be a key in avoiding the production of mutated transcripts that could lead to cancerous phenotypes

The study was based on the analysis of e-governance for administration in Pakistan related to public trust. The study was based on a quantitative approach and a literature review from a theoretical perspective. The quantitative approach was based on the survey from purposive sampling. The survey questionnaire was based on different variables based on the theoretical framework of public trust and the role of e-governance. The results from the survey have shown that e-governance is an emerging platform in Pakistan. However, it needs to work on accountability, responsiveness, clarity, transparency, laws, and regulations to become effective in maintaining public trust in maintaining Pakistan administration
