    How to improve Drenchrite

    Understanding the strengths and weaknesses of DrenchRite and how it relates to faecal egg count reduction tests (FECRT) will lead to clearer interpretation of DrenchRite results and a better perspective on both tests. In previous studies efficacy was estimated for worms resistant to BZ, LEV and BZILEV combinations by FECRT, DrenchRite and post mortem worm counts. Correlations of in vivo efficacy with FECRT determined by them were: Trichostrongylus colubriformis 88%, Ostertagia circumcincta 56% and Haemonchus contortus 69%. The low correlation for O. circumcincta was associated with the variability in estimating species composition when O. circumcincta was present at low levels. Correlations of DrenchRite with in vivo efficacy were: O. circumcincta 87%, T. colubriformis 82% and H. contortus 80%. The correlation between resistance determined by Larval Development Assay (LDA) and FECRT was not high at 61% (averaged over drugs and species). Similar data for Macrocyclic Lactones (ML) resistant worms are not yet available. Previous studies have shown for some LDA systems that eggs are more likely to be inhibited in development to L3s early (2-3 weeks post infection (PI)) and late (beyond 10 weeks PI) after infection and are most resilient to the effect of drugs at about 7 weeks PI. No systematic changes in DrenchRite results were observed over age of worm infection when tested weekly for 2 to 14 week old worms. Large bowel worms of sheep have not generally been examined under LDA conditions so a study was undertaken to determine the LD50s of susceptible Oesophagostomum columbianum (nodule worm) and Chabertia ovina (large mouthed bowel worm) on DrenchRite plates. LD50s for BZ and LEV were approximately 0.04 and 0.8 uM respectively for both worms. For the ML O. columbianum had a much higher LD50 (45 nM) than C. ovina (8 nM). In relation to the other common sheep worms, development characteristics of C. ovina in LDA is like H. contortus while O. columbianum is more like O. circumcincta. Both large bowel worms were very sensitive to BZILEV combinations, with LD50s 4 to 8 fold lower than those observed for H. contortus and O. circumcincta

    Charge Fluctuations on Membrane Surfaces in Water

    We generalize the predictions for attractions between over-all neutral surfaces induced by charge fluctuations/correlations to non-uniform systems that include dielectric discontinuities, as is the case for mixed charged lipid membranes in an aqueous solution. We show that the induced interactions depend in a non-trivial way on the dielectric constants of membrane and water and show different scaling with distance depending on these properties. The generality of the calculations also allows us to predict under which dielectric conditions the interaction will change sign and become repulsive

    Preliminary data on increased reactivity towards children in distress after testosterone administration in women: a matter of protection?

    Emotional reactivity to others' distress is a vital prerequisite for a caring response. Testosterone, in contrast, is mostly associated with protection of personal dominance and decreased responsiveness to others' needs. However, experimental work also indicates that rising testosterone levels in response to infant distress can potentially facilitate protection. We assessed the impact of testosterone administration on participants' emotional reactivity to infants in distress, measuring their facial responses on the corrugator supercilii forehead muscle ('frowning') and the zygomaticus major ('smiling') as an index of emotional responses towards children. Moreover, we probed whether the effect of testosterone is moderated by participants' self-reported nurturance and protective tendencies. Our preliminary results showed that testosterone not only increased emotional reactivity to empathy eliciting images of children, but that this increase was strongest in participants with strong protective tendencies.     Effects of testosterone administration on threat and escape anticipation in the orbitofrontal cortex.

    Recent evidence suggests that the steroid hormone testosterone can decrease the functional coupling between orbitofrontal cortex (OFC) and amygdala. Theoretically this decoupling has been linked to a testosterone-driven increase of goal-directed behaviour in case of threat, but this has never been studied directly. Therefore, we placed twenty-two women in dynamically changing situations of escapable and inescapable threat after a within-subject placebo controlled testosterone administration. Using functional magnetic resonance imaging (fMRI) we provide evidence that testosterone activates the left lateral OFC (LOFC) in preparation of active goal-directed escape and decouples this OFC area from a subcortical threat system including the central-medial amygdala, hypothalamus and periaqueductal gray. This LOFC decoupling was specific to threatening situations, a point that was further emphasized by an absence of such decoupling in a second experiment focused on resting-state connectivity. These results not only confirm that testosterone administration decouples the LOFC from the subcortical threat system, but also show that this is specifically the case in response to acute threat, and ultimately leads to an increase in LOFC activity when the participant prepares a goal-directed action to escape. Together these results for the first time provide a detailed understanding of functional brain alterations induced by testosterone under threat conditions, and corroborate and extend the view that testosterone prepares the brain for goal-directed action in case of threat.FSW – Publicaties zonder aanstelling Universiteit Leide

    Ginzburg-Landau functional for nearly antiferromagnetic perfect and disordered Kondo lattices

    Interplay between Kondo effect and trends to antiferromagnetic and spin glass ordering in perfect and disordered bipartite Kondo lattices is considered. Ginzburg-Landau equation is derived from the microscopic effective action written in three mode representation (Kondo screening, antiferromagnetic correlations and spin liquid correlations). The problem of local constraint is resolved by means of Popov-Fedotov representation for localized spin operators. It is shown that the Kondo screening enhances the trend to a spin liquid crossover and suppresses antiferromagnetic ordering in perfect Kondo lattices and spin glass ordering in doped Kondo lattices.     Mass‐loading the Earth's dayside magnetopause boundary layer and its effect on magnetic reconnection

    When the interplanetary magnetic field is northward for a period of time, O+ from the high‐latitude ionosphere escapes along reconnected magnetic field lines into the dayside magnetopause boundary layer. Dual‐lobe reconnection closes these field lines, which traps O+ and mass loads the boundary layer. This O+ is an additional source of magnetospheric plasma that interacts with magnetosheath plasma through magnetic reconnection. This mass loading and interaction is illustrated through analysis of a magnetopause crossing by the Magnetospheric Multiscale spacecraft. While in the O+‐rich boundary layer, the interplanetary magnetic field turns southward. As the Magnetospheric Multiscale spacecraft cross the high‐shear magnetopause, reconnection signatures are observed. While the reconnection rate is likely reduced by the mass loading, reconnection is not suppressed at the magnetopause. The high‐latitude dayside ionosphere is therefore a source of magnetospheric ions that contributes often to transient reduction in the reconnection rate at the dayside magnetopause.publishedVersio

    Amygdala responses to emotional faces in twins discordant or concordant for the risk for anxiety and depression.

    Background: Functional brain imaging studies have shown deviant amygdala responses to emotional stimuli in subjects suffering from anxiety and depressive disorder, but both hyperactivity and hypoactivity compared to healthy controls have been reported. To account for these discrepant findings, we hypothesize that genetic and environmental risk factors differently impact on amygdala functioning. Methods: To test this hypothesis, we assessed amygdala responses to an emotional faces paradigm during functional magnetic resonance imaging in monozygotic twin pairs discordant for the risk of anxiety and depression (n = 10 pairs) and in monozygotic twin pairs concordant for high (n = 7 pairs) or low (n = 15 pairs) risk for anxiety and depression. Results: Main effects (all faces vs. baseline) revealed robust bilateral amygdala activity across groups. In discordant twins, increased amygdala responses were found for negatively valenced stimuli (angry/anxious faces) in high-risk twins compared to their low-risk co-twins. In contrast, concordant high-risk pairs revealed blunted amygdala reactivity to both positive and negative faces compared with concordant low-risk pairs. Post-hoc analyses showed that these findings were independent of 5-HTTLPR genotype. Conclusions: Our findings indicate amygdala hyperactivity in subjects who are at high risk for anxiety and depression through environmental factors and amygdala hypoactivity in those at risk mainly through genetic factors. We suggest that this result reflects a difference in baseline amygdala activation in these groups. Future imaging studies on anxiety and depression should aim to avoid admixture of subjects who are at genetic risk with those at risk due to environmental factors. © 2008 Elsevier Inc.
