    Ethylene supports colonization of plant roots by the mutualistic fungus Piriformospora indica

    The mutualistic basidiomycete Piriformospora indica colonizes roots of mono- and dicotyledonous plants, and thereby improves plant health and yield. Given the capability of P. indica to colonize a broad range of hosts, it must be anticipated that the fungus has evolved efficient strategies to overcome plant immunity and to establish a proper environment for nutrient acquisition and reproduction. Global gene expression studies in barley identified various ethylene synthesis and signaling components that were differentially regulated in P. indica-colonized roots. Based on these findings we examined the impact of ethylene in the symbiotic association. The data presented here suggest that P. indica induces ethylene synthesis in barley and Arabidopsis roots during colonization. Moreover, impaired ethylene signaling resulted in reduced root colonization, Arabidopsis mutants exhibiting constitutive ethylene signaling, -synthesis or ethylene-related defense were hyper-susceptible to P. indica.     Die Fragebogenforschung belegt, dass Respondenten durch Kontextinformationen eines Fragebogens systematisch in ihrem Antwortverhalten beeinflusst werden. So zeigten Norenzayan und Schwarz (1999), dass Probanden bei freier Antwortmöglichkeit eher persönlichkeitsbezogene Ursachen zur ErklĂ€rung von Straftaten nennen, wenn der Fragebogen scheinbar von einem Institut fĂŒr Persönlichkeitsforschung (verglichen mit einem Institut fĂŒr Sozialforschung) erstellt wurde. Hierzu diskutierte ErklĂ€rungen sind einerseits Konversationsmaximen, die einen Bezug zwischen Adressat und Gesagtem induzieren, andererseits kognitive Primings, die selektive kognitive Aktivierungen und damit VerfĂŒgbarkeiten bedingen sollen. Die vorliegende Studie untersucht diese ErklĂ€rungsalternativen, indem sie erstmals in einem analogen Studiendesign persönlichkeitsbezogene und soziale GrĂŒnde in geschlossenen Antwortformaten vorgibt und gewichten lĂ€sst. Mögliche Gewichtungsunterschiede sind somit nicht mittels kognitiver VerfĂŒgbarkeit erklĂ€rbar.     A call to arms: setting the framework for a code of practice for mosquito management in European wetlands

    1. Wetlands provide multiple services to human societies. Despite policies dedicated to their protection, current European policies do not address the need to balance mosquito management approaches to mitigate dis‐services to human health and well‐being while ensuring that wetland conservation goals are met. 2. Herein, we outline criteria for consideration when developing mosquito control programmes in European wetlands that will allow managers and public health authorities to adopt effective and ecologically sound approaches. 3. Synthesis and applications . The proposed code of practice provides practical advice to local authorities and those involved in mosquito control in order to design an integrated mosquito management strategy that aligns with current environmental legislation. Although this code of practice was developed by European experts, it is transferable to other geographical contexts, integrating the expertise and knowledge of local stakeholders and researchers from the fields of medical entomology, human and animal health and ecology

    Cell-Type-Specific Complement Expression in the Healthy and Diseased Retina

    Complement dysregulation is a feature of many retinal diseases, yet mechanistic understanding at the cellular level is limited. Given this knowledge gap about which retinal cells express complement, we performed single-cell RNA sequencing on similar to 92,000 mouse retinal cells and validated our results in five major purified retinal cell types. We found evidence for a distributed cell-type-specific complement expression across 11 cell types. Notably, Muller cells are the major contributor of complement activators c1s, c3, c4, and cfb. Retinal pigment epithelium (RPE) mainly expresses cfh and the terminal complement components, whereas cfi and cfp transcripts are most abundant in neurons. Aging enhances c1s, cfb, cfp, and cfi expression, while cfh expression decreases. Transient retinal ischemia increases complement expression in microglia, Muller cells, and RPE. In summary, we report a unique complement expression signature for murine retinal cell types suggesting a well-orchestrated regulation of local complement expression in the retinal microenvironment

    The cellular events that dictate the initiation of the complement pathway in ocular degeneration, such as age-related macular degeneration (AMD), is poorly understood. Using gene expression analysis (single cell and bulk), mass spectrometry, and immunohistochemistry, we dissected the role of multiple retinal and choroidal cell types in determining the complement homeostasis. Our scRNA-seq data show that the cellular response to early AMD is more robust in the choroid, particularly in fibroblasts, pericytes and endothelial cells. In late AMD, complement changes were more prominent in the retina especially with the expression of the classical pathway initiators. Notably, we found a spatial preference for these differences. Overall, this study provides insights into the heterogeneity of cellular responses for complement expression and the cooperation of neighboring cells to complete the pathway in healthy and AMD eyes.     West Nile virus vector Culex modestus established in southern England

    Background: The risk posed to the United Kingdom by West Nile virus (WNV) has previously been considered low, due to the absence or scarcity of the main Culex sp. bridge vectors. The mosquito Culex modestus is widespread in southern Europe, where it acts as the principle bridge vector of WNV. This species was not previously thought to be present in the United Kingdom. Findings: Mosquito larval surveys carried out in 2010 identified substantial populations of Cx. modestus at two sites in marshland in southeast England. Host-seeking-adult traps placed at a third site indicate that the relative seasonal abundance of Cx. modestus peaks in early August. DNA barcoding of these specimens from the United Kingdom and material from southern France confirmed the morphological identification. Conclusions: Cx. modestus appears to be established in the North Kent Marshes, possibly as the result of a recent introduction. The addition of this species to the United Kingdom’s mosquito fauna may increase the risk posed to the United Kingdom by WNV

    Tumor-derived GDF-15 blocks LFA-1 dependent T cell recruitment and suppresses responses to anti-PD-1 treatment

    Immune checkpoint blockade therapy is beneficial and even curative for some cancer patients. However, the majority don't respond to immune therapy. Across different tumor types, pre-existing T cell infiltrates predict response to checkpoint-based immunotherapy. Based on in vitro pharmacological studies, mouse models and analyses of human melanoma patients, we show that the cytokine GDF-15 impairs LFA-1/ÎČ2-integrin-mediated adhesion of T cells to activated endothelial cells, which is a pre-requisite of T cell extravasation. In melanoma patients, GDF-15 serum levels strongly correlate with failure of PD-1-based immune checkpoint blockade therapy. Neutralization of GDF-15 improves both T cell trafficking and therapy efficiency in murine tumor models. Thus GDF-15, beside its known role in cancer-related anorexia and cachexia, emerges as a regulator of T cell extravasation into the tumor microenvironment, which provides an even stronger rationale for therapeutic anti-GDF-15 antibody development
