Dobbiamo anche fare un po' di distinzione quando parliamo di doppiaggi in generale. Se ci riferiamo ai film che vengono importati – e qui siamo in una scuola per traduttori, e presumo che avrete tanto lavoro in questo settore – questi devono subire una lavorazione fatta di tantissime fasi, e tutte delicate. Bisogna fare in modo di rispettare le intenzioni del linguaggio, e quello è veramente un campo dove i bravi non sono tanti e a volte possono anche rovinare un film, quando l'opera non viene finita nel rispetto di come è nata

    Gamma-Sarcoglycan Mediated Mechanotransduction in Skeletal Muscle

    Loss of gamma-sarcoglycan (g-SG) is common to both Duchenne and Limb Girdle muscular dystrophies, and is sufficient to induce severe muscle degeneration and signaling defects in response to mechanical load without causing susceptibility to contractile damage. This suggests that disease occurs not by structural deficits, but through aberrant signaling, namely, disruption of normal mechanotransduction signaling through the SG complex. However, the mechanisms of g-SG-mediated mechanical signaling are poorly understood. Here, we identified the muscle-specific protein archvillin as a g-SG and dystrophic interacting protein. Archvillin expression is dependent on g-SG, where levels are significantly unregulated in human LGMD2C patient muscle and at the sarcolemma of murine g-SG-null (gsg-/-) muscle. However, archvillin is delocalized in mdx muscle, indicating dystrophin-dependent sarcolemmal localization. In situ eccentric contraction (ECC) of C57 mouse tibialis anterior (TA) muscles causes ERK1/2 phosphorylation, nuclear activation of P-ERK1/2, and stimulus-dependent archvillin association with P-ERK1/2, whereas TA muscles from gsg-/- and mdx mice exhibit heightened P-ERK1/2 and increased nuclear P-ERK1/2 localization following ECCs, but the archvillin-P-ERK1/2 association is completely ablated. These results position archvillin as a mechanically sensitive component of the dystrophin complex, and demonstrate that signaling defects caused by loss of g-SG occur both at the sarcolemma and in the nucleus. We also extended our previous studies on mechanosensitive, g-SG dependent ERK1/2 phosphorylation to determine whether additional pathways are altered with loss of g-SG. Using a passive stretching protocol to isolate the effects of externally applied tension, we found that in isolated EDL muscles, Akt, S6RP, and p70S6K activation increases with stretch in both C57 and gsg-/- isolated muscles. Treatment of muscles with or without rapamycin during stretch blocks p70S6K activation in stretched C57 muscles, and reduces downstream S6RP phosphorylation, indicating mTOR dependence. However, even though rapamycin treatment decreases p70S6K activation in stretched gsg-/- muscles, S6RP phosphorylation remains elevated. These results suggest that p70S6K is an important component of g-SG-dependent mechanotransduction in skeletal muscle, where loss of g-SG uncouples the response of p70S6K to stretch and implies that g-SG is important for inactivation of this pathway. In sum, this study demonstrates altered load-sensing mechanisms in muscular dystrophies where the sarcoglycans are absent

    Bottom-Up Initiatives for Sustainable Mountain Development in Italy: An Interregional Explorative Survey

    In recent years, citizen-led initiatives have emerged to complement top-down development policies, particularly to pursue sustainability objectives and benefit traditionally left-behind communities and places. Existing research on this phenomenon in Italian mountain areas suggests a primary emphasis on revitalizing the natural environment, preserving local culture and traditions, and delivering social services to address the absence of public facilities. However, there is still a lack of understanding regarding their profiles, key features, social and environmental impacts, and challenges at the national level. As part of a broader research project, this empirical work contributes to the literature on bottom-up sustainability-oriented initiatives in mountain regions by conducting an explorative interregional online survey. We gathered original data on 196 initiatives from the mountain areas of the Italian regions of Basilicata, Lombardia, Marche, Sicilia, and Veneto. The findings indicate that our sample shares common traits, visions, and values, making a positive contribution to the sustainable development of their respective territories. However, these initiatives must contend with internal and external challenges to enhance their resilience and impact. Importantly, the research may support policymakers at various levels in the design of public policies that harness the full potential of bottom-up initiatives, ultimately enhancing the development and sustainability of these lands

    Analysis on the impact of additives on space charge behavior of thermally aged XLPE plaques

    This article investigates the space charge properties of XLPE-based materials characterized by different concentration and types of additives and fillers inside the polymeric compound. Materials were aged under three different temperatures (87 °C, 110 °C and 130 °C) for 24, 18 and 12 months, respectively. Space charge profiles of both unaged and aged materials were obtained through the Pulsed Electro Acoustic (PEA) method. Additives and fillers are proven to significantly impact the space charge behavior of the insulating material both in the unaged and aged states. The impact of antioxidants, together with their kinetics under thermal aging conditions, is analyzed and claims an effective containment of the degradation kinetics, keeping the accumulated space charge to low values

    Aberrant ribonucleotide incorporation and multiple deletions in mitochondrial DNA of the murine MPV17 disease model

    All DNA polymerases misincorporate ribonucleotides despite their preference for deoxyribonucleotides, and analysis of cultured cells indicates that mammalian mitochondrial DNA (mtDNA) tolerates such replication errors. However, it is not clear to what extent misincorporation occurs in tissues, or whether this plays a role in human disease. Here, we show that mtDNA of solid tissues contains many more embedded ribonucleotides than that of cultured cells, consistent with the high ratio of ribonucleotide to deoxynucleotide triphosphates in tissues, and that riboadenosines account for three-quarters of them. The pattern of embedded ribonucleotides changes in a mouse model of Mpv17 deficiency, which displays a marked increase in rGMPs in mtDNA. However, while the mitochondrial dGTP is low in the Mpv17−/− liver, the brain shows no change in the overall dGTP pool, leading us to suggest that Mpv17 determines the local concentration or quality of dGTP. Embedded rGMPs are expected to distort the mtDNA and impede its replication, and elevated rGMP incorporation is associated with early-onset mtDNA depletion in liver and late-onset multiple deletions in brain of Mpv17−/− mice. These findings suggest aberrant ribonucleotide incorporation is a primary mtDNA abnormality that can result in pathology

Una propuesta que toma como premisa la interdisciplinariedad, la apreciación de los residentes, destacando sus usos y costumbres, y la importancia de la memoria como formadora y transformadora de la identidad local, con una reflexión directa sobre el desarrollo social de la población misma.Tendo como contexto o estudo de caso da Estância Hidromineral de Monte Alegre do Sul, este artigo apresenta uma proposta de intervenção urbana que destaca a transformação da paisagem de borda dos corpos d’agua em um lugar sensível, de qualidade ambiental e com um conteúdo museológico que gere uma apreensão sobre questões locais assim como sobre toda a sociedade. Uma proposta que adota como premissa a interdisciplinaridade, a valorização dos moradores, destacando seus usos e costumes, e a importância da memória como formadora e transformadora da identidade local, com reflexo direto no desenvolvimento social da própria população.Taking as context the case study of the Estância Hidromineral of Monte Alegre do Sul, this paper presents a proposal of urban intervention that highlights the transformation of the border landscape of water bodies in a sensitive place, of environmental quality and of museological content that engenders apprehension about local issues and about the society as well. A proposal that adopts as a premise the interdisciplinarity, the valorization of the residents, highlighting their usages and customs, and the importance of memory as a transformation of local identity, with a direct development of the population itself
