    Sensitivity to CPT-11 of xenografted human colorectal cancers as a function of microsatellite instability and p53 status

    Biological parameters influencing the response of human colorectal cancers (CRCs) to CPT-11, a topoisomerase 1 (top1) inhibitor, were investigated using a panel of nine CRCs xenografted into nude mice. CRC xenografts differed in their p53 status (wt or mut) and in their microsatellite instability phenotype (MSI+when altered). Five CRC xenografts were established from clinical samples. All five had a functional p53, two were MSI+and three were MSI–. Tumour-bearing nude mice were treated intraperitonealy (i.p.) with CPT-11. At 10 mg kg–1of CPT-11, four injections at 4-day intervals, four of the five xenografts responded to CPT-11 (growth delay of up to 10 days); the non-responder tumour was MSI−. At 40 mg kg−1of CPT-11, six injections at 4-day intervals, the five CRCs displayed variable but marked responses with complete regressions. In order to assess the role of p53 status in CPT-11 response, four CRC lines were used. HT29 cell line was MSI−/ Ala273-mutp53, its subclone HT29A3 being transfected by wtp53. LoVo cell line was MSI+/ wtp53, its subclone X17LoVo dominantly expressed Ala273-mutp53 after transfection. LoVo tumours (MSI+/ mutp53) were more sensitive than X17LoVo (MSI+/ mutp53. HT 29 tumours (MSI−Imutp53), were refractory to CPT-11 while HT29A3 tumours (MSI−/ wtp53) were sensitive, showing that wtp53 improves the drug-response in these MSI−tumours. Levels of mRNA expression of top1, fasR, TP53 and mdr1 were semi-quantified by reverse transcription polymerase chain reaction. None of these parameters correlated with CPT-11 response. Taken together, these observations indicate that MSI and p53 alterations could be associated with different CPT-11 sensitivities; MSI phenotype moderately influences the CPT-11 sensitivity, MSI+being more sensitive than MSI−CRC freshly obtained from patients, mutp53 status being associated with a poor response to CPT-11. © 2000 Cancer Research Campaig

    Aproximación a las comunidades de carófitos que existieron en la Albufera de Valencia a partir del estudio de las oósporas del sedimento

    Charophyte oospores and their features (size, shape, colour, outer wall ornamentation, etc.) are able to persist in seed banks of aquatic ecosystems. Thus, these characteristics can be used for taxonomic species identification, thereby providing knowledge about the specific composition of the charophyte community in a habitat that currently lacks, or has greatly altered, submerged vegetation. Several years ago, l’Albufera de València lost the charophyte meadows that covered the lake bottom in the past, which had never been studied in detail. Several sediment cores were extracted from the lagoon at three sites and the oospores and gyrogonites were isolated and characterized. The oospore/gyrogonite densities in the sediment layers were estimated. Ten different taxa have been identified: six belonging to genus Chara [Ch. vulgaris, Ch. Hispida var. major, Ch. fragilis (Ch. globularis), Ch. Hispida var. baltica, Ch. aspera, Ch. tomentosa]; one Nitella species (N. hyalina), two Tolypella species (T. glomerata and T. hispanica) and one Lamprothamnium species (L. papulosum). The latter, which indicates the period when water salinity was high in the lagoon, was found in the deepest sediment layers. An increase in the oospore abundance and diversity was observed in line with sediment depth. This indicates, on the one hand, that the lagoon went through different ecological stages (with important changes in salinity, mainly) and, on the other hand, the eutrophication process that the lagoon has been undergoing for at least the last 45 years, has progressively affected the abundance and diversity of the charophyte community.Las oósporas de carófitos y sus características (tamaño, forma, color, ornamentación de la pared, etc.) pueden persistir en los bancos de semillas del sedimento de ecosistemas acuáticos, y pueden ser utilizadas para identificar los diferentes táxones y obtener de este modo una aproximación a la composición florística de las comunidades de carófitos que colonizaron ambientes acuáticos en los que actualmente no existe vegetación acuática o se encuentra muy alterada. La Albufera de Valencia perdió hace muchos años las praderas de carófitos que cubrían sus fondos sin tener un estudio detallado de las mismas. Se han extraído diversos testigos de sedimento en tres puntos de esta laguna, se han caracterizado las oósporas y girogonitos encontrados y se ha estimado la densidad de oósporas-girogonitos en diferentes profundidades del sedimento. En total se han identificado 10 táxones, que corresponden: 6 al género Chara [Ch. vulgaris, Ch. hispida var. major, Ch. fragilis (Ch. globularis), Ch. hispida var. baltica, Ch. aspera y Ch. tomentosa]; una especie de Nitella (N. hyalina); 2 de Tolypella (T. glomerata y T. hispanica), y una de Lamprothamnium (L. papulosum). Esta última, indicadora de la época en la que la laguna poseía una mayor salinidad, se encontró en las capas más profundas del sedimento. Se ha observado un aumento del número y de la diversidad de oósporas en capas más profundas del sedimento, lo que indica, por un lado, que la laguna atravesó distintas fases ecológicas (con importantes cambios de salinidad principalmente) y, por otro, que el proceso de eutrofización que padece la laguna durante al menos los últimos 45 años afectó progresivamente a la abundancia y diversidad de carófitos

    Qui meurt après une néphrectomie pour cancer ? Étude des facteurs de risque de décès, des causes de décès et des réunions de morbi-mortalité (étude UroCCR-33)

    BACKGROUND AND METHODS: Nephrectomy is the treatment for renal cell cancer from T1-4 tumors but remains at risk. To determine the thirty-day mortality rate after nephrectomy for cancer and to identify causes and risk factors of death in order to find clinical applications. From 2014 to 2017, we performed a retrospective multicentric analysis of prospectively collected data study involving the French network for research on kidney cancer (UroCCR). All patients who died after nephrectomy for cancer during the first thirty days were identified. Patients\u27 characteristics, causes of death and morbidity and mortality reviews reports were analyzed for each death. RESULTS AND LIMITATIONS: In total, 2578 patients underwent nephrectomy and 35 deaths occurred. The thirty-day mortality rate was 1.4%. In univariate analysis, symptoms at diagnosis (P=0.006, OR=2.56 IC (1.3-5.03)), c stage superior to cT1 (P<0.0001, OR=6.13 IC (2.8-13.2)), cT stage superior to cT2 (P<0.0001, OR=8.8 IC (4.39-17.8)), nodal invasion (P<0.0001, OR=4.6 IC (1.9-10.7)), distant metastasis (P=0.001, OR=4.01 IC (1.7-8.9)), open surgery (P<0.0001, OR=0.272 IC (0.13-0.54)) and radical nephrectomy (P=0.007, OR=2.737 IC (1.3-5.7)) were risk factors of thirty-day mortality. In a multivariable model, only cT stage superior to T2 (P=0.015, OR=3.55 IC (1.27-10.01)) was a risk factor of thirty-day mortality. The main cause of postoperative death was pulmonary (n=15; 43%). The second cause was postoperative digestive sepsis for 7 patients (20%). Only 2 morbidity and mortality reviews had been done for the 35 deaths. Limitations are related to the thirty-day mortality criteria and descriptive study design. CONCLUSIONS: Symptomatic patients, stage cTNM and type and techniques of surgery are determinants of thirty-day mortality after nephrectomy for cancer. The first cause of postoperative death is pulmonary. Morbidity and mortality reviews should be considered to better understand causes of death and to reduce early mortality after nephrectomy for cancer. LEVEL OF EVIDENCE: 4

    High prevalence of vertebral fractures in women with breast cancer starting aromatase inhibitor therapy

    Background: The purpose of this study was to describe bone status in a large cohort of postmenopausal women with nonmetastatic breast cancer, at the initiation of aromatase inhibitor therapy.Patients and methods: A prospective, transversal and clinical study was conducted. Each woman had an extensive medical history, a biological evaluation, a bone mineral density (BMD) measurement and spinal X-rays. Results: Four hundred and ninety-seven women aged 63.8 ± 9.6 years were included in this study. Eighty-five percent of these women had a 25-OH vitamin D concentration <75 nmol/l. One hundred and fifty-six women (31.4%) had a T-score < −2 at one of the three site measurements. Ninety-five women (19.1%) had a history of nonvertebral fracture with a total of 120 fractures. Spine X-rays evaluation revealed that 20% of the women had at least one vertebral fracture. The presence of vertebral fracture was associated with nonvertebral fracture history [odds ratio (OR) 1.6, 95% confidence interval (CI) 1.1–2.4] and with spine BMD (OR 1.4, 95% CI 1.1–1.7). The prevalence of vertebral fracture reached 62.9% in women with age above 70 years and femoral T-score < −2.5. Conclusion: Before starting aromatase inhibitor therapy for breast cancer, a large proportion of women had a vitamin D insufficiency and vertebral fractures

    Quasiprojectile breakup and isospin equilibration at Fermi energies: an indication of longer projectile-target contact times?

    An investigation of the quasiprojectile breakup channel in semiperipheral and peripheral collisions of 58,64^{58,64}Ni+58,64^{58,64}Ni at 32 and 52 MeV/nucleon is presented. Data have been acquired in the first experimental campaign of the INDRA-FAZIA apparatus in GANIL. The effect of isospin diffusion between projectile and target in the two asymmetric reactions has been highlighted by means of the isospin transport ratio technique, exploiting the neutron-to-proton ratio of the quasiprojectile reconstructed from the two breakup fragments. We found evidence that, for the same reaction centrality, a higher degree of relaxation of the initial isospin imbalance is achieved in the breakup channel with respect to the more populated binary output, possibly indicating the indirect selection of specific dynamical features. We have proposed an interpretation based on different average projectile-target contact times related to the two exit channels under investigation, with a longer interaction for the breakup channel. The time information has been extracted from AMD simulations of the studied systems coupled to GEMINI++: the model calculations support the hypothesis hereby presented
