    Cooperative Research As a Strategy for University Teacher Training. A Case Study of Lesson and Learning Study

    This paper presents the possibilities offered by Lesson and Learning Studies for training and for improving and generating knowledge by reconstructing the practical knowledge of teachers. Firstly, we provide a summary of the concept of practical knowledge and the tradition of teachers researching their own practice. This is followed by some principles of Lesson and Learning Studies, with examples of their possibilities for university teacher training taken from a case study of our own practice during a university master's degree

    Lesson study as a tool for rebuilding teachers' practical thinking: a bridge between espoused theories and theories-in-use

    This Lesson Study forms part of the research work carried out by the Spanish team, in order to analyse the potential of this professional teacher development methodology for the reconstruction of teachers' practical thinking. It is a qualitative research process which involves the monitoring of seven infant school teachers during two school cycles. The teachers developed a Lesson Study over this period, with a practical thinking case study being carried out both before and after. This paper focuses on the Lesson Study case study. The Lesson Study was developed independently by the group of teachers, who were advised throughout by the undersigned researchers. At the same time, the researchers carried out a case study focusing on the Lesson Study being developed by the teachers. Findings/Conclusions: - From the start of the Lesson Study, we can find evidence of changes in the practical thinking of the teachers involved, which led to immediate changes in their classroom practice. - The development of the Lesson Study brought about significant reflection with regards to the Infant Education curriculum, in some cases resulting in substantial modification to the way it is implemented in their different classrooms. - The teachers pointed out the value of cooperative work on several different occasions. - They also emphasised the high level of learning they achieved by observing how their colleagues developed the lesson designed by the group using different strategies and data collection instruments, such as documentation inspired by the Reggio Emilia approach, when reflecting on their own practice. On the whole, we can state that the cooperative design work of this lesson showed the contrast between the espoused theories of the participants and their theories-in-use, conditioned fundamentally by their work contexts and previous experience. Lesson Study is a way to bring these two poles closer together, enriching teachers with strategies to help detect contradictions in their practice and to resolve them and feel more comfortable with what they do.Universidad de Málaga.     Lesson Study and the development of teaching competences: from practical knowledge to practical thinking

    Este artículo pretende mostrar la prometedora relación entre los procesos generados por las Lesson Studies y el desarrollo del pensamiento práctico en la formación docente, tanto inicial como permanente, a través de dos investigaciones . Para ello proponemos ampliar el foco de interés de las Lesson Study no sólo a la mejora de la “lección” o de la “enseñanza” (Lesson), en definitiva de la práctica, sino también a la reconstrucción y mejora del conocimiento práctico de los docentes. Creemos que esta propuesta metodológica reúne una serie de estrategias básicas que con algunos matices o variaciones desarrollados en el contexto español , permite también reconstruir el pensamiento práctico de los docentes tanto en el marco de la formación inicial como permanente, precisamente cuando trabajan cooperativamente sobre una lección en un proyecto de enseñanza que pretende provocar el aprendizaje de los estudiantes.Universidad de Málaga.     High Prevalence of Systemic Autoimmune Diseases in Patients with Menière's Disease

    BACKGROUND: Autoimmunity appears to be associated with the pathophysiology of Meniere's disease (MD), an inner ear disorder characterized by episodes of vertigo associated with hearing loss and tinnitus. However, the prevalence of autoimmune diseases (AD) in patients with MD has not been studied in individuals with uni or bilateral sensorineural hearing loss (SNHL). METHODS AND FINDINGS: We estimated the prevalence of AD in 690 outpatients with MD with uni or bilateral SNHL from otoneurology clinics at six tertiary referral hospitals by using clinica criteria and an immune panel (lymphocyte populations, antinuclear antibodies, C3, C4 and proinflammatory cytokines TNFα, INFγ). The observed prevalence of rheumatoid arthritis (RA), systemic lupus erythematosus (SLE) and ankylosing spondylitis (AS) was higher than expected for the general population (1.39 for RA, 0.87 for SLE and 0.70 for AS, respectively). Systemic AD were more frequently observed in patients with MD and diagnostic criteria for migraine than cases with MD and tension-type headache (p = 0.007). There were clinical differences between patients with uni or bilateral SNHL, but no differences were found in the immune profile. Multiple linear regression showed that changes in lymphocytes subpopulations were associated with hearing loss and persistence of vertigo, suggesting a role for the immune response in MD. CONCLUSIONS: Despite some limitations, MD displays an elevated prevalence of systemic AD such as RA, SLE and AS. This finding, which suggests an autoimmune background in a subset of patients with MD, has important implications for the treatment of MD

    Low threshold acquisition controller for Skipper charge-coupled devices

    The development of the Skipper-charge-coupled devices (Skipper-CCDs) has been a major technological breakthrough for sensing very weak ionizing particles. The sensor allows to reach the ultimate sensitivity of silicon material as a charge signal sensor by unambiguous determination of the charge signal collected by each cell or pixel, even for single electron-hole pair ionization. Extensive use of the technology was limited by the lack of specific equipment to operate the sensor at the ultimate performance. A simple, single-board Skipper-CCD controller designed by the authors is presented and aimed for the operation of the detector in high sensitivity scientific applications. Our article describes the main components and functionality of the so-called low threshold acquisition controller together with experimental results when connected to a Skipper-CCD sensor.     Burden of rare variants in synaptic genes in patients with severe tinnitus: An exome based extreme phenotype study

    Background: tinnitus is a heterogeneous condition associated with audiological and/or mental disorders. Chronic, severe tinnitus is reported in 1% of the population and it shows a relevant heritability, according to twins, adoptees and familial aggregation studies. The genetic contribution to severe tinnitus is unknown since large genomic studies include individuals with self-reported tinnitus and large heterogeneity in the phenotype. The aim of this study was to identify genes for severe tinnitus in patients with extreme phenotype. Methods: for this extreme phenotype study, we used three different cohorts with European ancestry (Spanish with Meniere disease (MD), Swedes tinnitus and European generalized epilepsy). In addition, four independent control datasets were also used for comparisons. Whole-exome sequencing was performed for the MD and epilepsy cohorts and whole-genome sequencing was carried out in Swedes with tinnitus. Findings: we found an enrichment of rare missense variants in 24 synaptic genes in a Spanish cohort, the most significant being PRUNE2, AKAP9, SORBS1, ITGAX, ANK2, KIF20B and TSC2 (p < 2E 04), when they were compared with reference datasets. This burden was replicated for ANK2 gene in a Swedish cohort with 97 tinnitus individuals, and in a subset of 34 Swedish patients with severe tinnitus for ANK2, AKAP9 and TSC2 genes (p < 2E 02). However, these associations were not significant in a third cohort of 701 generalized epilepsy individuals without tinnitus. Gene ontology (GO) and gene-set enrichment analyses revealed several pathways and biological processes involved in severe tinnitus, including membrane trafficking and cytoskeletal protein binding in neurons. Interpretation: a burden of rare variants in ANK2, AKAP9 and TSC2 is associated with severe tinnitus. ANK2, encodes a cytoskeleton scaffolding protein that coordinates the assembly of several proteins, drives axonal branching and influences connectivity in neurons

    4th booster-dose SARS-CoV-2 heterologous and homologous vaccination in rheumatological patients

    Objectiveto evaluate the immune response to the SARS-CoV-2 vaccines in adults with immune-mediated rheumatic diseases (IMRDs) in comparison to healthy individuals, observed 1-20 weeks following the fourth vaccine dose. Additionally, to evaluate the impact of immunosuppressive therapies, vaccination schedules, the time interval between vaccination and sample collection on the vaccine’s immune response.MethodsWe designed a longitudinal observational study conducted at the rheumatology department of Hospital de Copiapó. Neutralizing antibodies (Nabs) titers against the Wuhan and Omicron variant were analyzed between 1-20 weeks after administration of the fourth dose of the SARS-CoV-2 vaccine to 341 participants (218 IMRD patients and 123 healthy controls). 218 IMRD patients with rheumatoid arthritis (RA), psoriatic arthritis (PsA), ankylosing spondylitis (AS), systemic lupus erythematosus (SLE), systemic vasculitis (VS) and systemic scleroderma (SS) were analyzed.ResultsPerforming a comparison between the variants, Wuhan vs Omicron, we noticed that there were significant differences (p&lt;0.05) in the level of the ID50, both for healthy controls and for patients with IMRDs. The humoral response of patients with IMRDs is significantly lower compared to healthy controls for the Omicron variant of SARS-CoV-2 (p = 0.0015). The humoral response of patients with IMRDs decreases significantly when the time interval between vaccination and sample collection is greater than 35 days. This difference was observed in the response, both for the Wuhan variant and for the Omicron variant.ConclusionThe IMRDs patients, the humoral response variation in the SARS-CoV-2 vaccine depends on doses and type of vaccine administered, the humoral response times and the treatment that these patients are receiving

    El presente estudio se realizó para determinar la estabilidad de una pomada con actividad antiinflamatoria de uso tópico a partir del extracto etanólico de Muehlenbeckia volcánica (Benth) Endl. . Se hizo una selección cuidadosa de las partes aéreas de Muehlenbeckia volcánica (Benth.) Endl. La droga fue secada bajo sombra en condiciones especiales evitando la humedad, el polvo y el sol directo. Luego fue  estabilizada  en la estufa a 35°C. El material seco fue  fragmentado en un molino manual obteniéndose un tamaño de  partículas de 5 mm para asegurar que el proceso de extracción cumpla  con los requisitos de calidad. A partir del material vegetal (50g) seco y fragmentado se procedió a realizar el proceso de  la extracción por el método de la percolación. Las pruebas de estabilidad se realizaron con los protocolos de Estabilidad &lt;1047&gt; USP 41. El extracto fluido de las partes aéreas de la especie Muehlenbeckia volcánica (Benth), presentaron triterpenos y o esteroides, taninos, flavonoides, saponinas y catequinas. La pomada o/w, fue formulada con  una concentración del 10% de extracto fluido. Las características organolépticas de la pomada obtenida fueron, aspecto homogéneo, con brillo sin grumos, olor inodoro, la respuesta táctil  fue fluida y muy agradable, no hubo presencia de exudado y la extensibilidad de la pomada obtenida a partir del extracto fluido se incrementó levemente con el aumento de la temperatura. Se concluye que la pomada formulada a una concentración de 10% a partir del extracto fluido de la especie Muehlenbeckia volcánica (Benth) Endl, presentó  características organolépticas, controles fisicoquímicos  y estabilidad acelerada dentro de los valores permitidos

    Evidence for classification of c.1852_1853AA>GC in MLH1 as a neutral variant for Lynch syndrome

    Background: Lynch syndrome (LS) is an autosomal dominant inherited cancer syndrome characterized by early onset cancers of the colorectum, endometrium and other tumours. A significant proportion of DNA variants in LS patients are unclassified. Reports on the pathogenicity of the c.1852_1853AA>GC (p.Lys618Ala) variant of the MLH1 gene are conflicting. In this study, we provide new evidence indicating that this variant has no significant implications for LS. Methods: The following approach was used to assess the clinical significance of the p.Lys618Ala variant: frequency in a control population, case-control comparison, co-occurrence of the p.Lys618Ala variant with a pathogenic mutation, co-segregation with the disease and microsatellite instability in tumours from carriers of the variant. We genotyped p.Lys618Ala in 1034 individuals (373 sporadic colorectal cancer [CRC] patients, 250 index subjects from families suspected of having LS [revised Bethesda guidelines] and 411 controls). Three well-characterized LS families that fulfilled the Amsterdam II Criteria and consisted of members with the p.Lys618Ala variant were included to assess co-occurrence and co-segregation. A subset of colorectal tumour DNA samples from 17 patients carrying the p.Lys618Ala variant was screened for microsatellite instability using five mononucleotide markers. Results: Twenty-seven individuals were heterozygous for the p.Lys618Ala variant; nine had sporadic CRC (2.41%), seven were suspected of having hereditary CRC (2.8%) and 11 were controls (2.68%). There were no significant associations in the case-control and case-case studies. The p.Lys618Ala variant was co-existent with pathogenic mutations in two unrelated LS families. In one family, the allele distribution of the pathogenic and unclassified variant was in trans, in the other family the pathogenic variant was detected in the MSH6 gene and only the deleterious variant co-segregated with the disease in both families. Only two positive cases of microsatellite instability (2/17, 11.8%) were detected in tumours from p.Lys618Ala carriers, indicating that this variant does not play a role in functional inactivation of MLH1 in CRC patients. Conclusions: The p.Lys618Ala variant should be considered a neutral variant for LS.
