    Mites as parasites infesting fish have been described in a few case reports involving Histiostoma anguillarum, H. papillata, and Schwiebea estradai. We describe the unexpected occurrence of oribatid mites of the genus Trhypochthoniellus on farmed tilapia Oreochromis niloticus. The fish had mites on the skin, fins, and gills, as well as in the mouth. The morphological characteristics of the mites, observed by optical and scanning electron microscopy, were consistent with those described for T. longisetus longisetus.     Evaluation of an integrated system for classification, assessment and comparison of services for long-term care in Europe: the eDESDE-LTC study

    The harmonization of European health systems brings with it a need for tools to allow the standardized collection of information about medical care. A common coding system and standards for the description of services are needed to allow local data to be incorporated into evidence-informed policy, and to permit equity and mobility to be assessed. The aim of this project has been to design such a classification and a related tool for the coding of services for Long Term Care (DESDE-LTC), based on the European Service Mapping Schedule (ESMS). Methods. The development of DESDE-LTC followed an iterative process using nominal groups in 6 European countries. 54 researchers and stakeholders in health and social services contributed to this process. In order to classify services, we use the minimal organization unit or "Basic Stable Input of Care" (BSIC), coded by its principal function or "Main Type of Care" (MTC). The evaluation of the tool included an analysis of feasibility, consistency, ontology, inter-rater reliability, Boolean Factor Analysis, and a preliminary impact analysis (screening, scoping and appraisal). Results: DESDE-LTC includes an alpha-numerical coding system, a glossary and an assessment instrument for mapping and counting LTC. It shows high feasibility, consistency, inter-rater reliability and face, content and construct validity. DESDE-LTC is ontologically consistent. It is regarded by experts as useful and relevant for evidence-informed decision making. Conclusion: DESDE-LTC contributes to establishing a common terminology, taxonomy and coding of LTC services in a European context, and a standard procedure for data collection and international comparison

    Influência do sombreamento artificial e da adubação química na produção de mudas de Adenanthera pavonina L.

    The aim of this study was to evaluate the effects of artificial shading and chemical fertilization on the seedling growth of Adenanthera pavonina L. The growth  was analysed in plants growing in a green house under 30 and 60% of artificial shading, in "cerrado" soil with or without addition of NPK (4,14,18). The measurements were made monthly, from 30 until 180 days after emerging (D.A.E.). The analysis of the biometrical and physiological parameters indicated that from 30 until 120 D.A.E., the edaphic and light conditions did not promote significant differences in plant growth. Therefore, from 150 D.A.E.,  the best growing conditions were the use of "cerrado" soil with additions of NPK and under high light intensity.O objetivo deste trabalho foi avaliar a influência do sombreamento artificial e da adubação química na produção de mudas de Adenanthera pavonina L. O crescimento dessa espécie foi avaliado em mudas cultivadas em casa de vegetação sob 30 e 60% de sombreamento artificial, em solo de cerrado com e sem adição de NPK (4-14-8) com avaliações mensais a partir de 30 até 180 dias após a emergência (D.A.E.). A análise dos parâmetros biométricos e fisiológicos revelou que até 120 D.A.E. as condições edáficas e de luminosidade, sob as quais foram cultivadas as plantas de Adenanthera pavonina L., não interferiram significativamente no crescimento dessa espécie. Porém, partindo de 150 D.A.E., as melhores condições de cultivo foram o uso de solo de cerrado com adição de NPK e manutenção sob alta intensidade luminosa

    Current Regulatory Requirements for Biosimilars in Six Member Countries of BRICS-TM: Challenges and Opportunities

This is an open-access article distributed under the terms of the Creative Commons Attribution License (CC BY). https://creativecommons.org/licenses/by/4.0/Background: The aim of the study was to identify, interpret, and compare the current perspectives of regulatory agencies in six member countries of BRICS-TM (Brazil, Russia, India, China, South Africa, Turkey, and Mexico) on the different criteria used for biosimilar development and marketing authorisation process. Methods: A semi-quantitative questionnaire was developed covering the organisation of agency, biosimilar development criteria and marketing authorisation process and sent to seven regulatory agencies covering the BRICS-TM countries. All data was kept anonymous and confidential. Data processing and analysis was carried out; descriptive statistics were used for quantitative data and content analysis was employed to generate themes for qualitative data. Results: Out of the seven regulatory agencies included in the study, six representatives provided the responses. The perspectives of these six regulatory agencies varied on a number of aspects relating to the review criteria for biosimilar development and licencing process. The most prevalent model for data assessment is the “full review” of a marketing authorisation application. There is lack of a standard approach across the agencies on sourcing of the reference biological product, in vivo toxicity studies and confirmatory clinical studies. Most agencies restrict interaction with biosimilar developers and any scientific advice is non-binding. The marketing authorisation approval depends on scientific assessment of the dossier, sample analysis and GMP certification. The agencies do not issue any public assessment report specifying the summary basis of biosimilar approval. Conclusion: Regulatory agencies across the six emerging economies are steadily improving the regulatory mechanism in the area of biosimilars. However, there remains scope for increasing the effectiveness and efficiency of the processes by encouraging open and transparent interaction with developers, adopting a flexible approach toward accepting advanced analytical data in lieu of clinical studies and enhancing regulatory reliance amongst agencies. This will help to simplify the new biosimilar development programmes and make them more cost-effective.Peer reviewe

    Resonant elastic X-ray scattering of antiferromagnetic superstructures in EuPtSi3_{3}

    We report resonant elastic X-ray scattering (REXS) of long-range magnetic order in EuPtSi3_{\text{3}}, combining different scattering geometries with full linear polarization analysis to unambiguously identify magnetic scattering contributions. At low temperatures, EuPtSi3_{\text{3}} stabilizes type A antiferromagnetism featuring various long-wavelength modulations. For magnetic fields applied in the hard magnetic basal plane, well-defined regimes of cycloidal, conical, and fan-like superstructures may be distinguished that encompass a pocket of commensurate type A order without superstructure. For magnetic field applied along the easy axis, the phase diagram comprises the cycloidal and conical superstructures only. Highlighting the power of polarized REXS, our results reveal a combination of magnetic phases that suggest a highly unusual competition between antiferromagnetic exchange interactions with Dzyaloshinsky--Moriya spin--orbit coupling of similar strength

    Pranlukast Antagonizes CD49f and Reduces Sternness in Triple-Negative Breast Cancer Cells

    Introduction: Cancer stem cells (CSCs) drive the initiation, maintenance, and therapy response of breast tumors. CD49f is expressed in breast CSCs and functions in the maintenance of stemness. Thus, blockade of CD49f is a potential therapeutic approach for targeting breast CSCs. In the present study, we aimed to repurpose drugs as CD49f antagonists. Materials and Methods: We performed consensus molecular docking using a subdomain of CD49f that is critical for heterodimerization and a collection of pharmochemicals clini-cally tested. Molecular dynamics simulations were employed to further characterize drug-target binding. Using MDA-MB-231 cells, we evaluated the effects of potential CD49f antagonists on 1) cell adhesion to laminin; 2) mammosphere formation; and 3) cell viability. We analyzed the effects of the drug with better CSC-selectivity on the activation of CD49f-downstream signaling by Western blot (WB) and co-immunoprecipitation. Expressions of the stem cell markers CD44 and SOX2 were analyzed by flow cytometry and WB, respectively. Transactivation of SOX2 promoter was evaluated by luciferase reporter assays. Changes in the number of CSCs were assessed by limiting-dilution xenotransplantation. Results: Pranlukast, a drug used to treat asthma, bound to CD49f in silico and inhibited the adhesion of CD49f+ MDA-MB-231 cells to laminin, indicating that it antagonizes CD49f-containing integrins. Molecular dynamics analysis showed that pranlukast binding induces con-formational changes in CD49f that affect its interaction with β1-integrin subunit and constrained the conformational dynamics of the heterodimer. Pranlukast decreased the clonogenicity of breast cancer cells on mammosphere formation assay but had no impact on the viability of bulk tumor cells. Brief exposure of MDA-MB-231 cells to pranlukast altered CD49f-dependent signaling, reducing focal adhesion kinase (FAK) and phosphatidylinositol 3-kinase (PI3K) activation. Further, pranlukast-treated cells showed decreased CD44 and SOX2 expression, SOX2 promoter transacti-vation, and in vivo tumorigenicity, supporting that this drug reduces the frequency of CSC. Conclusion: Our results support the function of pranlukast as a CD49f antagonist that reduces the CSC population in triple-negative breast cancer cells. The pharmacokinetics and toxicology of this drug have already been established, rendering a potential adjuvant therapy for breast cancer patients

    Satellite and Ground Remote Sensing Techniques to Trace the Hidden Growth of a Lava Flow Field: The 2014-15 Effusive Eruption at Fogo Volcano (Cape Verde)

    Fogo volcano erupted in 2014–2015 producing an extensive lava flow field in the summit caldera that destroyed two villages, Portela and Bangaeira. The eruption started with powerful explosive activity, lava fountains, and a substantial ash column accompanying the opening of an eruptive fissure. Lava flows spreading from the base of the eruptive fissure produced three arterial lava flows. By a week after the start of the eruption, a master lava tube had already developed within the eruptive fissure and along the arterial flow. In this paper, we analyze the emplacement processes based on observations carried out directly on the lava flow field, remote sensing measurements carried out with a thermal camera, SO2 fluxes, and satellite images, to unravel the key factors leading to the development of lava tubes. These were responsible for the rapid expansion of lava for the ~7.9 km length of the flow field, as well as the destruction of the Portela and Bangaeira villages. The key factors leading to the development of tubes were the low topography and the steady magma supply rate along the arterial lava flow. Comparing time-averaged discharge rates (TADR) obtained from satellite and Supply Rate (SR) derived from SO2 flux data, we estimate the amount and timing of the lava flow field endogenous growth, with the aim of developing a tool that could be used for hazard assessment and risk mitigation at this and other volcanoes.This research received no external funding.Published11156V.
