    Spatiotemporal distribution of the magnetotactic multicellular prokaryote Candidatus Magnetoglobus multicellularis in a Brazilian hypersaline lagoon and in microcosms

    Candidatus Magnetoglobus multicellularis is an unusual morphotype of magnetotactic prokaryotes. These microorganisms are composed of a spherical assemblage of gram-negative prokaryotic cells capable of swimming as a unitaligned along a magnetic field. While they occur in many aquatic habitats around the world, high numbers of Ca. M. multicellularishave been detected in Araruama Lagoon, a large hypersaline lagoon near the city of Rio de Janeiro, in Brazil. Here,we report on the spatiotemporal distribution of one such population in sediments of Araruama Lagoon, including its annualdistribution and its abundance compared with the total bacterial community. In microcosm experiments, Ca. M. multicellulariswas unable to survive for more than 45 days: the population density gradually decreased coinciding with a shift to theupper layers of the sediment. Nonetheless, Ca. M. multicellularis was detected throughout the year in all sites studied. Changes in the population density seemed to be related to the input of organic matter as well as to salinity. The populationdensity of Ca. M. multicellularis did not correlate with the total bacterial counts; instead, changes in the microbial communitystructure altered their counts in the environment. [Int Microbiol 2012; 15(3):141-149

    The usefulness of an Online Simplified Screening Questionnaire (SSQ) in identifying work-related cancers

    To obtain a history of occupational exposure in the workplace, the questionnaire is one of the main sources of information. The aim of this study was to develop an online questionnaire using the REDCap data management platform based on the Work-Related Cancer Surveillance Guidelines, reported by the Brazilian National Cancer Institute. Several issues were taken into consideration for its routine application. It should be simple, easy, capable of being applied in a short time and used in the clinical setting of collecting information on the occupational history of the cancer patient. Consequently, this could enable the compulsory notification of work-related cancer. The questionnaire was developed based on questions about the use of and exposure to carcinogenic factors at work and due to smoking. An entirely electronic version of the cancer patient interview was performed using tablets. The online questionnaire was applied at the Barretos Cancer Hospital, Barretos, to newly diagnosed patients from July 2016 to 2018. A total of 1063 patients were included, and 550 indicated positively when asked “Do you work, or have you worked with this substance and/or in this function?/job?” Of these potentially notified patients, 38 subsequently had compulsorily reported work-related cancer. Another important result of this study was the creation and development of a website. In conclusion, we developed an online tool that could facilitate hospital routines, contributing to generating data for the compulsory notification of work-related cancer and triggering investigations and surveillance actions in Brazil.This research was funded by Barretos Cancer Hospital and the Public Ministry of Labor Campinas (Research, Prevention and Education of Occupational Cancer).     Three-Dimensional and Biomimetic Technology in Cardiac Injury After Myocardial Infarction: Effect of Acellular Devices on Ventricular Function and Cardiac Remodelling

    Dilated cardiomyopathy (DMC) of ischemic or non-ischemic aetiology remains a lethal condition nowadays. Despite early percutaneous or medical revascularization after an acute myocardial infarct (AMI), many patients still develop DMC and severe heart failure due to cardiac remodelling. Possibility of regenerating myocardium already damaged or at least inducing a more positive cardiac remodelling with use of biodegradable scaffolds has been attempted in many experimental studies, which can be cellular or acellular. In the cellular scaffolds, the cells are incorporated in the structure prior to implantation of the same into the injured tissue. Acellular scaffolds, in turn, are composites that use one or more biomaterials present in the extracellular matrix (ECM), such as proteoglycans non-proteoglycan polysaccharide, proteins and glycoproteins to stimulate the chemotaxis of cellular/molecular complexes as growth factors to initiate specific regeneration. For the development of scaffold, the choice of biomaterials to be used must meet specific biological, chemical and architectural requirements like ECM of the tissue of interest. In acute myocardial infarction, treating the root of the problem by repairing injured tissue is more beneficial to the patient. Inducing more constructive forms of endogenous repair. Thus, patches of acellular scaffolds capable of mimicking the epicardium and ECM should be able to attenuate both cardiac remodelling and adverse cardiac dysfunction

    Efeito da época de preparo do camalhão no desenvolvimento de plantas de soja em terras baixas/ Effect of the time of preparing the camalhão on the development of soybean plants

    Objetivou-se com o presente estudo avaliar o estabelecimento e o desempenho de crescimento de plantas de soja, quando cultivadas em sistema de sulco-camalhão, preparado antecipadamente ou na hora da semeadura, em terras baixas de clima temperado. O experimento foi conduzido a campo, na área experimental da Embrapa Clima Temperado, Estação Experimental Terras Baixas, no município do Capão do Leão-RS, em delineamento experimental em blocos casualizados, com parcelas dispostas em faixas, com seis repetições. Os tratamentos constaram da época de preparo do camalhão. Os camalhões antecipados (tratamento 1 - TCA), foram construídos em abril de 2018, com camalhoeira, sendo imediatamente semeado a lanço o azevém cv. BRS Ponteio, na densidade de 15 kg ha?1 de sementes. Previamente a semeadura da soja, a área foi dessecada com 1440 ge.a. ha?1 de glyphosate, e a semeadura foi realizada com semeadeira Vence Tudo. Os camalhões confeccionados concomitantemente ao plantio (tratamento 2 - TCFH), foram feitos pela própria semeadeira, que possuía pé de pato específicos para tal operação. Foram avaliadas a área foliar, diâmetro do caule, altura de planta e conteúdo de água das plantas de soja em função dos dias após emergência (DAE). As avaliações foram efetuadas quinzenalmente da emergência à maturação dos grãos. A soja semeada no camalhão feito concomitante à semeadura apresentou melhor desenvolvimento vegetativo, comparativamente ao plantio sobre sulco-camalhão antecipado, principalmente aumentando a área foliar e diâmetro de caule. Supõe-se que condições diferenciais de adensamento e fertilidade do solo e consequente desenvolvimento radicular possam ter contribuído para esse resultado, mas isso deve ser investigado em maiores detalhes futuramente para confirmar ou refutar esses resultados

    Is intrauterine exposure to acetaminophen associated with emotional and hyperactivity problems during childhood? : findings from the 2004 Pelotas birth cohort

    Background: Longitudinal studies have consistently reported that prenatal exposure to acetaminophen can to lead to an increased risk of attention deficit-hyperactivity disorder during childhood. This study aimed to investigate the association between intrauterine exposure to acetaminophen and the presence of emotional and behavioral problems at the ages of 6 and 11 years in a low-middle income country. Methods: We performed a prospective longitudinal population-based study using data from the 2004 Pelotas birth cohort. From the 4231 initial cohort participants, 3722 and 3566 children were assessed at 6 and 11 years of age, respectively. The outcomes were assessed using the parent version of Strengths and Difficulties Questionnaire (SDQ). The cut-off points established for the Brazilian population were used to categorize the outcomes. Crude and adjusted odds ratio were obtained through logistic regression. Results: Acetaminophen was used by 27.5% (95% confidence interval [CI]: 26.1–28.9) of the mothers at least once during pregnancy. The prevalence of emotional problems at 6 and 11 years was 13.6 and 19.9%, respectively. For hyperactivity problems, prevalence was 13.9 and 16.1%, respectively. Intrauterine exposure to acetaminophen increased the odds of having emotional (odds ratio [OR] = 1.47; 95% CI: 1.07–2.02) and hyperactivity/inattention (OR = 1.42; 95% CI: 1.06–1.92) problems in 6-year-old boys. At the age of 11, a small decrease in the effect was observed for both outcomes after adjustment: OR = 1.31 (95% CI: 0.99–1.73) for emotional problems and OR = 1.25 (95% CI: 0.95–1.65) for hyperactivity/inattention in boys. No association for any phenotypes at both ages was observed for girls. Conclusion: The effect of intrauterine exposure to acetaminophen in emotional and hyperactivity symptoms was dependent on sex in a Brazilian cohort. While it seemed to be important for boys, mainly at 6 years of age, for girls, no association was observed

    Genome of the Avirulent Human-Infective Trypanosome—Trypanosoma rangeli

    Background: Trypanosoma rangeli is a hemoflagellate protozoan parasite infecting humans and other wild and domestic mammals across Central and South America. It does not cause human disease, but it can be mistaken for the etiologic agent of Chagas disease, Trypanosoma cruzi. We have sequenced the T. rangeli genome to provide new tools for elucidating the distinct and intriguing biology of this species and the key pathways related to interaction with its arthropod and mammalian hosts.  Methodology/Principal Findings: The T. rangeli haploid genome is ,24 Mb in length, and is the smallest and least repetitive trypanosomatid genome sequenced thus far. This parasite genome has shorter subtelomeric sequences compared to those of T. cruzi and T. brucei; displays intraspecific karyotype variability and lacks minichromosomes. Of the predicted 7,613 protein coding sequences, functional annotations could be determined for 2,415, while 5,043 are hypothetical proteins, some with evidence of protein expression. 7,101 genes (93%) are shared with other trypanosomatids that infect humans. An ortholog of the dcl2 gene involved in the T. brucei RNAi pathway was found in T. rangeli, but the RNAi machinery is non-functional since the other genes in this pathway are pseudogenized. T. rangeli is highly susceptible to oxidative stress, a phenotype that may be explained by a smaller number of anti-oxidant defense enzymes and heatshock proteins.  Conclusions/Significance: Phylogenetic comparison of nuclear and mitochondrial genes indicates that T. rangeli and T. cruzi are equidistant from T. brucei. In addition to revealing new aspects of trypanosome co-evolution within the vertebrate and invertebrate hosts, comparative genomic analysis with pathogenic trypanosomatids provides valuable new information that can be further explored with the aim of developing better diagnostic tools and/or therapeutic targets
