    RNA-Sequencing Analysis of 5' Capped RNAs Identifies Many New Differentially Expressed Genes in Acute Hepatitis C Virus Infection

    We describe the first report of RNA sequencing of 5' capped (Pol II) RNAs isolated from acutely hepatitis C virus (HCV) infected Huh 7.5 cells that provides a general approach to identifying differentially expressed annotated and unannotated genes that participate in viral-host interactions. We identified 100, 684, and 1,844 significantly differentially expressed annotated genes in acutely infected proliferative Huh 7.5 cells at 6, 48, and 72 hours, respectively (fold change ≥ 1.5 and Bonferroni adjusted p-values < 0.05). Most of the differentially expressed genes (>80%) and biological pathways (such as adipocytokine, Notch, Hedgehog and NOD-like receptor signaling) were not identified by previous gene array studies. These genes are critical components of host immune, inflammatory and oncogenic pathways and provide new information regarding changes that may benefit the virus or mediate HCV induced pathology. RNAi knockdown studies of newly identified highly upregulated FUT1 and KLHDC7B genes provide evidence that their gene products regulate and facilitate HCV replication in hepatocytes. Our approach also identified novel Pol II unannotated transcripts that were upregulated. Results further identify new pathways that regulate HCV replication in hepatocytes and suggest that our approach will have general applications in studying viral-host interactions in model systems and clinical biospecimens

    Dietary resveratrol attenuates oxidative stress in Pacific whiteleg shrimp, Litopenaeus vannamei, in response to ammonia stress

    To study the effect of resveratrol on the antioxidant system of Litopenaeus vannamei, resveratrol (0, 40, 80, 120 and 180 mg/kg) was added to the feed. After 7 days of culture, 10 tails of L. vannamei were randomly selected and exposure to ammonia stress at the sublethal concentration of ammonia (20 mg/L) for 24h. The results showed that the malondialdehyde (MDA) level in serum of shrimp fed with resveratrol decreased significantly compare to the control group (p< 0.05). After the ammonia stress, the MDA levels in serum in the Res 120 group and the Res 160 group was lower significantly than that in the control group (p< 0.05). The activities of superoxide dismutase (SOD) in the hepatopancreas and the serum, and glutathione peroxidase (GPX) in the serum of shrimps fed with resveratrol showed a decrease trend before the ammonia stress. Conversely, the activities of SOD and the total antioxidant competence (T-AOC) levels in the hepatopancreas in the Res 80 group and Res 120 group were significantly higher than that in the control group after the ammonia stress (p<0.05). Dietary resveratrol decreased oxidative damage, increased antioxidant enzyme activities under the ammonia stress. Our results demonstrated that resveratrol attenuated oxidative stress and improved the resistance to the ammonia stress

    <p>(A) The effect of tannic acid on a HCV replicon, that displays intracellular replication events, was determined. HCV1b replicon RNA was electroporated into Huh7.5 cells followed by seeding of cells into six-well plates. After 48 hours of culture cells were trypsinized and seeded into twelve-well plates and tannic acid was added at concentrations of 0 μM, 3.1 μM, 6.3 μM, 12.5 μM and 25 μM and cells were cultured for 48 hours as described in Materials and Methods. Mock electroporation (no HCV1b replicon RNA) of cells provided controls. Cells were lysed and Western blots were done with anti-NS3 monoclonal antibodies as described in Materials and Methods. Experiments were performed three times and representative examples are shown. (B) The levels of NS3 were normalized to β-actin and the ratios of NS3/β-actin relative to 0 μM tannic acid (no inhibitor, 100% level of NS3) were plotted against inhibitor concentrations. The viability of Huh7.5 cells was determined using a CellTiter 96 Aqueous One solution cell proliferation assay as described in Materials and Methods. Each experiment was performed in triplicate and repeated at least three times. The CC<sub>50</sub> based on these studies was 146.1 ± 6.2 μM. (C) The direct effect of tannic acid on Renilla luciferase (Rluc) assays: HCV JFH1-AM120-Rluc infected cell lysates containing Rluc protein was mixed with increasing concentrations of tannic acid for ten minutes at room temperature and luciferase activity was measured as described in Materials and Methods. Assays were done in triplicate and experiments done three times. Results are presented as the mean ± standard deviation (SD) (n = 9) of relative light units (RLU). No significant difference (Student’s t-test p>0.05) in Rluc assay results were observed with tannic acid concentrations ≤ 50 μM.</p

    <p>(A) The effect of tannic acid on cell-to-cell spread of HCV as measured by viral protein staining. Huh7.5 cells were infected with HCV using an MOI of 0.005 at 37°C. The inoculum was removed two hours post infection and replaced with a 1% agarose medium overlay containing 25 μM tannic acid. Controls had no tannic acid. Cells were incubated at 37°C for 72 hours as described in Materials and Methods. Infected cells were labeled by indirect immuno-fluorescence using an anti-HCV NS5A monoclonal antibody (red), nuclei were stained with DAPI (blue) and photographs were taken with a fluorescence microscope as described in Materials and Methods. (B) The mean and standard deviation of infected cells/focus was determined by visual counting of 30 foci present on randomly selected fields of cover slips for each condition. P-values were calculated using the Student’s ttest (asterisk indicates P = 0.000019). Experiments were performed three times and representative examples are shown in A and B.</p

Inhibition of HCV JFH1-AM120-Rluc entry into Huh7.5 cells by tannic acid (25 μM) and heparin controls (200 μg/ml) was compared using four different experimental conditions (indicated by I through IV). Virus binding to cells was performed for 1 h at 4°C in the absence (II to IV) or presence (I) of compounds. Subsequently, cells were washed with PBS and shifted to 37°C to allow HCV entry to proceed. Infectivity is expressed as the percentage of the control (no tannic acid) which was assigned a 100% value. Mean values ± SD of three independent experiments done in triplicate are presented.
