    An oxygen isotope record of lacustrine opal from a European Maar indicates climatic stability during the Last Interglacial

    The penultimate temperate period, 127–110 ka before present (BP), bracketed by abrupt shifts of the global climate system initiating and terminating it, is considered as an analogue of the Holocene because of a similar low global ice‐volume. Ice core records as well as continental and marine records exhibit conflicting evidence concerning the climate variability within this period, the Last Interglacial. We present, for the first time, a high‐resolution record of oxygen isotopes in diatom opal of the Last Interglacial obtained from the Ribains Maar in France (44°50′09″N 3°49′16″E). Our results indicate that the Last Interglacial in southwestern Europe was generally a period of climatic stability. The record shows that the temperate period was initiated by an abrupt warm event followed midway by a minor climatic transition to a colder climate. An abrupt isotopic depletion that occurs simultaneously with abrupt changes in pollen and diatom assemblages marks the end of the temperate period, and is correlative with the Melisey I stadial.     Polarization sensitive spectroscopy of charged Quantum Dots

    We present an experimental and theoretical study of the polarized photoluminescence spectrum of single semiconductor quantum dots in various charge states. We compare our high resolution polarization sensitive spectral measurements with a new many-carrier theoretical model, which was developed for this purpose. The model considers both the isotropic and anisotropic exchange interactions between all participating electron-hole pairs. With this addition, we calculate both the energies and polarizations of all optical transitions between collective, quantum dot confined charge carrier states. We succeed in identifying most of the measured spectral lines. In particular, the lines resulting from singly-, doubly- and triply- negatively charged excitons and biexcitons. We demonstrate that lines emanating from evenly charged states are linearly polarized. Their polarization direction does not necessarily coincide with the traditional crystallographic direction.     A rapid co-culture stamping device for studying intercellular communication.

    Regulation of tissue development and repair depends on communication between neighbouring cells. Recent advances in cell micro-contact printing and microfluidics have facilitated the in-vitro study of homotypic and heterotypic cell-cell interaction. Nonetheless, these techniques are still complicated to perform and as a result, are seldom used by biologists. We report here development of a temporarily sealed microfluidic stamping device which utilizes a novel valve design for patterning two adherent cell lines with well-defined interlacing configurations to study cell-cell interactions. We demonstrate post-stamping cell viability of >95%, the stamping of multiple adherent cell types, and the ability to control the seeded cell density. We also show viability, proliferation and migration of cultured cells, enabling analysis of co-culture boundary conditions on cell fate. We also developed an in-vitro model of endothelial and cardiac stem cell interactions, which are thought to regulate coronary repair after myocardial injury. The stamp is fabricated using microfabrication techniques, is operated with a lab pipettor and uses very low reagent volumes of 20 μl with cell injection efficiency of >70%. This easy-to-use device provides a general strategy for micro-patterning of multiple cell types and will be important for studying cell-cell interactions in a multitude of applications

    Second-Hand Tobacco Smoke in Never Smokers Is a Significant Risk Factor for Coronary Artery Calcification

    ObjectivesThe aim of this study was to assess the relationship of the extent of subclinical atherosclerosis measured by coronary artery calcification (CAC) to the extent of second-hand tobacco smoke (SHTS) exposure in asymptomatic people who never smoked.BackgroundAn association between SHTS and CAC was recently reported in a single study, but the quantitative aspects of the relationship are not known.MethodsA cohort of 3,098 never smokers 40 to 80 years of age, enrolled in the FAMRI-IELCAP (Flight Attendant Medical Research Institute International Early Lung Cancer Action Program) screening program, completed a SHTS questionnaire, and had a low-dose nongated computed tomography scan. The questionnaire provided a quantitative score for total SHTS exposure, as well as separately as a child and as an adult at home and at work; 4 categories of exposure to SHTS were identified (minimal, low, moderate, and high exposure). CAC was graded using a previously validated ordinal scale score that ranged from 0 to 12. Logistic regression analysis of the prevalence and ordered logistic regression analysis of the extent of CAC were performed to assess the independent contribution of SHTS adjusted for age, sex, diabetes, hypercholesterolemia, hypertension, and renal disease. Linear and quadratic regression analyses of CAC and SHTS were performed.ResultsThe prevalence of CAC was 24.3% (n = 754) and was significantly higher in those with more than minimal SHTS exposure compared with those with minimal SHTS exposure (26.4% vs. 18.5%, p < 0.0001). The adjusted odds ratios for CAC prevalence were 1.54 (95% confidence interval: 1.17 to 2.20) for low SHTS exposure, 1.60 (95% confidence interval: 1.21 to 2.10) for moderate exposure, and 1.93 (95% confidence interval: 1.49 to 2.51) for high exposure. The association of the extent of SHTS with the extent of CAC was confirmed by the adjusted odds ratio (p < 0.0001).ConclusionsThe presence and extent of CAC were associated with extent of SHTS exposure even when adjusted for other risk factors for CAC, suggesting that SHTS exposure causes CAC

    Oceanic distribution of inorganic germanium relative to silicon: Germanium discrimination by diatoms

    Seventeen inorganic germanium and silicon concentration profiles collected from the Atlantic, southwest Pacific, and Southern oceans are presented. A plot of germanium concentration versus silicon concentration produced a near-linear line with a slope of 0.760 × 10−6 (±0.004) and an intercept of 1.27 (±0.24) pmol L−1 (r2 = 0.993, p < 0.001). When the germanium-to-silicon ratios (Ge/Si) were plotted versus depth and/or silicon concentrations, higher values are observed in surface waters (low in silicon) and decreased with depth (high in silicon). Germanium-to-silicon ratios in diatoms (0.608–1.03 × 10−6) and coupled seawater samples (0.471–7.46 × 10−6) collected from the Southern Ocean are also presented and show clear evidence for Ge/Si fractionation between the water and opal phases. Using a 10 box model (based on PANDORA), Ge/Si fractionation was modeled using three assumptions: (1) no fractionation, (2) fractionation using a constant distribution coefficient (KD) between the water and solid phase, and (3) fractionation simulated using Michaelis-Menten uptake kinetics for germanium and silicon via the silicon uptake system. Model runs indicated that only Ge/Si fractionation based on differences in the Michaelis-Menten uptake kinetics for germanium and silicon can adequately describe the data. The model output using this fractionation process produced a near linear line with a slope of 0.76 × 10−6 and an intercept of 0.92 (±0.28) pmol L−1, thus reflecting the oceanic data set. This result indicates that Ge/Si fractionation in the global ocean occurs as a result of subtle differences in the uptake of germanium and silicon via diatoms in surface waters

    Distress related to myocardial infarction and cardiovascular outcome: a retrospective observational study

    Background During acute coronary syndromes patients perceive intense distress. We hypothesized that retrospective ratings of patients' MI-related fear of dying, helplessness, or pain, all assessed within the first year post-MI, are associated with poor cardiovascular outcome. Methods We studied 304 patients (61 ± 11 years, 85% men) who after a median of 52 days (range 12-365 days) after index MI retrospectively rated the level of distress in the form of fear of dying, helplessness, or pain they had perceived at the time of MI on a numeric scale ranging from 0 ("no distress") to 10 ("extreme distress"). Non-fatal hospital readmissions due to cardiovascular disease (CVD) related events (i.e., recurrent MI, elective and non-elective stent implantation, bypass surgery, pacemaker implantation, cerebrovascular incidents) were assessed at follow-up. The relative CVD event risk was computed for a (clinically meaningful) 2-point increase of distress using Cox proportional hazard models. Results During a median follow-up of 32 months (range 16-45), 45 patients (14.8%) experienced a CVD-related event requiring hospital readmission. Greater fear of dying (HR 1.21, 95% CI 1.03-1.43), helplessness (HR 1.22, 95% CI 1.04-1.44), or pain (HR 1.27, 95% CI 1.02-1.58) were significantly associated with an increased CVD risk without adjustment for covariates. A similarly increased relative risk emerged in patients with an unscheduled CVD-related hospital readmission, i.e., when excluding patients with elective stenting (fear of dying: HR 1.26, 95% CI 1.05-1.51; helplessness: 1.26, 95% CI 1.05-1.52; pain: HR 1.30, 95% CI 1.01-1.66). In the fully-adjusted models controlling for age, the number of diseased coronary vessels, hypertension, and smoking, HRs were 1.24 (95% CI 1.04-1.46) for fear of dying, 1.26 (95% CI 1.06-1.50) for helplessness, and 1.26 (95% CI 1.01-1.57) for pain. Conclusions Retrospectively perceived MI-related distress in the form of fear of dying, helplessness, or pain was associated with non-fatal cardiovascular outcome independent of other important prognostic factors

    Urbanization comprehensively impairs biological rhythms in coral holobionts

    Coral reefs are in global decline due to climate change and anthropogenic influences (Hughes et al., Conservation Biology, 27: 261–269, 2013). Near coastal cities or other densely populated areas, coral reefs face a range of additional challenges. While considerable progress has been made in understanding coral responses to acute individual stressors (Dominoni et al., Nature Ecology & Evolution, 4: 502–511, 2020), the impacts of chronic exposure to varying combinations of sensory pollutants are largely unknown. To investigate the impacts of urban proximity on corals, we conducted a year-long in-natura study—incorporating sampling at diel, monthly, and seasonal time points—in which we compared corals from an urban area to corals from a proximal non-urban area. Here we reveal that despite appearing relatively healthy, natural biorhythms and environmental sensory systems were extensively disturbed in corals from the urban environment. Transcriptomic data indicated poor symbiont performance, disturbance to gametogenic cycles, and loss or shifted seasonality of vital biological processes. Altered seasonality patterns were also observed in the microbiomes of the urban coral population, signifying the impact of urbanization on the holobiont, rather than the coral host alone. These results should raise alarm regarding the largely unknown long-term impacts of sensory pollution on the resilience and survival of coral reefs close to coastal communities
