    Polish Adults’ Knowledge, Perceptions and Attitudes Concerning Legionellosis

    Despite the growing prevalence of legionellosis in Poland and worldwide, little is known about the extent of public awareness regarding the seriousness of this disease and the appropriate preventive measures. The aim of this work is to assess the Polish adults’ knowledge, perceptions, and beliefs about legionellosis and its causative agents, risk factors, exposure, and other relevant facts. Data for this cross-sectional study were gathered via a questionnaire that was constructed and validated by the study investigators before commencing the survey, which lasted from January to March 2022. Knowledge, attitude and practice towards legionella were measured and quantified. One-way ANOVA and chi square tests were used to compare between demographic variables and the level of knowledge. Regression analysis was conducted to examine the predictors for higher knowledge among study participants. A total of 251 participants with a mean age of 28.26 ± 9.6 were enrolled in the current study. Over two thirds (74%) were females, with higher education (62%). Older age was associated with less knowledge about legionellosis (B = -0.049, p < 0.001), while higher education was associated with more knowledge (B = 1.656, p < 0.001). No significant differences were found between genders (p = 0.066). A knowledge gap was present for diagnostic tests regarding legionella. On the other hand, knowledge about prevention procedures was quite high among study participants. This study showed that overall knowledge about legionellosis in Polish adults was quite low. In particular, older age groups and the less educated are in need of more awareness of legionellosis disease. A knowledge gap was particularly present regarding how the disease is diagnosed. Awareness campaigns containing simple, easy-to-understand information could prove useful in combating the disease

    Investigation into the prophylactic and therapeutic activity of coenzyme Q10 against COVID-19

    Purpose: To evaluate the anti-SARS CoV-2 effect of Coenzyme Q 10, Ubiquinol-10, and idebenone, which have beneficial therapeutic applications against diverse virus types, using molecular docking approach.Methods: The potential activity of Coenzyme Q10, Ubiquinol-10, and Idebenone against viral infections was explored through the collection of data from relevant literature, and by modelling these compounds virtually, using in silico investigation methods.Results: Coenzyme Q10 and ubiquinol-10 showed significant docking performance. They interacted with numerous amino acid residues of the main protease of SARS-CoV-2 ACE2 (7C8J), Alpha thrombin (1AE8), TYRO (4TS1) protein targets sides, SARS-coronavirus Orf7a accessory protein (1XAK), TNF (1RJ8), and Cytokine/receptor (1I1R).Conclusion: The findings of our study showed promising inhibitory activities of the selected compounds against the main proteases of SARS-CoV-2. Consequently, these compounds have theoretical effects on inhibiting the viral entry, reproduction, and ultimately the prevention and/or treatment of the SARSCoV2 infection

    Stability-Indicating HPLC Determination of Gemcitabine in Pharmaceutical Formulations

    A simple, sensitive, inexpensive, and rapid stability indicating high performance liquid chromatographic method has been developed for determination of gemcitabine in injectable dosage forms using theophylline as internal standard. Chromatographic separation was achieved on a Phenomenex Luna C-18 column (250 mm × 4.6 mm; 5μ) with a mobile phase consisting of 90% water and 10% acetonitrile (pH 7.00±0.05). The signals of gemcitabine and theophylline were recorded at 275 nm. Calibration curves were linear in the concentration range of 0.5–50 μg/mL. The correlation coefficient was 0.999 or higher. The limit of detection and limit of quantitation were 0.1498 and 0.4541 μg/mL, respectively. The inter- and intraday precision were less than 2%. Accuracy of the method ranged from 100.2% to 100.4%. Stability studies indicate that the drug was stable to sunlight and UV light. The drug gives 6 different hydrolytic products under alkaline stress and 3 in acidic condition. Aqueous and oxidative stress conditions also degrade the drug. Degradation was higher in the alkaline condition compared to other stress conditions. The robustness of the methods was evaluated using design of experiments. Validation reveals that the proposed method is specific, accurate, precise, reliable, robust, reproducible, and suitable for the quantitative analysis

    Phenolic Content and Antioxidant and Antimicrobial Activities of Malva sylvestris L., Malva oxyloba Boiss., Malva parviflora L., and Malva aegyptia L. Leaves Extract

    Background. The plants brought by Arabs were of real therapeutic values. Ibn Al-Baitar, an Islamic scholar (pharmacist, botanist, and physician), in his encyclopedia wrote the detailed characterization of more than one thousand herbs describing their medicinal value, methods of preparation, and their route of administration. Objectives. The current investigation points towards the quantitative characterization of the phenolic contents among the four edible Malva plants species (Malva sylvestris L., Malva oxyloba Boiss., Malva parviflora L., and Malva aegyptia L.) and also towards assessing their antibacterial activity against one Gram-positive isolate (Staphylococcus aureus) and four Gram-negative strains Escherichia coli, Pseudomonas aeruginosa, Shigella sonnei, and Proteus vulgaris. It also aimed to evaluate the free radical scavenging activity of hexane, methanolic, aqueous, and acetone extracts of four Malva species. Methods. By utilizing the Folin–Ciocalteu procedure and gallic acid as a reference molecule, the phenolic contents were estimated. In addition, the broth microdilution method was used to evaluate four plants’ 16 extracts, and the DPPH (2,2-diphenyl-1-picrylhydrazyl) method was utilized to assess the abovementioned extracts against oxidative stress. Results. The results showed that the methanolic extract of M. oxyloba has the highest contents of phenols (191.54 ± 2.84 mg of GAE/g) and has the best antioxidant capacity with an IC50 value of 1.94 ± 1.84 µg/ml, which is very close to Trolox. Regarding the ferrous ion chelating activity of the extract, the methanolic extract of M. sylvestris exhibits appreciable activity with IC50 values of 52.7 ± 1.8 µg/ml. In addition, the plant extract and acetone extract of M. sylvestris showed a strong antibacterial activity with an MIC value of 0.0078 mg/ml. Conclusion. The methanolic extract of M. oxyloba has a pharmacological potential as a valuable natural product that can be utilized as a main ingredient in the design and development of new therapeutic formulations. It exerts multiple inhibitory properties against oxidative stress and bacterial growth. As such, it is emerging as a promising therapeutic agent for the treatment of various neurodegenerative diseases and many types of human infectious diseases
