    Spatial market expansion through mergers

    In this paper we present a model that studies firm mergers in a spatial setting. A new model is formulated that addresses the issue of finding the number of branches that have to be eliminated by a firm after merging with another one, in order to maximize profits. The model is then applied to an example of bank mergers in the city of Barcelona.     Behavioural facial animation using motion graphs and mind maps

    We present a new behavioural animation method that combines motion graphs for synthesis of animation and mind maps as behaviour controllers for the choice of motions, significantly reducing the cost of animating secondary characters. Motion graphs are created for each facial region from the analysis of a motion database, while synthesis occurs by minimizing the path distance that connects automatically chosen nodes. A Mind map is a hierarchical graph built on top of the motion graphs, where the user visually chooses how a stimulus affects the character's mood, which in turn will trigger motion synthesis. Different personality traits add more emotional complexity to the chosen reactions. Combining behaviour simulation and procedural animation leads to more emphatic and autonomous characters that react differently in each interaction, shifting the task of animating a character to one of defining its behaviour.</p

    Intervenção Social - Arte ou Ciência?: O Assistente Social como agente de mudança numa autarquia de interior.

    O presente trabalho baseia-se na pesquisa documental e na análise crítico-reflexiva sobre o serviço social no Poder Local, a sua evolução, influências e implicações no trabalho desenvolvido no (pelo) Gabinete de Ação Social do Município de Pampilhosa da Serra. O serviço social assume, durante o Estado Novo, um papel de controle, com uma prática associada essencialmente ao Estado e a algumas entidades da sociedade civil, nomeadamente à Igreja Católica. Com a Era Democrática, assiste-se ao crescimento da intervenção do Estado e à descentralização da sua atuação para as Autarquias, assumindo-se o SAAL (1974) como um marco histórico. A intervenção ganha, assim, um caráter “territorial” crescendo as responsabilidades das autarquias, quer no âmbito das transferências de competências por parte do Poder Central, quer ao nível da dinamização de serviços de âmbito municipal e da criação de respostas sociais específicas. O PRS (1997) e a RLIS (2013), apresentam-se como exemplos dessa territorialização, reforçando as atribuições das Autarquias ao nível da ação social. O primeiro, implementado a nível nacional, visa a articulação de recursos e uma intervenção integrada. Tem como princípios, a promoção a participação da população e dos agentes locais quer na elaboração de diagnósticos sociais, quer na criação de respostas adequadas às necessidades. A segunda, em fase de implementação, visa a criação de uma metodologia de trabalho ao nível do atendimento e acompanhamento social. Destaca-se o papel das Autarquias e da Sociedade Civil, na efetivação de uma intervenção social de proximidade. A crescente desresponsabilização do Estado na intervenção social, em particular do Poder Central, motivada pela Crise Económica, provoca alterações nas políticas sociais. Surgem as políticas de inserção direcionadas para públicos específicos, tendencialmente contratualizadas e centradas no sujeito. Estas alterações tiveram, também, implicações na atuação do Município de Pampilhosa da Serra, onde o serviço social se desenvolveu a par da implementação dos programas de âmbito comunitário, nomeadamente do PDIAS e PLCP (1996) e do projecto-piloto do RMG (1997). Sistematizou-se com a implementação das Redes Sociais e operacionaliza-se no Gabinete de Ação Social, atuando em três dimensões: mediação, promoção e execução. Da análise e reflexão em torno da atuação do GAS, no qual inscrevemos a nossa intervenção profissional, consideramos que esta se desenvolve numa relação sociopolítica e operacional, assumindo a Autarquia um papel ativo ao criar e/ou reforçar medidas de apoio socioeducativo e económico, de forma a garantir o bem-estar social e a qualidade de vida dos cidadãos Pampilhosenses. O município assume-se como o patamar de atuação de proximidade por excelência, onde o local se perspetiva como o espaço onde a intervenção social se operacionaliza, enquanto que o Poder Local, em conjunto com a sociedade Civil, assumem o poder de co construir a mudança social. / This work is based on documentary research and critical and reflective analysis of the social service in Local Government, its evolution, influences and implications on the work of the (at) Social Action Office of the municipality of Pampilhosa da Serra. The social service assumes, during the Estado Novo, a paper control, primarily associated with a practice the state and some civil society organizations, including the Catholic Church. With the Democratic Era, we are witnessing the growth of state intervention and the decentralization of its activities to the local authorities, assuming the SAAL (1974) as a historical landmark. Intervention win, so a character "territorial" growing responsibilities of local authorities, or within the transfer of responsibilities from the Central Power, both in terms of promotion of municipal services and the creation of specific social responses. The PRS (1997) and the RLIS (2013), are presented as examples of territorial, strengthening the powers of local authorities to the level of social action. The first, implemented nationally, aimed at articulating features and an integrated intervention. Its principles, promoting the participation of the population and local actors when developing social diagnosis, whether the creation of appropriate responses to the needs. The second, under implementation, aims to create a working methodology in terms of care and social support. It highlights the role of local authorities and civil society in the execution of a social intervention proximity. The growing irresponsibility of the state in social intervention, in particular the Central Power, motivated by the economic crisis, causes changes in social policies. Arise inclusion policies targeting specific audiences tend contracted and centered on the subject. These changes have also implications for the work of the municipality of Pampilhosa da Serra, where the social work developed together with the implementation of Community-wide programs, including the PDIAS and PLCP (1996) and the pilot project of RMG (1997). Systematized with the implementation of Social Networks and made operational in the Social Action Office, working in three dimensions: mediation, promotion and implementation. Analysis and reflection around the GAS operation, in which we inscribe our professional intervention, we believe that this is developed in a socio-political and operational relationship, assuming the Municipality an active role to create and / or strengthen measures of socio-educational and economic support, to ensure the welfare and quality of life of citizens Pampilhosenses.     Widespread infection with hemotropic mycoplasmas in bats in Spain, including a hemoplasma closely related to 'Candidatus Mycolasma hemohominis'

    Molecular analyses of blood samples revealed infection with hemoplasmas in 97% of 31 cave bats captured in three caves in North-Eastern Spain. The characterization of 1250 bp of the 16S rRNA gene in 29 of the positive bats identified two different groups of sequences. Twenty-two Schreibers' bats (Miniopterus schreibersii) and one long-eared bat (Myotis capaccinii) shared one group, composed of seven closely related sequences. These sequences showed an identity of about 97% with "Candidatus Mycoplasma hemohominis" and the phylogenetic branch including bat and human sequences showed a 100% bootstrap value, supporting a close phylogenetic relationship between these hemoplasmas. The second group, representing a potentially novel species, was composed of a single sequence shared by six Schreibers' bats that had 91% identity with the recently reported hemoplasma from little brown bats in North America.     Purification of Arabinoxylans from corn fiber and preparation of bioactive films for food packaging

    Corn fiber, a by-product of the starch industry, is presently incorporated in animal feed. However, it has arabinoxylans as added-value components (besides ferulic acid) that should be valorized. In this work, the raw material, a fraction enriched in arabinoxylans from corn fiber, previously produced by alkaline extraction from corn fiber and pre-concentrated by ultrafiltration, was further purified. The use of ultrafiltration operated in diafiltration mode (dia-ultrafiltration) was evaluated for the purification of the arabinoxylans fraction. The objective was to maximize the removal of the small contaminants from the fraction and to maximize the permeability and/or the permeate flux, by selecting the relevant operating conditions involved in this process. The removal of contaminants (%) was estimated when their apparent rejection stabilized. Edible films were produced, from the resultant purified arabinoxylans fraction, using glycerol as plasticizer (30% dry basis). Additionally, films with the incorporation of ferulic acid were developed, in order to obtain barriers with antioxidant activity. The films were characterized in terms of mechanical properties, antioxidant activity and permeability to water vapor. Publisher Copyright: © 2023 by the authors.Arabinoxylan has prebiotic properties, as it is able to resist digestion in the small intestine and undergoes fermentation in the large intestine. In this work, arabinoxylan was extracted from corn fiber using an alkaline solution and further purified with membrane processing. It was found that the extracts were mainly composed of xylose (50–52%), arabinose (37–39%), galactose (9%) and glucose (1–4%), with an A/X ratio of 0.72–0.77. All the extracts were composed of phenolic compounds, including ferulic acid derivatives such as dimers, trimers and tetramers. The purified extract had a lower concentration of ferulic and p-coumaric acid (0.004 and 0.02 mg/mgdry_weight, respectively) when compared to raw extract (19.30 and 2.74 mg/mgdry_weight, respectively). The same effect was observed for the antioxidant activity, with purified extracts having a lower value (0.17 ± 0.02 µmol TEAC/mg) when compared to the raw extract (2.20 ± 0.35 µmol TEAC/mg). The purified extract showed a greater antiproliferative effect against the HT29 cell line with EC50 = 0.12 ± 0.02 mg/mL when compared to the raw extract (EC50 = 5.60 ± 1.6 mg/mL). Both raw and purified extracts did not show any cytotoxicity to the Caco-2 cell line in the maximum concentration tested (10 mg/mL).publishersversionpublishe

    The 4q25 variant rs13143308T links risk of atrial fibrillation to defective calcium homoeostasis

    Aims: Single nucleotide polymorphisms on chromosome 4q25 have been associated with risk of atrial fibrillation (AF) but the exiguous knowledge of the mechanistic links between these risk variants and underlying electrophysiological alterations hampers their clinical utility. Here, we tested the hypothesis that 4q25 risk variants cause alterations in the intracellular calcium homoeostasis that predispose to spontaneous electrical activity. Methods and results: Western blotting, confocal calcium imaging, and patch-clamp techniques were used to identify mechanisms linking the 4q25 risk variants rs2200733T and rs13143308T to defects in the calcium homoeostasis in human atrial myocytes. Our findings revealed that the rs13143308T variant was more frequent in patients with AF and that myocytes from carriers of this variant had a significantly higher density of calcium sparks (14.1¿±¿4.5 vs. 3.1¿±¿1.3 events/min, P¿=¿0.02), frequency of transient inward currents (ITI) (1.33¿±¿0.24 vs. 0.26¿±¿0.09 events/min, P¿<¿0.001) and incidence of spontaneous membrane depolarizations (1.22¿±¿0.26 vs. 0.56¿±¿0.17 events/min, P¿=¿0.001) than myocytes from patients with the normal rs13143308G variant. These alterations were linked to higher sarcoplasmic reticulum calcium loading (10.2¿±¿1.4 vs. 7.3¿±¿0.5¿amol/pF, P¿=¿0.01), SERCA2 expression (1.37¿±¿0.13 fold, P¿=¿0.03), and RyR2 phosphorylation at ser2808 (0.67¿±¿0.08 vs. 0.47¿±¿0.03, P¿=¿0.01) but not at ser2814 (0.28¿±¿0.14 vs. 0.31¿±¿0.14, P¿=¿0.61) in patients carrying the rs13143308T risk variant. Furthermore, the presence of a risk variant or AF independently increased the ITI frequency and the increase in the ITI frequency observed in carriers of the risk variants was exacerbated in those with AF. By contrast, the presence of a risk variant did not affect the amplitude or properties of the L-type calcium current in patients with or without AF. Conclusions: Here, we identify the 4q25 variant rs13143308T as a genetic risk marker for AF, specifically associated with excessive calcium release and spontaneous electrical activity linked to increased SERCA2 expression and RyR2 phosphorylation.Peer ReviewedPostprint (author's final draft

    Transcriptome-based SNP discovery and validation in the hybrid zone of the neotropical annual fish genus Austrolebias

    The genus Austrolebias (Cyprinodontiformes: Rivulidae) represents a specious group of taxa following annual life cycles in the neotropical ichthyofauna. They live in temporary ponds and each generation must be completed in a few months, depending on environmental stochasticity. Annual fish survive the dry season through diapausing eggs buried in the substrate of these ponds. A hypothesized bimodal hybrid zone between two taxa of the genus, A. charrua and A. reicherti from Dos Patos Merin lagoon system, was recently proposed based on genetics and morphological analyses. However, hundreds of additional nuclear molecular markers should be used to strongly support this hypothesized bimodal pattern. In the present paper, we conducted RNA-seq-based sequencing of the transcriptomes from pools of individuals of A. charrua, A. reicherti and their putative natural hybrids from the previously characterized hybrid zone. As a result, we identified a set of 111,725 SNP (single nucleotide polymorphism) markers, representing presumably fixed allelic di erences among the two species. The present study provided the first panel of 106 SNP markers as a single diagnostic multiplex assay and validated their capacity to reconstruct the patterns of the hybrid zone between both taxa. These nuclear markers combined with Cytb gene and morphological analyses detected a population structure in which some groups among the hybrid swarms showed di erent level of introgression towards one or the other parental species according to their geographic distribution. High-quality transcriptomes and a large set of gene-linked SNPs should greatly facilitate functional and population genomics studies in the hybrid zone of these endangered species

    A stable temperature may favour continuous reproduction by Theodoxus fluviatilis and explain its high densities in some karstic springs

    Theodoxus fluviatilis is a common gastropod in many karstic springs in central Portugal. We investigated the possible reasons for the near-total restriction of this species to these springs. We first determined the spatial distribution of the species within a spring (Anços) and related the densities at sampling-patch scales to selected physical and chemical variables. We then determined the densities at several locations downstream from the spring and related these densities to selected physical and chemical variables. Finally, we assessed the population dynamics of the gastropod in the spring. In the spring, T. fluviatilis was more abundant in shallow areas with a rapid current and cobble-boulder substrates. In June-July 2006, the mean densities of T. fluviatilis in the spring varied from ~ 10 to ~ 9000 individuals m-2 but decreased to zero 3800 m downstream. The physical and chemical changes along the stretch studied were minor; no significant correlations (Spearman rank correlation; p > 0.05) were observed between the gastropod abundances and the measured environmental variables or the PCA axes. In the spring, juveniles and egg masses were observed year-round, suggesting continuous reproduction. The temperature at the spring was fairly constant, ranging from 15.3 to 16.6 ºC during the year (daily variation 0.05) entre la abundancia de T. fluviatilis y las variables ambientales o los ejes de Análisis de Componentes Principales. En el manantial, juveniles y masas de huevos fueron observadas durante todo el año y no se obsevaron cohortes, sugeriendo reproducción contínua. La temperatura en el manantial varió entre 15.3 y 16.6 ºC durante todo el año (variación diaria < 0.1 ºC). Novecientos metros río abajo las variaciones térmicas diarias llegaron a los 5 ºC. Proponemos pues que la elevada densidad de T. fluviatilis en el manantial kárstico de Anços se mantiene gracias a las temperaturas elevadas y constantes que favorecen su continua reproducción

    Fundamentos e instrumentos do financiamento público em investigação e desenvolvimento - caso português

    Mestrado em Economia e Gestão de Ciência, Tecnologia e InovaçãoO financiamento público em investigação e desenvolvimento tem vindo a tornar-se um elemento fulcral para os Estados contribuírem para o desenvolvimento dos respectivos países, quer este seja perspectivado nos planos cultural, social ou económico. Ao longo deste trabalho, enquadram-se os fundamentos do financiamento das actividades de investigação e desenvolvimento em Portugal, tentando identificá-los a partir da caracterização dos observados noutros países, constatando as possíveis (directas e indirectas) influências na política científica portuguesa. A partir de tal perspectiva é observada também a evolução dos organismos tutelares das políticas científicas e fontes de financiamento, identificando os principais programas para o efeito. Neste contexto, são igualmente realçados alguns dos momentos relevantes do desenvolvimento do Sistema Científico e Tecnológico Nacional (SCTN), enquadrado por uma abordagem do financiamento público das actividades de Investigação e Desenvolvimento (I&D), tendo em vista perceber muitas das caracterizações a que o SCTN tem sido sujeito em múltiplos trabalhos sobre o tema. Conclui-se que as políticas para o desenvolvimento científico e tecnológico português têm sido maioritariamente influenciado por acontecimentos e realidades externas, sendo particularmente importante o enquadramento resultante da integração do país em organizações internacionais onde se destacam a OTAN, OCDE e União Europeia.The public funding of research and development has been a key element for the States to contribute to the development of their countries, whether it is viewed in cultural, social or economic fields. Throughout this work, it is provided the fundamentals of funding research and development activities in Portugal, trying to identify them from the characterization of those observed in other countries, noting the potential (direct and indirect) influence on science policy in Portugal. From such a perspective it is also observed the evolution of the institutions responsible for science policies and funding sources and identifying the main programs for this purpose. In this context, it is also highlighted main of the relevant moments of the development of National Scientific and Technological System (NSTS), framed by an approach of public funding of research and development in order to realize many of the characterizations that the NSTS has been subject to multiple work on the topic. These conclude that the Portuguese scientific and technological development is largely influenced by external events and realities, and it is particularly important framework results from the country integration in international organizations like NATO, OECD and European Union
