    Nest cover and faecal glucocorticoid metabolites are linked to hatching success and telomere length in breeding Common Eiders (Somateria mollissima)

    Habitat-associated crypsis may affect perceived predation vulnerability, selecting for different predator avoidance strategies. Glucocorticoids could mediate the adjustment of escape responses to the extent of crypsis, introducing an overlooked source of variation in glucocorticoid-fitness relationships. However, prolonged exposure to elevated glucocorticoids may be costly, leading to accelerated telomere loss and, consequently, senescence. Here, we examined how nest cover and immunoreactive faecal glucocorticoid metabolite (fGCM) levels are linked to hatching success and telomere length in breeding female Common Eiders (Somateria mollissima (L., 1758)). We hypothesized that the degree of nest crypsis, reflecting differences in perceived predation risk, would moderate the relationship between reproductive success and fGCM levels. We also expected that telomere length would be shorter in birds with higher glucocorticoid concentration. Results showed that individuals with high fGCM levels had higher hatching success in nests with low cover, while low fGCM levels were more successful in well-concealed nests. We found that shorter telomeres were associated with high fGCM in nesting sites offering little cover and with low fGCM in well-concealed ones. This study provides the first evidence of habitat-dependent moderation of the relationships between stress physiology, telomere length and hatching success.Peer reviewe

    Keratins Stabilize Hemidesmosomes through Regulation of β4-Integrin Turnover

    Epidermal integrity and wound healing depend on remodeling of cell-matrix contacts including hemidesmosomes. Mutations in β4-integrin and plectin lead to severe epidermolysis bullosa (EB). Whether mutations in keratins K5 or K14, which cause EB simplex, also compromise cell-matrix adhesion through altering hemidesmosomal components is not well investigated. In particular, the dependence of β4-integrin endocytosis and turnover on keratins remains incompletely understood. Here, we show that the absence of keratins causes loss of plectin-β4-integrin interaction and elevated β4-integrin phosphorylation at Ser1354 and Ser1362. This triggered a caveolin-dependent endocytosis of β4-integrin but not of other integrins through Rab5 and Rab11 compartments in keratinocytes. Expressing a phospho–deficient β4-integrin mutant reduces β4-integrin endocytosis and rescues plectin localization in keratin–free cells. β4-integrin phosphorylation in the absence of keratins resulted from elevated Erk1/2 activity downstream of increased EGFR and PKCα signaling. Further, increased Erk1/2 phosphorylation and altered plectin localization occur in keratin–deficient mouse epidermis in vivo. Strikingly, expression of the K14-R125P EBS mutant also resulted in plectin mislocalization and elevated β4-integrin turnover, suggesting disease relevance. Our data underscore a major role of keratins in controlling β4-integrin endocytosis involving a plectin-Erk1/2-dependent mechanism relevant for epidermal differentiation and pathogenesis

    Sources of fluids and metals and evolution models of skarn deposits in the Qimantagh metallogenic belt: A case study from the Weibao deposit, East Kunlun Mountains, northern Tibetan Plateau.

The final version is available from Elsevier via the DOI in this record.The Weibao Cu-Pb-Zn deposit is typical of skarn deposits in the Qimantagh metallogenic belt (QMB), EastKunlun Mountains, northern Tibetan Plateau. It comprises three ore blocks from west to east, known as Weixi,Main and Weidong. Mineralization in the Weibao deposit is intimately related to Late Triassic intrusions oc-curring at Weixi and Weidong, and orebodies are predominantly hosted by the Langyashan Formation (marinecarbonate rocks), and to a lesser extent the Devonian volcanic rocks. Skarns from Weixi and Weidong arecharacterized by a high garnet/pyroxene ratio and diopside- and andradite-rich composition of pyroxenes andgarnets, indicating a proximal, oxidized type. In contrast, skarn mineralogy of Main indicates a slightly reducedcondition, typical of Pb-Zn skarn deposits. At least five hydrothermal mineralization stages can be identified andthe microthermometric study indicates a general trend of cooling and dilution of the magmatic-hydrothermalfluids. Significant precipitation of Cu-Fe sulfides commenced from the fluid with the temperature of 340–448 °Cand the salinity of 2.1–15.0 wt% NaCl equiv. Pb-Zn sulfides however mainly precipitated when the temperaturesdeclined to < 370 °C and the salinity declined to < 7.6 wt% NaCl equiv. Carbon, hydrogen and oxygen isotopiccomposition of the fluids suggests a predominantly igneous source for the initial fluids, which were modifiedoutward by fluid-rock interaction and mixing with meteoric waters. Both sulfur and lead isotope composition ofsulfides supports a mixed sulfur and metal reservoir consisting of Triassic intrusive rocks and wall rocks.Compared to early skarn-forming and late quartz-carbonate stages, two ore-forming stages show clear evidenceof fluid boiling and fluid-rock interaction. Combined with microthermometric data of ore-forming stages, it canbe deduced that fluid cooling, boiling and fluid-rock interaction were responsible for the significant metalprecipitation.     Adakite-like granitoids of Songkultau: A relic of juvenile Cambrian arc in Kyrgyz Tien Shan

    The early Paleozoic Terskey Suture zone, located in the southern part of the Northern Tien Shan domain in Kyrgyzstan, comprises tectonic slivers of dismembered ophiolites and associated primitive volcanics and deep-marine sediments. In the Lake Songkul area, early-middle Cambrian pillow basalts are crosscut by the Songkultau intrusion of coarse-grained gneissose quartz diorites and tonalites with geochemical characteristics typical for high-SiO2 adakites (SiO2 ​> ​56 ​wt.%, Al2O3 ​> ​15 ​wt.%, Na2O ​> ​3.5 ​wt.% and high Sr/Y and La/Yb ratios). The Songkultau granitoids have positive initial εNd (+3.8 to +6.4) and εHf (+12.3 to +13.5) values indicating derivation from sources with MORB-like isotopic signature. Volcanic formations, surrounding the Songkultau intrusion, have geochemical affinities varying from ocean floor to island arc series. This rock assemblage is interpreted as a relic of an early-middle Cambrian primitive arc where the adakite-like granitoids were derived from partial melting of young and hot subducted oceanic crust. An age of 505 ​Ma, obtained for the Songkultau intrusion, shows that hot subduction under the Northern Tien Shan continued until middle Cambrian. The primitive arc complexes were obducted onto the Northern Tien Shan domain, where the Andean type continental magmatic arc developed in Cambrian and Ordovician. Formation of the Andean type arc was accompanied by uplift, erosion and deposition of coarse clastic sediments. A depositional age of ca. 470 Ma, obtained for the gravellites in the Lake Songkul area, is in agreement with the timing of deposition for lower Ordovician conglomerates elsewhere in the Northern Tien Shan, and corresponds to the main phase of the Andean type magmatism. The Songkultau adakites in association with surrounding ocean floor and island arc formations constitute a relic of a primitive Cambrian arc and represent a juvenile domain of substantial size identified so far within the predominantly crustal-derived terranes of Tien Shan. On a regional scale this primitive arc can be compared with juvenile Cambrian arcs of Kazakhstan, Gorny Altai and Mongolia.©2020 China University of Geosciences (Beijing) and Peking University.     Evaluating the personality structure of semi-captive Asian elephants living in their natural habitat

    Data on personality for long-lived, highly social wild mammals with high cognitive abilities are rare. We investigated the personality structure of Asian elephants (Elephas maximus) by using a large sample of semi-captive timber elephants in Myanmar. Data were collected during 2014-2017 using questionnaires, for which elephant riders (mahouts) rated 28 behavioural adjectives of elephants. Repeated questionnaires were obtained for each elephant from several raters whenever possible, resulting in 690 ratings of 150 female and 107 male elephants. We started by performing a confirmatory factor analysis to compare the fit of our data to a previously published captive elephant personality structure. Owing to a poor fit of this model to our data, we proceeded by performing explanatory factor analysis to determine the personality structure in our study population. This model suggested that personality in these elephants was manifested as three factors that we labelled as Attentiveness, Sociability and Aggressiveness. This structure did not differ between the sexes. These results provide the basis for future research on the link between personality and reproductive success in this endangered species and more generally, help to resolve the selective pressures on personalities in long-lived, highly social species

    Preliminary evaluation of the CellFinder literature curation pipeline for gene expression in kidney cells and anatomical parts

    Biomedical literature curation is the process of automatically and/or manually deriving knowledge from scientific publications and recording it into specialized databases for structured delivery to users. It is a slow, error-prone, complex, costly and, yet, highly important task. Previous experiences have proven that text mining can assist in its many phases, especially, in triage of relevant documents and extraction of named entities and biological events. Here, we present the curation pipeline of the CellFinder database, a repository of cell research, which includes data derived from literature curation and microarrays to identify cell types, cell lines, organs and so forth, and especially patterns in gene expression. The curation pipeline is based on freely available tools in all text mining steps, as well as the manual validation of extracted data. Preliminary results are presented for a data set of 2376 full texts from which >4500 gene expression events in cell or anatomical part have been extracted. Validation of half of this data resulted in a precision of ~50% of the extracted data, which indicates that we are on the right track with our pipeline for the proposed task. However, evaluation of the methods shows that there is still room for improvement in the named-entity recognition and that a larger and more robust corpus is needed to achieve a better performance for event extraction.     CellFinder: a cell data repository

    CellFinder (http://www.cellfinder.org) is a comprehensive one-stop resource for molecular data characterizing mammalian cells in different tissues and in different development stages. It is built from carefully selected data sets stemming from other curated databases and the biomedical literature. To date, CellFinder describes 3394 cell types and 50 951 cell lines. The database currently contains 3055 microscopic and anatomical images, 205 whole-genome expression profiles of 194 cell/tissue types from RNA-seq and microarrays and 553 905 protein expressions for 535 cells/tissues. Text mining of a corpus of >2000 publications followed by manual curation confirmed expression information on ∼900 proteins and genes. CellFinder's data model is capable to seamlessly represent entities from single cells to the organ level, to incorporate mappings between homologous entities in different species and to describe processes of cell development and differentiation. Its ontological backbone currently consists of 204 741 ontology terms incorporated from 10 different ontologies unified under the novel CELDA ontology. CellFinder's web portal allows searching, browsing and comparing the stored data, interactive construction of developmental trees and navigating the partonomic hierarchy of cells and tissues through a unique body browser designed for life scientists and clinicians

    Angle resolved photoelectron spectroscopy of two-color XUV-NIR ionization with polarization control

    Electron emission caused by extreme ultraviolet (XUV) radiation in the presence of a strong near infrared (NIR) field leads to multiphoton interactions that depend on several parameters. Here, a comprehensive study of the influence of the angle between the polarization directions of the NIR and XUV fields on the two-color angle-resolved photoelectron spectra of He and Ne is presented. The resulting photoelectron angular distribution strongly depends on the orientation of the NIR polarization plane with respect to that of the XUV field. The prevailing influence of the intense NIR field over the angular emission characteristics for He(1s) and Ne(2p) ionization lines is shown. The underlying processes are modeled in the frame of the strong field approximation (SFA) which shows very consistent agreement with the experiment reaffirming the power of the SFA for multicolor-multiphoton ionization in this regime

    X-Ray Photoabsorption of Density-sensitive Metastable States in Ne VII, Fe XXII, and Fe XXIII

    Metastable states of ions can be sufficiently populated in absorbing and emitting astrophysical media, enabling spectroscopic plasma-density diagnostics. Long-lived states appear in many isoelectronic sequences with an even number of electrons, and can be fed at large rates by various photonic and electronic mechanisms. Here, we experimentally investigate beryllium-like and carbon-like ions of neon and iron that have been predicted to exhibit detectable features in astrophysical soft X-ray absorption spectra. An ion population generated and excited by electron impact is subjected to highly monochromatic X-rays from a synchrotron beamline, allowing us to identify Kα transitions from metastable states. We compare their energies and natural line widths with state-of-the-art theory and benchmark level population calculations at electron densities of 1010.5 cm-3
