    What contributes to depression in Parkinson's disease?

    Background: Depression is a common problem in patients with Parkinson's disease, but its mechanism is poorly understood. It is thought that neurochemical changes contribute to its occurrence, but it is unclear why some patients develop depression and others do not. Using a community-based sample of patients with Parkinson's disease, we investigated the contributions of impairment, disability and handicap to depression in Parkinson's disease. Methods: Ninety-seven patients seen in a population-based study on the prevalence of Parkinson's disease completed the Beck Depression Inventory (BDI). Clinical and historical information on symptoms and complications of Parkinson's disease were obtained from the patients by a neurologist. In addition, clinician and patient ratings of disability on the Schwab and England scale were obtained and a quality of life questionnaire was completed. Results: Moderate to severe depression (BDI [gt-or-equal, slanted] 18) was reported by 19·6% of the patients. Higher depression scores were associated with advancing disease severity, recent self-reported deterioration, higher akinesia scores, a mini-mental score of 50% of the variance of depression scores. Conclusions: Depression in patients with Parkinson's disease is associated with advancing disease severity, recent disease deterioration and occurrence of falls. Regression analysis suggests that depression in Parkinson's disease is more strongly influenced by the patients' perceptions of handicap than by actual disability. The treatment of depression should therefore be targeted independently of treatment of the motor symptoms of Parkinson's disease, and consider the patients' own perception of their disease

    An absolute reference frame for clumped isotope thermometry

    Analysis of multiply substituted isotopologues of molecules (‘clumped isotope geochemistry’) presents special challenges to both precision and accuracy. Previous discussions have focused on mass spectrometric precision for these rare species and intralaboratory reference frames. This discipline has spread, demanding interlaboratory standardization. We present a four-laboratory study of the calibration of mass-47 anomalies (Δ_(47) values) in CO_2 (especially extracted from carbonate). We consider: instrument linearity, source fragmentation/recombination reactions (which vary between mass spectrometers and with time and instrument settings), and differences in methods, materials and conditions for sample preparation. We address these problems by developing a method for standardizing Δ_(47) measurements to an absolute reference frame based on theoretical predictions of the abundances of multiply-substituted isotopologues of gaseous CO_2 that has reached a thermodynamic equilibrium at a known temperature. By analyzing CO_2 gases that have been subjected to established laboratory procedures known to promote isotopic equilibration (i.e., heated gases and water-equilibrated CO_2), and by reference to the statistical thermodynamic predictions of equilibrium isotopic distributions, it is possible to construct an empirical transfer function that can then be applied to CO_2 samples with unknown Δ_(47) values. This reference frame may be unique in that it is based on thermodynamic equilibrium, rather than the isotopic composition of an arbitrary reference material. We describe the protocol necessary to construct such a reference frame, the method for converting gases with unknown clumped isotope compositions to this frame, and suggest a protocol for ensuring that reported Δ_(47) values can be compared among different laboratories, independent of laboratory-specific analytical or methodological artefacts. Application of this approach to measurements of CO_2 extracted from several carbonate reference materials results in interlaboratory agreement on their Δ_(47) values to within est. ±0.01 ‰, 1σ. Finally, we present a revised paleotemperature scale that applies when using the absolute reference frame described here, as opposed to the previous paleotemperature equation based on data from a single laboratory. More generally, this study presents a model for how interlaboratory standardization might be approached for other ‘clumped isotope’ measurements

    Zusammenfassung: Hintergrund: Schwerbrandverletzte (mehr als 20% verbrannter Körperoberfläche bei Erwachsenen) weisen in der ersten Phase (8-48h) einen durch das massive Kapillarleck bedingten Verbrennungsschock auf, der einer Infusionstherapie bedarf, um die Hämodynamik wieder herzustellen. Bis in die 80erJahre stellte eine unzureichende Flüssigkeitstherapie (Unterinfusion) die Haupttodesursache von Verbrennungspatienten dar. Seither ist die übermäßige Flüssigkeitstherapie (Überinfusion) zu einer beachtenswerten Quelle von Komplikationen geworden: abdominales Kompartmentsyndrom, Entlastungsschnitte (Escharotomie), Verschlechterung des Gasaustauschs, Verlängerung der künstlichen Beatmung und des Spitalaufenthalts. Die Überinfusion hat Ende der 90erJahre begonnen, wo innerhalb der ersten 24h Flüssigkeitsmengen zugeführt wurden, die weit über den 4ml/kg/%BSA ("burn surface area") der Parkland-Formel lagen.     Contribution of H. pylori and Smoking Trends to US Incidence of Intestinal-Type Noncardia Gastric Adenocarcinoma: A Microsimulation Model

    Background: Although gastric cancer has declined dramatically in the US, the disease remains the second leading cause of cancer mortality worldwide. A better understanding of reasons for the decline can provide important insights into effective preventive strategies. We sought to estimate the contribution of risk factor trends on past and future intestinal-type noncardia gastric adenocarcinoma (NCGA) incidence. Methods and Findings: We developed a population-based microsimulation model of intestinal-type NCGA and calibrated it to US epidemiologic data on precancerous lesions and cancer. The model explicitly incorporated the impact of Helicobacter pylori and smoking on disease natural history, for which birth cohort-specific trends were derived from the National Health and Nutrition Examination Survey (NHANES) and National Health Interview Survey (NHIS). Between 1978 and 2008, the model estimated that intestinal-type NCGA incidence declined 60% from 11.0 to 4.4 per 100,000 men, <3% discrepancy from national statistics. H. pylori and smoking trends combined accounted for 47% (range = 30%–58%) of the observed decline. With no tobacco control, incidence would have declined only 56%, suggesting that lower smoking initiation and higher cessation rates observed after the 1960s accelerated the relative decline in cancer incidence by 7% (range = 0%–21%). With continued risk factor trends, incidence is projected to decline an additional 47% between 2008 and 2040, the majority of which will be attributable to H. pylori and smoking (81%; range = 61%–100%). Limitations include assuming all other risk factors influenced gastric carcinogenesis as one factor and restricting the analysis to men. Conclusions: Trends in modifiable risk factors explain a significant proportion of the decline of intestinal-type NCGA incidence in the US, and are projected to continue. Although past tobacco control efforts have hastened the decline, full benefits will take decades to be realized, and further discouragement of smoking and reduction of H. pylori should be priorities for gastric cancer control efforts.     Finite size corrections to random Boolean networks

    Since their introduction, Boolean networks have been traditionally studied in view of their rich dynamical behavior under different update protocols and for their qualitative analogy with cell regulatory networks. More recently, tools borrowed from statistical physics of disordered systems and from computer science have provided a more complete characterization of their equilibrium behavior. However, the largest part of the results have been obtained in the thermodynamic limit, which is often far from being reached when dealing with realistic instances of the problem. The numerical analysis presented here aims at comparing - for a specific family of models - the outcomes given by the heuristic belief propagation algorithm with those given by exhaustive enumeration. In the second part of the paper some analytical considerations on the validity of the annealed approximation are discussed.Comment: Minor correction

    Repositioning the Catalytic Triad Aspartic Acid of Haloalkane Dehalogenase: Effects on Stability, Kinetics, and Structure

    Haloalkane dehalogenase (DhlA) catalyzes the hydrolysis of haloalkanes via an alkyl-enzyme intermediate. The covalent intermediate, which is formed by nucleophilic substitution with Asp124, is hydrolyzed by a water molecule that is activated by His289. The role of Asp260, which is the third member of the catalytic triad, was studied by site-directed mutagenesis. Mutation of Asp260 to asparagine resulted in a catalytically inactive D260N mutant, which demonstrates that the triad acid Asp260 is essential for dehalogenase activity. Furthermore, Asp260 has an important structural role, since the D260N enzyme accumulated mainly in inclusion bodies during expression, and neither substrate nor product could bind in the active-site cavity. Activity for brominated substrates was restored to D260N by replacing Asn148 with an aspartic or glutamic acid. Both double mutants D260N+N148D and D260N+N148E had a 10-fold reduced kcat and 40-fold higher Km values for 1,2-dibromoethane compared to the wild-type enzyme. Pre-steady-state kinetic analysis of the D260N+N148E double mutant showed that the decrease in kcat was mainly caused by a 220-fold reduction of the rate of carbon-bromine bond cleavage and a 10-fold decrease in the rate of hydrolysis of the alkyl-enzyme intermediate. On the other hand, bromide was released 12-fold faster and via a different pathway than in the wild-type enzyme. Molecular modeling of the mutant showed that Glu148 indeed could take over the interaction with His289 and that there was a change in charge distribution in the tunnel region that connects the active site with the solvent. On the basis of primary structure similarity between DhlA and other α/β-hydrolase fold dehalogenases, we propose that a conserved acidic residue at the equivalent position of Asn148 in DhlA is the third catalytic triad residue in the latter enzymes.

    Assessing cognitive dysfunction in Parkinson's disease: An online tool to detect visuo-perceptual deficits.

    BackgroundPeople with Parkinson's disease (PD) who develop visuo-perceptual deficits are at higher risk of dementia, but we lack tests that detect subtle visuo-perceptual deficits and can be performed by untrained personnel. Hallucinations are associated with cognitive impairment and typically involve perception of complex objects. Changes in object perception may therefore be a sensitive marker of visuo-perceptual deficits in PD.ObjectiveWe developed an online platform to test visuo-perceptual function. We hypothesised that (1) visuo-perceptual deficits in PD could be detected using online tests, (2) object perception would be preferentially affected, and (3) these deficits would be caused by changes in perception rather than response bias.MethodsWe assessed 91 people with PD and 275 controls. Performance was compared using classical frequentist statistics. We then fitted a hierarchical Bayesian signal detection theory model to a subset of tasks.ResultsPeople with PD were worse than controls at object recognition, showing no deficits in other visuo-perceptual tests. Specifically, they were worse at identifying skewed images (P &lt; .0001); at detecting hidden objects (P = .0039); at identifying objects in peripheral vision (P &lt; .0001); and at detecting biological motion (P = .0065). In contrast, people with PD were not worse at mental rotation or subjective size perception. Using signal detection modelling, we found this effect was driven by change in perceptual sensitivity rather than response bias.ConclusionsOnline tests can detect visuo-perceptual deficits in people with PD, with object recognition particularly affected. Ultimately, visuo-perceptual tests may be developed to identify at-risk patients for clinical trials to slow PD dementia. © 2018 The Authors.     A dynamic explanation for the origin of the western Mediterranean organic-rich layers

    The eastern Mediterranean sapropels are among the most intensively investigated phenomena in the paleoceanographic record, but relatively little has been written regarding the origin of the equivalent of the sapropels in the western Mediterranean, the organic-rich layers (ORLs). ORLs are recognized as sediment layers containing enhanced total organic carbon that extend throughout the deep basins of the western Mediterranean and are associated with enhanced total barium concentration and a reduced diversity (dysoxic but not anoxic) benthic foraminiferal assemblage. Consequently, it has been suggested that ORLs represent periods of enhanced productivity coupled with reduced deep ventilation, presumably related to increased continental runoff, in close analogy to the sapropels. We demonstrate that despite their superficial similarity, the timing of the deposition of the most recent ORL in the Alboran Sea is different than that of the approximately coincident sapropel, indicating that there are important differences between their modes of formation. We go on to demonstrate, through physical arguments, that a likely explanation for the origin of the Alboran ORLs lies in the response of the western Mediterranean basin to a strong reduction in surface water density and a shoaling of the interface between intermediate and deep water during the deglacial period. Furthermore, we provide evidence that deep convection had already slowed by the time of Heinrich Event 1 and explore this event as a potential agent for preconditioning deep convection collapse. Important differences between Heinrich-like and deglacial-like influences are highlighted, giving new insights into the response of the western Mediterranean system to external forcing

    Abiotic formation of O2 and O3 in high-CO2 terrestrial atmospheres

    Previous research has indicated that high amounts of ozone (O3) and oxygen (O2) may be produced abiotically in atmospheres with high concentrations of CO2. The abiotic production of these two gases, which are also characteristic of photosynthetic life processes, could pose a potential "false-positive" for remote-sensing detection of life on planets around other stars.We show here that such false positives are unlikely on any planet that possesses abundant liquid water, as rainout of oxidized species onto a reduced planetary surface should ensure that atmospheric H2 concentrations remain relatively high, and that O2 and O3 remain low. Our aim is to determine the amount of O3 and O2 formed in a high CO2 atmosphere for a habitable planet without life. We use a photochemical model that considers hydrogen (H2) escape and a detailed hydrogen balance to calculate the O2 and O3 formed on planets with 0.2 of CO2 around the Sun, and 0.02, 0.2 and 2 bars of CO2 around a young Sun-like star with higher UV radiation. The concentrations obtained by the photochemical model were used as input in a radiative transfer model that calculated the spectra of the modeled planets. The O3 and O2 concentrations in the simulated planets are extremely small, and unlikely to produce a detectable signature in the spectra of those planets. We conclude that with a balanced hydrogen budget, and for planets with an active hydrological cycle, abiotic formation of O2 and O3 is unlikely to create a possible false positive for life detection in either the visible/near-infrared or mid-infrared wavelength regimes.Comment: 27 pages, 15 figures, Astronomy & Astrophysics accepte
