    The Structure of a Receptor with Two Associating Transmembrane Domains on the Cell Surface: Integrin α\u3csub\u3eIIb\u3c/sub\u3eβ\u3csub\u3e3\u3c/sub\u3e

    Structures of intact receptors with single-pass transmembrane domains are essential to understand how extracellular and cytoplasmic domains regulate association and signaling through transmembrane domains. A chemical and computational method to determine structures of the membrane regions of such receptors on the cell surface is developed here and validated with glycophorin A. An integrin heterodimer structure reveals association over most of the lengths of the α and β transmembrane domains and shows that the principles governing association of hetero and homo transmembrane dimers differ. A turn at the Gly of the juxtamembrane GFFKR motif caps the α TM helix and brings the two Phe of GFFKR into the α/β interface. A juxtamembrane Lys residue in β also has an important role in the interface. The structure shows how transmembrane association/dissociation regulates integrin signaling. A joint ectodomain and membrane structure shows that substantial flexibility between the extracellular and TM domains is compatible with TM signaling. © 2009 Elsevier Inc. All rights reserved

    Dye exclusion microfluidic microscopy

    We present an optical system to measure height maps of nonadherent cells as they flow through a microfluidic channel. The cells are suspended in an index-matching absorbing buffer, where cell height is evaluated by measuring the difference in absorption between the cell and the background. Unlike interferometric microscopes, the measured cell height is nearly independent of the cell's optical properties. The height maps are captured using a single exposure of a color camera, and consequently the system is capable of high-throughput characterization of large collections of cells. Using this system, we have measured more than 1600 height maps and volumes of three different leukemia cell lines

    Negative-Index Metamaterials: Second-Harmonic Generation, Manley-Rowe Relations and Parametric Amplification

    Second harmonic generation and optical parametric amplification in negative-index metamaterials (NIMs) are studied. The opposite directions of the wave vector and the Poynting vector in NIMs results in a "backward" phase-matching condition, causing significant changes in the Manley-Rowe relations and spatial distributions of the coupled field intensities. It is shown that absorption in NIMs can be compensated by backward optical parametric amplification. The possibility of distributed-feedback parametric oscillation with no cavity has been demonstrated. The feasibility of the generation of entangled pairs of left- and right-handed counter-propagating photons is discussed.Comment: 7 pages, 6 figure

    Negative Refractive Index in Optics of Metal-Dielectric Composites

    Specially designed metal-dielectric composites can have a negative refractive index in the optical range. Specifically, it is shown that arrays of single and paired nanorods can provide such negative refraction. For pairs of metal rods, a negative refractive index has been observed at 1.5 micrometer. The inverted structure of paired voids in metal films may also exhibit a negative refractive index. A similar effect can be accomplished with metal strips in which the refractive index can reach -2. The refractive index retrieval procedure and the critical role of light phases in determining the refractive index is discussed.Comment: 39 pages, 17 figures, 24 equation

    Facilitating return to work through early specialist health-based interventions (FRESH): protocol for a feasibility randomised controlled trial

    Background Over one million people sustain traumatic brain injury each year in the UK and more than 10 % of these are moderate or severe injuries, resulting in cognitive and psychological problems that affect the ability to work. Returning to work is a primary rehabilitation goal but fewer than half of traumatic brain injury survivors achieve this. Work is a recognised health service outcome, yet UK service provision varies widely and there is little robust evidence to inform rehabilitation practice. A single-centre cohort comparison suggested better work outcomes may be achieved through early occupational therapy targeted at job retention. This study aims to determine whether this intervention can be delivered in three new trauma centres and to conduct a feasibility, randomised controlled trial to determine whether its effects and cost effectiveness can be measured to inform a definitive trial. Methods/design Mixed methods study, including feasibility randomised controlled trial, embedded qualitative studies and feasibility economic evaluation will recruit 102 people with traumatic brain injury and their nominated carers from three English UK National Health Service (NHS) trauma centres. Participants will be randomised to receive either usual NHS rehabilitation or usual rehabilitation plus early specialist traumatic brain injury vocational rehabilitation delivered by an occupational therapist. The primary objective is to assess the feasibility of conducting a definitive trial; secondary objectives include measurement of protocol integrity (inclusion/exclusion criteria, intervention adherence, reasons for non-adherence) recruitment rate, the proportion of eligible patients recruited, reasons for non-recruitment, spectrum of TBI severity, proportion of and reasons for loss to follow-up, completeness of data collection, gains in face-to-face Vs postal data collection and the most appropriate methods of measuring primary outcomes (return to work, retention) to determine the sample size for a larger trial. Discussion To our knowledge, this is the first feasibility randomised controlled trial of a vocational rehabilitation health intervention specific to traumatic brain injury. The results will inform the design of a definitive trial
