    Characterization of type II toxin anti-toxin systems in Aggregatibacter actinomycetemcomitans.

    Microbes express many protective mechanisms in response to environmental stress. Toxin/anti-toxin systems encode a biologically active toxin and a labile anti-toxin that inhibits the toxin’s activity. These systems are known to contribute to persister cell and biofilm formation. A. actinomycetemcomitans thrives in the complex oral microbial community and is subjected to continual environmental flux. Little is known regarding the presence and function of TA systems in this organism or their contribution survival in the oral environment. Using BLAST searches and other informatics tools, we identified 11 intact TA systems that are conserved across all seven serotypes of A. actinomycetemcomitans and represent the RelBE, MazEF and HipAB families of TA systems. The A. actinomycetemcomitans TA systems identified selectively responded to various environmental conditions that exist in the oral cavity. Transcription of two putative RelBE-like TA systems, D11S_1194-1195 and D11S_1718-1719, were induced in response to low pH, and were selected for further study. Deletion of D11S_1718-1719 significantly reduced metabolic activity of stationary phase A. actinomycetemcomitans cells during prolonged exposure to acidic conditions. The mutant also exhibited reduced biofilm biomass when cultured under acidic conditions. The D11S_1194 and D11S_1718 toxins inhibited in vitro translation of dihydrofolate reductase (DHFR), and degraded ribosome-associated, but not free mRNA. In contrast, the corresponding antitoxins or equimolar mixtures of toxin and antitoxin had no effect on DHFR production or RNA degradation. Preliminary results comparing the proteomes of acid stressed mutants to the acid stressed wild-type suggest that metabolism proteins are the most affected by these two TA systems. Among these, proteins involved in nucleotide metabolism are largely over-represented in the mutants. Other identified proteins are directly involved in quorum-sensing, iron transport and virulence (e.g. leukotoxin). Results of these studies indicate that the anti-toxin proteins inhibit the activity of the corresponding toxins and suggest that D11S_1194-1195 and D11S_1718-1719 are RelBE-like type II TA systems that are activated under acidic conditions. The toxins of both systems may function to cleave ribosome-associated mRNA to inhibit translation in A. actinomycetemcomitans

    Multienvironment Quantitative Trait Loci Analysis for Photosynthate Acquisition, Accumulation, and Remobilization Traits in Common Bean Under Drought Stress

    Many of the world’s common bean (Phaseolus vulgaris L.) growing regions are prone to either intermittent or terminal drought stress, making drought the primary cause of yield loss under farmers’ field conditions. Improved photosynthate acquisition, accumulation, and then remobilization have been observed as important mechanisms for adaptation to drought stress. The objective of this study was to tag quantitative trait loci (QTL) for photosynthate acquisition, accumulation, and remobilization to grain by using a recombinant inbred line population developed from the Mesoamerican intragenepool cross of drought-susceptible DOR364 and drought-tolerant BAT477 grown under eight environments differing in drought stress across two continents: Africa and South America. The recombinant inbred line population expressed quantitative variation and transgressive segregation for 11 traits associated with drought tolerance. QTL were detected by both a mixed multienvironment model and by composite interval mapping for each environment using a linkage map constructed with 165 genetic markers that covered 11 linkage groups of the common bean genome. In the multienvironment, mixed model, nine QTL were detected for 10 drought stress tolerance mechanism traits found on six of the 11 linkage groups. Significant QTL × environment interaction was observed for six of the nine QTL. QTL × environment interaction was of the cross-over type for three of the six significant QTL with contrasting effect of the parental alleles across different environments. In the composite interval mapping, we found 69 QTL in total. The majority of these were found for Palmira (47) or Awassa (18), with fewer in Malawi (4). Phenotypic variation explained by QTL in single environments ranged up to 37%, and the most consistent QTL were for Soil Plant Analysis Development (SPAD) leaf chlorophyll reading and pod partitioning traits. QTL alignment between the two detection methods showed that yield QTL on b08 and stem carbohydrate QTL on b05 were most consistent between the multilocation model and the single environment detection. Our results indicate the relevance of QTL detection in the sites in which bean breeding will be undertaken and the importance of photosynthate accumulation as a trait for common bean drought tolerance

    Diversity and equality in honours and awards programs – steps towards a fair representation of membership

    Honours and awards bestowed by professional societies recognize and reward members who have advanced the goals and values of that society. All too often, however, awards reflect a small network of people who know about the awards and participate in the process. This network works wonderfully for the people lucky enough to be in it, but typically neglects the full range and breadth of scholarship and service within the society. We represent a combined 15C years’ experience on the honours’ committee for a large professional society (the American Geophysical Union) and here offer strategies to increase the representation of honourees.Women represented less than 20%of awardees when we first became committee members in 2008; women represented 50% of awardees in 2019. There is still much to do to ensure that members from other typically underrepresented groups (non-US members, members from underrepresented races/ethnicities) are truly represented and honoured for outstanding science and service. We recommend forming canvassing committees that will scour the literature, conferences, and membership lists for appropriate and otherwise overlooked nominees; providing implicit bias training to selection committees; and ensuring selection committees focus on the criteria for the award rather than non-pertinent, often personal, information, as well as additional strategies that allow us to recognize our worthy colleagues

    GPR Imaging of Prehistoric Animal Bone-beds

    This research investigates the detection capabilities of Ground-penetrating radar for imaging prehistoric animal bone-beds. The first step of this investigation was to determine the dielectric properties of modern animal bone as a proxy for applying non-invasive ground-penetrating radar (GPR) for detecting prehistoric animal remains. Over 90 thin section samples were cut from four different modern faunal skeleton remains: bison, cow, deer, and elk. One sample of prehistoric mammoth core was also analyzed. Sample dielectric properties (relative permittivity, loss factor, and loss-tangent values) were measured with an impedance analyzer over frequencies ranging from 10 MHz to 1 GHz. The results reveal statistically significant dielectric-property differences among different animal fauna, as well as variation as a function of frequency. The measured sample permittivity values were then compared to modeled sample permittivity values using common dielectric-mixing models. The dielectric mixing models were used to report out new reported values of dry bone mineral of 3-5 in the frequency range of 10 MHz to 1 GHz. The second half of this research collected controlled GPR experiments over a sandbox containing buried bison bone elements to evaluate GPR detection capabilities of buried animal bone. The results of the controlled GPR sandbox tests were then compared to numerical models in order to predict the ability of GPR to detect buried animal bone given a variety of different depositional factors, the size and orientation of the bone target and the degree of bone weathering. The radar profiles show that GPR is an effective method for imaging the horizontal and vertical extent of buried animal bone. However, increased bone weathering and increased bone dip were both found to affect GPR reflection signal strength. Finally, the controlled sandbox experiments were also utilized to investigate the impact of survey design for imaging buried animal bone. In particular, the effects of GPR antenna orientation relative to the survey line (broad-side mode versus end-fire mode) and polarization effects of the buried bone targets were investigated. The results reveal that animal bone does exhibit polarization effects. However, the polarization results are greatly affected by the irregular shape and size of the bone, which ultimately limits the potential usefulness of trying to utilize polarization data to determine the orientation of buried bone targets. In regard to antenna orientation, end-fire mode was found to have little difference in amplitude response as compared to the more commonly used broad-side mode and in fact sometimes outperformed the broad-side mode.     Counter-stereotypical pictures as a strategy for overcoming spontaneous gender stereotypes

    The present research investigated the use of counter-stereotypical pictures as a strategy for overcoming spontaneous gender stereotypes when certain social role nouns and professional terms are read. Across two experiments, participants completed a judgment task in which they were presented with word pairs comprised of a role noun with a stereotypical gender bias (e.g., beautician) and a kinship term with definitional gender (e.g., brother). Their task was to quickly decide whether or not both terms could refer to one person. In each experiment they completed two blocks of such judgment trials separated by a training session in which they were presented with pictures of people working in gender counter-stereotypical (Experiment 1) or gender stereotypical roles (Experiment 2). To ensure participants were focused on the pictures, they were also required to answer four questions on each one relating to the character’s leisure activities, earnings, job satisfaction, and personal life. Accuracy of judgments to stereotype incongruent pairings was found to improve significantly across blocks when participants were exposed to counter-stereotype images (9.87%) as opposed to stereotypical images (0.12%), while response times decreased significantly across blocks in both studies. It is concluded that exposure to counter-stereotypical pictures is a valuable strategy for overcoming spontaneous gender stereotype biases in the short term

    Detection methods for non-Gaussian gravitational wave stochastic backgrounds

    We address the issue of finding an optimal detection method for a discontinuous or intermittent gravitational wave stochastic background. Such a signal might sound something like popcorn popping. We derive an appropriate version of the maximum likelihood detection statistic, and compare its performance to that of the standard cross-correlation statistic both analytically and with Monte Carlo simulations. The maximum likelihood statistic performs better than the cross-correlation statistic when the background is sufficiently non-Gaussian. For both ground and space based detectors, this results in a gain factor, ranging roughly from 1 to 3, in the minimum gravitational-wave energy density necessary for detection, depending on the duty cycle of the background. Our analysis is exploratory, as we assume that the time structure of the events cannot be resolved, and we assume white, Gaussian noise in two collocated, aligned detectors. Before this detection method can be used in practice with real detector data, further work is required to generalize our analysis to accommodate separated, misaligned detectors with realistic, colored, non-Gaussian noise.Comment: 25 pages, 12 figures, submitted to physical review D, added revisions in response to reviewers comment

    The X-ray Properties of z>4 Quasars

    We report on a search for X-ray emission from quasars with redshifts greater than four using the ROSAT public database. Our search has doubled the number of z>4 quasars detected in X-rays from 6 to 12. Most of those known prior to this work were radio-loud and X-ray selected sources; our study increases the number of X-ray detected, optically selected z>4 quasars from one to seven. We present their basic X-ray properties and compare these to those of lower redshift quasars.     Patient-reported outcomes on empagliflozin treatment in glycogen storage disease type Ib:An international questionnaire study

    In patients with glycogen storage disease type Ib (GSD Ib), quality of life is severely hampered by neutropenia and neutropenia-associated symptoms. SGLT2 inhibitors are a new treatment option and have shown improved medical outcomes in more than 120 patients so far. The aim of this international questionnaire study was to assess patient-reported outcomes of this new treatment in GSD Ib patients. Patients and caregivers of pediatric patients were invited to complete a web-based questionnaire. This was designed to evaluate treatment effects of the SGLT2 inhibitor empagliflozin on clinical symptoms and important aspects of daily life including physical performance, sleep, social and work life, traveling, socioeconomic aspects, and quality of life. The questionnaire was completed by 73 respondents from 17 different countries. The mean duration of treatment was 15 months, the cumulative treatment time was 94.8 years. More than 80% of patients reported an improved quality of life. The number of hospitalizations was reduced (66% of patients), as well as the number of days absent from school or work. Granulocyte colony-stimulating factor (G-CSF) treatment could be stopped in 49% of patients and reduced in another 42%. Clear improvement of neutropenia and all neutropenia-associated symptoms was reported by the majority of patients. Additionally, patients or caregivers reported positive effects on appetite (63%), level of activity (75%), overall well-being (96%), and sleep (63%). Empagliflozin positively impacts many aspects of daily life including work and social life and thereby significantly improves quality of life of patients and caregivers.</p

    A Blind Search for Magnetospheric Emissions from Planetary Companions to Nearby Solar-type Stars

    This paper reports a blind search for magnetospheric emissions from planets around nearby stars. Young stars are likely to have much stronger stellar winds than the Sun, and because planetary magnetospheric emissions are powered by stellar winds, stronger stellar winds may enhance the radio luminosity of any orbiting planets. Using various stellar catalogs, we selected nearby stars (<~ 30 pc) with relatively young age estimates (< 3 Gyr). We constructed different samples from the stellar catalogs, finding between 100 and several hundred stars. We stacked images from the 74-MHz (4-m wavelength) VLA Low-frequency Sky Survey (VLSS), obtaining 3\sigma limits on planetary emission in the stacked images of between 10 and 33 mJy. These flux density limits correspond to average planetary luminosities less than 5--10 x 10^{23} erg/s. Using recent models for the scaling of stellar wind velocity, density, and magnetic field with stellar age, we estimate scaling factors for the strength of stellar winds, relative to the Sun, in our samples. The typical kinetic energy carried by the stellar winds in our samples is 15--50 times larger than that of the Sun, and the typical magnetic energy is 5--10 times larger. If we assume that every star is orbited by a Jupiter-like planet with a luminosity larger than that of the Jovian decametric radiation by the above factors, our limits on planetary luminosities from the stacking analysis are likely to be a factor of 10--100 above what would be required to detect the planets in a statistical sense. Similar statistical analyses with observations by future instruments, such as the Low Frequency Array (LOFAR) and the Long Wavelength Array (LWA), offer the promise of improvements by factors of 10--100.Comment: 11 pages; AASTeX; accepted for publication in A

    The arrival of New Labour into Government witnessed the prominent re-emergence of place onto the policy agenda. This heralded a range of area-based-initiatives designed to both tackle neighbourhood forms of deprivation and to re-establish a sense of identity and connection between individuals and their local community. In terms of place-making, effective and inclusive participation, representation and leadership were all identified as prerequisites for the creation of sustainable communities . But how important is local leadership and strategic vision within local public service organisations in achieving the desired place-making outcomes? This paper examines the extent to which local leadership and strategic vision represents a significant factor in promoting higher levels of satisfaction, belonging, cohesion and participation across single tier councils in England. The ensuing empirical evidence raises significant questions not only about the importance of local leadership in place-making, but also the environmental and organizational factors that shape local places
