    Crosstalk between H2A variant-specific modifications impacts vital cell functions

    Selection of C-terminal motifs participated in evolution of distinct histone H2A variants. Hybrid types of variants combining motifs from distinct H2A classes are extremely rare. This suggests that the proximity between the motif cases interferes with their function. We studied this question in flowering plants that evolved sporadically a hybrid H2A variant combining the SQ motif of H2A.X that participates in the DNA damage response with the KSPK motif of H2A.W that stabilizes heterochromatin. Our inventory of PTMs of H2A.W variants showed that in vivo the cell cycle-dependent kinase CDKA phosphorylates the KSPK motif of H2A. W but only in absence of an SQ motif. Phosphomimicry of KSPK prevented DNA damage response by the SQ motif of the hybrid H2A.W/X variant. In a synthetic yeast expressing the hybrid H2A.W/X variant, phosphorylation of KSPK prevented binding of the BRCT-domain protein Mdb1 to phosphorylated SQ and impaired response to DNA damage. Our findings illustrate that PTMs mediate interference between the function of H2A variant specific C-terminal motifs.     POOL development status and production experience

    The pool of persistent objects for LHC (POOL) project, part of the large Hadron collider (LHC) computing grid (LCG), is now entering its third year of active development. POOL provides the baseline persistency framework for three LHC experiments. It is based on a strict component model, insulating experiment software from a variety of storage technologies. This paper gives a brief overview of the POOL architecture, its main design principles and the experience gained with integration into LHC experiment frameworks. It also presents recent developments in the POOL works areas of relational database abstraction and object storage into relational database management systems (RDBMS) systems

    The genetic and epigenetic landscape of the Arabidopsis centromeres.

    Centromeres attach chromosomes to spindle microtubules during cell division and, despite this conserved role, show paradoxically rapid evolution and are typified by complex repeats. We used longread sequencing to generate the Col-CEN Arabidopsis thaliana genome assembly that resolves all five centromeres. The centromeres consist of megabase-scale tandemly repeated satellite arrays, which support CENH3 occupancy and are densely DNA methylated, with satellite variants private to each chromosome. CENH3 preferentially occupies satellites that show least divergence and occur in higherorder repeats. The centromeres are invaded by ATHILA retrotransposons, which disrupt genetic and epigenetic organization. Centromeric crossover recombination is suppressed, yet low levels of meiotic DSBs occur that are regulated by DNA methylation. We propose that Arabidopsis centromeres are evolving via cycles of satellite homogenization and retrotransposon-driven diversification.BBSRC grants BB/S006842/1, BB/S020012/1 and BB/V003984/1

    A Purity Monitoring System for the H1 Liquid Argon Calorimeter

    The ionization probes used for monitoring the liquid argon purity in the H1 calorimeter are described and results of their operation in tests at CERN and during the period 1992 to the end of 1998 at HERA are given. The high sensitivity of the charge measurements leads to refined charge collection models, and to the observation of a variation of the ionization yield of our electron sources with temperature.Comment: 26 pages, 14 figure
