    Developments and challenges in the European pork sector

    This paper aims to give insight into the structure of and variety in the European pork system and suggests topics for further research of the European pork sector. It provides an overview of the different types of pork chains, their quality systems. governance structures and supporting technology. The paper further describes the concentration and up-scaling found in all links of the chain in most of the countries investigated. Moreover, a development towards chain-wide quality management systems and new collaborative structures in the various chains can be recognised. However, there is also a trend towards development of pork chains that aim at high-quality production for regional and niche markets, in particular in Southern European countries. Although the paper tries to give a European picture, it focuses specifically on five countries: the Netherlands, Germany, Greece, Spain and Hungary. The paper concludes with major bottlenecks and opportunities for European pork chains, after which new research issues are raised

    New materials for strontium removal from nuclear waste streams

    The primary aim of this project is to investigate potential new materials for application in ion exchange processes to remove 9^90^0Sr from nuclear waste streams. This work can be broadly split in to two sections, work on attempts to synthesise new materials and work to investigate ion exchange properties of two recently prepared materials AV-7, a synthetic analogue of tin-kostylvite and AV-3, a synthetic analogue of petarasite. Synthesis on new materials was focused on metal silicate materials, in particular titanium, zirconium and tin silicates containing exchangeable group I and II cations. These synthesis attempts initially were focus on targeted mineral phases such as noonkanbahite, BaKNaTi2_2(Si4_4O1_12_2)O2_2, followed by a series of brief surveys examining the effects of various changes to precursor gels such as concentration of bases such as NaOH, metal to silicon ratios and the presence of mineralizing agents such as sodium fluoride. The synthesis of two synthetic mineral phases potentially interesting for ion exchange is also reported here, titanite and fresnoite. Ion exchange studies focused mainly on AV-7 and AV-3 but also included well known ion exchange materials for comparison such as clinoptilolite and Nb-doped crystalline silicotitanate and brief investigations in to the ion exchange of fresnoite and titanite. Ion exchange was followed using X-Ray fluorescence, ion chromatography and radioactive 8^85^5Sr exchanges measured using scintillation counters

    Nicotinamide enhances myelin production after demyelination through reduction of astrogliosis and microgliosis

    Caloric restriction is the chronic reduction of total caloric intake without malnutrition and has attracted a lot of attention as, among multiple other effects, it attenuates demyelination and stimulates remyelination. In this study we have evaluated the effect of nicotinamide (NAM), a well-known caloric restriction mimetic, on myelin production upon demyelinating conditions. NAM is the derivative of nicotinic acid (vitamin B3) and a precursor of nicotinamide adenine dinucleotide (NAD+), a ubiquitous metabolic cofactor. Here, we use cortical slices ex vivo subjected to demyelination or cultured upon normal conditions, a lysolecithin (LPC)-induced focal demyelination mouse model as well as primary glial cultures. Our data show that NAM enhances both myelination and remyelination ex vivo, while it also induces myelin production after LPC-induced focal demyelination ex vivo and in vivo. The increased myelin production is accompanied by reduction in both astrogliosis and microgliosis in vivo. There is no direct effect of NAM on the oligodendrocyte lineage, as no differences are observed in oligodendrocyte precursor cell proliferation or differentiation or in the number of mature oligodendrocytes. On the other hand, NAM affects both microglia and astrocytes as it decreases the population of M1-activated microglia, while reducing the pro-inflammatory phenotype of astrocytes as assayed by the reduction of TNF-α. Overall, we show that the increased myelin production that follows NAM treatment in vivo is accompanied by a decrease in both astrocyte and microglia accumulation at the lesion site. Our data indicate that NAM influences astrocytes and microglia directly, in favor of the remyelination process by promoting a less inflammatory environment

    Agri-food business: Global challenges â Innovative solutions

    The rise of a western-style middle class in many successful emerging economies like China currently is inducing deep structural changes on agricultural world markets and within the global agri-food business. As a result of both higher incomes and concerns over product safety and quality the global demand for high-quality and safe food products is increasing significantly. In order to meet the new required quality, globally minimum quality standards are rising and private standards emerging. All over the world these developments cause adjustments at the enterprise, chain and market levels. At the same time, the tremendously increasing demand for renewable energy has led to the emergence of a highly promising market for biomass production. This has far-reaching consequences for resource allocation in the agri-food business, for the environment, for the poor in developing countries and for agricultural policy reforms. The challenges increase with ongoing liberalisation, globalisation and standardisation, all of which change trade patterns for agricultural and food commodities, and influence production costs and commodity prices. The objective of the IAMO Forum is to show opportunities as well as risks for all participants of the food economy in the ongoing globalisation process: for small peasants in developing countries, farmers in Europe and globally active food enterprises and retailers. The success of enterprises depends on the ability to find innovative solutions with regard to the organisation of enterprises, chains, and markets, as well as future policy design. Concerning bio-energy strategies has to be identified to combat global warming most efficiently and concurrently attenuate the competition between "tank and table" on farmland.     Ablation of CNTN2+Pyramidal Neurons During Development Results in Defects in Neocortical Size and Axonal Tract Formation

    Corticothalamic axons express Contactin-2 (CNTN2/TAG-1), a neuronal recognition molecule of the immunoglobulin superfamily involved in neurogenesis, neurite outgrowth, and fasciculation. TAG-1, which is expressed transiently by cortical pyramidal neurons during embryonic development, has been shown to be fundamental for axonal recognition, cellular migration, and neuronal proliferation in the developing cortex. Although Tag-1(-/-) mice do not exhibit any obvious defects in the corticofugal system, the role of TAG-1+ neurons during the development of the cortex remains elusive. We have generated a mouse model expressing EGFP under the Tag-1 promoter and encompassing the coding sequence of Diptheria Toxin subunit A (DTA) under quiescence with no effect on the expression of endogenous Tag-1. We show that while the line recapitulates the expression pattern of the molecule, it highlights an extended expression in the forebrain, including multiple axonal tracts and neuronal populations, both spatially and temporally. Crossing these mice to the Emx1-Cre strain, we ablated the vast majority of TAG-1+ cortical neurons. Among the observed defects were a significantly smaller cortex, a reduction of corticothalamic axons as well as callosal and commissural defects. Such defects are common in neurodevelopmental disorders, thus this mouse could serve as a useful model to study physiological and pathophysiological cortical development

    A Forward Genetic Screen in Mice Identifies Mutants with Abnormal Cortical Patterning

    Formation of a 6-layered cortical plate and axon tract patterning are key features of cerebral cortex development. Abnormalities of these processes may be the underlying cause for a range of functional disabilities seen in human neurodevelopmental disorders. To identify mouse mutants with defects in cortical lamination or corticofugal axon guidance, N-ethyl-N-nitrosourea (ENU) mutagenesis was performed using mice expressing LacZ reporter genes in layers II/III and V of the cortex (Rgs4-lacZ) or in corticofugal axons (TAG1-tau-lacZ). Four lines with abnormal cortical lamination have been identified. One of these was a splice site mutation in reelin (Reln) that results in a premature stop codon and the truncation of the C-terminal region (CTR) domain of reelin. Interestingly, this novel allele of Reln did not display cerebellar malformation or ataxia, and this is the first report of a Reln mutant without a cerebellar defect. Four lines with abnormal cortical axon development were also identified, one of which was found by whole-genome resequencing to carry a mutation in Lrp2. These findings demonstrated that the application of ENU mutagenesis to mice carrying transgenic reporters marking cortical anatomy is a sensitive and specific method to identify mutations that disrupt patterning of the developing brain
