Optimization of rRNA Depletion by Testing Various Kits
RNA sequencing (RNAseq) can be used to compare gene expression between different groups, identify mutations, and confirm gene knockouts. In RNAseq, RNA is extracted and built into a sequenceable library to analyze the transcriptome. Ribosomal RNA (rRNA) makes up roughly 90% of the total RNA extracted and does not provide useful transcriptome information, thus it must be removed in order to perform transcriptional analysis. At MBP Titan, the protocol used KAPA’s RiboErase Depletion kit using custom methanotroph oligonucleotides to specifically bind to rRNA for depletion. On average, the amount of rRNA remaining in the libraries ranged from 10-30%. Several depletion methods and kits were tested to develop a more effective and robust depletion method that can be high-throughput and automated while keeping library preparation costs low and cut down on hands-on time. The KAPA RiboErase kit skipping intermediate purification and the Lucigen RNase H method resulted in an average of 1-5% remaining rRNA
Reading and school libraries today
RESUMEN: Con este trabajo se pretende profundizar en la importancia que tiene la lectura y la necesidad de fomentarla en Educación Primaria, a través del contexto familiar y escolar del alumnado. Los estudiantes no nacen lectores, por ello, es fundamental desarrollar su interés por los libros y generar experiencias lectoras satisfactorias y significativas para ellos. Asimismo, se habla de la relación entre la lectura y las nuevas tecnologías que amplían las posibilidades lectoras ante los nuevos formatos digitales que se adaptan a la realidad tecnológica actual. Además, se resalta la relevancia que tienen las bibliotecas escolares en el fomento de la lectura. Para ello, deben estar incluidas en los proyectos del centro, trabajar de manera coordinada con los docentes para fomentar su uso (no solo en la asignatura de Lengua Castellana y Literatura) y ofrecer formatos y contenidos variados para la comunidad educativa, teniendo en cuenta los gustos, demandas, formación y preferencias. Para finalizar, se desarrollan una serie de propuestas (club de lectura, booktrailer, booktube y podcast) para fomentar el hábito lector entre los más jóvenes, con la participación de toda la comunidad educativa y la presencia de las nuevas tecnologías.ABSTRACT: The aim of this work is to look into the importance of reading and the necessity of improving it in Primary Education, through the children’s family and school environment. Students are not born readers, for this reason, it is fundamental to develop their interest for books and create satisfying and significant Reading experiences for them. Moreover, I have talked about the relationship between reading and new technologies that broaden the reading possibilities due to the new digital formats that adapt to the current technological reality. Furthermore, it is highlighted the relevance school libraries have in the Reading encouragement. Therefore, they should be included in the school projects, teachers ought to work in a coordinated way in order to promote their use (not only in the Spanish Language and Literature subject) and they must offer diverse formats and contents to the educational community, taking into account their likes, demands, education and preferences. Last but not least, several proposals have been developed (reading club, booktrailer, booktube and podcast) in order to promote the reading habit among the youths, with the collaboration of the educational community and the presence of new technologies.Grado en Magisterio en Educación Primari
Case Histories of Widespread Liquefaction and Lateral Spread Induced by the 2007 Pisco, Peru Earthquake
Case histories of widespread liquefaction and lateral spread induced by the Mw 8.0, 2007 Pisco, Peru earthquake and observed during a post-earthquake GEER reconnaissance are presented. A long duration of the earthquake over 200 seconds and two phases of strong ground motion induced widespread liquefaction and lateral spread of sand coastal deposits and road embankments over a total length of approximately 100 km of coastal region. Six case histories of liquefaction are presented and discussed including a massive lateral spread of a marine terrace believed to be as large or even larger than that reported along the Shinano River during the 1964 Niigata earthquake in Japan
Attenuation of Peak Ground Acceleration with Distance of the June 15, 1999, Tehuacán, México, Earthquake
A seismic event of moment magnitude 7.0 struck the central region of Mexico on June 15, 1999 at 15:41 hrs (local time) between the states of Puebla and Oaxaca. The epicenter of the earthquake was located approximately 20 km to the south-southeast of the town of Tehuacán, Puebla and 55 km to the northeast of Huajuapan de León, Oaxaca. A reconnaissance team consisting of a group of researchers from the University of California at Berkeley (UCB) and the Autonomous University of Mexico (UNAM) visited the area and gathered some preliminary information on the geotechnical aspects of this earthquake. This paper briefly presents some key geotechnical observations and then focuses on the attenuation of peak ground acceleration with distance. A total of 29 strong-motion recordings over a variety of geologic site conditions were compiled and used to develop the observed attenuation of the peak ground acceleration (PGA) with distance for the main event. The results obtained were compared with estimations of PGA using North American attenuation relationships
Estrategia para la retención en carrera de ingeniería: Ingeniería Electrónica de la Universidad Nacional de Tucumán
This paper aims at presenting a retention strategy used by the Electronic Engineer career from the Universidad Nacional de Tucumán in order to motivate, integrate and keep their students. The procedure used is the implementation of practical workshops aimed at students of all years of the career who have as premise “to learn by doing”. The workshops seek to bring the student in all aspects of the exercise of the profession. They are taught one at a time in sequence and are assessed by completing a simple project. The results of the procedure used were measured by the number of enrolled students and surveys verifying satisfaction.Este trabajo tiene como objetivo presentar una estrategia de retención empleada por la carrera de Ingeniería Electrónica de la Universidad Nacional de Tucumán como una herramienta para motivar, integrar y retener a sus estudiantes. Se emplea como procedimiento la implementación de talleres prácticos orientados a estudiantes de todos los años de la carrera que tienen como premisa “aprender haciendo”. Buscan acercar al estudiante en aspectos propios del ejercicio de la profesión. Se imparten de uno en uno en forma secuencial y se evalúan mediante la realización de un proyecto simple. Los resultados de la metodología empleada se midieron mediante el número de estudiantes inscriptos y encuestas comprobando la satisfacción
Aspectos geotécnicos del sismo de Pisco, Perú del 15 de agosto del 2007
Este artículo presenta un resumen de las observaciones realizadas durante un reconocimiento de los efectos geotécnicos del sismo de Pisco ocurrido el 15 de agosto del 2007 en la zona central de la costa del Perú. Este sismo de magnitud (Mw) 8.0 causó daños importantes a obras de ingeniería civil en una amplia zona que incluye las ciudades de Ica, Pisco, Cañete y Chincha. Desde una perspectiva geotécnica, los aspectos más relevantes de este sismo son la gran extensión de licuación de suelo observada en la zona mesosísmica y el considerable daño que causó en las zonas urbanas y en la infraestructura de transporte. En particular se destaca la documentación de un deslizamiento lateral que involucró un área de al menos 3 km2 de terreno y asentamientos de hasta 0.9 m observados en estructuras livianas. El sismo también causó varios deslizamientos de talud que resultaron en bloqueos de carreteras. También se presenta un análisis preliminar de los registros de aceleraciones obtenidos durante el sismo
