    Threshold temperatures for subjective heat stress in urban apartments — analysing nocturnal bedroom temperatures during a heat wave in Germany

    As climate change progresses, it is causing more frequent and severe heat waves, resulting in higher indoor temperatures. Various temperature thresholds for indicating indoor overheating have been proposed in different contexts, extending from reduced comfort in buildings to subjective heat stress and onset of first or serious health problems. This study reviews these thresholds and identifies threshold values for subjective heat stress of occupants in the city of Augsburg, Germany, distinguishing between vulnerable and non-vulnerable households. Survey data from 427 private households are analysed using unpaired analysis of variances (ANOVA), t-tests and regression analysis to identify factors related to subjective heat stress at home during night-time. The findings imply that health implications during heat waves, age, local climate zones favouring the urban heat island effect and higher indoor temperature represent significant factors for subjective heat stress. A significant difference in subjective heat stress among different groups related to temperature could be identified for thresholds of 24.8 °C (people living alone) and 26.7 °C (people with chronic disease). As WHO threshold for health risk from overheating is 24 °C, people are apparently at heat-related risk without feeling that they are at risk, especially when they have chronic diseases; thus they may not see the urgency of taking adaptation measures

    Heat adaptation measures in private households: an application and adaptation of the protective action decision model

    Extreme heatwaves will occur more frequently and with higher intensity in future. Their consequences for human health can be fatal if adaptation measures will not be taken. This study analyses factors related to heat adaptation measures in private households in Germany. During the summer months of 2019, indoor temperatures were measured in over 500 private households in the City of Augsburg, Germany, accompanied by a survey to find out about heat perception and adaptation measures. Hypotheses deducted from the Protective Action Decision Model were tested using one-way ANOVAs, regression analysis and in the end a multiple hierarchical regression model. The results of the hypotheses tested imply an influence of knowledge and heat risk perception of heat adaptation behaviour and an influence of age on heat risk perception. The results of the regression model show an influence of the efficacy-related attribute, of age, indoor temperature, subjective heat stress and health implications to heat adaptation behaviour. In the end, this study proposes adjustments to the PADM according to the results of the hierarchical regression analysis

    Make your brand heard: A qualitative study on the use of corporate podcasts as a branding tool

    As an increasingly popular on-demand medium, podcasts have become more professional and commercialized in recent times. This is also evident from a look at corporate communications, where many companies add podcasts into their communication mix. To align these corporate podcasts with other communication channels, the concept of corporate branding can be helpful. This approach has not been considered by empirical studies so far. Our qualitative interview study wants to investigate the role of corporate podcasts in corporate branding and examines their integration into the communication strategy of nationally and internationally operating companies. 13 experts from large companies responsible for the respective podcast have been interviewed. The research results show that podcasts are mostly integrated into the company’s overarching communications strategy and incorporate company-specific branding aspects to varying degrees. Corporate podcasts are primarily used to highlight innovative and modern aspects of the brands, especially through the tone of voice. The intended impact of corporate podcasts is often a personal and emotional connection, interaction, and resonance with listeners. Increasing reputation, visibility, and conveying authenticity is also targeted. As a branding tool, podcasts are evaluated rather implicitly. The general evaluation is perceived as challenging and tends to focus on qualitative performance measurement. This study underscores the high need for research on corporate podcasts as a branding tool as well as for key performance indicators (KPIs) of podcasts

    Accurate measurement of 5-methylcytosine and 5-hydroxymethylcytosine in human cerebellum DNA by oxidative bisulfite on an array (OxBS-array).

    The Infinium 450K Methylation array is an established tool for measuring methylation. However, the bisulfite (BS) reaction commonly used with the 450K array cannot distinguish between 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC). The oxidative-bisulfite assay disambiguates 5mC and 5hmC. We describe the use of oxBS in conjunction with the 450K array (oxBS-array) to analyse 5hmC/5mC in cerebellum DNA. The "methylation" level derived by the BS reaction is the combined level of 5mC and 5hmC at a given base, while the oxBS reaction gives the level of 5mC alone. The level of 5hmC is derived by subtracting the oxBS level from the BS level. Here we present an analysis method that distinguishes genuine positive levels of 5hmC at levels as low as 3%. We performed four replicates of the same sample of cerebellum and found a high level of reproducibility (average r for BS = 98.3, and average r for oxBS = 96.8). In total, 114,734 probes showed a significant positive measurement for 5hmC. The range at which we were able to distinguish 5hmC occupancy was between 3% and 42%. In order to investigate the effects of multiple replicates on 5hmC detection we also simulated fewer replicates and found that decreasing the number of replicates to two reduced the number of positive probes identified by > 50%. We validated our results using qPCR in conjunction with glucosylation of 5hmC sites followed by MspI digestion and we found good concordance with the array estimates (r = 0.94). This experiment provides a map of 5hmC in the cerebellum and a robust dataset for use as a standard in future 5hmC analyses. We also provide a novel method for validating the presence of 5hmC at low levels, and highlight some of the pitfalls associated with measuring 5hmC and 5mC.S.     Does Feedback-Related Brain Response during Reinforcement Learning Predict Socio-motivational (In-)dependence in Adolescence?

    This multi-methodological study applied functional magnetic resonance imaging to investigate neural activation in a group of adolescent students (N = 88) during a probabilistic reinforcement learning task. We related patterns of emerging brain activity and individual learning rates to socio-motivational (in-)dependence manifested in four different motivation types (MTs): (1) peer-dependent MT, (2) teacher-dependent MT, (3) peer-and-teacher-dependent MT, (4) peer-and-teacher-independent MT. A multinomial regression analysis revealed that the individual learning rate predicts students’ membership to the independent MT, or the peer-and-teacher-dependent MT. Additionally, the striatum, a brain region associated with behavioral adaptation and flexibility, showed increased learning-related activation in students with motivational independence. Moreover, the prefrontal cortex, which is involved in behavioral control, was more active in students of the peer-and-teacher-dependent MT. Overall, this study offers new insights into the interplay of motivation and learning with (1) a focus on inter-individual differences in the role of peers and teachers as source of students’ individual motivation and (2) its potential neurobiological basis

    Individual differences in the perception of similarity and difference.

    Thematically related concepts like coffee and milk are judged to be more similar than thematically unrelated concepts like coffee and lemonade. We investigated whether thematic relations exert a small effect that occurs consistently across participants (i.e., a generalized model), or a large effect that occurs inconsistently across participants (i.e., an individualized model). We also examined whether difference judgments mirrored similarity or whether these judgments were, in fact, non-inverse. Five studies demonstrated the necessity of an individualized model for both perceived similarity and difference, and additionally provided evidence that thematic relations affect similarity more than difference. Results suggest that models of similarity and difference must be attuned to large and consistent individual variability in the weighting of thematic relations

    Protease-Triggered Release of Stabilized CXCL12 from Coated Scaffolds in an Ex Vivo Wound Model

    Biomaterials are designed to improve impaired healing of injured tissue. To accomplish better cell integration, we suggest to coat biomaterial surfaces with bio-functional proteins. Here, a mussel-derived surface-binding peptide is used and coupled to CXCL12 (stromal cell-derived factor 1α), a chemokine that activates CXCR4 and consequently recruits tissue-specific stem and progenitor cells. CXCL12 variants with either non-releasable or protease-mediated-release properties were designed and compared. Whereas CXCL12 was stabilized at the N-terminus for protease resistance, a C-terminal linker was designed that allowed for specific cleavage-mediated release by matrix metalloproteinase 9 and 2, since both enzymes are frequently found in wound fluid. These surface adhesive CXCL12 derivatives were produced by expressed protein ligation. Functionality of the modified chemokines was assessed by inositol phosphate accumulation and cell migration assays. Increased migration of keratinocytes and primary mesenchymal stem cells was demonstrated. Immobilization and release were studied for bioresorbable PCL-co-LC scaffolds, and accelerated wound closure was demonstrated in an ex vivo wound healing assay on porcine skin grafts. After 24 h, a significantly improved CXCL12-specific growth stimulation of the epithelial tips was already observed.     Mobile Diary App Versus Paper-Based Diary Cards for Patients With Borderline Personality Disorder:Economic Evaluation

    BACKGROUND: The cost-effectiveness of using a mobile diary app as an adjunct in dialectical behavior therapy (DBT) in patients with borderline personality disorder is unknown. OBJECTIVE: This study aims to perform an economic evaluation of a mobile diary app compared with paper-based diary cards in DBT treatment for patients with borderline personality disorder in a psychiatric outpatient facility. METHODS: This study was conducted alongside a pragmatic, multicenter, randomized controlled trial. The participants were recruited at 5 Danish psychiatric outpatient facilities and were randomized to register the emotions, urges, and skills used in a mobile diary app or on paper-based diary cards. The participants in both groups received DBT delivered by the therapists. A cost-consequence analysis with a time horizon of 12 months was performed. Consequences included quality-adjusted life years (QALYs), depression severity, borderline severity, suicidal behavior, health care use, treatment compliance, and system usability. All relevant costs were included. Focus group interviews were conducted with patients, therapists, researchers, and industry representatives to discuss the potential advantages and disadvantages of using a mobile diary app. RESULTS: A total of 78 participants were included in the analysis. An insignificantly higher number of participants in the paper group dropped out before the start of treatment (P=.07). Of those starting treatment, participants in the app group had an average of 37.1 (SE 27.55) more days of treatment and recorded an average of 3.16 (SE 5.10) more skills per week than participants in the paper group. Participants in both groups had a QALY gain and a decrease in depression severity, borderline severity, and suicidal behavior. Significant differences were found in favor of the paper group for both QALY gain (adjusted difference −0.054; SE 0.03) and reduction in depression severity (adjusted difference −1.11; SE 1.57). The between-group difference in total costs ranged from US 107.37toUS107.37 to US 322.10 per participant during the 12 months. The use of services in the health care sector was similar across both time points and groups (difference: psychiatric hospitalization <5 and <5; general practice −1.32; SE 3.68 and 2.02; SE 3.19). Overall, the patients showed high acceptability and considered the app as being easy to use. Therapists worried about potential negative influences on the therapist-patient interaction from new work tasks accompanying the introduction of the new technology but pointed at innovation potential from digital database registrations. CONCLUSIONS: This study suggests both positive and negative consequences of mobile diary apps as adjuncts to DBT compared with paper diary cards. More research is needed to draw conclusions regarding its cost-effectiveness.
