    In vivo generation of thrombin in patients with liver disease without apparent evidence of activation of the intrinsic or extrinsic pathway of coagulation

    Background: Patients with liver diseases are in a hypercoagulable state, as evidenced by enhanced in vitro thrombin generating capacity and elevated plasma levels of markers of in vivo thrombin generation. However, it is unknown by which mechanism in vivo activation of coagulation occurs. Objectives: We aimed to clarify the mechanisms underlying enhanced in vivo thrombin generation to provide a rationale for targeted anticoagulant therapy. Patients/Methods: Overall, 191 patients diagnosed with stable or acutely decompensated cirrhosis, acute liver failure or injury, acute-on-chronic liver failure, or sepsis without underlying chronic liver disease were recruited from King's College Hospital, London, from 2017 to 2021 and compared with reference values of 41 healthy controls. We measured levels of markers of in vivo activation of coagulation and activation of the intrinsic and extrinsic pathways, their respective zymogens, and natural anticoagulants. Results: Thrombin-antithrombin complexes, prothrombin fragment 1+2 (F1+2), and D-dimer levels were increased in acute and chronic liver disease, proportional to disease severity. Plasma levels of free activated factor XII (FXIIa), C1-esterase-inhibitor (C1inh)-FXIIa, C1inh-factor XI, C1inh-plasma kallikrein, factor-VIIa-antithrombin-complexes, and activated FVII were reduced in acute and chronic liver disease, even after adjusting for zymogen levels, which were also substantially reduced. Natural anticoagulants antithrombin and protein C were profoundly reduced in liver patients. Conclusions: This study provides evidence of enhanced thrombin generation in liver disease without detectable activation of the intrinsic or extrinsic pathway. We propose that defective anticoagulant mechanisms highly amplify the low-grade activation of coagulation by either pathway.</p

    Von Willebrand factor is an independent predictor of short-term mortality in acutely ill patients with cirrhosis

    Background and Aims: Levels of von Willebrand factor (VWF) are elevated in patients with cirrhosis, and correlate well with disease severity. In patients with decompensated cirrhosis (DC), plasma VWF is associated with mortality. The value of VWF in predicting short-term mortality risk in patients with acute-on-chronic liver failure (ACLF) is, however, unclear.Methods: We included patients with DC (n = 111) and ACLF (n = 105). We measured VWF levels and correlated these with other laboratory parameters and prediction models for mortality. Also, we assessed the predictive value of VWF in the prediction of 90- and 30-day mortality in patients with DC and ACLF, respectively, and compared this to the predictive value of clinically used prediction models. Finally, we determined the optimal cut-off value for VWF in patients with ACLF.Results: Sixteen of 111 (14%) patients with DC and 35 of 105 (33%) with ACLF died within 90 and 30 days, respectively. VWF was associated with mortality and correlated closely with other prediction models. In patients with ACLF, VWF levels had a discrimination for 30-day mortality comparable with these models and accurately identified ACLF patients with high 30-day mortality risk.Conclusions: Levels of VWF associate closely with risk of mortality in patients with DC and ACLF, and may have predictive utility as a laboratory marker of prognosis. Further research is warranted to assess the additional value of VWF in the prediction of mortality and associated complications in chronic liver failure syndromes.</p

    New Mediterranean Biodiversity Records (July 2016)

    This contribution forms part of a series of collective articles published regularly in Mediterranean Marine Science that report on new biodiversity records from the Mediterranean basin. The current article presents 51 geographically distinct records for 21 taxa belonging to 6 Phyla, extending from the western Mediterranean to the Levantine. The new records, per country, are as follows: Spain: the cryptogenic calcareous sponge Paraleucilla magna is reported from a new location in the Alicante region. Algeria: the rare Atlanto-Mediterranean bivalve Cardium indicum is reported from Annaba. Tunisia: new distribution records for the Indo-Pacific lionfish Pterois miles from Zembra Island and Cape Bon. Italy: the ark clam Anadara transversa is reported from mussel cultures in the Gulf of Naples, while the amphipod Caprella scaura and the isopods Paracerceis sculpta and Paranthura japonica are reported as associated to the –also allochthonous–bryozoan Amathia verticillata in the Adriatic Sea; in the latter region, the cosmopolitan Atlantic tripletail Lobotes surinamensisis also reported, a rare finding for the Mediterranean. Slovenia: a new record of the non-indigenous nudibranch Polycera hedgpethi in the Adriatic. Greece: several new reports of the introduced scleractinian Oculina patagonica, the fangtooth moray Enchelycore anatina, the blunthead puffer Sphoeroides pachygaster (all Atlantic), and the lionfish Pterois miles (Indo-Pacific) suggest their ongoing establishment in the Aegean Sea; the deepest bathymetric record of the invasive alga Caulerpa cylindracea in the Mediterranean Sea is also registered in the Kyklades, at depths exceeding 70 m. Turkey: new distribution records for two non indigenous crustaceans, the blue crab Callinectes sapidus (Atlantic origin) and the moon crab Matuta victor (Indo-Pacific origin) from the Bay of Izmir and Antalya, respectively; in the latter region, the Red Sea goatfish Parupeneus forsskali, is also reported. Lebanon: an array of records of 5 alien and one native Mediterranean species is reported by citizen-scientists; the Pacific jellyfish Phyllorhiza punctata and the Indo-Pacific teleosteans Tylerius spinosissimus, Ostracion cubicus, and Lutjanus argentimaculatus are reported from the Lebanese coast, the latter notably being the second record for the species in the Mediterranean Sea since 1977; the native sand snake-eel Ophisurus serpens, rare in the eastern Mediterranean, is reported for the first time from Lebanon, this being its easternmost distribution range; finally, a substantial number of sightings of the lionfish Pterois miles further confirm the current establishment of this lessepsian species in the Levantine

    The “New Mediterranean Biodiversity Records” series includes new records of marine species found in the Mediterranean basin and/or information on the spatial distribution of already established species of particular interest. The current article presents information on 20 marine taxa classified per country according to their geographic position in the Mediterranean, from west to east. The new records per ecoregion are as follows: Algeria: the first record of the fish Etrumeus golanii is reported along the Algerian coast. Tunisia: the alien jellyfish Phyllorhiza punctata is reported for the first time in the Gulf of Gabès. Italy: the first record of Siganus rivulatus in the Strait of Sicily and a new record of Katsuwonus pelamis from the central Tyrrhenian Sea are reported. The establishment of the isopod of the genus Mesanthura in the northern Tyrrhenian with some notes on its ecology are also included. Croatia: signs of establishment of the Lessepsian Siganus luridus and the occurrence of the alien mollusc Rapana venosa are reported. Albania: the first record of the elasmobranch Alopias superciliosus and a recent sighting of the rare monk seal Monachus monachus in Albanian waters are given. Greece: signs of the establishment of the fish Parupeneus forsskali and of the ascidian Hermania momus in Hellenic Aegean waters are reported. Turkey: a new record of the fish P. forsskali and of the Acarea of the genus Acaromantis and Simognathus are given, while the first case of Remora australis in association with delphinids and the occurrence of the sea star Coscinasterias tenuispina are reported. Also, the establishment of the two alien species Isognomon legumen and Viriola sp. [cf. corrugata] are presented. Egypt: the fish Bathygobius cyclopterus is reported for the first time in Mediterranean Sea waters. Also, a new record of Pagellus bogaraveo and a first record of Seriola fasciata in Egyptian Mediterranean waters are reported. Lebanon: the first record of Dondice banyulensis is presented

    In this Collective Article on “New Mediterranean Biodiversity Records”, we present additional records of species found in the Mediterranean Sea. These records refer to eight different countries mainly throughout the northern part of the basin, and include 28 species, belonging to five Phyla. The findings per country include the following species: Spain: Callinectes sapidus and Chelidonura fulvipunctata; Monaco: Aplysia dactylomela; Italy: Charybdis (Charybdis) feriata, Carcharodon carcharias, Seriola fasciata, and Siganus rivulatus; Malta: Pomacanthus asfur; Croatia: Lagocephalus sceleratus and Pomadasys incisus; Montenegro: Lagocephalus sceleratus; Greece: Amathia (Zoobotryon) verticillata, Atys macandrewii, Cerithium scabridum, Chama pacifica, Dendostrea cf. folium, Ergalatax junionae, Septifer cumingii, Syphonota geographica, Syrnola fasciata, Oxyu- richthys petersi, Scarus ghobban, Scorpaena maderensis, Solea aegyptiaca and Upeneus pori; Turkey: Lobotes surinamensis, Ruvettus pretiosus and Ophiocten abyssicolum. In the current article, the presence of Taractes rubescens (Jordan & Evermann, 1887) is recorded for the first time in the Mediterranean from Italy. The great contribution of citizen scientists in monitoring biodiversity records is reflected herein, as 10% of the authors are citizen scientists, and contributed 37.5% of the new findings.peer-reviewe

    Self-assembled monolayer of light-harvesting core complexes of photosynthetic bacteria on an amino-terminated ITO electrode

    Light-harvesting antenna core (LH1-RC) complexes isolated from Rhodospirillum rubrum and Rhodopseudomonas palustris were successfully self-assembled on an ITO electrode modified with 3-aminopropyltriethoxysilane. Near infra-red (NIR) absorption, fluorescence, and IR spectra of these LH1-RC complexes indicated that these LH1-RC complexes on the electrode were stable on the electrode. An efficient energy transfer and photocurrent responses of these LH1-RC complexes on the electrode were observed upon illumination of the LH1 complex at 880 nm

    Mutation of a single residue, β-glutamate-20, alters protein–lipid interactions of light harvesting complex II

    It is well established that assembly of the peripheral antenna complex, LH2, is required for proper photosynthetic membrane biogenesis in the purple bacterium Rhodobacter sphaeroides. The underlying interactions are, as yet, not understood. Here we examined the relationship between the morphology of the photosynthetic membrane and the lipid–protein interactions at the LH2–lipid interface. The non-bilayer lipid, phosphatidylethanolamine, is shown to be highly enriched in the boundary lipid phase of LH2. Sequence alignments indicate a putative lipid binding site, which includes β-glutamate-20 and the adjacent carotenoid end group. Replacement of β-glutamate-20 with alanine results in significant reduction of phosphatidylethanolamine and concomitant raise in phosphatidylcholine in the boundary lipid phase of LH2 without altering the lipid composition of the bulk phase. The morphology of the LH2 housing membrane is, however, unaffected by the amino acid replacement. In contrast, simultaneous modification of glutamate-20 and exchange of the carotenoid sphaeroidenone with neurosporene results in significant enlargement of the vesicular membrane invaginations. These findings suggest that the LH2 complex, specifically β-glutamate-20 and the carotenoids' polar head group, contribute to the shaping of the photosynthetic membrane by specific interactions with surrounding lipid molecules
