    Genotoxic and Cytotoxic Safety Evaluation of Papain (Carica papaya L.) Using In Vitro Assays

    Papain, a phytotherapeutic agent, has been used in the treatment of eschars and as a debriding chemical agent to remove damaged or necrotic tissue of pressure ulcers and gangrene. Its benefits in these treatments are deemed effective, since more than 5000 patients, at the public university hospital at Rio de Janeiro, Brazil, have undergone papain treatment and presented satisfactory results. Despite its extensive use, there is little information about toxic and mutagenic properties of papain. This work evaluated the toxic and mutagenic potential of papain and its potential antioxidant activity against induced-H2O2 oxidative stress in Escherichia coli strains. Cytotoxicity assay, Growth inhibition test, WP2-Mutoxitest and Plasmid-DNA treatment, and agarose gel electrophoresis were used to investigate if papain would present any toxic or mutagenic potential as well as if papain would display antioxidant properties. Papain exhibited negative results for all tests. This agent presented an activity protecting cells against H2O2-induced mutagenesis

    Xyloglucan and Concanavalin A based dressings in the topical treatment of mice wound healing process

    For medical biomaterials, xyloglucan dispersions can form films or gels to be applied as a wound dressing. For this purpose, the structural characterization of xyloglucan dressing (XG) and xyloglucan dressing containing 0.5 mg/mL of concanavalin A (XGL) was performed. The lectin release capacity and stability, cytotoxicity, and pro-wound healing effects were also investigated. XG and XGL films were prepared by mixing 0.5 % (w/v) xyloglucan with 0.3 % (v/v) glycerol. The ConA incorporated in the xyloglucan dressing maintained its biological activity for fourteen days in a controlled-release manner. The films were non-toxic, homogeneous, flexible, and accelerated the wound contraction compared with the control group, promoting less infiltration of inflammatory cells, angiogenesis, remodeling, and early epithelization. The films also alleviate the inflammation phase by reducing the production of pro-inflammatory cytokines (IFN-, TNF-, IL-1, IL-6, and IL-12), especially the XGL film, which promoted the up- and down-regulation of important proteins associated with the wound repair.     Clinical and Urodynamic results of the Argus T® sling in moderate and severe male stress urinary incontinence after radical prostatectomy – a 5 year prospective study.

    ABSTRACT Purpose: Sling as a therapeutic option for male stress urinary incontinence (SUI) has been reviewed in the last two decades, as it is a relatively simpliest surgery compared to artificial urinary sphincter and has the ability to modulate urethral compression. This study aims to evaluate the efficacy, rate of complications, quality of life and the effects on bladder emptying of the Argus T® compressive and ajustable sling in moderate and severe male SUI treatment. Materials and Methods: Men eligible for stress urinary incontinence treatment after radical prostatectomy were recruited and prospectively evaluated, from March 2010 to November 2016. It was selected outpatient men with moderate and severe SUI, after 12 months of radical prostatectomy, who have failed conservative treatment. All patients had a complete clinical and urodynamic pre and post treatment evaluation, by means of clinical history, physical examination, urine culture, 1-hour pad test and ICIq-SF questionnaire. The UDS was performed after 12, 18 and 24 months postoperatively. Results: Thirty-seven men underwent sling surgery, 19 patients (51.4%) with moderate and 18 (48.6%) with severe SUI. The minimum follow-up time was 5 years. Overall, we had a success rate of 56.7% at 60 months follow-up. After surgery, we did not observe significant changes in the urodynamic parameters evaluated during the follow-up. No patient had urodynamic bladder outlet obstruction (BOO) after sling implantation. Readjustment of the Argus T® sling was performed in 16 (41%) of the patients and 51% of the patients reported some adverse event. Conclusion: We demonstrate a long-term efficacy and safety of Sling Argus T® as an alternative to moderate and severe male SUI treatment.     Professional activities of parents of High School students applicants to Health Sciences Courses

    Modelo do estudo: Estudo baseado em coleta de informações por meio de Questionário Estruturado. Objetivo do estudo: Verificar indicadores da situação econômica e cultural das famílias de alunos do segundo grau que pleiteiam o ingresso em cursos destinados a formar profissionais da saúde. Metodologia: Questionários auto-administrados foram respondidos por 256 estudantes do segundo grau, visitantes de Feira das Profissões da USP (FPUSP-interior). Esses questionários continham perguntas relativas ao tipo de escola que frequentavam e as atividades laborais de seus genitores. Os 108 estudantes de Escolas Públicas constituíram o Grupo 1 e os 148 alunos de Escolas Particulares formaram o Grupo 2. Resultados: A atividade profissional paterna mais frequente no Grupo 1 foi a de “Técnico Manual”(42,15%) e no Grupo 2 a de “Profissional de Nível Universitário” (24,11%). O maior grupo de mães do Grupo 1 foi composto por “Donas de Casa” (22,33%) seguido pelo das que exerciam “Outras Profissões Manuais”( 21,36%). No Grupo 2 a categoria mais frequente para as mães foi “Profissionais de Nível Universitário” (23,18%) seguida por “Professores e Diretores de Escolas” (19,56%). Cinco participantes do Grupo 1 e 27 do Grupo 2 eram filhos de Profissionais da Saúde. Conclusões: Apesar das diferenças das atividades laborais predominantes em cada um dos grupos estudados, os resultados confirmam que aspiração de se tornar um Profissional da Saúde independe da formação ou das profissões dos pais.Study design: Study based on collection of information by means of a Structured Questionnaire. Methodology: Self-report questionnaires were answered by 256 high school students who were visiting the Professions Fair of USP (FPUSP- inland). The questionnaires contained questions about the type of school the students frequented and the work activities of their parents. Group 1 consisted of 108 Public School students and Group 2 consisted of 148 Private School students. Results: The most frequent paternal professional activity was “Manual Technician” (42.15%) for Group 1 and “University Level Professional” (24.11%) for Group 2 . The largest group of mothers in Group 1 consisted of “Housewives” (22.33%), followed by the group of mothers who had “Other Manual Professions” (21.36%). In Group 2, the most frequent category for the mothers was “University Level Professional” (23.18%), followed by “Teachers and Directors of Schools” (19.56%). Five participants in Group 1 and 27 in Group 2 were children of Health Professionals. Conclusions: Despite the differences in the work activities predominating in each group studied, the results confirm the ambition to become a Health Professional regardless of the training or profession of the parent

    Caryocar brasiliense camb protects against genomic and oxidative damage in urethane-induced lung carcinogenesis

    The antioxidant effects of Caryocar brasiliense Camb, commonly known as the pequi fruit, have not been evaluated to determine their protective effects against oxidative damage in lung carcinogenesis. In the present study, we evaluated the role of pequi fruit against urethane-induced DNA damage and oxidative stress in forty 8-12 week old male BALB/C mice. An in vivo comet assay was performed to assess DNA damage in lung tissues and changes in lipid peroxidation and redox cycle antioxidants were monitored for oxidative stress. Prior supplementation with pequi oil or its extract (15 µL, 60 days) significantly reduced urethane-induced oxidative stress. A protective effect against DNA damage was associated with the modulation of lipid peroxidation and low protein and gene expression of nitric oxide synthase. These findings suggest that the intake of pequi fruit might protect against in vivo genotoxicity and oxidative stress

    A utilização de áudios em sala de aula não tem sido muito utilizada nos últimos anos, entretanto existem portais que disponibilizam esse recurso como material didático. Um destes portais é o Dia a Dia Educação da Secretaria do Estado do Paraná-SEED. Assim este artigo apresenta uma análise realizada a partir de áudios que estão disponibilizados nesse portal. Nossa intenção foi verificar a funcionalidade e a recepção dos estudantes para este tipo de recurso. As avaliações demonstram que é possível utilizar esse recurso em sala de aula, entretanto é preciso superar algumas dificuldades técnicas impostas pelos recursos tecnológicos oferecidos à professores e estudantes nas escolas do Paraná

    Career self: a longitudinal study with college students

    O self de carreira constituí um subconjunto organizado do universo cognitivo de uma pessoa, responsável pelo carácter subjetivo que a mesma confere à carreira. Este estudo pretende avaliar mudanças no conteúdo do self de carreira de estudantes universitários, do início para o final do último ano de graduação. Para tal, recorreu-se a medidas repetidas dos índices da Grelha de Repertório da Carreira (Silva & Taveira, 2005; Silva, 2008). Na investigação, participaram 80 estudantes, dos quais 49 são mulheres (61,25%) e 31 são homens (38,75%), com idades entre os 21 e os 45 anos (M= 23,9, DP= 4,31). Os resultados indicam que, no final da licenciatura, os estudantes diminuem a distância como se constrõem em relação aos outros e mantêm uma construção positiva do self de carreira.Fundação para a Ciência e a Tecnologia (FCT

    Azimuthal anisotropy of neutral pion production in Au+Au collisions at sqrt(s_NN) = 200 GeV: Path-length dependence of jet quenching and the role of initial geometry

    We have measured the azimuthal anisotropy of pi0's for 1 < pT < 18 GeV/c for Au+Au collisions at sqrt s_NN = 200 GeV. The observed anisotropy shows a gradual decrease in 3 < pT < 7 - 10 GeV/c, but remains positive beyond 10 GeV/c. The magnitude of this anisotropy is under-predicted, up to at least 10 GeV/c, by current perturbative QCD (pQCD) energy-loss model calculations. An estimate of the increase in anisotropy expected from initial-geometry modification due to gluon saturation effects and initial-geometry fluctuations is insufficient to account for this discrepancy. Calculations which implement a path length dependence steeper than what is implied by current pQCD energy-loss models, show reasonable agreement with the data.Comment: 384 authors, 6 pages text, 3 figures.
