Mutual Zonated Interactions of Wnt and Hh Signaling Are Orchestrating the Metabolism of the Adult Liver in Mice and Human
The Hedgehog (Hh) and Wnt/β-Catenin (Wnt) cascades are morphogen pathways whose pronounced influence on adult liver metabolism has been identified in recent years. How both pathways communicate and control liver metabolic functions are largely unknown. Detecting core components of Wnt and Hh signaling and mathematical modeling showed that both pathways in healthy liver act largely complementary to each other in the pericentral (Wnt) and the periportal zone (Hh) and communicate mainly by mutual repression. The Wnt/Hh module inversely controls the spatiotemporal operation of various liver metabolic pathways, as revealed by transcriptome, proteome, and metabolome analyses. Shifting the balance to Wnt (activation) or Hh (inhibition) causes pericentralization and periportalization of liver functions, respectively. Thus, homeostasis of the Wnt/Hh module is essential for maintaining proper liver metabolism and to avoid the development of certain metabolic diseases. With caution due to minor species-specific differences, these conclusions may hold for human liver as well
Formation of Ejecta and Dust Pond Deposits on Asteroid Vesta
Dust and melt ponds have been studied on planetary bodies including Eros, Itokawa, and the Moon. However, depending on the nature of the regolith material properties and the location of the planetary body, the formation mechanism of the ponded features varies. On Eros and Itokawa, ponded features are formed from dry regolith materials whereas on the Moon similar features are thought to be produced by ejecta melt. On the surface of Vesta, we have identified type 1, ejecta ponds, and type 2, dust ponds. On Vesta type 1 pond are located in the vicinity of ejecta melt of large impact craters. The material is uniformly distributed across the crater floor producing smooth pond surfaces which have a constant slope and shallow depth. The hosting crater of melt-like ponds has a low raised rim and is located on relatively low elevated regions. Whereas, the type 2 ponds on Vesta reveal an undulating surface that is frequently displaced from the crater center or extends toward the crater wall with an abruptly changing slope. We suggested that for the production of the type 2 ponds, localized seismic diffusion and volatile-induced fluidization may be responsible for Vesta. Due to Vesta's large size (in comparison to Eros and Itokawa), the surface may have experienced local-scale rare high-amplitude seismic diffusion which was sufficient to drift fine material. Similarly, short-lived volatile activities were capable to transfer dusty material on to the surface. Segregation and smoothing of transferred material lack further surface activities, hindering the formation of smooth morphology
Observations of GRB 990123 by the Compton Gamma-Ray Observatory
GRB 990123 was the first burst from which simultaneous optical, X-ray and
gamma-ray emission was detected; its afterglow has been followed by an
extensive set of radio, optical and X-ray observations. We have studied the
gamma-ray burst itself as observed by the CGRO detectors. We find that
gamma-ray fluxes are not correlated with the simultaneous optical observations,
and the gamma-ray spectra cannot be extrapolated simply to the optical fluxes.
The burst is well fit by the standard four-parameter GRB function, with the
exception that excess emission compared to this function is observed below ~15
keV during some time intervals. The burst is characterized by the typical
hard-to-soft and hardness-intensity correlation spectral evolution patterns.
The energy of the peak of the nu f_nu spectrum, E_p, reaches an unusually high
value during the first intensity spike, 1470 +/- 110 keV, and then falls to
\~300 keV during the tail of the burst. The high-energy spectrum above ~MeV is
consistent with a power law with a photon index of about -3. By fluence, GRB
990123 is brighter than all but 0.4% of the GRBs observed with BATSE, clearly
placing it on the -3/2 power-law portion of the intensity distribution.
However, the redshift measured for the afterglow is inconsistent with the
Euclidean interpretation of the -3/2 power-law. Using the redshift value of >=
1.61 and assuming isotropic emission, the gamma-ray fluence exceeds 10E54 ergs.Comment: Submitted to The Astrophysical Journal. 16 pages including 4 figure
The Dynamic Exponent of the Two-Dimensional Ising Model and Monte Carlo Computation of the Sub-Dominant Eigenvalue of the Stochastic Matrix
We introduce a novel variance-reducing Monte Carlo algorithm for accurate
determination of autocorrelation times. We apply this method to two-dimensional
Ising systems with sizes up to , using single-spin flip dynamics,
random site selection and transition probabilities according to the heat-bath
method. From a finite-size scaling analysis of these autocorrelation times, the
dynamical critical exponent is determined as (12)
Organized B cell sites in cartilaginous fishes reveal the evolutionary foundation of germinal centers
The absence of germinal centers (GCs) in cartilaginous fishes lies at odds with data showing that nurse sharks can produce robust antigen-specific responses and affinity mature their B cell repertoires. To investigate this apparent incongruity, we performed RNA sequencing on single nuclei, allowing us to characterize the cell types present in the nurse shark spleen, and RNAscope to provide in situ cellular resolution of key marker gene expression following immunization with R-phycoerythrin (PE). We tracked PE to the splenic follicles where it co-localizes with CXCR5high centrocyte-like B cells and a population of putative T follicular helper (Tfh) cells, surrounded by a peripheral ring of Ki67+ AID+ CXCR4+ centroblast-like B cells. Further, we reveal selection of mutations in B cell clones dissected from these follicles. We propose that the B cell sites iden tified here represent the evolutionary foundation of GCs, dating back to the jawed vertebrate ancestor
Pseudomonas aeruginosa Adaptation to Lungs of Cystic Fibrosis Patients Leads to Lowered Resistance to Phage and Protist Enemies
Pathogenic life styles can lead to highly specialized interactions with host species, potentially resulting in fitness trade-offs in other ecological contexts. Here we studied how adaptation of the environmentally transmitted bacterial pathogen, Pseudomonas aeruginosa, to cystic fibrosis (CF) patients affects its survival in the presence of natural phage (14/1, ΦKZ, PNM and PT7) and protist (Tetrahymena thermophila and Acanthamoebae polyphaga) enemies. We found that most of the bacteria isolated from relatively recently intermittently colonised patients (1-25 months), were innately phage-resistant and highly toxic for protists. In contrast, bacteria isolated from long time chronically infected patients (2-23 years), were less efficient in both resisting phages and killing protists. Moreover, chronic isolates showed reduced killing of wax moth larvae (Galleria mellonella) probably due to weaker in vitro growth and protease expression. These results suggest that P. aeruginosa long-term adaptation to CF-lungs could trade off with its survival in aquatic environmental reservoirs in the presence of microbial enemies, while lowered virulence could reduce pathogen opportunities to infect insect vectors; factors that are both likely to result in poorer environmental transmission. From an applied perspective, phage therapy could be useful against chronic P. aeruginosa lung infections that are often characterized by multidrug resistance: chronic isolates were least resistant to phages and their poor growth will likely slow down the emergence of beneficial resistance mutations
A One-sided, Highly Relativistic Jet from Cygnus X-3
Very Long Baseline Array images of the X-ray binary, Cygnus X-3, were
obtained 2, 4 and 7 days after the peak of a 10 Jy flare on 4 February 1997.
The first two images show a curved one-sided jet, the third a scatter-broadened
disc, presumably at the position of the core. The jet curvature changes from
the first to the second epoch, which strongly suggests a precessing jet. The
ratio of the flux density in the approaching to that in the (undetected)
receding jet is > 330; if this asymmetry is due to Doppler boosting, the
implied jet speed is > 0.81c. Precessing jet model fits, together with the
assumptions that the jet is intrinsically symmetric and was ejected during or
after the major flare, yield the following constraints: the jet inclination to
the line of sight must be < 14 degrees; the cone opening angle must be < 12

Characterizing College Science Assessments: The Three-Dimensional Learning Assessment Protocol

Meanwhile, science education at the K-12 level is undergoing significant changes as a result of the emphasis on scientific and engineering practices, crosscutting concepts, and disciplinary core ideas. This framework of "three-dimensional learning" is based on the literature about how people learn science and how we can help students put their knowledge to use. Recently, similar changes are underway in higher education by incorporating three-dimensional learning into college science courses. As these transformations move forward, it will become important to assess three-dimensional learning both to align assessments with the learning environment, and to assess the extent of the transformations. In this paper we introduce the Three-Dimensional Learning Assessment Protocol (3D-LAP), which is designed to characterize and support the development of assessment tasks in biology, chemistry, and physics that align with transformation efforts. We describe the development process used by our interdisciplinary team, discuss the validity and reliability of the protocol, and provide evidence that the protocol can distinguish between assessments that have the potential to elicit evidence of three-dimensional learning and those that do not
