    Objective: The aim of this study is to investigate the degree of variation of hydronephrosis by hydration in patients with ureteropelvic junction obstruction (UPJO). Material and Methods: Patients with antenatally diagnosed UPJO who admitted in a three months period were evaluated in terms of variation in sonographic findings for hydronephrosis. Serial renal ultrasounds were carried out before and after hydration considering parameters for hydronephrosis. Parameters evaluated on each ultrasound included degree of hydronephrosis, maximum anteroposterior diameter of the renal pelvis (mAPD) and anteroposterior pelvic diameter at hilum (hAPD), renal parenchymal thickness, dimensions of both kidneys, grade of hydronephrosis and the volume of the bladder. Results: Eleven children with unilateral hydronephrosis due to UPJO were evaluated. There were 10 males and 1 female at a mean age of 10.3 (3-30) months. The mean value of mAPD before hydration was 19.3±4.3 and increased to 21.3±4.6 mm (9.6% increase) after hydration showing a statistically significant difference (p=0.006). The corresponding measurements for hAPD before and after hydration were 14.5±3.6 mm and 14.6±3.4 mm respectively (p=0.846). Parenchymal thickness before and after hydration were 7.2±0.53 and 6.9±0.44 respectively which also showed no statistical significance (p=0.335). Hydration was found to have no significant effect on the parameters examined of contralateral kidney. Conclusion: The commonly used ultrasonographic parameters in the follow-up examination of hydronephrotic kidneys are prone to hydration effects. Among these parameters hAPD seems to be less affected.     Polymorphism in seed endosperm proteins (gliadins and glutenins) of Turkish cultivated einkorn wheat [Triticum monococcum ssp. monococcum] landraces

    The objective of this study is the analysis of polymorphism in seed endosperm proteins (gliadins and glutenins) of Turkish cultivated einkorn wheat [Triticum monococcum ssp. monococcum] landraces. The genetic diversity of high-molecular-weight (HMW) glutenin subunits and the gliadin proteins in 10 landrace populations of cultivated einkorn wheat, originating from Turkey, was investigated using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and ammonium lactic acid polyacrylamide gel electrophoresis (A-PAGE), respectively. For glutenins, the mean number of alleles, the mean number of effective alleles, the mean value of genetic diversity and the mean value of average genetic diversity were detected as 3.50, 2.98, 0.65 and 0.28, respectively. The genetic differentiation was 0.57, while gene flow was 0.19 between populations. For gliadins, the mean number of alleles, the mean number of effective alleles, the mean value of total genetic diversity and the genetic diversity within population were detected as 2.00, 1.21, 0.17 and 0.15, respectively. The genetic differentiation was 0.08, whereas gene flow was 6.15 between populations. STRUCTURE is a software package program for population genetic analysis, was used to infer population structures of landraces populations. The optimum value for K was obtained as 10. Considering the high number of proteins and genetic variation, and increased interest in organic products, the farming of einkorn wheat should be supported and conservation of germplasm in landraces should be maintained as important genetic resources. The landraces germplasm should be conserved for future crop improvement processes

    Gas sensing properties of carbon nanotubes modified with calixarene molecules measured by QCM techniques

Calixarene molecules were synthesized individually by considering their functional groups to attract the gas. Calixarene modified carbon nanotube based sensors were fabricated using drop-casting method on a quartz crystal microbalance gold electrode. Carbon monoxide, carbondioxide, oxygen and dry air were used as active gases for adsorption process, while high-purity nitrogen gas was used for desorption process. The selectivity and sensitivity of calixarene modified carbon nanotube are investigated in detail. Our experimental results show that functional calixarene modified carbon nanotube coated quartz crystal microbalance sensors are very sensitive and selective to gas of CO 2 at room temperature operation.This research was supported by TÜBİTAK under project numbers TBAG 109 T240 and 110 M349 and IYTE research project number 2010IYTE2

    Extracción asistida por microondas de compuestos no polares de cáscaras de pistacho y caracterización de los extractos

    Soxhlet and microwave assisted extraction (MAE) methods were used to obtain non-polar compounds from pistachio hull. MAE parameters (liquid to solid ratio, microwave power, and extraction time) were studied to obtain maximum extraction yield. The optimal conditions were found to be liquid to solid ratio of 15:1 (v/w), microwave power of 250 W and extraction time of 12.5 min. The extraction yields were 9.81 and 9.50% for MAE and Soxhlet methods, respectively. The total phenolic content, antioxidant activity and tocopherol content of the extract obtained by MAE was found to be significantly higher than those of the Soxhlet extract (p < 0.05). The results showed that the extract contained α-tocopherols (567.65 mg/kg) and oleic acid (48.46%) as the major tocopherols and fatty acids. These findings propose that hull extracts can be considered as a good source of natural bioactive compounds and MAE can be a good alternative to the traditional Soxhlet method.Se utilizó la extracción mediante Soxhlet y métodos de extracción asistida mediante microondas (MAE) para obtener compuestos no polares de las cascaras de pistacho. Se estudiaron los parámetros para la MAE (relación líquido-sólido, potencia de microondas y tiempo de extracción) para obtener el máximo rendimiento de la extracción. Se encontró que las condiciones óptimas eran una relación líquido a sólido de 15:1 (v/p), potencia de microondas de 250 W y un tiempo de extracción de 12,5 minutos. Los rendimientos de extracción fueron 9.81 y 9.50% para los métodos MAE y Soxhlet, respectivamente. El contenido fenólico total, la actividad antioxidante y el contenido de tocoferoles de los extractos obtenidos por MAE fueron significativamente más altos que los de los extractos de Soxhlet (p < 0,05). Los resultados muestran que el extracto contiene α-tocoferol (567.65 mg/kg) y ácido oleico (48.46%) como los principales tocoferoles y ácidos grasos, respectivamente. Estos hallazgos proponen que los extractos de las cascaras pueden considerarse como una buena fuente de compuestos bioactivos naturales y MAE puede ser una buena alternativa al método Soxhlet tradicional

    Microbial arms race: Ballistic "nematocysts" in dinoflagellates represent a new extreme in organelle complexity

    We examine the origin of harpoon-like secretory organelles (nematocysts) in dinoflagellate protists. These ballistic organelles have been hypothesized to be homologous to similarly complex structures in animals (cnidarians); but we show, using structural, functional, and phylogenomic data, that nematocysts evolved independently in both lineages. We also recorded the first high-resolution videos of nematocyst discharge in dinoflagellates. Unexpectedly, our data suggest that different types of dinoflagellate nematocysts use two fundamentally different types of ballistic mechanisms: one type relies on a single pressurized capsule for propulsion, whereas the other type launches 11 to 15 projectiles froman arrangement similar to a Gatling gun.Despite their radical structural differences, these nematocysts share a single origin within dinoflagellates and both potentially use a contraction-based mechanism to generate ballistic force. The diversity of traits in dinoflagellate nematocysts demonstrates a stepwise route by which simple secretory structures diversified to yield elaborate subcellular weaponry

    Secreted Frizzled-related Protein 2 (sFRP2) Redirects Non-canonical Wnt Signaling from Fz7 to Ror2 during Vertebrate Gastrulation

Available from American Society for Biochemistry and Molecular Biology via the DOI in this record.Convergent extension movements during vertebrate gastrulation require a balanced activity of non-canonical Wnt signaling pathways, but the factors regulating this interplay on the molecular level are poorly characterized. Here we show that sFRP2, a member of the secreted frizzled-related protein (sFRP) family, is required for morphogenesis and papc expression during Xenopus gastrulation. We further provide evidence that sFRP2 redirects non-canonical Wnt signaling from Frizzled 7 (Fz7) to the receptor tyrosine kinase-like orphan receptor 2 (Ror2). During this process, sFRP2 promotes Ror2 signal transduction by stabilizing Wnt5a-Ror2 complexes at the membrane, whereas it inhibits Fz7 signaling, probably by blocking Fz7 receptor endocytosis. The cysteine-rich domain of sFRP2 is sufficient for Ror2 activation, and related sFRPs can substitute for this function. Notably, direct interaction of the two receptors via their cysteine-rich domains also promotes Ror2-mediated papc expression but inhibits Fz7 signaling. We propose that sFRPs can act as a molecular switch, channeling the signal input for different non-canonical Wnt pathways during vertebrate gastrulation
