    Viability and tissue quality of cartilage flaps from patients with femoroacetabular hip impingement: A matched-control comparison

    Background: Chondrolabral damage is commonly observed in patients with cam-type femoroacetabular impingement (FAI). Chondral flap reattachment has recently been proposed as a possible preservation technique. Purpose/Hypothesis: The purpose of this study was to determine the viability and tissue quality of chondral flaps from patients with FAI at the time of arthroscopy. It was hypothesized that chondral flaps from patients with cam lesions of the hip would exhibit less viability and greater tissue degeneration than would those of a matched control group. Study Design: Cohort study; Level of evidence, 2. Methods: Patients with cam-type FAI who were treated with hip arthroscopy between 2014 and 2016 were asked to participate in this study. The cartilage lesions were localized and classified intraoperatively according to Beck classification. A chondral flap (study group) and a cartilage sample (control group) were obtained from each patient for histologic evaluation. Cellular viability and tissue quality were examined and compared in both groups. Cellular viability was determined with live/dead staining, and tissue quality was evaluated using safranin O/fast green, hematoxylin and eosin (H&amp;E) staining, and immunohistochemistry for collagen II. Osteoarthritis Research Society International (OARSI) grading was used for quality assessment, and Image J software was used to calculate the percentage of tissue viability and Col II stain. Results: A total of 10 male patients with a mean age of 38.4 years (range, 30-55 years) were enrolled. All chondral flaps were classified as Beck grade 4. The mean cellular viability of the chondral flaps was reduced (54.6% ± 25.6%), and they were found to be degenerated (OARSI grade, 4 ± 1.27). Control samples also had reduced viability (38.8% ± 30.3%) and were degenerative (OARSI grade, 3.5 ± 1.38). There was no statistically significant intergroup difference for viability ( P = .203) or OARSI grade ( P = .645), nor was there an intragroup correlation between viability and OARSI grade ( P &gt; .05). A significant negative correlation ( r = −0.9, P = .035) was found between OARSI grade and collagen II percentage scale in 5 selected samples. Conclusion: Despite appearing normal macroscopically, the chondral flaps from patients with cam-type FAI displayed loss of viability and tissue degeneration. In addition, control samples obtained away from the injury area also displayed cartilage damage and degeneration. Careful consideration should be taken when attempting to reattach the chondral flap. </jats:sec

    Interplay of the two ancient metabolites auxin and MEcPP regulates adaptive growth.

    The ancient morphoregulatory hormone auxin dynamically realigns dedicated cellular processes that shape plant growth under prevailing environmental conditions. However, the nature of the stress-responsive signal altering auxin homeostasis remains elusive. Here we establish that the evolutionarily conserved plastidial retrograde signaling metabolite methylerythritol cyclodiphosphate (MEcPP) controls adaptive growth by dual transcriptional and post-translational regulatory inputs that modulate auxin levels and distribution patterns in response to stress. We demonstrate that in vivo accumulation or exogenous application of MEcPP alters the expression of two auxin reporters, DR5:GFP and DII-VENUS, and reduces the abundance of the auxin-efflux carrier PIN-FORMED1 (PIN1) at the plasma membrane. However, pharmacological intervention with clathrin-mediated endocytosis blocks the PIN1 reduction. This study provides insight into the interplay between these two indispensable signaling metabolites by establishing the mode of MEcPP action in altering auxin homeostasis, and as such, positioning plastidial function as the primary driver of adaptive growth

    Effect of polyvinyl alcohol ligands on supported gold nano-catalysts: Morphological and kinetics studies

    The effect of polyvinyl alcohol (PVA) stabilizers and gold nanoparticles supported on active carbon (AuNPs/AC) was investigated in this article. Polymers with different molecular weights and hydrolysis degrees have been synthesized and used, like the stabilizing agent of Au nano-catalysts obtained by the sol-immobilization method. The reduction of 4-nitrophenol with NaBH4 has been used as a model reaction to investigate the catalytic activity of synthesized Au/AC catalysts. In addition, we report several characterization techniques such as ultraviolet-visible spectroscopy (UV-Vis), dynamic light scattering (DLS), X-ray diffraction (XRD), transmission electron microscopy (TEM), and X-ray photoelectron spectroscopy (XPS) in order to correlate the properties of the polymer with the metal nanoparticle size and the catalytic activity. A volcano plot was observed linking the catalytic performance with hydrolysis degree and the maximum of the curve was identified at a value of 60%. The Au:PVA-60 weight ratio was changed in order to explain how the amount of the polymer can influence catalytic properties. The effect of nitroaromatic ring substituents on the catalytic mechanism was examined by the Hammett theory. Moreover, the reusability of the catalyst was investigated, with little to no decrease in activity observed over five catalytic cycles. Morphological and kinetic studies reported in this paper reveal the effect of the PVA polymeric stabilizer properties on the size and catalytic activity of supported gold nanoparticles

    Gestión turística y eventos meteorológicos extremos en destinos litorales

    El trabajo que se presenta a continuación aborda el impacto de fenómenos meteorológicos extremos (olas de frío, tormentas eléctricas, vientos fuertes, etcétera) en los destinos turísticos de la costa bonaerense para el período comprendido entre 2005-2015. Se realiza la identificación e inventario de los eventos meteorológicos de tipo extremo que tuvieron lugar en invierno, otoño y primavera. De esta manera se continua con el análisis realizado por Gil et al. (2015). Los resultados servirán para mejorar la gestión pública de los destinos turísticos en términos de la prevención y mitigación de eventos extremos.The work presented below addresses the impact of extreme weather (cold waves, thunderstorms, high winds, etc.) in tourist destinations in the Buenos Aires coast for the period 2005-2015. Identification and inventory of extreme weather events type that took place in winter, autumn and spring is performed. Thus we continue with the analysis by Gil et al. (2015). The results will serve to improve public management of tourist destinations, in terms of prevention and mitigation of extreme events

    RESUMEN El objetivo del siguiente trabajo consiste en identificar e inventariar los eventos climatológicos extremos en los balnearios del litoral bonaerense y la gestión realizada para la mitigación del peligro. La importancia de esto radica en que estos eventos son cada vez más frecuentes e intensos y afectan a los destinos turísticos principalmente en verano. Se identificaron y catalogaron eventos climáticos extremos (2005 - 2015) y la gestión pública realizada en los destinos para su mitigación. Los resultados indican que el incremento de fenómenos como la caída de rayos impulsó desde la gestión pública el desarrollo de acciones preventivas. Mientras que no existen medidas para los eventos climáticos extremos habituales como las tormentas, vientos fuertes, granizo, etc

    A Comparison of Perturbed Initial Conditions and Multiphysics Ensembles in a Severe Weather Episode in Spain

    Ensembles of numerical model forecasts are of interest to operational early warning forecasters as the spread of the ensemble provides an indication of the uncertainty of the alerts, and the mean value is deemed to outperform the forecasts of the individual models. This paper explores two ensembles on a severe weather episode in Spain, aiming to ascertain the relative usefulness of each one. One ensemble uses sensible choices of physical parameterizations (precipitation microphysics, land surface physics, and cumulus physics) while the other follows a perturbed initial conditions approach. The results show that, depending on the parameterizations, large differences can be expected in terms of storm location, spatial structure of the precipitation field, and rain intensity. It is also found that the spread of the perturbed initial conditions ensemble is smaller than the dispersion due to physical parameterizations. This confirms that in severe weather situations operational forecasts should address moist physics deficiencies to realize the full benefits of the ensemble approach, in addition to optimizing initial conditions. The results also provide insights into differences in simulations arising from ensembles of weather models using several combinations of different physical parameterizations

    Effects of Curing on Photosensitive Resins in {SLA} Additive Manufacturing

    Different mechanical properties characterise the materials of 3D printed components, depending on the specific additive manufacturing (AM) process, its parameters, and the post-treatment adopted. Specifically, stereolithography (SLA) uses a photopolymerisation technique that creates solid components through selective solidification. In this study, 72 specimens were 3D printed using 12 commercial-grade methacrylate resins and tested under uniaxial tensile loads. The resin specimens were evaluated before and after curing. The recommended cure temperature and time were followed for all materials. The stress-strain curves measured during the testing campaign were evaluated in terms of maximum tensile strength, Young’s modulus, ductility, resilience, and toughness. The results reveal that the curing process increases the material stiffness and resistance to tensile loads. However, it was found that the curing process generally reduces the plasticity of the resins, causing a more or less marked brittle behaviour. This represents a potential limitation to the use of SLA 3D printing for structural elements which require some plasticity to avoid dangerous sudden failures

    Influence of stabilisers on the catalytic activity of supported Au colloidal nanoparticles for the liquid phase oxidation of glucose to glucaric acid: understanding the catalyst performance from NMR relaxation and computational studies

    Supported Au colloidal nanoparticles have been prepared in the presence of stabilising polymers, such as, PVA, PVP and PEG (polyvinylalcohol, polyvinylpyrrolidone, polyethylene glycol). The effect of the polymer to Au weight ratio was investigated, for the synthesis of Au nanoparticles with varying particle size and particle size distribution. By varying the polymer/Au wt/wt ratio, Au nanoparticles with mean diameters from 3 to 8 nm were synthesised. The synthesised Au catalysts were studied in the liquid phase oxidation of glucose to glucaric acid under alkaline conditions. We demonstrated that the choice of polymer and polymer to Au weight ratio, have an important influence in terms of catalytic activity and yield to glucaric acid. The highest yield to glucaric acid (22%) was obtained using Au–PVA catalysts. A strong deactivation was observed using Au catalysts. Further evaluation of the possible reasons for deactivation were investigated using experimental, computational and NMR relaxation studies
