OBJECTIVE: To analyze the epidemiological characteristics of adolescents with cancer referred to an oncologic center, between 2000 and 2006. METHODS: A retrospective descriptive study was carried out in order to evaluate the epidemiological data of patients aged between ten and 19 years at diagnosis and admitted at the Institute of Oncology (IOP/Graacc) of Universidade Federal de São Paulo (UNIFESP), Brazil, between 2000 and 2006. RESULTS: Among 2,362 patients admitted during this period with a diagnosis of cancer, 629 (26.6%) were adolescents. Mean age was 13.7 years, being 56.8% male. Regarding race, 60.7% of the patients were white, 30% mulattoes, 6.5% blacks, and 2.5% of patients had no characterization of race in the medical records. The most frequent types of tumors were: central nervous system tumors (22.1%), osteosarcoma (14.6%), lymphoma (14.5%), and leukemia (14.5%). The overall survival at five years was 73.7%. Adolescents with the diagnosis of rhabdomyosarcoma presented disseminated disease and histology of worse prognosis, which contributed for increasing the mortality rate of this group of patients. CONCLUSIONS: Adolescents with cancer are a group of patients with distinct characteristics when compared to other cancer groups. Our results show differences in the prevalence of tumors during adolescence and childhood, in the later leukemias and central nervous system tumors predominate. Eight fluorescent compounds containing a naphthotriazole moiety substituted at position 2 by a (vinylsulfonyl)aryl group or its precursors, containing hydroxyl or sulphonic groups or N-methylglycine, were prepared and characterized. The products were recovered in moderate yields after column chromatography or recrystallization and identified by proton and carbon nuclear magnetic resonance spectroscopy. Double resonance, heteronuclear multiple quantum coherence and heteronuclear multiple bond correlation experiments were carried out for complete assignment of proton and carbon signals. Absorption and emission spectra were obtained, in acetonitrile, for all the compounds and the fluorescence quantum yields determined. All compounds are promising fluorescent probes due to their high fluorescence quantum yields.Research grant VZ MSMT-0021627501, Czech RepublicFundação para a Ciência e a Tecnologia (FCT) - REEQ/ 630/QUI/2005; REDE/1517/RMN/2005FEDER - REEQ/ 630/QUI/2005; REDE/1517/RMN/200

Antiproliferative effect of colonic fermented phenolic compounds from jaboticaba (Myrciaria trunciflora) fruit peel in a 3D cell model of colorectal cancer

Jaboticaba is a Brazilian native berry described as a rich source of phenolic compounds (PC) with health promoting effects. PC from jaboticaba peel powder (JPP) have low intestinal bio-accessibility and are catabolized by gut microbiota. However, the biological implication of PCderived metabolites produced during JPP digestion remains unclear. This study aimed to evaluate the antiproliferative effects of colonic fermented JPP (FJPP) in a 3D model of colorectal cancer (CRC) composed by HT29 spheroids. JPP samples fermented with human feces during 0, 2, 8, 24 or 48 h were incubated (10,000 µg mL−1 ) with spheroids, and cell viability was assessed after 72 h. Chemometric analyses (cluster and principal component analyses) were used to identify the main compounds responsible for the bioactive effect. The antiproliferative effect of FJPP in the CRC 3D model was increased between 8 h and 24 h of incubation, and this effect was associated with HHDP-digalloylglucose isomer and dihydroxyphenyl-γ-valerolactone. At 48 h of fermentation, the antiproliferative effect of FJPP was negligible, indicating that the presence of urolithins did not improve the bioactivity of JPP. These findings provide relevant knowledge on the role of colonic microbiota fermentation to generate active phenolic metabolites from JPP with positive impact on CRC

This work aimed at identifying heat protective mechanisms promoted by CO2 in Coffea arabica (cv. Icatu and IPR108) and Coffea canephora cv. Conilon CL153. Plants were grown at 25/20 degrees C (day/night), under 380 or 700 mu L CO2 L-1, and then gradually submitted to 31/25, 37/30, and 42/34 degrees C. Relevant heat tolerance up to 37/30 degrees C for both [CO2] and all coffee genotypes was observed, likely supported by the maintenance or increase of the pools of several protective molecules (neoxanthin, lutein, carotenes, ohtocopherol, HSP70, raffinose), activities of antioxidant enzymes, such as superoxide dismutase (SOD), ascorbate peroxidase (APX), glutathione reductase (GR), catalase (CAT), and the upregulated expression of some genes (ELIP, Chaperonin 20). However, at 42/34 degrees C a tolerance threshold was reached, mostly in the 380 -plants and Icatu. Adjustments in raffinose, lutein, beta-carotene, alpha-tocopherol and HSP70 pools, and the upregulated expression of genes related to protective (FLIPS, HSP70, Chape 20, and 60) and antioxidant (CAT, CuSOD2, APX Cyt, APX ChI) proteins were largely driven by temperature. However, enhanced [CO2] maintained higher activities of GR (Icatu) and CAT (Icatu and IPR108), kept (or even increased) the Cu,Zn-SOD, APX, and CAT activities, and promoted a greater upregulation of those enzyme genes, as well as those related to HSP70, ELIPs, Chaperonins in CL153, and Icatu. These changes likely favored the maintenance of reactive oxygen species (ROS) at controlled levels and contributed to mitigate of photosystem II photoinhibition at the highest temperature. Overall, our results highlighted the important role of enhanced [CO2] on the coffee crop acclimation and sustainability under predicted future global warming scenarios.publishersversionpublishe
