    Genome comparison and context analysis reveals putative mobile forms of restriction–modification systems and related rearrangements

    The mobility of restriction–modification (RM) gene complexes and their association with genome rearrangements is a subject of active investigation. Here we conducted systematic genome comparisons and genome context analysis on fully sequenced prokaryotic genomes to detect RM-linked genome rearrangements. RM genes were frequently found to be linked to mobility-related genes such as integrase and transposase homologs. They were flanked by direct and inverted repeats at a significantly high frequency. Insertion by long target duplication was observed for I, II, III and IV restriction types. We found several RM genes flanked by long inverted repeats, some of which had apparently inserted into a genome with a short target duplication. In some cases, only a portion of an apparently complete RM system was flanked by inverted repeats. We also found a unit composed of RM genes and an integrase homolog that integrated into a tRNA gene. An allelic substitution of a Type III system with a linked Type I and IV system pair, and allelic diversity in the putative target recognition domain of Type IIG systems were observed. This study revealed the possible mobility of all types of RM systems, and the diversity in their mobility-related organization

    Rapid Synthesis of L-Idosyl Glycosyl Donors from α- Thioglucosides for the Preparation of Heparin Disaccharides

    A new methodology for the synthesis of the most challenging heparin building block has been developed. Orthogonally protected L-idosyl glycosyl donors were prepared by C5 epimerization of the corresponding thioglucosides using the hydroboration/oxidation method followed by a 4,6-acetal formation. The -anomeric configuration was crucial and the bulky C4 substituent was advantageous for the high L-ido diastereoselectivity. The 4,6-arylmethylene group proved to be a directing element in glycosylation thereby stereoselective -idosylation could be achieved by using idosyl donors without a C-2 participating group

    L'hĂ©mophilie A acquise (HA) est une maladie hĂ©morragique acquise rare par auto-anticorps anti-FVIII, idiopathique ou associĂ©e Ă  une autre pathologie (post-partum, nĂ©oplasie, maladie auto-immune). Le traitement consiste en la prise en charge du syndrome hĂ©morragique, et en l'Ă©radication de l inhibiteur par un traitement immunosuppresseur. Nous rapportons une sĂ©rie de 36 patients consĂ©cutifs pris en charge au CHU de Nantes entre 1993 et 2010 pour HA. Ce travail rĂ©trospectif mono-centrique retrouve des donnĂ©es conformes Ă  celles de la littĂ©rature. Il permet de rĂ©-insister sur la nĂ©cessitĂ© d'un diagnostic et d'un traitement prĂ©coces, ce qui nĂ©cessite une sensibilisation des mĂ©decins Ă  cette maladie. Il insiste sur la iatrogĂ©nie trĂšs importante du traitement de fond, et sur la nĂ©cessitĂ© d'Ă©valuer la place du rituximab. Il permet de rĂ©flĂ©chir Ă  l'Ă©tablissement d un score de gravitĂ© pour guider ce traitement.     Thanks to the introduction of legumes and cover crops, which enable crop diversification whilst qualifying for coupled aids, this innovative system has proved to enhance agronomic, environmental and economic performances. On a 3 years average basis, the 2-years rotation has proven to release 32% less GHG than maize monocropping. This is mainly due to a significant decrease of operating costs enabled by lower nitrogen inputs (-40%) thanks to the soybeans. It also brings economic satisfaction as soybean is well adapted in this region and a biogas plant gives additional added value to the energy catch crop. Even with higher mechanisation costs due to the ECC, the net margin with aid remains higher than that of the reference monocropping (+ 37€/hectare on average over 3 years, although similar margin per hour worked)
