It accounts for more pain and functional disability than any other musculoskeletal disease and is an important source of high societal and economic costs. Although the pathophysiology of OA is poorly understood, the risk factors associated with disease development are well-established. They include age, obesity, sex, previous incidence of joint injuries, meniscal damage, joint instability, malalignment, genetics, bone shape (including anatomical deformities), muscle weakness and sarcopenia, and metabolic disease.     Cell sources of inflammatory mediators present in bone marrow areas inside the meniscus

Methods: Mice subjected to transection of the medial collateral and anterior cruciate ligaments and meniscotomy (osteoarthritis model) or to a sham procedure, as well as non-operated (naive) mice and rats, had knee joints excised. Tissues were stained with hematoxylin-eosin and tartrate-resistant acid phosphatase (TRAP). CD68+ cells, inducible nitric oxide synthase (iNOS), interleukin (IL)-1β, and tumor necrosis factor (TNF) expression were detected using immunohistochemistry. Results: Lamellar ossified areas, bone-entrapped osteocytes and bone marrow spaces were found inside menisci of one week up to 6 months-old naïve mice, regardless of gender. Menisci from naive rats also showed the same pattern with bone marrow areas. CD68+ cells were identified in bone marrow areas inside the meniscus of mice. TRAP+ osteoclasts, and hematogenous precursors expressing IL-1β, TNF, and iNOS were identified inside bone marrow areas in meniscal samples from both naïve and sham operated mice. Quantitative immunoexpression of IL-1 β, TNF and iNOS was more intense, P = 0.0194, 0.0293, 0.0124, respectively, in mouse knees from mice sacrificed 49 days after being subjected to an osteoarthritis (OA) model as compared to sham operated animals. Conclusion: We provide novel data showing that rodent menisci display bone marrow areas with cells able to produce inflammatory mediators.     A artrite reumatoide (AR) é uma doença inflamatória autoimune, sistêmica, de curso progressivo, caracterizada por exuberante sinovite crônica, que pode gerar deformidades e incapacidade funcional, cujo tratamento precoce minimiza o dano às juntas. Sua etiopatogenia ainda não está completamente elucidada, mas compreende respostas imunológicas com a participação de células T auxiliares (Th1). Uma aparente menor gravidade da AR em pacientes de regiões com menor renda poderia estar associada a maior prevalência de parasitoses intestinais, especialmente as helmintíases. A rigor, um desvio na resposta imune para o predomínio de células T auxiliares (Th2), decorrente da exposição crônica a helmintos, modularia negativamente a inflamação em doentes com AR, e levaria a menor gravidade e dano articular. A revisão de aspectos da influência da reposta imunológica nas parasitoses intestinais, especialmente as helmintíases, em pacientes com artrite reumatoide é o objetivo desse trabalho.Rheumatoid arthritis (RA) is a systemic autoimmune inflammatory disease, with a progressive course, characterized by chronic synovitis that may evolve with deformities and functional disability, and whose early treatment minimizes joint damage. Its etiopathogenesis is not fully elucidated but comprises immunologic responses mediated by T helper cells (Th1). An apparent minor severity of RA in patients from regions with lower income could be associated with a higher prevalence of gut parasites, especially helminths. Strictly, a shift in the immune response toward the predominance of T helper cells (Th2), due to the chronic exposure to helminths, could modulate negatively the inflammation in RA patients, resulting in lower severity/joint injury.     Heat Stress and Body Temperature in Brown Swiss Cows Raised in Semi-Arid Climate of Ceará State, Brazil

    Background: In tropical countries like Brazil, air temperature and relative humidity have a significant effect on animal physiology; there is a great impact of solar radiation on physiological parameters, especially on body temperature. This study evaluated the occurrence of heat stress in Brown Swiss cows in a tropical semi-arid climate, and checked for the correlation between internal body temperatures [rectal temperature (RT) and vaginal temperature (VT)] with surface temperature (ST) to determine if these variables are associated.Materials, Methods & Results: Twenty-eight Brown Swiss cows at three stages of the lactation cycle were used in this study: 10 nonpregant lactating (NPL) cows, 8 dry pregnant (DP) cows, and 10 pregnant lactating (PL) cows. These animals were between the second and third calving, weighed between 346 and 720 kg, and had ages between 2 and 13 years. During the experimental period, air temperature and relative humidity (RH) at the experimental site were measured using a digital thermohygrometer. The temperature and humidity index (THI) was calculated according to methodology described by Thom (1958), and was used as an environmental comfort parameter. For the evaluation of RT and VT, two digital clinical thermometers, one inserted in the vagina and the other in the rectum, were used simultaneously to minimize stress. Surface temperature (ST) was assessed using a digital infrared laser thermometer at a distance of 50 cm from the animal. Surface temperature was measured in the forehead (FST), thorax (TST), flank (FLST), and legs (LST). During the study period, the ambient temperature (AT) was significantly higher outside (in the sun) than inside of the facilities (in the shade) (P < 0.05). RH was inversely proportional to AT, and was significantly higher inside than outside the facilities (P < 0.05). Like AT, THI was significantly higher outside (in the sun) than inside the facilities (in the shade), with significant differences between these locations (P < 0.05). The amplitudes of the differences between the locations were as follows: 3.8°C for AT, 6.2% for RH, and 2.6 for THI. Mean values of rectal and vaginal temperatures were not significantly different from each other independent of lactation cycle stage, and were almost always significantly higher than the measured surface temperatures (P < 0.05). The maximum values obtained for each lactation cycle stage in this study were higher when compared to the mean values, showing that they are more representative of the occurrence of heat stress. Correlations between internal temperatures (RT and VT) and surface temperatures (TSF, TST, TSFL, and TSP) were weak and non-significant. ST values exhibited mostly weak, non-significant correlations, with the exception of FST with FLST and LST, which had moderate, significant correlations, as shown by the following coefficient factors: FST x FLST, 0.34; LST x FST, 0.415; and LST x FLST, 0.37.Discussion: A temperature of 34°C with RH ranging from 46% to 80% (i.e., a THI between 83 and 89) has been reported to have a significant thermal impact on dairy Brown Swiss cows; the THI values found in the present experiment were close to those. Body temperatures exhibited significant variations depending on the lactation cycle stage of the cows. Lactation concomitant with pregnancy significantly increases internal temperatures (RT and VT) in DP and NPL cows. This finding may be explained by the fact that pregnancy concomitant with lactation accelerates the metabolism, which results in higher food intake and increased production of body heat with consequent increase in internal body temperature. The maximum values of internal body temperatures (RT and TV) at all stages of the lactation cycle were higher than their corresponding average values, which indicates that some cows exhibited, at given moments, hyperthermia

    Current epidemiological studies have showed a growing number of obese patients worldwide. New surgical treatments for obesity that can provide consistent weight loss and effective management of associated comorbidities have been increasingly investigated. The effects of bariatric surgery include long-term benefits such as lower blood glucose levels with good management of type 2 diabetes, hyperlipidemia, metabolic syndrome, reduced risk of coronary heart disease, better quality of life and improved life expectancy. However, bariatric surgery is a procedure that implies malabsorption of nutrients and can result in nutritional deficiencies, anemia, hypocalcemia, low absorption of vitamin D and hyperparathyroidism, causing decreased bone mineral density and increased risk of osteoporosis and may play a major role in the progression and severity of periodontal disease

    Biochemical Characterization of an In-House Coccidioides Antigen: Perspectives for the Immunodiagnosis of Coccidioidomycosis

    The objective of this study was to evaluate the reactivity of an in-house antigen, extracted from a strain of C. posadasii isolated in northeastern Brazil, by radial immunodiffusion and Western blotting, as well as to establish its biochemical characterization. the protein antigen was initially extracted with the use of solid ammonium sulfate and characterized by 1-D electrophoresis. Subsequently, it was tested by means of double radial immunodiffusion and Western blotting. A positive reaction was observed against the antigen by both immunodiagnostic techniques tested on sera from patients suffering from coccidioidomycosis. Besides this, two immunoreactive protein bands were observed and were revealed to be a beta-glucosidase and a glutamine synthetase after sequencing of the respective N-terminal regions.     Genetic potential of segregating cowpea progenies regarding inflorescence type

    O objetivo deste trabalho foi avaliar o potencial genético produtivo de progênies de feijão-caupi (Vigna unguiculata) segregantes quanto ao tipo da inflorescência. Foram avaliadas 68 progênies F4 obtidas dos retrocruzamentos: (TV x 5058‑09C x Cacheado‑roxo) x TV x 5058‑09C e (AU94‑MOB‑816 x Cacheado‑roxo) x AU94‑MOB‑816, com os genitores. Dois experimentos foram conduzidos no delineamento de blocos ao acaso, tendo‑se avaliado 17 progênies, com quatro repetições, em parcelas subdivididas quanto à inflorescência: simples e composta. A análise estatística foi realizada por modelos mistos via procedimento REML/BLUP. As estimativas das variâncias genéticas foram significativas para todos os caracteres estudados. Os caracteres comprimento do pedúnculo, número médio de vagens por pedúnculo e floração inicial apresentaram alta variabilidade e expressão do componente genético para a inflorescência composta. As progênies de inflorescência simples apresentam potencial genético produtivo similar às progênies de inflorescência composta. As progênies resultantes do retrocruzamento (AU94‑MOB‑816 x Cacheado‑roxo) x AU94‑MOB‑816 são promissoras como estratégia para aumentar os níveis atuais de produtividade do feijão-caupi.The objective of this work was to evaluate the genetic yield potential of segregating cowpea progenies regarding inflorescence type. Sixty‑eight F4 progenies obtained from the backcrosses (TV x 5058‑09C x Curly‑purple) x TV x 5058‑09C, and (AU94‑MOB‑816 x Curly‑purple) x AU94‑MOB‑816, with parents, were evaluated. Two experiments were carried out in a randomized complete block design with 17 progenies and four replicates, in a split‑plot arrangement regarding inflorescence: simple and composite. The statistical analysis was carried out by mixed models via REML/BLUP procedure. The estimates of genetic variance were significant for all evaluated traits. The traits peduncle length, average number of pods per peduncle, and initial flowering showed high variability and expression of the composite inflorescence genetic component. Progenies with simple inflorescence show similar genetic yield potential to the progenies with composite inflorescence. The progenies resulting from the backcrosses (AU94‑MOB‑816 x Curly‑purple) x AU94‑MOB‑816 are promising as a strategy to increase the current levels of grain yield in cowpea

    Low prevalence of renal, cardiac, pulmonary, and neurological extra-articular clinical manifestations in spondyloarthritis: analysis of the Brazilian Registry of Spondyloarthritis

    OBJECTIVE: To describe the extra-articular manifestations (cardiac, renal, pulmonary, and neurological), usually not related to spondyloarthritis (SpA), in a large cohort of Brazilian patients. MATERIALS AND METHODS: This retrospective study analyzed 1,472 patients diagnosed with SpA and cared for at 29 health care centers distributed in the five major geographic regions in the country, participating in the Brazilian Registry of Spondyloarthritis (BRS). All patients were assessed for the prevalence of major extra-articular manifestations (cardiac, renal, pulmonary, and neurological), classified according to the diagnosis [ankylosing spondylitis (AS), psoriatic arthritis (PsA), reactive arthritis (ReA), arthritis associated with inflammatory bowel disease (IBD), undifferentiated spondyloarthritis (uSpA), and juvenile SpA], and according to the clinical presentation (axial, peripheral, mixed, and enthesitis). RESULTS: Of the patients with SpA assessed, 963 had AS, 271 PsA, 49 ReA, 48 arthritis associated with IBD, 98 uSpA, and 43 juvenile SpA. Cardiac involvement was reported in 44 patients (3.0%), pulmonary involvement in 19 (1.3%), renal involvement in 17 (1.2%), and neurological involvement in 13 patients (0.9%). Most patients with visceral involvement had AS or PsA, and the mixed (axial + peripheral) and/or predominantly axial clinical form. CONCLUSION: Cardiac, renal, pulmonary, and neurological extra-articular manifestations are quite infrequent in SpA, ranging from 0.9% to 3% in this large Brazilian cohort, and affected predominantly patients with AS and PsA.OBJETIVO: Descrever as manifestações extra-articulares (cardíacas, renais, pulmonares e neurológicas) geralmente não relacionadas às espondiloartrites (EpA) em uma grande coorte de pacientes brasileiros. MÉTODOS: Este estudo retrospectivo analisou 1.472 pacientes com o diagnóstico de EpA atendidos em 29 centros distribuídos pelas cinco principais regiões geográficas do Brasil, integrantes do Registro Brasileiro de Espondiloartrites. Todos os pacientes foram avaliados para a prevalência das principais manifestações extra-articulares (cardíacas, renais, pulmonares e neurológicas), divididas por diagnóstico [espondilite anquilosante (EA), artrite psoriásica (AP), artrite reativa (ARe), artrite associada a doença inflamatória intestinal (DII), EpA indiferenciada (EI) e EpA juvenil] e por forma clínica (axial, periférica, mista e entesítica). RESULTADOS: Dentre os pacientes avaliados com EpA, 963 apresentavam EA, 271 AP, 49 ARe, 48 artrite associada a DII, 98 EI e 43 EpA juvenil. Acometimento cardíaco foi observado em 44 pacientes (3,0%), seguido por acometimento pulmonar em 19 (1,3%), renal em 17 (1,2%) e neurológico em 13 pacientes (0,9%). A maioria dos casos de acometimento visceral ocorreu nos pacientes com EA ou AP e naqueles com forma clínica mista (axial e periférica) e/ou predominantemente axial. CONCLUSÃO: As manifestações extra-articulares cardíacas, renais, pulmonares e neurológicas são muito pouco frequentes nas EpA, variando de 0,9%-3% nesta grande coorte brasileira, estando mais associadas a EA e AP.37938
