On the effects of variable leg-spring properties during hopping

A mountain ridge model for quantifying oblique mountain wave propagation and distribution
Following the current understanding of gravity waves (GWs) and especially mountain waves (MWs), they have high potential of horizontal propagation from their source. This horizontal propagation and therefore the transport of energy is usually not well represented in MW parameterizations of numerical weather prediction and general circulation models. The lack thereof possibly leads to shortcomings in the model's prediction as e.g. the cold pole bias in the Southern Hemisphere and the polar vortex breaking down too late. In this study we present a mountain wave model (MWM) for quantification of the horizontal propagation of orographic gravity waves. This model determines MW source location and associates their parameters from a fit of idealized Gaussian shaped mountains to topography data. Propagation and refraction of these MWs in the atmosphere is modeled using the ray-tracer GROGRAT. Ray-tracing each MW individually allows for an estimation of momentum transport due to both vertical and horizontal propagation. This study presents the MWM itself and gives validations of MW induced temperature perturbations to ECMWF IFS numerical weather prediction data and estimations of gravity wave momentum flux (GWMF) compared to HIRDLS satellite observations. The MWM is capable of reproducing the general features and amplitudes of both of these data sets and, in addition, is used to explain some observational features by investigating MW parameters along their trajectories.</p
Smartphone-Experimente zu gleichmĂ€Ăig beschleunigten Bewegungen mit der App phyphox
Die Bedeutung von Smartphones in der heutigen Gesellschaft nimmt stetig zu. Fast alle Jugendlichen besitzen ein eigenes Smartphone und nutzen dies tĂ€glich. Vielen ist nicht bewusst, welche verschiedenen Sensoren in den modernen Smartphones integriert sind, die insbesondere zur Erfassung verschiedenster (physikalischer) Daten verwendet werden können. Fast alle Sensoren können ĂŒber die an der RWTH Aachen entwickelte App phyphox (phyphox.org) ausgelesen werden. Zudem bietet diese App die Möglichkeit, Experimente zu editieren, die individuell auf Lerngruppen angepasst werden können. Im Beitrag werden zwei Experimente vorgestellt, die im Bereich der Mechanik fĂŒr die Sekundarstufe II des Gymnasiums entwickelt wurden und dabei den Einsatz des Smartphones mit der App phyphox nutzen. Konkret sind ein Lehrerdemonstrationsexperiment auf der Luftkissenbahn und ein SchĂŒlerexperiment fĂŒr den freien Fall konzipiert worden, in denen das Smartphone zur Erfassung von Weg-Zeit-Daten dient. In diesem Beitrag werden die App phyphox und die beiden konzipierten Experimente aus physikalischer und didaktischer Sicht dargestellt

Neben einer Reihe von vordefinierten Experimenten besteht mit einem Editor die Möglichkeit, eigene Experimente zu kreieren und bei Bedarf ĂŒber einen Fernzugriff zu steuern. Exemplarisch wurden zwei Smartphone-Experimente zu harmonischen Schwingungen fĂŒr die Sekundarstufe II entwickelt. Observations of Gravity Wave Refraction and Its Causes and Consequences
Horizontal gravity wave (GW) refraction was observed around the Andes and Drake Passage during the SouthTRAC campaign. GWs interact with the background wind through refraction and dissipation. This interaction helps to drive midatmospheric circulations and slows down the polar vortex by taking GW momentum flux (GWMF) from one location to another. The SouthTRAC campaign was composed to gain improved understanding of the propagation and dissipation of GWs. This study uses observational data from this campaign collected by the German High Altitude Long Range research aircraft on 12 September 2019. During the campaign a minor sudden stratospheric warming in the southern hemisphere occurred, which heavily influenced GW propagation and refraction and thus also the location and amount of GWMF deposition. Observations include measurements from below the aircraft by Gimballed Limb Observer for Radiance Imaging of the Atmosphere and above the aircraft by Airborne Lidar for the Middle Atmosphere. Refraction is identified in two different GW packets as low as â4 km and as high as 58 km. One GW packet of orographic origin and one of nonorographic origin is used to investigate refraction. Observations are supplemented by the Gravity-wave Regional Or Global Ray Tracer, a simplified mountain wave model, ERA5 data and high-resolution (3 km) WRF data. Contrary to some previous studies we find that refraction makes a noteworthy contribution in the amount and the location of GWMF deposition. This case study highlights the importance of refraction and provides compelling arguments that models should account for this
The von Hippel-Lindau Tumor Suppressor Protein Promotes c-Cbl-Independent Poly-Ubiquitylation and Degradation of the Activated EGFR
Somatic mutations or reduced expression of the von Hippel-Lindau (VHL) tumor suppressor occurs in the majority of the clear cell renal cell carcinoma (ccRCC) and is a causal factor for the pathogenesis of ccRCC. pVHL was reported to suppress the oncogenic activity of Epidermal Growth Factor Receptor (EGFR) by reducing the expression of the EGFR agonist TGF-α and by reducing the translation efficiency of EGFR itself. Furthermore, it was reported that pVHL down-regulates activated EGFR by promoting efficient lysosomal degradation of the receptor. These modes of negative regulation of EGFR by pVHL were dependent on Hypoxia Inducible Factor (HIF). In this study, we report that HIF was not the only factor stabilizing the activated EGFR in VHL-deficient ccRCC cells. Down-regulation of endogenous HIF in these cells had little effect on the turnover rates of the activated EGFR. Furthermore, neither pretreatment with lysomomal inhibitors pretreatment nor down-regulation of c-Cbl, a major E3 ubiquitin ligase that targets the activated EGFR for lysosomal degradation, significantly increased the stabilities of EGFR in VHL-expressing ccRCC cells. In contrast, pretreatment with proteasomal inhibitors extended EGFR lifetime and led to similar EGFR half-lives in VHL-expressing and VHL-deficient ccRCC cells. Down-regulation of c-Cbl in VHL-deficient ccRCC cells revealed that the c-Cbl and pVHL collaborated to down-regulate the activated EGFR. Finally, we found that pVHL promoted the poly-ubiquitylation of the activated EGFR, and this function was c-Cbl-independent. Thus these results indicate that pVHL limits EGFR signaling by promoting c-Cbl-independent poly-ubiquitylation of the activated receptor, which likely results in its degradation by proteasome
Tyrosine kinase signalling in breast cancer: ErbB family receptor tyrosine kinases
ERBB family receptor tyrosine kinases are overexpressed in a significant subset of breast cancers. One of these receptors, HER2/neu, or ErbB-2, is the target for a new rational therapeutic antibody, Herceptin. Other inhibitors that target this receptor, and another family member, the epidermal growth factor (EGF) receptor, are moving into clinical trials. Both of these receptors are sometimes overexpressed in breast cancer, and still subject to regulation by hormones and other physiological regulators. Optimal use of therapeutics targeting these receptors will require consideration of the several modes of regulation of these receptors and their interactions with steroid receptors

Leukozyten aus dem Blutstrom in entzĂŒndetes Gewebe. Bei der Auswanderung
(Extravasation) werden Leukozyten zuerst eingefangen und verlangsamt
(initialer Kontakt), um anschlieĂend auf dem Gewebeendothel zu rollen, fest zu
adhĂ€rieren und schlieĂlich durch das Endothel hindurch zu wandern, um den
EntzĂŒndungsherd zu erreichen. Diese gezielte Auswanderung ist ein
physiologischer Prozess. Da es durch Persistenz des Antigens bei chronischen
Erkrankungen zu einem unkontrollierten Auswandern der Leukozyten mit der Folge
eines sekundÀren Gewebeschadens kommt, ist die Möglichkeit der gezielten
Hemmung der Extravasation von groĂem medizinischen Interesse. Ein möglicher
Ansatzpunkt ist dabei die Modulierung der L-SelektinâLigand-Interaktion, da
L-Selektin, das konstitutiv auf Leukozyten exprimiert wird, eine entscheidende
Rolle wÀhrend des initialen Kontaktes der Leukozyten mit dem Endothel spielt.
Diese Interaktion wird durch Bindung der LektindomÀne des L-Selektins an sLex-
Motive und Sulfatgruppen endothelialer Liganden vermittelt. In der
vorliegenden Arbeit wurde die L-SelektinâLigand-Interaktion auf zwei
verschiedenen Wegen moduliert, wobei fĂŒr die Interaktionsstudien initial ein
Flusskammer-Assay etabliert wurde, mit dem erfolgreich die Zell / Ligand-
Interaktion quantifiziert werden konnte. Erstens wurden in intakten Zellen die
NeuraminsÀurereste der zellulÀren Glykoproteine durch Behandlung der Zellen
sowohl mit dem natĂŒrlichen Mannosamin-VorlĂ€uferderivat peracMNAc, als auch mit
den artifiziellen Derivaten peracMNProp und peracMNCycloProp verÀndert. In
diesen modifizierten Zellen wurde untersucht, ob die verÀnderte Sialylierung
zu einer verÀnderten Interaktion zwischen Selektinen und dem Liganden PSGL-1
kommt. Die Untersuchungen zeigten, dass es bei allen Behandlungen zu einer
Hypersialylierung der Zellen kam, wobei die Behandlung mit peracMNProp bzw.
peracMNCycloProp zum Einbau der strukturell verÀnderten NeuraminsÀure
N-PropanoylneuraminsĂ€ure bzw. N-Cyclopropyl-carbonyl-NeuraminsĂ€ure fĂŒhrte.
Weiterhin konnte eine verstÀrkte AdhÀsion der PSGL-1-exprimierenden Zellen an
den Liganden P-Selektin nach Supplementierung gemessen werden, die durch den
Einbau der artifiziellen SialinsÀuren noch verstÀrkt wurde. Dahingegen zeigten
die entsprechend behandelten L-Selektin-exprimierenden Zellen aufgrund der
Hypersialylierung eine verminderte Interaktion mit dem Liganden PSGL-1. Durch
die Behandlung mit den artifiziellen Derivaten kam es dabei zu einer weiteren
Verminderung der Interaktion mit PSGL-1. Zweitens wurden unterschiedliche
potentielle L-Selektin-Inhibitoren zur Modulation der Interaktion verwendet:
zwei L-Selektin-Aptamere, welche sich in der Anzahl der Bindungs-domÀne
unterschieden (Monomer und Dimer) und drei Vertreter aus der Gruppe der
dendritischen Polyglycerolsulfate (dPG2 kDaS23, dPG6 kDaS67 und dPG480
kDaS5185), die sich in der GröĂe des PolyglycerolgerĂŒstes und in der Anzahl
der funktionalisierten Sulfate unterschieden. Die dendritischen
Polyglycerolsulfate zeigten eine inhibitorische Wirkung auf die L-Selektin
âLigand-Interaktion, wobei sich die gewĂŒnschte inhibitorische Wirkung in
AbhÀngigkeit der Anzahl bindungsrelevanter Sulfate einstellte. So konnte eine
eindrucksvolle Verbesserung der Inhibition bei Verwendung von dPG480 kDaS5185
um das 1000fache gegenĂŒber dPG6 kDaS67 erzielt werden. Damit konnte gezeigt
werden, dass eine multivalente PrÀsentation der Bindungs-epitope eine
entscheidende Rolle fĂŒr die Bindung an L-Selektin spielt. Die Bindungsstudien
an den DNA-Aptameren konnten dieses Ergebnis untermauern, da die dimere
PrÀsentation des Bindungsepitops zu einer 50fachen besseren inhibitorischen
Wirkung gegenĂŒber dem monomeren Aptamer fĂŒhrt. Diese Resultate zeigen
eindeutig, dass eine gezielte Modulation der L-SelektinâLigand-Interaktion
möglich ist und dass mit den dendritschen Polyglycerolsulfaten und den DNA-
Aptamer-Varianten neuartige Inhibitoren charakterisiert wurden, die das
Potential fĂŒr eine therapeutische Anwendung besitzen.During inflammation leukocytes exit the bloodstream into the site of
inflammation (extravasation). Therefore, free floating cells are captured by
endothelial ligands, slowed down by transient interactions (initial contact)
and start rolling on the vascular endothelium and attach firmly to pass
through the endothelium and to finally access the center of inflammation. This
is a desired process during the innate immune response, but in chronic
diseases extravasation of leukocytes is misregulated and leads to tissue
damage. Therefore, it is desirable to regulate the uncontrolled leukocyte
migration, e.g. by modulation of the L-selectinâligand interaction.
L-selectin, which is constitutively expressed on leukocytes, plays an
important role for the initial contact of leukocytes to the endothelium. The
binding of L-selectin depends on the interaction of its lectin domain to the
endothel ligands containing the sLex-motif and sulfate groups. In the present
study, the modulation of the L-selectinâligand interaction was studied in two
different ways. Initially a flow chamber assay was established to quantify the
cell / ligand interaction under defined conditions. In a first step the
neuraminic acids of glycoproteins of intact cells were modified by
incorporation of the natural mannosamine derivative peracMNAc and the
unnatural derivatives peracMNProp and peracMNCycloProp. These modified cells
were examined concerning the changes in the interaction between selectins and
their ligand PSGL-1. Analyses confirmed the successful incorporation of the
sialic acid derivatives and therefore the hypersialylation of the cells. The
treatment with the unnatural derivatives peracMNProp and peracMNCycloProp lead
to the integration of the artificial neuraminic acids N-propanoyl neuraminic
acid and N-cyclopropylcarbonyl neuraminic acid into the glycan structures. The
modifications lead to improved adhesion of PSGL-1 expressing cells to
P-selectin due to a higher incorporation of sialic acids and its derivatives
in glycan structures. In contrast, the modification of the L-selectin glycans
with the sialic acid derivatives leads to a reduced interaction of L-selectin
expressing cells with the ligand PSGL-1. The second way to modulate L-selectin
interactions relied on different potential inhibitors: three different
dendritic polyglycerol sulfates (dPG2 kDaS23, dPG6 kDaS67 and dPG480
kDaS5185), which differed in the size of the polyglycerol core and in the
amount of sulfate groups, and two L-selectin-aptamers, which differed in the
presentation of the L-selectin specific binding epitop (monomer and dimer).
The application of the polyglycerol sulfates showed significant inhibition
dependent on the amount of the binding sulfates on the L-selectin-ligand
interaction. This work demonstrates thus an impressive 1000-fold stronger
inhibition by dPG480 kDaS5185 compared to dPG6 kDaS67. This reflects the
important role of multivalent presentation of the binding epitopes for the
L-selectinâligand interaction. The interaction studies with the DNA-aptamers
confirmed this observation. Compared to the monomeric aptamer, the dimeric
epitope presentation shows a 50-fold (dimer-flex) better inhibition. These
results clearly demonstrate the possibility to modulate the L-selectin-ligand
interaction specifically and present the dendritic polyglycerol sulfates and
DNA-aptamer variants as novel inhibitors with the potential for a successful
therapeutic application
