    Drivers of bird diversity in an understudied African centre of endemism:The Angolan Central Escarpment Forest

    Natural habitats are being rapidly lost due to human activities. It is therefore vital to understand how these activities influence biodiversity so that suitable guidelines can be established for conservation. This is particularly important in understudied, high biodiversity, areas such as the Angolan Escarpment. Here we examine which habitat characteristics drive bird diversity and endemic species presence at Kumbira Forest, a key site in the Central Escarpment Forest. Bird diversity was sampled by 10 min bird point counts, whereas habitat characteristics were measured by a combination of ground-based vegetation surveys and remotely sensed data modelling of Landsat images. GLM, multi-model inference and model averaging were used to determine the most important variables driving species richness and the presence of endemics. The remote sensing variables performed poorly in predicting presence of Red-crested Turaco Tauraco erythrolophus and Gabela Bushshrike Laniarius amboimensis but they contributed significantly to explain species richness and Gabela Akalat Sheppardia gabela presence, both of which were associated with greater canopy cover. Liana density and elevation were also important explanatory variables in certain cases. Conservation actions at Kumbira should focus on increasing canopy cover and maintaining forest integrity (as measured by liana density), as these actions are likely to have the most positive outcomes for the avifauna

    Effect of sequential medium on in vitro culture of goat ovarian cortical tissue

    A sequential medium was evaluated on the survival, activation and growth rates of caprine preantral follicles submitted to a long-term culture period, aiming to establish an ideal in vitro culture system. Ovarian fragments were cultured for 16 days in α-MEM+ alone or supplemented with hormones (GH and/or FSH) added sequentially on different days of culture. Ovarian fragments were cultured in the first (days 0–8) and second (days 8–16) halves of the culture period, generating 10 treatments: α-MEM+/α-MEM+, FSH/FSH, FSH/GH, FSH/FSH + GH, GH/GH, GH/FSH, GH/FSH + GH, FSH + GH/FSH + GH, FSH + GH/FSH and FSH + GH/GH. Follicle morphology, viability and ultrastructure were analyzed. After day 1 of culture, FSH treatments maintained the percentage of normal follicles similar to the fresh control. At day 16 of culture, the treatment FSH/GH showed the highest (P < 0.05) percentage of normal follicles. The ultrastructure of follicles was preserved in the fresh control and FSH/GH treatment. Follicles cultured with FSH/GH had a higher (P < 0.05) viability than α-MEM+; however the viability was lower (P < 0.05) when compared to the fresh control. The FSH/GH treatment showed the highest (P < 0.05) percentage of follicular activation and secondary follicle formation and produced the largest (P < 0.05) mean follicular diameter after 16 days of culture. In conclusion, a sequential medium supplemented with FSH followed by GH during a long-term culture maintains the survival, viability and ultrastructure of goat preantral follicles, and promotes activation and secondary follicles

    Response of patients with acute respiratory failure caused by COVID-19 to awake-prone position outside the intensive care unit based on pulmonary involvement

    OBJECTIVES: Since there are difficulties in establishing effective treatments for COVID-19, a vital way to reduce mortality is an early intervention to prevent disease progression. This study aimed to evaluate the performance of patients with COVID-19 with acute hypoxic respiratory failure according to pulmonary impairment in the awake-prone position, outside of the intensive care unit (ICU). METHODS: A prospective observational cohort study was conducted on COVID-19 patients under noninvasive respiratory support. Clinical and laboratory data were obtained for each patient before the treatment and after they were placed in the awake-prone position. To identify responders and non-responders after the first prone maneuver, receiver operating characteristic curves with sensitivity and specificity of the PaO2/FiO2&nbsp;and SpO2/FiO2&nbsp;indices were analyzed. The maneuver was considered positive if the patient did not require endotracheal intubation for ventilatory assistance. RESULTS: Forty-eight patients were included, and 64.6% were categorized as responders. The SpO2/FiO2&nbsp;index was effective for predicting endotracheal intubation in COVID-19 patients regardless of lung parenchymal damage (area under the curve 0.84, cutoff point 165, sensitivity 85%, specificity 75%). Responders had better outcomes with lower hospital mortality (hazard ratio [HR]=0.107, 95% confidence interval [CI]: 0.012-0.93) and a shorter length of stay (median difference 6 days, HR=0.30, 95% CI: 0.13-0.66) after adjusting for age, body mass index, sex, and comorbidities. CONCLUSIONS: The awake-prone position for COVID-19 patients outside the ICU can improve oxygenation and clinical outcomes regardless of the extent of pulmonary impairment. Furthermore, the SpO2/FiO2&nbsp;index discriminates responders from non-responders to the prone maneuver predicting endotracheal intubation with a cutoff under or below 165. &nbsp

    Activation of Type 1 Cannabinoid Receptor (CB1R) promotes neurogenesis in murine subventricular zone cell cultures

    The endocannabinoid system has been implicated in the modulation of adult neurogenesis. Here, we describe the effect of type 1 cannabinoid receptor (CB1R) activation on self-renewal, proliferation and neuronal differentiation in mouse neonatal subventricular zone (SVZ) stem/progenitor cell cultures. Expression of CB1R was detected in SVZ-derived immature cells (Nestin-positive), neurons and astrocytes. Stimulation of the CB1R by (R)-(+)-Methanandamide (R-m-AEA) increased self-renewal of SVZ cells, as assessed by counting the number of secondary neurospheres and the number of Sox2+/+ cell pairs, an effect blocked by Notch pathway inhibition. Moreover, R-m-AEA treatment for 48 h, increased proliferation as assessed by BrdU incorporation assay, an effect mediated by activation of MAPK-ERK and AKT pathways. Surprisingly, stimulation of CB1R by R-m-AEA also promoted neuronal differentiation (without affecting glial differentiation), at 7 days, as shown by counting the number of NeuN-positive neurons in the cultures. Moreover, by monitoring intracellular calcium concentrations ([Ca2+](i)) in single cells following KCl and histamine stimuli, a method that allows the functional evaluation of neuronal differentiation, we observed an increase in neuronal-like cells. This proneurogenic effect was blocked when SVZ cells were co-incubated with R-m-AEA and the CB1R antagonist AM 251, for 7 days, thus indicating that this effect involves CB1R activation. In accordance with an effect on neuronal differentiation and maturation, R-m-AEA also increased neurite growth, as evaluated by quantifying and measuring the number of MAP2-positive processes.     Incidence and risk factors for Preeclampsia in a cohort of healthy nulliparous pregnant women: a nested case-control study

    The objective of this study is to determine the incidence, socio-demographic and clinical risk factors for preeclampsia and associated maternal and perinatal adverse outcomes. This is a nested case-control derived from the multicentre cohort study Preterm SAMBA, in five different centres in Brazil, with nulliparous healthy pregnant women. Clinical data were prospectively collected, and risk factors were assessed comparatively between PE cases and controls using risk ratio (RR) (95% CI) plus multivariate analysis. Complete data were available for 1,165 participants. The incidence of preeclampsia was 7.5%. Body mass index determined at the first medical visit and diastolic blood pressure over 75 mmHg at 20 weeks of gestation were independently associated with the occurrence of preeclampsia. Women with preeclampsia sustained a higher incidence of adverse maternal outcomes, including C-section (3.5 fold), preterm birth below 34 weeks of gestation (3.9 fold) and hospital stay longer than 5 days (5.8 fold) than controls. They also had worse perinatal outcomes, including lower birthweight (a mean 379 g lower), small for gestational age babies (RR 2.45 [1.52-3.95]), 5-minute Apgar score less than 7 (RR 2.11 [1.03-4.29]), NICU admission (RR 3.34 [1.61-6.9]) and Neonatal Near Miss (3.65 [1.78-7.49]). Weight gain rate per week, obesity and diastolic blood pressure equal to or higher than 75 mmHg at 20 weeks of gestation were shown to be associated with preeclampsia.     Risk-factors for non-adherence to antiretroviral therapy

    Cross-sectional study analyzed as case-control to identify risk factors for non-adherence to antiretroviral therapy. We studied 412 out-clinics HIV infected subjects of three public hospitals of Recife, Pernambuco. The objective was to examine the association between non-adherence to the antiretroviral therapy and biological, social-behavior and demographics and economic factors, factors related to the disease and/or treatment, factors related to life habits and depression symptoms. Variables significantly associated with non-adherence to antiretroviral therapy were: time elapsed since HIV diagnosis (p = 0.002), daily dose (p = 0.046), use of alcohol (p = 0.030) and past drug use (p = 0.048), and borderline p-values were found for educational level (p = 0.093) and family monthly income (p = 0.08). In the multivariable analysis, the factors that remained in the final model were family monthly income, time period with HIV infection and use of alcohol. No association was observed between non-adherence to antiretroviral therapy and gender, age, sexual orientation, marital status, educational level and place of residence. Based on our results and the local situation we suggest: assessment of social needs; training of partners and/or families on supporting adherence, creation of "adherence groups" to motivate and to reassure patients on the benefits of treatment; counseling and/or psychotherapy for alcohol drinkers

    Cadastramento de doadores voluntários de Medula Óssea no REDOME

    Anais do 35º Seminário de Extensão Universitária da Região Sul - Área temática: SaúdeO transplante de medula óssea consiste na substituição de uma medula óssea doente ou deficitária por células normais, com o objetivo de reconstituição de uma nova medula. Um fator que dificulta a realização do procedimento é a falta de doador compatível. Assim, quanto maior o número de novos doadores voluntários cadastrados no REDOME (Registro Brasileiro de Doadores Voluntários de Medula Óssea), maiores são as possibilidades de o paciente encontrar um doador compatível. Nosso projeto de extensão tem como objetivo a conscientização da população sobre o cadastramento de doadores voluntários de medula óssea nas regiões de Maringá, Cianorte e Paranavaí, por meio de campanhas de captação de doadores, palestras de conscientização e divulgação sobre a doação e o transplante de medula óssea. O projeto vem colaborando no crescimento do REDOME com doadores mais conscientes sobre a responsabilidade do cadastro no banco. Assim, nossa equipe deve continuar realizando o trabalho de conscientização da população, pois quanto maior o número de doadores, maior é a possibilidade de se encontrar um doador compatíve
