    Dose-response effects of systemic anandamide administration in mice sequentially submitted to the open field and elevated plus-maze tests

    The endocannabinoid system is involved in the control of many physiological functions, including the control of emotional states. In rodents, previous exposure to an open field increases the anxiety-like behavior in the elevated plus-maze. Anxiolytic-like effects of pharmacological compounds that increase endocannabinoid levels have been well documented. However, these effects are more evident in animals with high anxiety levels. Several studies have described characteristic inverted U-shaped dose-response effects of drugs that modulate the endocannabinoid levels. However, there are no studies showing the effects of different doses of exogenous anandamide, an endocannabinoid, in animal models of anxiety. Thus, in the present study, we determined the dose-response effects of exogenous anandamide at doses of 0.01, 0.1, and 1.0 mg/kg in C57BL/6 mice (N = 10/group) sequentially submitted to the open field and elevated plus-maze. Anandamide was diluted in 0.9% saline, ethyl alcohol, Emulphor® (18:1:1) and administered ip (0.1 mL/10 g body weight); control animals received the same volume of anandamide vehicle. Anandamide at the dose of 0.1 mg/kg (but not of 0.01 or 1 mg/kg) increased (P < 0.05) the time spent and the distance covered in the central zone of the open field, as well as the exploration of the open arms of the elevated plus-maze. Thus, exogenous anandamide, like pharmacological compounds that increase endocannabinoid levels, promoted a characteristic inverted U-shaped dose-response effect in animal models of anxiety. Furthermore, anandamide (0.1 mg/kg) induced an anxiolytic-like effect in the elevated plus-maze (P < 0.05) after exposing the animals to the open field test.FAPESPCNP

    Deleção cromossómica intersticial em 14q “de novo”: apresentação de um caso

    Introdução: As deleções intersticiais são anomalias cromossómicas estruturais, desequilibradas, resultantes de dois pontos de quebra, frequentemente associadas a quadros clínicos anormais devido à perda de material genético ativo (eucromatina). As consequências fenotípicas dependem do segmento cromossómico perdido e do número de genes aí localizados. Material e Métodos: Os autores apresentam o caso de um indivíduo do sexo masculino, de 11 anos de idade, referenciado para estudo citogenético por apresentar um quadro clínico de atraso de desenvolvimento psicomotor, défice cognitivo e problemas de comportamento. Realizaram-se culturas sincronizadas de linfócitos de sangue periférico, bandas GTG de alta resolução e, posteriormente, estudos de hibridação in situ por fluorescência (FISH) com sondas de pintura cromossómica total e subtelomérica, específicas para o cromossoma 14. Resultados: A análise das metafases revelou a presença de uma anomalia estrutural no cromossoma 14, interpretada como uma deleção intersticial do segmento compreendido entre as bandas 14q24.3 e 14q32.1. A análise por FISH permitiu confirmar esta deleção intersticial. Como os cariótipos dos pais foram normais, conclui-se que esta anomalia cromossómica é “de novo”, estabelecendo-se o cariótipo do doente como: 46,XY,del(14)(q24.3q32.1).ish del(14)(wcp 14+,SHGC36156+)dn Discussão: A deleção intersticial encontrada no cromossoma 14 implica uma monossomia do segmento 14q24.3→14q32.1. As alterações descritas mais comuns, associadas a esta deleção, incluem ADPM e algumas malformações minor.     A case study undertaken recycling & reuse of glass fiber reinforced thermoset polumer wastes of composite materials industry

    Glass fibre-reinforced plastics (GFRP) have been considered inherently difficult to recycle due to both: crosslinked nature of thermoset resins, which cannot be remoulded, and complex composition of the composite itself. Presently, most of the GFRP waste is landfilled leading to negative environmental impacts and supplementary added costs. With an increasing awareness of environmental matters and the subsequent desire to save resources, recycling would convert an expensive waste disposal into a profitable reusable material. In this study, efforts were made in order to recycle grinded GFRP waste, proceeding from pultrusion production scrap, into new and sustainable composite materials. For this purpose, GFRP waste recyclates, were incorporated into polyester based mortars as fine aggregate and filler replacements at different load contents and particle size distributions. Potential recycling solution was assessed by mechanical behaviour of resultant GFRP waste modified polymer mortars. Results revealed that GFRP waste filled polymer mortars present improved flexural and compressive behaviour over unmodified polyester based mortars, thus indicating the feasibility of the GFRP industrial waste reuse into concrete-polymer composite materials

    Efeitos protetores do choque térmico na sepse e na síndrome da resposta inflamatória sistêmica

    Sepsis shock, multiorgan dysfunction and the acute respiratory distress syndrome are major contributors to morbidity and mortality in intensive care units. Animal studies have shown that these forms of injury can be attenuated or prevented if a phenomenon, called the stress response, is activated. The stress response, characterized by a transient downregulation of most cellular products and the upregulation of heat shockproteins (HSP), has been shown to provide protection to cells and experimental animals if triggered prior to or at an otherwise lethal injury. The mechanisms by which the stress response is protective are not known with certainty, but HSP appear to play an important role. This article summarizes the current literature on the effects of the stress response in protecting cells, organ systems and laboratory animals from lethal forms of systemicand organic damage.Sepse, disfunção orgânica múltipla e a síndrome da angústia respiratória aguda são as maiores causas e mortalidade nas unidades de terapia intensiva. Estudos em animais de laboratório demonstram que esta forma de injúria pode ser atenuada ou mesmo abolida se um fenômeno, conhecido como resposta ao estresse ou resposta ao choque térmico, for ativado. A resposta ao choque térmico, caracterizada pela suspensão transitória da produção da maioria das proteínas e pela ativação das proteínas do choque térmico (heat shock proteins - HSP), mostrou-se protetora a células e a animais de laboratório se desencadeada antes, ou imediatamente, que uma injúria letal. Os mecanismos pelos quais a resposta ao choque térmico é protetora não são claros,mas as proteínas do choque térmico parecem exercer um papel fundamental.     Sustainability Improvement of a composite materials’ industry through recycling and re-engineering process approaches

    Recent Advances in Mechanics and Materials in DesignThis case study was aimed at measuring and assessing the potential improvements that could be made on the eco-efficiency performance of a composite materials’ industry, specifically a glass fibre reinforced plastic (GFRP) pultrusion manufacturing company. For this purpose, all the issues involved in the pultrusion process of GFRP profiles were analysed, the current ecoefficiency performance of the company was determined, all the procedures applied in the production process were revised, and improvement strategies were planned and investigated with basis on the performed analysis. The new eco-efficiency ratios were estimated taking into account the implementation of new proceedings and procedures through re-engineering the manufacturing process and recycling approaches. These features lead to significant improvements on the sequent assessed eco-efficiency ratios, yielding to a more sustainable product and manufacturing process of pultruded GFRP profiles

    Enantioselective Determination of Fluoxetine and Norfluoxetine in Wastewater

    Microbial degradation of chiral compounds during wastewater treatment processes can be enantioselective and needs chiral analytical methodology to discriminate the biodegradation of both enantiomers. An enantioselective HPLC-FD method was developed and validated to monitor the degradation of fluoxetine (FLX) enantiomers by wastewater and the possible formation of its metabolite norfluoxetine (NFLX). The Solid Phase Extraction (SPE) of 50 mL of wastewater samples on 500 mg Oasis MCX cartridges was followed by the HPLC analysis using a Chirobiotic V chiral stationary phase under reversed mode. The developed method wasvalidated within the wastewater effluent used in microcosms laboratory assays. The chiral SPE-HPLC-FD method demonstrated to be selective, linear, sensitive, accurate and precise to quantify the enantiomers of FLX and of its metabolites NFLX in wastewaters. The limits of detection (0.8-2.0 ng mL -1 ) and quantification (2.0 – 4.0 ng mL -1 ) were adequate to monitoring the degradation assays at environmental level. The method proved to be robust to follow the biodegradation assays using real wastewater samplesspiked with FLX, during 46 days.     Identification of a mosaic non-inherited small supernumerary ring chromosome 2: cytogenetic-molecular studies and genotype-phenotype correlation

    Introduction: The identification of supernumerary marker chromosomes (SMCs) derived from all the autosomes is currently possible, but rarely by conventional cytogenetics alone. Supernumerary ring chromosomes (SRCs) account for about 10% of these cases. SRCs derived from chromosome 2 are unusual, and there are only a few cases reported in the literature. The severity of the phenotype depends on the type of the mosaicism, the percentage of cells affected by the genetic change and the chromosome involved. Methods: The authors report the case of a boy aged 8 referred for cytogenetic studies, presenting with behavior and learning problems, mental retardation with uncoordenated speech, attention deficit and hyperactivity (PHDA), as well as small slanting palpebral fissures. The karyotype was obtained from peripheral blood lymphocyte cultures using high resolution GTL banding and standard techniques. Fluorescence in situ hybridization (FISH) was performed using specific probes for the centromeric regions of all chromosomes (Chromoprobe Multiprobe - ISystem). Results: Cytogenetic analysis revealed two cell lines: one with a supernumerary marker ring chromosome, 47,XY,+r (52%), and a normal cell line, 46,XY (48%). The SRC was identified by FISH with the chromosome 2 centromeric probe. Since the parents had normal karyotypes, this abnormality was “de novo”. Final karyotype of the proband was: mos 47,XY,+r[26]/46,XY[24].ish r(2)(D2Z2+)dn. Discussion: The clinical description of this patient is in agreement with other reports of the literature. Molecular characterization by FISH analyses is an useful way of investigating the presence of euchromatin contained in a SMC and establishing new chromosomal syndromes. However, to better characterize this ring, in order to establish a more accurate genotype-phenotype correlation, more studies involving other technologies should be performed, thus allowing suitable genetic counsellin

    This special issue integrates eleven papers focussing on indicators able to convey the multiple expectations that society has concerning agricultural land. As scale issues have been so far overlooked within this research topic, a particular focus of this themed issue is to highlight the need for reconciling assessments across scales. The shared purpose of the contributions is to examine how the multiple societal expectations concerning agrarian landscapes might be incorporated into land use policy at different governance levels. A core set of these papers were presented in the symposium “Linking social indicators across scales”, to the European Congress of the International Association of Landscape Ecology (IALE) in September 2013 in Manchester, UK. This symposium was organized to create the time and space for a discussion on the issues of scale when capturing societal preferences concerning agrarian landscapes. While the landscape scale was specifically addressed by four papers, seven papers examined broader geographic units in addition to up-scaling and downscaling issues

    Enantioselective Degradation of Enantiomers of Fluoxetine Followed by HPLC- FD

    Environmental fate assessment of chiral pharmaceuticals is an important issue and little information is known about enantioselectivity in the environment. This kind of information is important for regulamentation of pharmaceutical industry and chiral switching processes. Fluoxetine (FLX), an anti-depressant worldwide used, is a chiral pharmaceutical prescribed in racemic form, and its main metabolite norfluoxetine (NFLX) is also chiral. In this study, enantioselective degradation of rac-FLX and degradation of its enantiomers separately, in a minimal salts medium inoculated by a bacterium consortium was examined both at light and dark conditions. Theassays were performed in a shaker at aerobic and ambient temperature conditions. The analytical method used was an enantioselective HPLC-FD method using a vancomycin-based chiral stationary phase in reversed mode to monitor enantiomers of FLX and NFLX. No degradation of enantiomers of FLX in the abiotic controls was observed. In theall assays (R)-FLX was degraded faster and totally until day 24th while (S)-FLX remained up to 20% of its initial concentration until the end of the experiment (38 days).
