    The genetic legacy of fragmentation and overexploitation in the threatened medicinal African pepper-bark tree Warburgia salutaris

    The pepper-bark tree (Warburgia salutaris) is one of the most highly valued medicinal plant species worldwide. Native to southern Africa, this species has been extensively harvested for the bark, which is widely used in traditional health practices. Illegal harvesting coupled with habitat degradation has contributed to fragmentation of populations and a severe decline in its distribution. Even though the species is included in the IUCN Red List as Endangered, genetic data that would help conservation efforts and future re-introductions are absent. We therefore developed new molecular markers to understand patterns of genetic diversity, structure, and gene flow of W. salutaris in one of its most important areas of occurrence (Mozambique). In this study, we have shown that, despite fragmentation and overexploitation, this species maintains a relatively high level of genetic diversity supporting the existence of random mating. Two genetic groups were found corresponding to the northern and southern locations. Our study suggests that, if local extinctions occurred in Mozambique, the pepper-bark tree persisted in sufficient numbers to retain a large proportion of genetic diversity.     Lexical, morphological and syntactic development in toddlers between 16 and 30 months old: a comparison across European Portuguese and Galician

    The main aims of this study were to investigate the relationship between the lexical size and the emergence of morphological and syntactic markers in toddlers between the ages of 16 and 30 months and to compare these results between Galician and European Portuguese. Parents of 3012 Portuguese toddlers and those of 1081 Galician toddlers completed the MacArthur–Bates Communicative Development Inventory: Words and Sentences. The results indicated that the number of words, the ability to combine words and the number of different morphemes produced increased with age. The ability to combine words was used as an indicator of syntactic development; this ability was also associated with the toddlers’ lexical size. In both samples, gender morphemes seemed to be the first to have their production generalized, followed by the plural and the past participle. The production of gender morphemes was accompanied by a small lexical size, whereas the imperfect tense and the person mark onset were associated with large lexical sizes.     Acute cardiometabolic effects of brief active breaks in sitting for patients with rheumatoid arthritis

    Exercise is a treatment in rheumatoid arthritis, but participation in moderate-to-vigorous exercise is challenging for some patients. Light-intensity breaks in sitting could be a promising alternative. We compared the acute effects of active breaks in sitting with those of moderate-to-vigorous exercise on cardiometabolic risk markers in patients with rheumatoid arthritis. In a crossover fashion, 15 women with rheumatoid arthritis underwent three 8-h experimental conditions: prolonged sitting (SIT), 30-min bout of moderate-to-vigorous exercise followed by prolonged sitting (EX), and 3-min bouts of light-intensity walking every 30 min of sitting (BR). Postprandial glucose, insulin, c-peptide, triglycerides, cytokines, lipid classes/subclasses (lipidomics), and blood pressure responses were assessed. Muscle biopsies were collected following each session to assess targeted proteins/genes. Glucose [−28% in area under the curve (AUC), P = 0.036], insulin (−28% in AUC, P = 0.016), and c-peptide (−27% in AUC, P = 0.006) postprandial responses were attenuated in BR versus SIT, whereas only c-peptide was lower in EX versus SIT (−20% in AUC, P = 0.002). IL-1β decreased during BR, but increased during EX and SIT (P = 0.027 and P = 0.085, respectively). IL-1ra was increased during EX versus BR (P = 0.002). TNF-α concentrations decreased during BR versus EX (P = 0.022). EX, but not BR, reduced systolic blood pressure (P = 0.013). Lipidomic analysis showed that 7 of 36 lipid classes/subclasses were significantly different between conditions, with greater changes being observed in EX. No differences were observed for protein/gene expression. Brief active breaks in sitting can offset markers of cardiometabolic disturbance, which may be particularly useful for patients who may find it difficult to adhere to exercise. NEW & NOTEWORTHY Exercise is a treatment in rheumatoid arthritis but is challenging for some patients. Light-intensity breaks in sitting could be a promising alternative. Our findings show beneficial, but differential, cardiometabolic effects of active breaks in sitting and exercise in patients with rheumatoid arthritis. Breaks in sitting mainly improved glycemic and inflammatory markers, whereas exercise improved lipidomic and hypotensive responses.     Estudos paleoambientais interdisciplinares: dinâmica da vegetação, do ambiente marinho e inferências climáticas milenares a atuais na Costa Norte do Espírito Santo, Brasil

    Estudos paleoambientais desde ~50.000 anos na costa do Brasil e, em particular, no litoral do Espírito Santo, são ainda insuficientes para servir de base a reconstituições da dinâmica da vegetação, de oscilações do nível relativo do mar e de flutuações climáticas e respectivas influências sobre a ação humana milenar. Para obter essas informações, uma equipe interdisciplinar, financiada por projetos temáticos FAPESP e CNPq, desenvolveu pesquisas correlatas na Reserva Natural Vale (RNV) e região. Para a caracterização da dinâmica da vegetação e marinha, com inferências climáticas, em locais de floresta de tabuleiros e campos naturais da RNV e região desde ~16.000 anos, utilizaram-se isótopos do C (12C, 13C e 14C) da matéria orgânica do solo e sedimentar, além de palinologia em sedimentos lacustres e terrestres. No estudo da dinâmica do ecótono floresta – campo, apresentam-se inferências preliminares sobre a evolução pedogenética dos Espodossolos associados ao campo, com ênfase às suas características físico-químicas, e também dos Argissolos, encontrados sob floresta.     Enhanced Leishmania braziliensis Infection Following Pre-Exposure to Sandfly Saliva

    Parasites of the genus Leishmania cause a variety of diseases known as leishmaniasis, that are transmitted by bites of female sand flies that, during blood-feeding, inject humans with parasites and saliva. It was shown that, in mice, immunity to sand-fly saliva is able to protect against the development of leishmaniasis. We have investigated, in the present study, whether this finding extends the sand fly species Lutzomyia intermedia, which is responsible for transmission of Leishmania braziliensis, a parasite species able to cause destructive skin lesions that can be fatal if left untreated. We observed that mice injected with sand fly saliva develop a specific immune response against salivary proteins. Most importantly, however, this immune response was unable to protect mice against a challenge infection with L. braziliensis, indicating that exposure to this sand fly saliva is harmful to the host. Indeed, subjects with cutaneous leishmaniasis have a higher immune response against L. intermedia saliva. These findings indicate that the anti-saliva immune response to sand fly saliva plays an important role in the outcome of leishmaniasis caused by L. braziliensis, in both mice and humans, and emphasize possible hurdles in the development of vaccines based on sand fly saliva

    Promover a fluência em leitura: um estudo com alunos do 2º ano de escolaridade

    Neste artigo são analisados os resultados de um Programa de Promoção da Fluência em Leitura (PPFL), implementado junto de 74 alunos do 2º ano de escolaridade de um Agrupamento Escolar do norte de Portugal. O PPFL é constituído por 22 sequências didáticas, elaboradas a partir de 22 textos (9 narrativos, 4 informativos e 9 poemas). Cada sequência foi operacionalizada em sessões de 10 a 15 minutos, durante 22 semanas, em ciclos de cinco dias (quinta-feira a quarta-feira). Em cada semana foi trabalhado apenas uma sequência didáctica (i.e. um texto). Foi utilizado um design quase experimental, com grupo experimental e grupo de controlo e com pré e pós-teste. Os sujeitos foram avaliados através de um teste de fluência de leitura – o Teste de Fluência em Leitura (TFL) –, considerando as variáveis velocidade e precisão. Os resultados evidenciam diferenças significativas a favor do grupo experimental, que superou as diferenças iniciais que se registavam no pré-teste.     PCR colorimetric dot-blot assay and clinical pretest probability for diagnosis of Pulmonary Tuberculosis in Smear-Negative patients

    <p>Abstract</p> <p>Background</p> <p>Smear-negative pulmonary tuberculosis (SNPTB) accounts for 30% of Pulmonary Tuberculosis (PTB) cases reported annually in developing nations. Polymerase chain reaction (PCR) may provide an alternative for the rapid detection of <it>Mycobacterium tuberculosis </it>(MTB); however little data are available regarding the clinical utility of PCR in SNPTB, in a setting with a high burden of TB/HIV co-infection.</p> <p>Methods</p> <p>To evaluate the performance of the PCR dot-blot in parallel with pretest probability (Clinical Suspicion) in patients suspected of having SNPTB, a prospective study of 213 individuals with clinical and radiological suspicion of SNPTB was carried out from May 2003 to May 2004, in a TB/HIV reference hospital. Respiratory specialists estimated the pretest probability of active disease into high, intermediate, low categories. Expectorated sputum was examined by direct microscopy (Ziehl-Neelsen staining), culture (Lowenstein Jensen) and PCR dot-blot. Gold standard was based on culture positivity combined with the clinical definition of PTB.</p> <p>Results</p> <p>In smear-negative and HIV subjects, active PTB was diagnosed in 28.4% (43/151) and 42.2% (19/45), respectively. In the high, intermediate and low pretest probability categories active PTB was diagnosed in 67.4% (31/46), 24% (6/25), 7.5% (6/80), respectively. PCR had sensitivity of 65% (CI 95%: 50%–78%) and specificity of 83% (CI 95%: 75%–89%). There was no difference in the sensitivity of PCR in relation to HIV status. PCR sensitivity and specificity among non-previously TB treated and those treated in the past were, respectively: 69%, 43%, 85% and 80%. The high pretest probability, when used as a diagnostic test, had sensitivity of 72% (CI 95%:57%–84%) and specificity of 86% (CI 95%:78%–92%). Using the PCR dot-blot in parallel with high pretest probability as a diagnostic test, sensitivity, specificity, positive and negative predictive values were: 90%, 71%, 75%, and 88%, respectively. Among non-previously TB treated and HIV subjects, this approach had sensitivity, specificity, positive and negative predictive values of 91%, 79%, 81%, 90%, and 90%, 65%, 72%, 88%, respectively.</p> <p>Conclusion</p> <p>PCR dot-blot associated with a high clinical suspicion may provide an important contribution to the diagnosis of SNPTB mainly in patients that have not been previously treated attended at a TB/HIV reference hospital.</p

    Effects of different types of verbal encouragement on ankle force and muscle activity

    The aim of this study is to investigate (i) the effect of live and recorded verbal encouragement on muscle activity and ankle force; (ii) the effect of communication/extroversion on the variables; (iii) the reliability intra and inter examiners of the variables. Twenty healthy-youngers were assessed by surface electromyography of tibialis anterior and ankle flexion force by an ergometer twice, with one week apart. No difference was found between ankle force (p = 0.373) and root mean square values (RMS) (p = 0.207) for any of the conditions assessed on day 1 nor between examiners 1 and 2 for both live and recorded conditions in RMS (p = 0.207) and force (p = 0.373). Between the 1st and 7th days, there were no differences for any of the conditions on RMS (main effect “Day” p = 0.261, “condition” p = 0.568, interaction p = 0.936) or force (main effect“Day” p = 0.889, “condition” p = 0.781, interaction p = 0.961). Intraclass correlation coefficients (ICCs) for the ankle force were, for without verbal encouragement (ICC2, k = 0.880), live verbal encouragement of examiner 1 (ICC2, k = 0.870), and recorded verbal encouragement of examiner 1 (ICC2, k = 0.920). RMS without verbal encouragement condition (ICC2, k = 0.860), live verbal encouragement of examiner 1 (ICC2, k = 0.930) and recorded verbal encouragement of examiner 1 (ICC2, k = 0.920). Reproducibility between the two examiner’s live encouragements for ankle force (ICC3, k = 0.981) and RMS (ICC3, k = 0.920). There was no effect of the presence or type of the augmented feedback in RMS and ankle force. We conclude that verbal encouragement does not influence ankle torque or muscle activity and there is good to excellent intra and inter rater reliability for subjects’ performance regardless of verbal encouragement modality.     A comprehensive assessment of the transcriptome of cork oak (Quercus suber) through EST sequencing

    Background: Cork oak (Quercus suber) is one of the rare trees with the ability to produce cork, a material widely used to make wine bottle stoppers, flooring and insulation materials, among many other uses. The molecular mechanisms of cork formation are still poorly understood, in great part due to the difficulty in studying a species with a long life-cycle and for which there is scarce molecular/genomic information. Cork oak forests are of great ecological importance and represent a major economic and social resource in Southern Europe and Northern Africa. However, global warming is threatening the cork oak forests by imposing thermal, hydric and many types of novel biotic stresses. Despite the economic and social value of the Q. suber species, few genomic resources have been developed, useful for biotechnological applications and improved forest management. Results: We generated in excess of 7 million sequence reads, by pyrosequencing 21 normalized cDNA libraries derived from multiple Q. suber tissues and organs, developmental stages and physiological conditions. We deployed a stringent sequence processing and assembly pipeline that resulted in the identification of ~159,000 unigenes. These were annotated according to their similarity to known plant genes, to known Interpro domains, GO classes and E.C. numbers. The phylogenetic extent of this ESTs set was investigated, and we found that cork oak revealed a significant new gene space that is not covered by other model species or EST sequencing projects. The raw data, as well as the full annotated assembly, are now available to the community in a dedicated web portal at http://www.corkoakdb.org. Conclusions: This genomic resource represents the first trancriptome study in a cork producing species. It can be explored to develop new tools and approaches to understand stress responses and developmental processes in forest trees, as well as the molecular cascades underlying cork differentiation and disease response.Peer Reviewe
