    S-glutathionylation of buccal cell proteins as biomarkers of exposure to hydrogen peroxide

    AbstractBackgroundExogenous or endogenous hydrogen peroxide (H2O2) is a reactive oxygen species (ROS) that can lead to oxidation of cellular nucleophiles, particularly cysteines in proteins. Commercial mouthwashes containing H2O2 provide the opportunity to determine clinically whether changes in S-glutathionylation of susceptible proteins in buccal mucosa cells can be used as biomarkers of ROS exposure.MethodsUsing an exploratory clinical protocol, 18 disease-free volunteers rinsed with a mouthwash containing 1.5% H2O2 (442mM) over four consecutive days. Exfoliated buccal cell samples were collected prior and post-treatment and proteomics were used to identify S-glutathionylated proteins.ResultsFour consecutive daily treatments with the H2O2-containing mouthwash induced significant dose and time-dependent increases in S-glutathionylation of buccal cell proteins, stable for at least 30min following treatments. Elevated levels of S-glutathionylation were maintained with subsequent daily exposure. Increased S-glutathionylation preceded and correlated with transcriptional activation of ROS sensitive genes, such as ATF3, and with the presence of 8-hydroxy deoxyguanosine. Data from a human buccal cell line TR146 were consistent with the trial results. We identified twelve proteins that were S-glutathionylated following H2O2 exposure.ConclusionsBuccal cells can predict exposure to ROS through increased levels of S-glutathionylation of proteins. These post-translationally modified proteins serve as biomarkers for the effects of H2O2 in the oral cavity and in the future, may be adaptable as extrapolated pharmacodynamic biomarkers for assessing the impact of other systemic drugs that cause ROS and/or impact redox homeostasis.General significanceS-glutathionylation of buccal cell proteins can be used as a quantitative measure of exposure to ROS

    Background: Periodontal disease is an inflammatory disease of the tissues supporting the teeth. Women who have periodontal disease while pregnant may be at risk of adverse pregnancy outcomes. Although the association between periodontal disease and adverse pregnancy outcomes has been addressed in a considerable number of systematic reviews and meta-analyses, there are important differences in the conclusions of these reviews. Systematic reviews assessing the effectivity of various therapeutic interventions to treat periodontal disease during pregnancy to try and reduce adverse pregnancy outcomes have also arrived at different conclusions. We aim to provide a systematic overview of systematic reviews comparing the frequency of adverse pregnancy outcomes between women with and without periodontal disease and/or evaluating the effect of preventive and therapeutic interventions for periodontal disease before or during pregnancy on adverse pregnancy outcomes. Methods: We will include systematic reviews reporting on studies comparing adverse pregnancy outcomes: (i) between women with or without periodontal disease before (<6 months) or during pregnancy and/or (ii) according to preventive or therapeutic interventions for periodontal disease. Eligible interventions include (combinations of) the following: oral hygiene education, use of antibiotics, subgingival scaling, and root planing. For preventive and/or therapeutic reviews, the following comparisons will be considered: no intervention, a placebo intervention, or an alternative intervent

    Maslinic Acid, a Natural Triterpene, Induces a Death Receptor-Mediated Apoptotic Mechanism in Caco-2 p53-Deficient Colon Adenocarcinoma Cells

    Maslinic acid (MA) is a natural triterpene present in high concentrations in the waxy skin of olives. We have previously reported that MA induces apoptotic cell death via the mitochondrial apoptotic pathway in HT29 colon cancer cells. Here, we show that MA induces apoptosis in Caco-2 colon cancer cells via the extrinsic apoptotic pathway in a dose-dependent manner. MA triggered a series of effects associated with apoptosis, including the cleavage of caspases -8 and -3, and increased the levels of t-Bid within a few hours of its addition to the culture medium. MA had no effect on the expression of the Bax protein, release of cytochrome-c or on the mitochondrial membrane potential. This suggests that MA triggered the extrinsic apoptotic pathway in this cell type, as opposed to the intrinsic pathway found in the HT29 colon-cancer cell line. Our results suggest that the apoptotic mechanism induced in Caco-2 may be different from that found in HT29 colon-cancer cells, and that in Caco-2 cells MA seems to work independently of p53.     Perspectiva de futuro, patrones de aprendizaje y rendimiento académico en estudiantes universitarios mexicanos

    The purpose of this study was to analyze the relationship between future perspective, the adoption of different learning patterns, and academic performance of university students. Participants were 175 students, average age 20, who responded to the Time Perspective Inventory (ZTPI) and the Inventory of Learning Styles (ILS). Results show that learning patterns play a mediating role between future perspectives and academic performance. We suggest the promotion of more equitable teaching contexts that affect the adoption of learning patterns aimed at understanding of meanings.Este estudio tuvo como propósito analizar la relación entre la perspectiva de futuro, la dominancia de distintos patrones de aprendizaje y el rendimiento académico en estudiantes universitarios. Participaron 175 estudiantes, con edad media de 20 años, que respondieron al Inventario de Perspectiva Temporal (ZTPI) y al Inventario de Estilos de Aprendizaje (ILS). Los resultados muestran que existe un papel mediador de los patrones de aprendizaje entre la perspectiva de futuro y el rendimiento académico. Se sugiere generar contextos de enseñanza que incidan en la adopción del patrón de aprendizaje dirigido a la comprensión de significados

    Diversidad genética y distribución regional de cepas de Mycobacterium bovis del ganado en México

    The molecular fingerprints of 878 isolates of Mycobacterium bovis from cattle, mostly dairy cattle, collected from cattle between 2009 and 2010 in different regions of Mexico were obtained by spoligotyping. Seventy-two percent (72 %) of the spoligotypes fell into nine clusters, and 27 % of the isolates fell into only two spoligotypes; 149 were orphan spoligotypes. The two predominant spoligotypes, arbitrarily designated as SP1 and SP2, were found in almost all States in Mexico, especially in central Mexico, where a concentration of dairy cattle is known. In spite of the wide distribution of spoligotypes observed, some show high regional preference, especially those in geographically distant regions. Only a few spoligotypes show patterns completely different from those shown by the most frequent spoligotypes, suggesting strange sources of infection or the formation of new genetic lines derived from non-lethal mutations. Most States with predominantly high dairy cattle populations showed similar spoligotypes, suggesting exchange of animals between regions. Some spoligotypes are common to dairy and beef cattle, suggesting transmission between populations, most probably due to the movement of dairy cattle to non-dairy regions.Se obtuvieron patrones moleculares (espoligotipos) de 878 aislados de Mycobacterium bovis de ganado de diferentes regiones de México entre los años 2009 y 2010. Setenta y dos por ciento (72 %) de los espoligotipos cayeron en nueve grupos y 27 % de los aislados dentro de sólo dos espoligotipos; 149 fueron espoligotipos individuales. Los dos espoligotipos predominantes, arbitrariamente identificados como SP1 y SP2, se distribuyen en la mayor parte del territorio nacional, en especial en la zona centro de México en ganado especializado en producción de leche. A pesar de la amplia distribución geográfica de los espoligotipos de mayor frecuencia, algunos muestran cierta localización, en especial los encontrados en zonas geográficas distantes, como es el caso de Chihuahua y Baja California. Aunque pocos, algunos espoligotipos muestran patrones moleculares distintos a los mostrados por los espoligotipos de mayor frecuencia, sugiriendo fuentes de infección desconocida. La mayoría de los Estados con ganadería predominantemente lechera muestran espoligotipos comunes, lo que sugiere intercambio regional frecuente de ganado. Algunos espoligotipos son comunes en ganado para leche y ganado para carne, lo que sugiere transmisión entre estas dos poblaciones; se desconoce, sin embargo, si los animales de carne infectados provienen de explotaciones extensivas o si son de engordas ubicadas dentro de la explotación lechera. Se propone la tipificación rutinaria de aislados de M. bovis obtenidos en todos los laboratorios de diagnóstico y mejorar la captura de información epidemiológica de los casos, para hacer mejores conclusiones epidemiológicas de la distribución espacial de las cepas de este agente en el territorio nacional

    Country-level gender inequality is associated with structural differences in the brains of women and men

    男女間の不平等と脳の性差 --男女間の不平等は脳構造の性差と関連する--. 京都大学プレスリリース. 2023-05-10.Gender inequality across the world has been associated with a higher risk to mental health problems and lower academic achievement in women compared to men. We also know that the brain is shaped by nurturing and adverse socio-environmental experiences. Therefore, unequal exposure to harsher conditions for women compared to men in gender-unequal countries might be reflected in differences in their brain structure, and this could be the neural mechanism partly explaining women’s worse outcomes in gender-unequal countries. We examined this through a random-effects meta-analysis on cortical thickness and surface area differences between adult healthy men and women, including a meta-regression in which country-level gender inequality acted as an explanatory variable for the observed differences. A total of 139 samples from 29 different countries, totaling 7, 876 MRI scans, were included. Thickness of the right hemisphere, and particularly the right caudal anterior cingulate, right medial orbitofrontal, and left lateral occipital cortex, presented no differences or even thicker regional cortices in women compared to men in gender-equal countries, reversing to thinner cortices in countries with greater gender inequality. These results point to the potentially hazardous effect of gender inequality on women’s brains and provide initial evidence for neuroscience-informed policies for gender equality

    The Helicobacter pylori Genome Project : insights into H. pylori population structure from analysis of a worldwide collection of complete genomes

    Helicobacter pylori, a dominant member of the gastric microbiota, shares co-evolutionary history with humans. This has led to the development of genetically distinct H. pylori subpopulations associated with the geographic origin of the host and with differential gastric disease risk. Here, we provide insights into H. pylori population structure as a part of the Helicobacter pylori Genome Project (HpGP), a multi-disciplinary initiative aimed at elucidating H. pylori pathogenesis and identifying new therapeutic targets. We collected 1011 well-characterized clinical strains from 50 countries and generated high-quality genome sequences. We analysed core genome diversity and population structure of the HpGP dataset and 255 worldwide reference genomes to outline the ancestral contribution to Eurasian, African, and American populations. We found evidence of substantial contribution of population hpNorthAsia and subpopulation hspUral in Northern European H. pylori. The genomes of H. pylori isolated from northern and southern Indigenous Americans differed in that bacteria isolated in northern Indigenous communities were more similar to North Asian H. pylori while the southern had higher relatedness to hpEastAsia. Notably, we also found a highly clonal yet geographically dispersed North American subpopulation, which is negative for the cag pathogenicity island, and present in 7% of sequenced US genomes. We expect the HpGP dataset and the corresponding strains to become a major asset for H. pylori genomics

    Reducing the environmental impact of surgery on a global scale: systematic review and co-prioritization with healthcare workers in 132 countries

    Abstract Background Healthcare cannot achieve net-zero carbon without addressing operating theatres. The aim of this study was to prioritize feasible interventions to reduce the environmental impact of operating theatres. Methods This study adopted a four-phase Delphi consensus co-prioritization methodology. In phase 1, a systematic review of published interventions and global consultation of perioperative healthcare professionals were used to longlist interventions. In phase 2, iterative thematic analysis consolidated comparable interventions into a shortlist. In phase 3, the shortlist was co-prioritized based on patient and clinician views on acceptability, feasibility, and safety. In phase 4, ranked lists of interventions were presented by their relevance to high-income countries and low–middle-income countries. Results In phase 1, 43 interventions were identified, which had low uptake in practice according to 3042 professionals globally. In phase 2, a shortlist of 15 intervention domains was generated. In phase 3, interventions were deemed acceptable for more than 90 per cent of patients except for reducing general anaesthesia (84 per cent) and re-sterilization of ‘single-use’ consumables (86 per cent). In phase 4, the top three shortlisted interventions for high-income countries were: introducing recycling; reducing use of anaesthetic gases; and appropriate clinical waste processing. In phase 4, the top three shortlisted interventions for low–middle-income countries were: introducing reusable surgical devices; reducing use of consumables; and reducing the use of general anaesthesia. Conclusion This is a step toward environmentally sustainable operating environments with actionable interventions applicable to both high– and low–middle–income countries
