    Constraining the nature of the accreting binary in CXOGBS J174623.5-310550

    We report optical and infrared observations of the X-ray source CXOGBS J174623.5-310550. This Galactic object was identified as a potential quiescent low-mass X-ray binary accreting from an M-type donor on the basis of optical spectroscopy and the broad Halpha emission line. The analysis of X-shooter spectroscopy covering 3 consecutive nights supports an M2/3-type spectral classification. Neither radial velocity variations nor rotational broadening is detected in the photospheric lines. No periodic variability is found in I- and r'-band light curves. We derive r' = 20.8, I = 19.2 and Ks = 16.6 for the optical and infrared counterparts with the M-type star contributing 90% to the I-band light. We estimate its distance to be 1.3-1.8 kpc. The lack of radial velocity variations implies that the M-type star is not the donor star in the X-ray binary. This could be an interloper or the outer body in a hierarchical triple. We constrain the accreting binary to be a < 2.2 hr orbital period eclipsing cataclysmic variable or a low-mass X-ray binary lying in the foreground of the Galactic Bulge.Comment: (9 pages, 5 figures, accepted for publication in MNRAS

    Estudio de las funciones dieléctricas del semiconductor sulfuro de plomo

    Hemos calculado la estructura electronica con la formulacion de Iii funcional densidad, del semiconductor PbS. A partir de ella, calculamos la variacion en frecuencia de las funciones dielectricas en el espectro comprendido entre 0 y 5 eV. Los picos obtenidos, asi como la forma general de las curvas correspondientes a las partes real epsilon1 e imaginaria epsilon2, resultan muy bien caracterizadas y entendidos en base a los diagram de bandas y densidades de estados calculados. El espectro dispersivo tiene su punto de inflexion en razonable acuerdo con la energia a la cual la absorcion aJcanza su maximo y comienza a disminuir, correspondiendo a un comportamiento metalico en el rango de las energfas mayores a 3 eV. Tambien presentamos resultados de la reflectividad de este material.We have performed a calculation of the electronic structure within the density functional formalism, of the semiconductor PbS. From it, the dependence in frecuency of the dielectric functions in the range between 0 and 5 eV, is calculated. The peaks, as well as the general shape of the curves corresponding to the real part Si and imaginary part s2, are very well described and understandable in terms of the calculated bands and density of states. The dispersive spectrum has its point of inflexion in reazonably agreement with the energy where the absorption is maximum, and starts decreasing in a metalic like behavior, for energies higher than 3 eV.     PUF60-related developmental disorder:A case series and phenotypic analysis of 10 additional patients with monoallelic PUF60 variants

    PUF60-related developmental disorder (also referred to as Verheij syndrome), resulting from haploinsufficiency of PUF60, is associated with multiple congenital anomalies affecting a wide range of body systems. These anomalies include ophthalmic coloboma, and congenital anomalies of the heart, kidney, and musculoskeletal system. Behavioral and intellectual difficulties are also observed. While less common than other features associated with PUF60-related developmental disorder, for instance hearing impairment and short stature, identification of specific anomalies such as ophthalmic coloboma can aid with diagnostic identification given the limited spectrum of genes linked with this feature. We describe 10 patients with PUF60 gene variants, bringing the total number reported in the literature, to varying levels of details, to 56 patients. Patients were recruited both via locally based exome sequencing from international sites and from the DDD study in the United Kingdom. Eight of the variants reported were novel PUF60 variants. The addition of a further patient with a reported c449-457del variant to the existing literature highlights this as a recurrent variant. One variant was inherited from an affected parent. This is the first example in the literature of an inherited variant resulting in PUF60-related developmental disorder. Two patients (20%) were reported to have a renal anomaly consistent with 22% of cases in previously reported literature. Two patients received specialist endocrine treatment. More commonly observed were clinical features such as: cardiac anomalies (40%), ocular abnormalities (70%), intellectual disability (60%), and skeletal abnormalities (80%). Facial features did not demonstrate a recognizable gestalt. Of note, but remaining of unclear causality, we describe a single pediatric patient with pineoblastoma. We recommend that stature and pubertal progress should be monitored in PUF60-related developmental disorder with a low threshold for endocrine investigations as hormone therapy may be indicated. Our study reports an inherited case with PUF60-related developmental disorder which has important genetic counseling implications for families.Published version, accepted version (12 month embargo)The article is available via Open Access.     Conjugation with L, L-diphenylalanine Self-Assemblies Enhances In Vitro Antitumor Activity of Phthalocyanine Photosensitizer

    We present the synthesis and characterization of new peptide conjugates obtained by hierarchical co-assembly of L,L-diphenylalanine (FF) and zinc phthalocyanine complexes (ZnPc) in water. Self-assembly capabilities under defined conditions were investigated by scanning electron microscopy, and photophysical properties were evaluated using UV-Vis and fluorescence spectroscopy. AFM observations demonstrated that these ZnPcs form different highly ordered arrays on the crystalline faces of the FF microplates and that surface roughness significantly changes with the presence of differently substituted phthalocyanine units. XRD assays showed that the overall molecular packing of the conjugates is organized according to a hexagonal symmetry, with ZnPcs hosted in the interstices of the peptide phase. In vitro photodynamic studies were conducted on human breast cancer MCF-7 cells to investigate both cellular uptake and cytotoxicity. It was shown that FF self-assemblies are not toxicity and enhance accumulation of ZnPc in MCF-7 cells, improving apoptotic cell death upon irradiation. Our findings demonstrate enhancement of ZnPc antitumor efficiency by FF conjugates and a proof-of-concept for new photosensitizer carriers based on peptide conjugates

    Public opinion and environmental policy output: a cross-national analysis of energy policies in Europe

    This article studies how public opinion is associated with the introduction of renewable energy policies in Europe. While research increasingly seeks to model the link between public opinion and environmental policies, the empirical evidence is largely based on a single case: the US. This limits the generalizability of findings and we argue accordingly for a systematic, quantitative study of how public opinion drives environmental policies in another context. Theoretically, we combine arguments behind the political survival of democratic leaders with electoral success and environmental politics. Ultimately, we suggest that office-seeking leaders introduce policies that seem favorable to the domestic audience; if the public prefers environmental protection, the government introduces such policies in turn. The main contribution of this research is the cross-country empirical analysis, where we combine data on the public's environmental attitudes and renewable energy policy outputs in a European context between 1974 and 2015. We show that as public opinion shifts towards prioritizing the environment, there is a significant and positive effect on the rate of renewable energy policy outputs by governments in Europe. To our knowledge, this is the first systematic, quantitative study of public opinion and environmental policies across a large set of countries, and we demonstrate that the mechanisms behind the introduction of renewable energy policies follow major trends across European states

    Benznidazole biotransformation and multiple targets in <i>Trypanosoma</i> cruzi revealed by metabolomics

    &lt;b&gt;Background&lt;/b&gt;&lt;p&gt;&lt;/p&gt; The first line treatment for Chagas disease, a neglected tropical disease caused by the protozoan parasite Trypanosoma cruzi, involves administration of benznidazole (Bzn). Bzn is a 2-nitroimidazole pro-drug which requires nitroreduction to become active, although its mode of action is not fully understood. In the present work we used a non-targeted MS-based metabolomics approach to study the metabolic response of T. cruzi to Bzn.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Methodology/Principal findings&lt;/b&gt;&lt;p&gt;&lt;/p&gt; Parasites treated with Bzn were minimally altered compared to untreated trypanosomes, although the redox active thiols trypanothione, homotrypanothione and cysteine were significantly diminished in abundance post-treatment. In addition, multiple Bzn-derived metabolites were detected after treatment. These metabolites included reduction products, fragments and covalent adducts of reduced Bzn linked to each of the major low molecular weight thiols: trypanothione, glutathione, γ-glutamylcysteine, glutathionylspermidine, cysteine and ovothiol A. Bzn products known to be generated in vitro by the unusual trypanosomal nitroreductase, TcNTRI, were found within the parasites, but low molecular weight adducts of glyoxal, a proposed toxic end-product of NTRI Bzn metabolism, were not detected.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Conclusions/significance&lt;/b&gt;&lt;p&gt;&lt;/p&gt; Our data is indicative of a major role of the thiol binding capacity of Bzn reduction products in the mechanism of Bzn toxicity against T. cruzi

    A phase II study of weekly cisplatin, 6S-stereoisomer leucovorin and fluorouracil as first-line chemotherapy for elderly patients with advanced gastric cancer

    The incidence of gastric cancer (GC) increases significantly after the fifth decade and palliative chemotherapy is the ultimate treatment in the majority of patients. We investigated safety and efficacy of a weekly regimen with cisplatin, fluorouracil and leucovorin as first-line chemotherapy for elderly patients with advanced GC. Chemotherapy-naive patients older than 65 years were considered eligible for study entry. Frail elderly patients were identified and excluded according to the following criteria: age >85 years, dependence in one or more activities of daily living (activities of daily living and instrumental activities of daily living scales), three or more comorbid conditions, one or more geriatric syndromes. Chemotherapy consisted of 1-day per week administration of intravenous cisplatin 35 mg m(-2), 6S-stereoisomer leucovorin 250 mg m(-2) and fluorouracil 500 mg m(-2) (PLF). Patients were re-evaluated after eight weekly cycles and six additional weekly administrations were planned for patients without disease progression. A 5-day subcutaneous filgrastim (5 mug Kg(-1) day(-1), days +1-+5) was used after the first treatment delay for neutropenia and maintained thereafter. In the whole group, the best intention-to-treat overall response rate was 43\% (95\% CI: 30-56\%). The time to disease progression and the median survival time were 5.3 and 8.6 months, respectively. Fatigue was the commonest nonhaematologic toxicity (71\% of the patients). Filgrastim was used in 30 patients who showed grade II (20 patients) or grade III (10 patients) neutropenia. Neither grade IV toxicity nor toxic deaths were observed. The weekly PLF regimen resulted safe and effective in elderly patients with advanced GC. This outpatient regimen is based on old and low-cost drugs and it may represent an alternative to new and more expensive combinations
