    Taxonomic notes on Encephalartos ferox (Cycadales : Zamiaceae), with the description of a new subspecies from Mozambique

    Continued work on the systematics of the genus Encephalartos has been conducted over a large area along the Mozambican coast to determine the infraspecific variation and taxonomic status of Encephalartos ferox populations. The northern extent of the species was also investigated based on anecdotal information that it might represent a new taxon. Several (45) vegetative and reproductive (ca. 40) characters were analysed and used to re-circumscribe E. ferox. Literature and iconographic citations are included, as is information on ecology, cytology and conservation as available. No specimens of E. ferox were located north of the Save River in the Sofala Province, Mozambique, and the northernmost population of the species was found to be anomalous. Accordingly a new subspecies, E. ferox subsp. emersus, is recognised based on its invariably emergent stems, shorter unarmed petiole length, shorter leaves, greater number and narrower leaflets (in seedlings as well) which are more closely spaced. Strobili and peduncles in subsp. emersus are shorter and narrower with strobili having a propensity towards yellow, while internally the megasporophylls are green, grading to white away from the outer surface. The IUCN Red List status is suggested as Critically Endangered A4d + B1a,b(v) + B2a,b(v), with the threat of illegal over-collection highlighted.Estudos contínuos sobre a sistemática do gênero Encephalartos já foram realizados sobre uma grande área ao longo da costa de Moçambique para determinar variações infra-específicas e estado taxonômico das populações de Encephalartos ferox.     Yellow and Red Supergiants in the Large Magellanic Cloud

    Due to their transitionary nature, yellow supergiants provide a critical challenge for evolutionary modeling. Previous studies within M31 and the SMC show that the Geneva evolutionary models do a poor job at predicting the lifetimes of these short-lived stars. Here we extend this study to the LMC while also investigating the galaxy's red supergiant content. This task is complicated by contamination by Galactic foreground stars that color and magnitude criteria alone cannot weed out. Therefore, we use proper motions and the LMC's large systemic radial velocity (\sim278 km/s) to separate out these foreground dwarfs. After observing nearly 2,000 stars, we identified 317 probable yellow supergiants, 6 possible yellow supergiants and 505 probable red supergiants. Foreground contamination of our yellow supergiant sample was \sim80%, while that of the the red supergiant sample was only 3%. By placing the yellow supergiants on the H-R diagram and comparing them against the evolutionary tracks, we find that new Geneva evolutionary models do an exemplary job at predicting both the locations and the lifetimes of these transitory objects.Comment: Accepted for publication in the Ap

    A Survey of Local Group Galaxies Currently Forming Stars: II. UBVRI Photometry of Stars in Seven Dwarfs and a Comparison of the Entire Sample

    We have obtained UBVRI images with the Kitt Peak and Cerro Tololo 4-m telescopes and Mosaic cameras of seven dwarfs in (or near) the Local Group, all of which have known evidence of recent star formation: IC10, NGC 6822, WLM, Sextans B, Sextans A, Pegasus,and Phoenix. We construct color-magnitude diagrams (CMDs) of these systems, as well as neighboring regions that can be used to evaluate the degree of foreground contamination by stars in the Milky Way. Inter-comparison of these CMDs with those of M31, M33, the LMC, and the SMC permits us to determine improved reddening values for a typical OB star found within these galaxies. All of the CMDs reveal a strong or modest number of blue supergiants. All but Pegasus and Phoenix also show the clear presence of red supergiants in the CMD, although IC10 appears to be deficient in these objects given its large WR population. The bright stars of intermediate color in the CMD are badly contaminated by foreground stars (30-100%), and considerable spectroscopy is needed before statistics on the yellow supergiants in these systems will be known. This study is intended to serve both as the impetus and "finding charts" for further space-based imaging, and for many spectroscopic programs at large aperture.Comment: Added Erratum.     Broad and Gag-Biased HIV-1 Epitope Repertoires Are Associated with Lower Viral Loads

    Background: HLA class-I alleles differ in their ability to control HIV replication through cell-mediated immune responses. No consistent associations have been found between the breadth of Cytotoxic T Lymphocytes (CTL) responses and the control of HIV-1, and it is unknown whether the size or distribution of the viral proteome-wide epitope repertoire, i.e., the intrinsic ability to present fewer, more or specific viral epitopes, could affect clinical markers of disease progression. Methodology/Principal Findings: We used an epitope prediction model to identify all epitope motifs in a set of 302 HIV-1 full-length proteomes according to each individual's HLA (Human Leukocyte Antigen) genotype. The epitope repertoire, i.e., the number of predicted epitopes per HIV-1 proteome, varied considerably between HLA alleles and thus among individual proteomes. In a subgroup of 270 chronically infected individuals, we found that lower viral loads and higher CD4 counts were associated with a larger predicted epitope repertoire. Additionally, in Gag and Rev only, more epitopes were restricted by alleles associated with low viral loads than by alleles associated with higher viral loads. Conclusions/Significance: This comprehensive analysis puts forth the epitope repertoire as a mechanistic component of the multi-faceted HIV-specific CTL response. The favorable impact on markers of disease status of the propensity to present more HLA binding peptides and specific proteins gives impetus to vaccine design strategies that seek to elicit responses to a broad array of HIV-1 epitopes, and suggest a particular focus on Gag

    A UBVR CCD Survey of the Magellanic Clouds

    We present photometry and a preliminary interpretation of a UBVR survey of the Large and Small Magellanic Clouds. We determine improved values for the relative number of blue and red supergiants.     Astroglial excitability and gliotransmission: an appraisal of Ca2+ as a signalling route

    Astroglial cells, due to their passive electrical properties, were long considered subservient to neurons and to merely provide the framework and metabolic support of the brain. Although astrocytes do play such structural and housekeeping roles in the brain, these glial cells also contribute to the brain's computational power and behavioural output. These more active functions are endowed by the Ca2+-based excitability displayed by astrocytes. An increase in cytosolic Ca2+ levels in astrocytes can lead to the release of signalling molecules, a process termed gliotransmission, via the process of regulated exocytosis. Dynamic components of astrocytic exocytosis include the vesicular-plasma membrane secretory machinery, as well as the vesicular traffic, which is governed not only by general cytoskeletal elements but also by astrocyte-specific IFs (intermediate filaments). Gliotransmitters released into the ECS (extracellular space) can exert their actions on neighbouring neurons, to modulate synaptic transmission and plasticity, and to affect behaviour by modulating the sleep homoeostat. Besides these novel physiological roles, astrocytic Ca2+ dynamics, Ca2+-dependent gliotransmission and astrocyte–neuron signalling have been also implicated in brain disorders, such as epilepsy. The aim of this review is to highlight the newer findings concerning Ca2+ signalling in astrocytes and exocytotic gliotransmission. For this we report on Ca2+ sources and sinks that are necessary and sufficient for regulating the exocytotic release of gliotransmitters and discuss secretory machinery, secretory vesicles and vesicle mobility regulation. Finally, we consider the exocytotic gliotransmission in the modulation of synaptic transmission and plasticity, as well as the astrocytic contribution to sleep behaviour and epilepsy

    Breakdown of the Endothelial Barrier Function in Tumor Cell Transmigration

    The ability of tumor cells to metastasize is associated with a poor prognosis for cancer. During the process of metastasis, tumor cells circulating in the blood or lymph vessels can adhere to, and potentially transmigrate through, the endothelium and invade the connective tissue. We studied the effectiveness of the endothelium as a barrier against the invasion of 51 tumor cell lines into a three-dimensional collagen matrix. Only nine tumor cell lines showed attenuated invasion in the presence of an endothelial cell monolayer, whereas 17 cell lines became invasive or showed a significantly increased invasion. Endothelial cells cocultured with invasive tumor cells increased chemokine gene expression of IL-8 and Gro-β. Expression of the IL-8 and Gro-β receptor, CXCR2, was upregulated in invasive tumor cells. Addition of IL-8 or Gro-β increased tumor cell invasiveness by more than twofold. Tumor cell variants selected for high CXCR2 expression were fourfold more invasive in the presence of an endothelial cell layer, whereas CXCR2 siRNA knock-down cells were fivefold less invasive. We demonstrate that Gro-β and IL-8 secreted by endothelial cells, together with CXCR2 receptor expression on invasive tumor cells, contribute to the breakdown of the endothelial barrier by enhancing tumor cell force generation and cytoskeletal remodeling dynamics

    The Jurassic–Cretaceous depositional and tectonic evolution of the southernwestern margin of the Neotethys Ocean, Northern Oman and United Arab Emirates

    The concept that the autochthonous, parautochthonous and allochthonous Permian–Cretaceous sequences in the United Arab Emirates (UAE) and Oman record the transition from platform, slope to basin sedimentation within the southern part of Neotethys has been fundamental to the interpretation of the geological history of the region. The results of a major geological mapping programme of the UAE, carried out by the British Geological Survey for the Federal Government of the UAE, coupled with the detailed examination of key sections within northern Oman has led to a re-evaluation of the geological evolution of this region. This detailed study has led to a greater appreciation of the sedimentology and depositional setting of the sediments laid down along the northeastern Arabian continental margin during the Jurassic to Cretaceous, allowing a more refined model of Neotethys Ocean basin evolution to be established. The model charts the progressive breakup of the Arabian continental margin and closure of Neotethys during the mid to late Cretaceous and is divided into three main stages: Stage 1—Initial rifting and formation of the Neotethys Ocean, followed by a prolonged period of stable, passive margin sedimentation which extended from the Permian to Late Jurassic times; Stage 2—Uplift and erosion of the shelf margin during the Late Jurassic to Early Cretaceous, coincident with increased carbonate-clastic sedimentation in the outer ramp, distal slope and basinal areas; Stage 3—Increased instability during the Late Cretaceous leading to the breakup of the platform margin and foreland basin sedimentation accompanying the obduction of the Oman-UAE ophiolite. Data obtained for the upper part of the platform and platform margin to slope successions has revealed that the topography of the “shelf”-slope-basinal margin was more subdued than previously thought, with this more gentle ramp margin morphology persisting until early to mid-Cretaceous times when the platform margin started to become unstable during ophiolite obduction. The thrust-repeated allochthonous sedimentary rocks of the Hamrat Duru Group were deposited on the outer platform margin/lower slope rise to basinal plain of this basin margin and includes the dismembered remains of two turbidite fan systems which fed carbonate-rich detritus into deeper parts of the ocean. A re-evaluation of the chert-rich sequences, previously equated with deposition on the abyssal plain of Neotethys, has led to the conclusion that they may record sedimentation at a much shallower level within a starved ocean basin, possibly in a mid-ramp (above storm wave base) to outer ramp setting. A marked change in basin dynamics occurred during the mid-Cretaceous leading to the development of a shallow ramp basin margin in Oman with terrestrial to shallow marine sedimentary rocks interdigitating with red siliceous mudstones. By contrast, the contemporaneous succession in the Dibba Zone of the UAE indicates considerable instability on a steep shelf break. This instability is recorded by the presence of several major olistostrome deposits within the Aruma Group of the UAE which are thought to have been generated in advance of the rapidly obducting Oman-UAE ophiolite

    TPL-2 negatively regulates interferon-β production in macrophages and myeloid dendritic cells

    Stimulation of Toll-like receptors (TLRs) on macrophages and dendritic cells (DCs) by pathogen-derived products induces the production of cytokines, which play an important role in immune responses. Here, we investigated the role of the TPL-2 signaling pathway in TLR induction of interferon-β (IFN-β) and interleukin-10 (IL-10) in these cell types. It has previously been suggested that IFN-β and IL-10 are coordinately regulated after TLR stimulation. However, in the absence of TPL-2 signaling, lipopolysaccharide (TLR4) and CpG (TLR9) stimulation resulted in increased production of IFN-β while decreasing IL-10 production by both macrophages and myeloid DCs. In contrast, CpG induction of both IFN-α and IFN-β by plasmacytoid DCs was decreased in the absence of TPL-2, although extracellular signal-regulated kinase (ERK) activation was blocked. Extracellular signal-related kinase–dependent negative regulation of IFN-β in macrophages was IL-10–independent, required protein synthesis, and was recapitulated in TPL-2–deficient myeloid DCs by retroviral transduction of the ERK-dependent transcription factor c-fos
