    Keratoderma Hereditaria Mutilans (Vohwinkel's Syndrome): A Trial of Isotretinoin

    An 8-year-old girl with the classic findings of keraloderma heredilaria mutilans (Vohwinkel's syndrome) was seen. Treatment with isotretinoin was instituted to decrease the hyperkeratosis and to prevent further autoamputation. After a 12-week course at 2 mg per kg per day, the patient had only minimal decrease in the amount of hyperkeratosis.     Study of evaluation of groundwater in Gadilam basin using hydrogeochemical and isotope data

    Gadilam river basin has gained its importance due to the presence of Neyveli Lignite open cast mines and other industrial complexes. It is also due to extensive depressurization of Cuddalore aquifer, and bore wells for New Veeranam Scheme are constructed downstream of the basin. Geochemical indicators of groundwater were used to identify the chemical processes that control hydrogeochemistry. Chemical parameters of groundwater such as pH, electrical conductivity, total dissolved solids, sodium (Na+), potassium (K+), calcium (Ca+), magnesium (Mg+), bicarbonate (HCO-3 ), sulfate (SO-4 ),phosphate (PO-4 ), and silica (H4SiO4) were determined. Interpretation of hydrogeochemical data suggests that leaching of ions followed by weathering and anthropogenic impact controls the chemistry of the groundwater. Isotopic study reveals that recharge from meteoric source in sedimentary terrain and rock-water interaction with significant evaporation prevails in hard rock region

    The Impact of the C-Terminal Domain on the Interaction of Human DNA Topoisomerase II ฮฑ and ฮฒ with DNA

    <b>Background</b> Type II DNA topoisomerases are essential, ubiquitous enzymes that act to relieve topological problems arising in DNA from normal cellular activity. Their mechanism of action involves the ATP-dependent transport of one DNA duplex through a transient break in a second DNA duplex; metal ions are essential for strand passage. Humans have two isoforms, topoisomerase IIฮฑ and topoisomerase IIฮฒ, that have distinct roles in the cell. The C-terminal domain has been linked to isoform specific differences in activity and DNA interaction. <b>Methodology/Principal Findings</b> We have investigated the role of the C-terminal domain in the binding of human topoisomerase IIฮฑ and topoisomerase IIฮฒ to DNA in fluorescence anisotropy assays using full length and C-terminally truncated enzymes. We find that the C-terminal domain of topoisomerase IIฮฒ but not topoisomerase IIฮฑ affects the binding of the enzyme to the DNA. The presence of metal ions has no effect on DNA binding. Additionally, we have examined strand passage of the full length and truncated enzymes in the presence of a number of supporting metal ions and find that there is no difference in relative decatenation between isoforms. We find that calcium and manganese, in addition to magnesium, can support strand passage by the human topoisomerase II enzymes. <b>Conclusions/Significance</b> The C-terminal domain of topoisomerase IIฮฒ, but not that of topoisomerase IIฮฑ, alters the enzyme's KD for DNA binding. This is consistent with previous data and may be related to the differential modes of action of the two isoforms in vivo. We also show strand passage with different supporting metal ions for human topoisomerase IIฮฑ or topoisomerase IIฮฒ, either full length or C-terminally truncated. They all show the same preferences, whereby Mg > Ca > Mn

    Control of Centrin Stability by Aurora A

    Aurora A is an oncogenic serine/threonine kinase which can cause cell transformation and centrosome amplification when over-expressed. Human breast tumors show excess Aurora A and phospho-centrin in amplified centrosomes. Here, we show that Aurora A mediates the phosphorylation of and localizes with centrin at the centrosome, with both proteins reaching maximum abundance from prophase through metaphase, followed by their precipitous loss in late stages of mitosis. Over-expression of Aurora A results in excess phospho-centrin and centrosome amplification. In contrast, centrosome amplification is not seen in cells over-expressing Aurora A in the presence of a recombinant centrin mutant lacking the serine phosphorylation site at residue 170. Expression of a kinase dead Aurora A results in a decrease in mitotic index and abrogation of centrin phosphorylation. Finally, a recombinant centrin mutation that mimics centrin phosphorylation increases centrin's stability against APC/C-mediated proteasomal degradation. Taken together, these results suggest that the stability of centrin is regulated in part by Aurora A, and that excess phosphorylated centrin may promote centrosome amplification in cancer

    Future global warming from atmospheric trace gases

    Human activity this century has increased the concentrations of atmospheric trace gases, which in turn has elevated global surface temperatures by blocking the escape of thermal infrared radiation. Natural climate variations are masking this temperature increase, but further additions of trace gases during the next 65 years could double or even quadruple the present effects, causing the global average temperature to rise by at least 1 ยฐC and possibly by more than 5 ยฐC. If the rise continues into the twenty-second century, the global average temperature may reach higher values than have occurred in the past 10 million years. ยฉ 1986 Nature Publishing Group

    Distinct Salmonella Enteritidis lineages associated with enterocolitis in high-income settings and invasive disease in low-income settings.

    An epidemiological paradox surrounds Salmonella enterica serovar Enteritidis. In high-income settings, it has been responsible for an epidemic of poultry-associated, self-limiting enterocolitis, whereas in sub-Saharan Africa it is a major cause of invasive nontyphoidal Salmonella disease, associated with high case fatality. By whole-genome sequence analysis of 675 isolates of S. Enteritidis from 45 countries, we show the existence of a global epidemic clade and two new clades of S. Enteritidis that are geographically restricted to distinct regions of Africa. The African isolates display genomic degradation, a novel prophage repertoire, and an expanded multidrug resistance plasmid. S. Enteritidis is a further example of a Salmonella serotype that displays niche plasticity, with distinct clades that enable it to become a prominent cause of gastroenteritis in association with the industrial production of eggs and of multidrug-resistant, bloodstream-invasive infection in Africa.This work was supported by the Wellcome Trust.     Indo-Pacific Warm Pool convection and ENSO since 1867 derived from Cambodian pine tree cellulose oxygen isotopes

    The Indo-Pacific Warm Pool (IPWP) is a major source of heat and moisture to the atmosphere and thus strongly influences the global climate. In this study, we investigate how moisture fluxes from the IPWP influence the stable isotope signature of precipitation over Southeast Asia by analyzing the oxygen isotopic composition (ฮด^18O) of tree cellulose from southern Cambodia. The cellulose ฮด^18O record, spanning AD 1867-2006, documents a regular seasonal cycle with an average amplitude of ~4โ€ฐ that is primarily controlled by seasonal differences in the isotopic composition of precipitation. Using the outputs from an isotope-enabled atmospheric model, we illustrate how the ฮด^18O of precipitation at our site is predominantly controlled by the amount of rainout that occurs over the moisture source region, the IPWP. This is verified by strong correlations of our cellulose ฮด^18O record with instrumental measurements of precipitation and outgoing longwave radiation over the IPWP, suggesting that the cellulose ฮด^18O could be used to reconstruct the convection intensity over the IPWP. Spectral analysis of the cellulose ฮด^18O reveals significant peaks at 2-7 years corresponding to ENSO frequencies. The variability of the cellulose ฮด^18O record on the ENSO band exhibits characteristics that match existing coral ฮด^18O records from the tropical Pacific, with reduced amplitude of variability in the 1920s through the 1960s, a period of weak ENSO activity

    Distinct Peripheral Blood RNA Responses to Salmonella in Pigs Differing in Salmonella Shedding Levels: Intersection of IFNG, TLR and miRNA Pathways

    Transcriptomic analysis of the response to bacterial pathogens has been reported for several species, yet few studies have investigated the transcriptional differences in whole blood in subjects that differ in their disease response phenotypes. Salmonella species infect many vertebrate species, and pigs colonized with Salmonella enterica serovar Typhimurium (ST) are usually asymptomatic, making detection of these Salmonella-carrier pigs difficult. The variable fecal shedding of Salmonella is an important cause of foodborne illness and zoonotic disease. To investigate gene pathways and biomarkers associated with the variance in Salmonella shedding following experimental inoculation, we initiated the first analysis of the whole blood transcriptional response induced by Salmonella. A population of pigs (nโ€Š=โ€Š40) was inoculated with ST and peripheral blood and fecal Salmonella counts were collected between 2 and 20 days post-inoculation (dpi). Two groups of pigs with either low shedding (LS) or persistent shedding (PS) phenotypes were identified. Global transcriptional changes in response to ST inoculation were identified by Affymetrix Genechipยฎ analysis of peripheral blood RNA at day 0 and 2 dpi. ST inoculation triggered substantial gene expression changes in the pigs and there was differential expression of many genes between LS and PS pigs. Analysis of the differential profiles of gene expression within and between PS and LS phenotypic classes identified distinct regulatory pathways mediated by IFN-ฮณ, TNF, NF-ฮบB, or one of several miRNAs. We confirmed the activation of two regulatory factors, SPI1 and CEBPB, and demonstrated that expression of miR-155 was decreased specifically in the PS animals. These data provide insight into specific pathways associated with extremes in Salmonella fecal shedding that can be targeted for further exploration on why some animals develop a carrier state. This knowledge can also be used to develop rational manipulations of genetics, pharmaceuticals, nutrition or husbandry methods to decrease Salmonella colonization, shedding and spread
