Understanding the evolution of native pinewoods in Scotland will benefit their future management and conservation
Scots pine (Pinus sylvestris L.) is a foundation species in Scottish highland forests and a national icon. Due to heavy exploitation, the current native pinewood coverage represents a small fraction of the postglacial maximum. To reverse this decline, various schemes have been initiated to promote planting of new and expansion of old pinewoods. This includes the designation of seed zones for control of the remaining genetic resources. The zoning was based mainly on biochemical similarity among pinewoods but, by definition, neutral molecular markers do not reflect local phenotypic adaptation. Environmental variation within Scotland is substantial and it is not yet clear to what extent this has shaped patterns of adaptive differentiation among Scottish populations. Systematic, rangewide common-environment trials can provide insights into the evolution of the native pinewoods, indicating how environment has influenced phenotypic variation and how variation is maintained. Careful design of such experiments can also provide data on the history and connectivity among populations, by molecular marker analysis. Together, phenotypic and molecular datasets from such trials can provide a robust basis for refining seed transfer guidelines for Scots pine in Scotland and should form the scientific basis for conservation action on this nationally important habitat

MLP: a MATLAB toolbox for rapid and reliable auditory threshold estimation
In this paper, we present MLP, a MATLAB toolbox enabling auditory
thresholds estimation via the adaptive Maximum Likelihood procedure proposed
by David Green (1990, 1993). This adaptive procedure is particularly appealing for
those psychologists that need to estimate thresholds with a good degree of accuracy
and in a short time. Together with a description of the toolbox, the current text
provides an introduction to the threshold estimation theory and a theoretical
explanation of the maximum likelihood adaptive procedure. MLP comes with a
graphical interface and it is provided with several built-in, classic psychoacoustics
experiments ready to use at a mouse click
Traditional closed-loop sanitation systems in peri-urban and rural Afghanistan: a SWOT analysis
The closed-loop-sanitation-system (CLSS), or sustainable sanitation system, has accelerated in recent years and been successfully implemented in many parts of the world. This study explored the strengths, weaknesses, opportunities, and threats (SWOT) of the traditional CLSS (T-CLSS) in both peri-urban and rural contexts within three different provinces in Afghanistan, the first study of its kind in this country. Participatory research tools such as transect walks, focus group discussions, and interactive workshops have been applied to assess the SWOT components of T- CLSS. The results show that T-CLSS is practiced historically in both peri-urban and rural areas by different generations using local and traditional knowledge, skills and technologies. Socio-cultural acceptance of the system is considered as one of the strengths in both rural and peri-urban areas. It is highly recommended that the feasibility of improved CLSS be assessed and implemented in the light of the T-CLSS system
Genetic Determinants of Variability in Glycated Hemoglobin (HbA1c) in Humans: Review of Recent Progress and Prospects for Use in Diabetes Care
Glycated hemoglobin A1c (HbA1c) indicates the percentage of total hemoglobin that is bound by glucose, produced from the nonenzymatic chemical modification by glucose of hemoglobin molecules carried in erythrocytes. HbA1c represents a surrogate marker of average blood glucose concentration over the previous 8 to 12 weeks, or the average lifespan of the erythrocyte, and thus represents a more stable indicator of glycemic status compared with fasting glucose. HbA1c levels are genetically determined, with heritability of 47% to 59%. Over the past few years, inroads into understanding genetic predisposition by glycemic and nonglycemic factors have been achieved through genome-wide analyses. Here I review current research aimed at discovering genetic determinants of HbA1c levels, discussing insights into biologic factors influencing variability in the general and diabetic population, and across different ethnicities. Furthermore, I discuss briefly the relevance of findings for diabetes monitoring and diagnosis
Results of the IGEC-2 search for gravitational wave bursts during 2005
The network of resonant bar detectors of gravitational waves resumed
coordinated observations within the International Gravitational Event
Collaboration (IGEC-2). Four detectors are taking part in this collaboration:
ALLEGRO, AURIGA, EXPLORER and NAUTILUS. We present here the results of the
search for gravitational wave bursts over 6 months during 2005, when IGEC-2 was
the only gravitational wave observatory in operation. The network data analysis
implemented is based on a time coincidence search among AURIGA, EXPLORER and
NAUTILUS, keeping the data from ALLEGRO for follow-up studies. With respect to
the previous IGEC 1997-2000 observations, the amplitude sensitivity of the
detectors to bursts improved by a factor about 3 and the sensitivity bandwidths
are wider, so that the data analysis was tuned considering a larger class of
detectable waveforms. Thanks to the higher duty cycles of the single detectors,
we decided to focus the analysis on three-fold observation, so to ensure the
identification of any single candidate of gravitational waves (gw) with high
statistical confidence. The achieved false detection rate is as low as 1 per

Global gene expression analysis of human erythroid progenitors

This article has an erratum: http://bloodjournal.hematologylibrary.org/content/118/26/6993.3.Understanding the pattern of gene expression during erythropoiesis is crucial for a synthesis of erythroid developmental biology. Here, we isolated 4 distinct populations at successive erythropoietin-dependent stages of erythropoiesis, including the terminal, pyknotic stage. The transcriptome was determined using Affymetrix arrays. First, we demonstrated the importance of using defined cell populations to identify lineage and temporally specific patterns of gene expression. Cells sorted by surface expression profile not only express significantly fewer genes than unsorted cells but also demonstrate significantly greater differences in the expression levels of particular genes between stages than unsorted cells. Second, using standard software, we identified more than 1000 transcripts not previously observed to be differentially expressed during erythroid maturation, 13 of which are highly significantly terminally regulated, including RFXAP and SMARCA4. Third, using matched filtering, we identified 12 transcripts not previously reported to be continuously up-regulated in maturing human primary erythroblasts. Finally, using transcription factor binding site analysis, we identified potential transcription factors that may regulate gene expression during terminal erythropoiesis. Our stringent lists of differentially regulated and continuously expressed transcripts containing many genes with undiscovered functions in erythroblasts are a resource for future functional studies of erythropoiesis. ALFRED: an allele frequency resource for research and teaching
ALFRED (http://alfred.med.yale.edu) is a free, web accessible, curated compilation of allele frequency data on DNA sequence polymorphisms in anthropologically defined human populations. Currently, ALFRED has allele frequency tables on over 663 400 polymorphic sites; 170 of them have frequency tables for more than 100 different population samples. In ALFRED, a population may have multiple samples with each ‘sample’ consisting of many individuals on which an allele frequency is based. There are 3566 population samples from 710 different populations with allele frequency tables on at least one polymorphism. Fifty of those population samples have allele frequency data for over 650 000 polymorphisms. Records also have active links to relevant resources (dbSNP, PharmGKB, OMIM, Ethnologue, etc.). The flexible search options and data display and download capabilities available through the web interface allow easy access to the large quantity of high-quality data in ALFRED
