GRIDCC: Real-time workflow system
The Grid is a concept which allows the sharing of resources between distributed communities, allowing each to progress towards potentially different goals. As adoption of the Grid increases so are the activities that people wish to conduct through it. The GRIDCC project is a European Union funded project addressing the issues of integrating instruments into the Grid. This increases the requirement of workflows and Quality of Service upon these workflows as many of these instruments have real-time requirements. In this paper we present the workflow management service within the GRIDCC project which is tasked with optimising the workflows and ensuring that they meet the pre-defined QoS requirements specified upon them
Integral equations for simple fluids in a general reference functional approach
The integral equations for the correlation functions of an inhomogeneous
fluid mixture are derived using a functional Taylor expansion of the free
energy around an inhomogeneous equilibrium distribution. The system of
equations is closed by the introduction of a reference functional for the
correlations beyond second order in the density difference from the equilibrium
distribution. Explicit expressions are obtained for energies required to insert
particles of the fluid mixture into the inhomogeneous system. The approach is
illustrated by the determination of the equation of state of a simple,
truncated Lennard--Jones fluid and the analysis of the behavior of this fluid
near a hard wall. The wall--fluid integral equation exhibits complete drying
and the corresponding coexisting densities are in good agreement with those
obtained from the standard (Maxwell) construction applied to the bulk fluid.
Self--consistency of the approach is examined by analyzing the
virial/compressibility routes to the equation of state and the Gibbs--Duhem
relation for the bulk fluid, and the contact density sum rule and the Gibbs
adsorption equation for the hard wall problem. For the bulk fluid, we find good
self--consistency for stable states outside the critical region. For the hard
wall problem, the Gibbs adsorption equation is fulfilled very well near phase
coexistence where the adsorption is large.For the contact density sum rule, we
find some deviationsnear coexistence due to a slight disagreement between the
coexisting density for the gas phase obtained from the Maxwell construction and
from complete drying at the hard wall.Comment: 29 page
Massively parallel landscape-evolution modelling using general purpose graphical processing units
As our expectations of what computer systems can do and our ability to capture data improves, the desire to perform ever more computationally intensive tasks increases. Often these tasks, comprising vast numbers of repeated computations, are highly interdependent on each other – a closely coupled problem. The process of Landscape-Evolution Modelling is an example of such a problem. In order to produce realistic models it is necessary to process landscapes containing millions of data points over time periods extending up to millions of years. This leads to non-tractable execution times, often in the order of years. Researchers therefore seek multiple orders of magnitude reduction in the execution time of these models. The massively parallel programming environment offered through General Purpose Graphical Processing Units offers the potential for multiple orders of magnitude speedup in code execution times. In this paper we demonstrate how the time dominant parts of a Landscape-Evolution Model can be recoded for a massively parallel architecture providing two orders of magnitude reduction in execution time
Severity scoring of manganese health effects for categorical regression
Characterizing the U-shaped exposure response relationship for manganese (Mn) is necessary for estimating the risk of adverse health from Mn toxicity due to excess or deficiency. Categorical regression has emerged as a powerful tool for exposure-response analysis because of its ability to synthesize relevant information across multiple studies and species into a single integrated analysis of all relevant data. This paper documents the development of a database on Mn toxicity designed to support the application of categorical regression techniques. Specifically, we describe (i) the conduct of a systematic search of the literature on Mn toxicity to gather data appropriate for dose-response assessment; (ii) the establishment of inclusion/exclusion criteria for data to be included in the categorical regression modeling database; (iii) the development of a categorical severity scoring matrix for Mn health effects to permit the inclusion of diverse health outcomes in a single categorical regression analysis using the severity score as the outcome variable; and (iv) the convening of an international expert panel to both review the severity scoring matrix and assign severity scores to health outcomes observed in studies (including case reports, epidemiological investigations, and in vivo experimental studies) selected for inclusion in the categorical regression database. Exposure information including route, concentration, duration, health endpoint(s), and characteristics of the exposed population was abstracted from included studies and stored in a computerized manganese database (MnDB), providing a comprehensive repository of exposure-response information with the ability to support categorical regression modeling of oral exposure data
A genomewide scan for Attention-Deficit/Hyperactivity Disorder in an extended sample: Suggestive linkage on 17p11
Attention-deficit/hyperactivity disorder (ADHD [MIM 143465]) is a common, highly heritable neurobehavioral disorder of childhood onset, characterized by hyperactivity, impulsivity, and/or inattention. As part of an ongoing study of the genetic etiology of ADHD, we have performed a genomewide linkage scan in 204 nuclear families comprising 853 individuals and 270 affected sibling pairs (ASPs). Previously, we reported genomewide linkage analysis of a “first wave” of these families composed of 126 ASPs. A follow-up investigation of one region on 16p yielded significant linkage in an extended sample. The current study extends the original sample of 126 ASPs to 270 ASPs and provides linkage analyses of the entire sample, using polymorphic microsatellite markers that define an ∼10-cM map across the genome. Maximum LOD score (MLS) analysis identified suggestive linkage for 17p11 (MLS=2.98) and four nominal regions with MLS values >1.0, including 5p13, 6q14, 11q25, and 20q13. These data, taken together with the fine mapping on 16p13, suggest two regions as highly likely to harbor risk genes for ADHD: 16p13 and 17p11. Interestingly, both regions, as well as 5p13, have been highlighted in genomewide scans for autism
Non-existence and uniqueness results for supercritical semilinear elliptic equations
Non-existence and uniqueness results are proved for several local and
non-local supercritical bifurcation problems involving a semilinear elliptic
equation depending on a parameter. The domain is star-shaped but no other
symmetry assumption is required. Uniqueness holds when the bifurcation
parameter is in a certain range. Our approach can be seen, in some cases, as an
extension of non-existence results for non-trivial solutions. It is based on
Rellich-Pohozaev type estimates. Semilinear elliptic equations naturally arise
in many applications, for instance in astrophysics, hydrodynamics or
thermodynamics. We simplify the proof of earlier results by K. Schmitt and R.
Schaaf in the so-called local multiplicative case, extend them to the case of a
non-local dependence on the bifurcation parameter and to the additive case,

Lnk Deficiency Leads to TPO-Mediated Osteoclastogenesis and Increased Bone Mass Phenotype
The Lnk adapter protein negatively regulates the signaling of thrombopoietin (TPO), the main megakaryocyte (MK) growth factor. Lnk-deficient (-/-) mice have increased TPO signaling and increased MK number. Interestingly, several mouse models exist in which increased MK number leads to a high bone mass phenotype. Here we report the bone phenotype of these mice. MicroCT and static histomorphometric analyses at 20 weeks showed the distal femur of Lnk-/- mice to have significantly higher bone volume fraction and trabecular number compared to wild-type (WT) mice. Notably, despite a significant increase in the number of osteoclasts (OC), and decreased bone formation rate in Lnk-/- mice compared to WT mice, Lnk-/- mice demonstrated a 2.5-fold greater BV/TV suggesting impaired OC function in vivo. Additionally, Lnk-/- mouse femurs exhibited non-significant increases in mid-shaft cross-sectional area, yet increased periosteal BFR compared to WT femurs was observed. Lnk-/- femurs also had non-significant increases in polar moment of inertia and decreased cortical bone area and thickness, resulting in reduced bone stiffness, modulus, and strength compared to WT femurs. Of note, Lnk is expressed by OC lineage cells and when Lnk-/- OC progenitors are cultured in the presence of TPO, significantly more OC are observed than in WT cultures. Lnk is also expressed in osteoblast (OB) cells and in vitro reduced alkaline phosphatase activity was observed in Lnk-/- cultures. A 13-hour laboratory school study of lisdexamfetamine dimesylate in school-aged children with attention-deficit/hyperactivity disorder
BackgroundLisdexamfetamine dimesylate (LDX) is indicated for the treatment of attention-deficit/hyperactivity disorder (ADHD) in children 6 to 12 years of age and in adults. In a previous laboratory school study, LDX demonstrated efficacy 2 hours postdose with duration of efficacy through 12 hours. The current study further characterizes the time course of effect of LDX.MethodsChildren aged 6 to 12 years with ADHD were enrolled in a laboratory school study. The multicenter study consisted of open-label, dose-optimization of LDX (30, 50, 70 mg/d, 4 weeks) followed by a randomized, placebo-controlled, 2-way crossover phase (1 week each). Efficacy measures included the SKAMP (deportment [primary] and attention [secondary]) and PERMP (attempted/correct) scales (secondary) measured at predose and at 1.5, 2.5, 5, 7.5, 10, 12, and 13 hours postdose. Safety measures included treatment-emergent adverse events (AEs), physical examination, vital signs, and ECGs.ResultsA total of 117 subjects were randomized and 111 completed the study. Compared with placebo, LDX demonstrated significantly greater efficacy at each postdose time point (1.5 hours to 13.0 hours), as measured by SKAMP deportment and attention scales and PERMP (P < .005). The most common treatment-emergent AEs during dose optimization were decreased appetite (47%), insomnia (27%), headache (17%), irritability (16%), upper abdominal pain (16%), and affect lability (10%), which were less frequent in the crossover phase (6%, 4%, 5%, 1%, 2%, and 0% respectively).ConclusionIn school-aged children (6 to 12 years) with ADHD, efficacy of LDX was maintained from the first time point (1.5 hours) up to the last time point assessed (13.0 hours).
