    Gender and rural history: A roundtable

    The aim of this debate article is to promote a discussion of a historiographical nature (not ideological, not political) about the meaning, place and role of gender in both the rural past and the rural historiography. The discussion revolves around a variety of questions, ranging from the relevance, the opportunity and the very history of the use of gender category in rural history, to the analysis of gender (im)balances in the community of historians working in this broadly defined field of studies, not to mention the very definition of what is meant by gender.     SiL: An Approach for Adjusting Applications to Heterogeneous Systems Under Perturbations

    Scientific applications consist of large and computationally-intensive loops. Dynamic loop scheduling (DLS) techniques are used to load balance the execution of such applications. Load imbalance can be caused by variations in loop iteration execution times due to problem, algorithmic, or systemic characteristics (also, perturbations). The following question motivates this work: "Given an application, a high-performance computing (HPC) system, and both their characteristics and interplay, which DLS technique will achieve improved performance under unpredictable perturbations?" Existing work only considers perturbations caused by variations in the HPC system delivered computational speeds. However, perturbations in available network bandwidth or latency are inevitable on production HPC systems. Simulator in the loop (SiL) is introduced, herein, as a new control-theoretic inspired approach to dynamically select DLS techniques that improve the performance of applications on heterogeneous HPC systems under perturbations. The present work examines the performance of six applications on a heterogeneous system under all above system perturbations. The SiL proof of concept is evaluated using simulation. The performance results confirm the initial hypothesis that no single DLS technique can deliver best performance in all scenarios, while the SiL-based DLS selection delivered improved application performance in most experiments

    The European Eel NCCβ Gene Encodes a Thiazide-Resistant Na-Cl Cotransporter

    The thiazide-sensitive Na-Cl cotransporter (NCC) is the major pathway for salt reabsorption in the mammalian distal convoluted tubule. NCC plays a key role in the regulation of blood pressure. Its inhibition with thiazides constitutes the primary baseline therapy for arterial hypertension. However, the thiazide-binding site in NCC is unknown. Mammals have only one gene encoding for NCC. The eel, however, contains a duplicate gene. NCCα is an ortholog of mammalian NCC and is expressed in the kidney. NCCβ is present in the apical membrane of the rectum. Here we cloned and functionally characterized NCCβ from the European eel. The cRNA encodes a 1043-amino acid membrane protein that, when expressed in Xenopus oocytes, functions as an Na-Cl cotransporter with two major characteristics, making it different from other known NCCs. First, eel NCCβ is resistant to thiazides. Single-point mutagenesis supports that the absence of thiazide inhibition is, at least in part, due to the substitution of a conserved serine for a cysteine at position 379. Second, NCCβ is not activated by low-chloride hypotonic stress, although the unique Ste20-related proline alanine-rich kinase (SPAK) binding site in the amino-terminal domain is conserved. Thus, NCCβ exhibits significant functional differences from NCCs that could be helpful in defining several aspects of the structure-function relationship of this important cotransporter

    Plural valuation of nature for equity and sustainability: Insights from the Global South

    Plural valuation is about eliciting the diverse values of nature articulated by different stakeholders in order to inform decision making and thus achieve more equitable and sustainable outcomes. We explore what approaches align with plural valuation on the ground, as well as how different social-ecological contexts play a role in translating plural valuation into decisions and outcomes. Based on a co-constructed analytical approach relying on empirical information from ten cases from the Global South, we find that plural valuation contributes to equitable and sustainable outcomes if the valuation process: 1) is based on participatory value elicitation approaches; 2) is framed with a clear action-oriented purpose; 3) provides space for marginalized stakeholders to articulate their values in ways that can be included in decisions; 4) is used as a tool to identify and help reconcile different cognitive models about human-nature relations; and 5) fosters open communication and collaboration among stakeholders. We also find that power asymmetries can hinder plural valuation. As interest and support for undertaking plural valuation grows, a deeper understanding is needed regarding how it can be adapted to different purposes, approaches, and social-ecological contexts in order to contribute to social equity and sustainability

    Thioredoxin 80-Activated-Monocytes (TAMs) Inhibit the Replication of Intracellular Pathogens

    BACKGROUND: Thioredoxin 80 (Trx80) is an 80 amino acid natural cleavage product of Trx, produced primarily by monocytes. Trx80 induces differentiation of human monocytes into a novel cell type, named Trx80-activated-monocytes (TAMs). PRINCIPAL FINDINGS: In this investigation we present evidence for a role of TAMs in the control of intracellular bacterial infections. As model pathogens we have chosen Listeria monocytogenes and Brucella abortus which replicate in the cytosol and the endoplasmic reticulum respectively. Our data indicate that TAMs efficiently inhibit intracellular growth of both L. monocytogenes and B. abortus. Further analysis shows that Trx80 activation prevents the escape of GFP-tagged L. monocytogenes into the cytosol, and induces accumulation of the bacteria within the lysosomes. Inhibition of the lysosomal activity by chloroquine treatment resulted in higher replication of bacteria in TAMs compared to that observed in control cells 24 h post-infection, indicating that TAMs kill bacteria by preventing their escape from the endosomal compartments, which progress into a highly degradative phagolysosome. SIGNIFICANCE: Our results show that Trx80 potentiates the bactericidal activities of professional phagocytes, and contributes to the first line of defense against intracellular bacteria

    Equitable and effective area‐based conservation: towards the conserved areas paradigm

    In 2018, the Parties to the Convention on Biological Diversity (CBD) adopted a decision on protected areas and other effective area-based conservation measures (OECMs). It contains the definition of an OECM and related scientific and technical advice that has broadened the scope of governance authorities and areas that can be engaged and recognised in global conservation efforts. The voluntary guidance on OECMs and protected areas, also included in the decision, promotes the use of diverse, effective and equitable governance models, the integration of protected areas and OECMs into wider landscapes and seascapes, and mainstreaming of biodiversity conservation across sectors. Taken as a whole, the advice and voluntary guidance provides further clarity about the CBD Parties’ understanding of what constitutes equitable and effective area-based conservation measures within and beyond protected areas and provides standardised criteria with which to measure and report areas’ attributes and performance. This policy perspective suggests that this CBD decision represents further evidence of the evolution from the ‘new paradigm for protected areas’ to a broader ‘conserved areas paradigm’ that embodies good governance, equity and effective conservation outcomes and is inclusive of a diversity of contributions to conservation within and beyond protected areas
