    Bogotá, capital de Colombia, es la ciudad que actualmente se encuentra más poblada dentro del país, puesto que aproximadamente consta de 7.363.782 habitantes, por lo mismo surge la imperiosa necesidad de que haya un servicio público que se encargue de la satisfacción de la necesidad de movilización masiva que existe a diario en la Capital. Para lo anterior desde el año de 1998 se inició con la construcción del Sistema Transmilenio, el que luego en el año 2000 el 4 de septiembre se inauguró y entró en operación 14 días más tarde, como opción de abastecimiento de la demanda de transporte público de la ciudad de Bogotá, sin embargo, tal como se puede ver en unas acciones populares instauradas por parte de la ciudadanía ante el Consejo de Estado, siendo este último quien da sentencia el día 11 de agosto del año 2011, se establece que efectivamente en la prestación del servicio de transporte público proporcionado por la empresa Transmilenio S.A, se vulneran los derechos de salubridad y seguridad ciudadana a los usuarios del sistema, por 9 condiciones de sobrecupo y hacinamiento, debidamente probado en el período que corresponde a los años de 2002 a 2011. Para lo anterior y evitar dicha situación de vulneración, el Consejo solicita a Transmilenio colocar en el interior de los articulados censores de peso y localización estratégica del personal de Misión Bogotá, pese a que se acogió las consideraciones de dicho órgano, la situación se sigue presentando hoy, año 2014

    Q4Q4Pacientes con HipofosfatasiaHypophosphatasia (HPP) is a rare inherited disorder characterized by low serum alkaline phosphatase. It affects bone and tooth mineralization, although extra-skeletal manifestations are frequent. HPP is caused by loss-of-function mutations in the ALPL gene, encoding the protein tissue-nonspecific alkaline phosphatase. The phenotype is broadly variable, from a subtype with exclusive odontological compromise (odontohypophosphatasia) to five subtypes with systemic involvement, classified according to the age of onset at first symptoms.     Spawning site distribution of a bluefin tuna reduces jellyfish predation on early life stages

    Bluefin tunas across the world migrate long distances to spawn in particularly warm and oligotrophic areas constrained by oceanographic fronts. The low abundance of predators in these areas increases survival chances of their early life stages, but its importance for choice of spawning habitat is unknown. Here, we use estimated clearance rates and data on spatial distributions of Atlantic bluefin tuna larvae Thunnus thynnus and metaephyrae of the jellyfish Pelagia noctiluca to quantify predation at a major spawning ground in the Mediterranean Sea. We found that high densities of P. noctiluca can rapidly deplete tuna eggs and preflexion larvae, but their patchy distribution and low spatial and temporal overlap results in overall low predation. The specific distribution of the spawning sites suggests that bluefin tunas may use local oceanography as cues to spawn outside areas with high predator densities.Postprint3,38

    Ultra high performance liquid chromatography–tandem mass spectrometry method for cyclosporine a quantification in biological samples and lipid nanosystems

    Cyclosporine A (CyA) is an immunosuppressant cyclic undecapeptide used for the prevention of organ transplant rejection and in the treatment of several autoimmune disorders. An ultra high performance liquid chromatography–tandem mass spectrometry method (UHPLC–MS/MS) to quantify CyA in lipid nanosystems and mouse biological matrices (whole blood, kidneys, lungs, spleen, liver, heart, brain, stomach and intestine) was developed and fully validated. Chromatographic separation was performed on an Acquity UPLC® BEH C18 column with a gradient elution consisting of methanol and 2 mM ammonium acetate aqueous solution containing 0.1% formic acid at a flow rate of 0.6 mL/min. Amiodarone was used as internal standard (IS). Retention times of IS and CyA were 0.69 min and 1.09 min, respectively. Mass spectrometer operated in electrospray ionization positive mode (ESI+) and multiple reaction monitoring (MRM) transitions were detected, m/z 1220.69 → 1203.7 for CyA and m/z 646 → 58 for IS. The extraction method from biological samples consisted of a simple protein precipitation with 10% trichloroacetic acid aqueous solution and acetonitrile and 5 μL of supernatant were directly injected into the UHPLC–MS/MS system. Linearity was observed between 0.001 μg/mL–2.5 μg/mL (r ≥ 0.99) in all matrices. The precision expressed in coefficient of variation (CV) was below 11.44% and accuracy in bias ranged from −12.78% to 7.99% including methanol and biological matrices. Recovery in all cases was above 70.54% and some matrix effect was observed. CyA was found to be stable in post-extraction whole blood and liver homogenate samples exposed for 6 h at room temperature and 72 h at 4 °C. The present method was successfully applied for quality control of lipid nanocarriers as well as in vivo studies in BALB/c mice

    In vitro intestinal co-culture cell model to evaluate intestinal absorption of edelfosine lipid nanoparticles

    Nanotechnology is providing a new therapeutic paradigm by enhancing drug efficacy and preventing side-effects. Edelfosine is a synthetic ether lipid analogue of platelet activating factor with high antitumor activity. The encapsulation of this potent antitumor drug in lipid nanoparticles increases its oral bioavailability; moreover, it prevents the hemolytic and gastrointestinal side-effects of the free drug. The literature points towards lymphatic absorption of lipid nanoparticles after oral administration, and previous in vitro and in vivo studies stress the protection against toxicity that these nanosystems provide. The present study is intended to assess the permeability of lipid nanoparticles across the intestinal barrier. Caco-2 monoculture and Caco-2/Raji co-culture were used as in vitro models of enterocytes and Microfold cells respectively. Results showed that free drug is internalized and possibly metabolized in enterocytes. These results do not correlate with those observed in vivo when edelfosine-lipid nanoparticles were administered orally in mice, which suggests that the microfold model is not a good model to study the absorption of edelfosine-lipid nanoparticles across the intestinal barrier in vitro

    A Systemic Framework to Evaluate Student Satisfaction in Latin American Universities under the COVID-19 Pandemic

    Latin American universities (LAUs) have been going through a serious lack of economic resources which has plunged them into a deep financial crisis. The COVID-19 pandemic has exacerbated this calamity. However, LAUs have implemented online teaching processes in order to mitigate the effects of scheduling and other classroom disruption. There is evidence that these modes of teaching have had a reasonable reception but the level of student satisfaction is yet unknown. This article takes a systemic view of the predicament facing LAUs. It represents the elements related to the disruption caused by COVID-19 in a rich picture, building a systemic framework to explore student satisfaction with remote teaching. Using a sample of 298 students from Brazil, Colombia, and Peru, the study analyzes their situation through: (a) Well-being, educational resources, and learning experience and; (b) General satisfaction with virtual classes. Applying exploratory factor analysis, this study identifies three dimensions: (a) satisfaction with support and adaptation in the virtual modality; (b) satisfaction with the interaction in the virtual classroom; and (c) satisfaction with the development of the study program. Medium/high scores for the dimensions indicate moderate/high levels of satisfaction. The findings suggest that there are still unsatisfied needs regarding access to digital resources and socio-emotional needs.
