    Effects of food quantity, dietary fatty acids and temperature on fitness of Daphnia magna

    The performance of zooplankton has a major impact on the efficiency in trophic transfer in pelagic food webs and is therefore examined in this study. I investigated the effect of different food quantity and food quality, as measured by omega-3 fatty acid (ω3-FA) content on survival, somatic growth and reproduction of Daphnia magna at high and low temperatures in laboratory growth experiments. I first investigated the response across a range from low to high food quantity (0.02, 0.07, 0.2, 0.7, 2.0 mg C l-1) of the green alga Scenedesmus acutus at 12.0˚C and 20.6°C. Food quantity constraints on somatic growth of Daphnia and increasing growth rate were found with increasing food concentration. An interaction between quantity and temperature showed a higher maximal growth at higher temperature and high food levels. Furthermore, the starvation point shifted to a lower food concentration at low temperatures. Subsequently I tested the response in survival, somatic and reproductive growth of D. magna to different quality of food in terms of FA content, temperature and food quantity. It was a 2x2x3 factorial design performed at two food levels (maximum growth at 2.0 mg C l-1 and close to the reproduction threshold concentration at 0.2 mg C l-1) and two temperatures (14.1˚C and 21.7°C). To address the question if 3-FA enrichment enhances fitness of D. magna, algal food suspensions were amended with eicosapentaenoic acid (EPA) or oleic acid (non-essential control treatment). The results show, besides the expected effects of food quantity and the interaction of temperature and food quantity, a significant effect of EPA enrichment on somatic growth rate and reproduction. EPA amendment improved somatic growth and egg production at both temperatures. The strongest effect of EPA enrichment was manifested on somatic and reproductive growth at low temperature and at high food concentration (2.0 mg C l-1). These results indicate that food quality is of greater ecological importance in cold freshwater systems, like at high latitudes and high altitudes. In temperate lakes, the effect of interaction between food quantity, quality and temperature is manifested in the seasonal as well as the vertical variation of these factors. When EPA content is high in surface waters of stratified lakes and zooplankton migrates vertically during night to colder deeper layers during night the combined effect of FA content and temperature can be expected to result in improved somatic growth

    How microbial food web interactions shape the arctic ocean bacterial community revealed by size fractionation experiments

    In the Arctic, seasonal changes are substantial, and as a result, the marine bacterial community composition and functions differ greatly between the dark winter and light-intensive summer. While light availability is, overall, the external driver of the seasonal changes, several internal biological interactions structure the bacterial community during shorter timescales. These include specific phytoplankton–bacteria associations, viral infections and other top-down controls. Here, we uncover these microbial interactions and their effects on the bacterial community composition during a full annual cycle by manipulating the microbial food web using size fractionation. The most profound community changes were detected during the spring, with ‘mutualistic phytoplankton’—Gammaproteobacteria interactions dominating in the pre-bloom phase and ‘substrate-dependent phytoplankton’—Flavobacteria interactions during blooming conditions. Bacterivores had an overall limited effect on the bacterial community composition most of the year. However, in the late summer, grazing was the main factor shaping the community composition and transferring carbon to higher trophic levels. Identifying these small-scale interactions improves our understanding of the Arctic marine microbial food web and its dynamics

    Comparing EPA production and fatty acid profiles of three Phaeodactylum tricornutum strains under western Norwegian climate conditions

    Microalgae could provide a sustainable alternative to fish oil as a source for the omega-3 polyunsaturated fatty acids eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). However, growing microalgae on a large-scale is still more cost-intensive than fish oil production, and outdoor productivities vary greatly with reactor type, geographic location, climate conditions and microalgae species or even strains. The diatom Phaeodactylum tricornutum has been intensively investigated for its potential in large-scale production, due to its robustness and comparatively high growth rates and EPA content. Yet, most research have been performed in southern countries and with a single commercial P. tricornutum strain, while information about productivities at higher latitudes and of local strains is scarce. We examined the potential of the climate conditions in Bergen, western Norway for outdoor cultivation of P. tricornutum in flat panel photobioreactors and cultivated three different strains simultaneously, one commercial strain from Spain (Fito) and two local isolates (M28 and B58), to assess and compare their biomass and EPA productivities, and fatty acid (FA) profiles. The three strains possessed similar biomass productivities (average volumetric productivities of 0.20, 0.18, and 0.21 g L− 1 d− 1), that were lower compared to productivities reported from southern latitudes. However, EPA productivities differed between the strains (average volumetric productivities of 9.8, 5.7 and 6.9 mg L− 1 d− 1), due to differing EPA contents (average of 4.4, 3.2 and 3.1% of dry weight), and were comparable to results from Italy. The EPA content of strain Fito of 4.4% is higher than earlier reported for P. tricornutum (2.6–3.1%) and was only apparent under outdoor conditions. A principal component analysis (PCA) of the relative FA composition revealed strain-specific profiles. However, including data from laboratory experiments, revealed more significant differences between outdoor and laboratory-grown cultures than between the strains, and higher EPA contents in outdoor grown cultures.publishedVersio

    Linking internal and external bacterial community control gives mechanistic framework for pelagic virus-to-bacteria ratios

    For more than 25 years, virus-to-bacteria ratios (VBR) have been measured and interpreted as indicators of the importance of viruses in aquatic ecosystems, yet a generally accepted theory for understanding mechanisms controlling VBR is still lacking. Assuming that the denominator (total bacterial abundance) is primarily predator controlled, while viral lysis compensates for host growth rates exceeding this grazing loss, the numerator (viral abundance) reflects activity differences between prokaryotic hosts. VBR is then a ratio between mechanisms generating structure within the bacterial community and interactions between different plankton functional types controlling bacterial community size. We here show how these arguments can be formalized by combining a recently published model for co-evolutionary host-virus interactions, with a previously published “minimum” model for the microbial food web. The result is a framework where viral lysis links bacterial diversity to microbial food web structure and function, creating relationships between different levels of organization that are strongly modified by organism-level properties such as cost of resistance

    The Response of Heterotrophic Prokaryote and Viral Communities to Labile Organic Carbon Inputs Is Controlled by the Predator Food Chain Structure

    Factors controlling the community composition of marine heterotrophic prokaryotes include organic-C, mineral nutrients, predation, and viral lysis. Two mesocosm experiments, performed at an Arctic location and bottom-up manipulated with organic-C, had very different results in community composition for both prokaryotes and viruses. Previously, we showed how a simple mathematical model could reproduce food web level dynamics observed in these mesocosms, demonstrating strong top-down control through the predator chain from copepods via ciliates and heterotrophic nanoflagellates. Here, we use a steady-state analysis to connect ciliate biomass to bacterial carbon demand. This gives a coupling of top-down and bottom-up factors whereby low initial densities of ciliates are associated with mineral nutrient-limited heterotrophic prokaryotes that do not respond to external supply of labile organic-C. In contrast, high initial densities of ciliates give carbon-limited growth and high responsiveness to organic-C. The differences observed in ciliate abundance, and in prokaryote abundance and community composition in the two experiments were in accordance with these predictions. Responsiveness in the viral community followed a pattern similar to that of prokaryotes. Our study provides a unique link between the structure of the predator chain in the microbial food web and viral abundance and diversity
